ABSTRACT

CONTEXT AND OBJECTIVE: Lasers are widely used in treating symptomatic benign prostatic hyperplasia. In current practice, potassium titanyl phosphate (KTP) lasers are the most common type of laser systems used. The aim here was to evaluate the rapid effect of high-power laser systems after application of hypericin. DESIGN AND SETTING: Experimental animal study conducted in the Department of Urology, Gülhane Military Medical Academy, Ankara, Turkey, in 2012. METHODS: Sixteen rats were randomized into four groups: 120 W KTP laser + hypericin; 120 W KTP laser alone; 80 W KTP laser + hypericin; and 80 W KTP laser alone. Hypericin was given intraperitoneally two hours prior to laser applications. The laser incisions were made through the quadriceps muscle of the rats. The depth and the width of the laser incisions were evaluated histologically and recorded. RESULTS: To standardize the effects of the laser, we used the ratio of depth to width. These new values showed us the depth of the laser application per unit width. The new values acquired were evaluated statistically. Mean depth/width values were 231.6, 173.6, 214.1 and 178.9 in groups 1, 2, 3 and 4, respectively. The most notable result was that higher degrees of tissue penetration were achieved in the groups with hypericin (P < 0.05). CONCLUSIONS: The encouraging results from our preliminary study demonstrated that hypericin may improve the effects of KTP laser applications. .


CONTEXTO E OBJETIVO: Lasers são amplamente utilizados no tratamento de hiperplasia benigna de próstata sintomática. Na prática atual, lasers de fosfato de titanilo de potássio (KTP) são os tipos mais comuns usados dos sistemas. O objetivo foi avaliar o efeito rápido do sistema laser de alta potência após a aplicação de hipericina. TIPO DE ESTUDO E LOCAL: Estudo experimental animal, realizado no Departamento de Urologia, Academia de Medicina Militar de Gülhane, Ancara, Turquia, em 2012. MÉTODOS: 16 ratos foram divididos aleatoriamente em 4 grupos: 120W KTP laser + hipericina; 120W KTP laser somente; 80W KTP laser + hipericina; 80W KTP laser somente. Hipericina foi dada intraperitonealmente duas horas antes da aplicação do laser. As incisões a laser foram feitas através do músculo quadríceps dos ratos. A profundidade e a largura das incisões a laser foram avaliadas histologicamente e registradas. RESULTADOS: Para padronizar o efeito do laser foi utilizada a razão entre profundidade e largura. Estes novos valores nos mostraram a profundidade da aplicação do laser de largura por unidade. Os novos valores adquiridos foram avaliados estatisticamente. Os valores da média de profundidade/largura foram 231,6, 173,6, 214,1 e 178,9 nos grupos 1, 2, 3 e 4, respectivamente. O resultado mais notável foi atingir altos graus de penetração tecidual nos grupos com hipericina (P < 0,05). CONCLUSÕES: Os resultados promissores do nosso estudo preliminar mostraram que hipericina pode melhorar os efeitos das aplicações do laser KTP. .

ABSTRACT

OBJETIVOS: Induzir a produção de membranas vitreorretinianas em modelo de trauma ocular animal. Avaliar a inibição do desenvolvimento da proliferação vitreorretiniana (PVR) com o uso de hiperecina. MÉTODOS: Estudo Experimental. Foram utilizados 19 coelhos machos pigmentados adultos com peso entre 2.000 e 3.000 gramas. Todos submetidos a modelo de trauma com dispase associada à diatermia da retina para indução de membranas de PVR. Separados randomicamente para receberem hiperecina (10 µM em 0,1 ml) ou solução salina (0,1 ml) como placebo. Avaliados clinicamente no sétimo, décimo quarto, vigésimo primeiro e vigésimo oitavo dias de pós-operatório com oftalmoscopia indireta e retinografia colorida digitalizada. O grau de PVR foi classificado em estágios (de 0 a 7) segundo Hida e colaboradores. RESULTADOS: A formação de membranas esteve presente em 79 por cento dos olhos, sendo 100 por cento nos olhos do grupo placebo e 60 por cento nos olhos do grupo tratamento (hiperecina). A comparação entre as médias dos estágios de PVR entre os grupos mostrou diferença estatisticamente significativa, com valor p=0,0321 pelo teste Wilcoxon. CONCLUSÕES: O modelo de trauma com uso de dispase e diatermia da retina produz membranas vitreorretinianas. A hiperecina mostrou-se eficaz na diminuição do aparecimento e progressão do PVR.


PURPOSE: To produce proliferative vitreoretinopathy (PVR) in an animal ocular trauma model. To evaluate the inhibition of (PVR) emergence and progression by hypericin. METHODS: Experimental Study. Nineteen pigmented male adult rabbits weighing between 2,000 and 3,000 grams were used in this study. All of them were submitted to trauma model with dispase and retinal diathermy to induce PVR membranes formation. They were randomly assigned to receive hypericin (10 µM in 0.1 ml) or saline solution (0.1 ml) as placebo. They were evaluated clinically in the seventh, fourteenth, twenty-first and twenty-eighth postoperative days with indirect ophthalmoscopy and digital color retinography. The PVR degree was classified according to Hida (0 to 7). RESULTS: Membranes formation was present in 79 percent of the eyes; being 100 percent in the eyes of placebo group and 60 percent in the eyes of treatment group (hypericin). The comparison between PVR phases averages within the groups showed a statistically significant difference between the two groups, with a p value of 0.0321 for Wilcoxon test. CONCLUSIONS: The trauma model with dispase and retinal diathermy produces vitreoretinal membranes. Hypericin was considered effective in PVR emergence and progression decrease.

ABSTRACT

Utilizing liquid chromatography-electro spray ionization-mass spectrometry [LC-[+,-]-ESI-MS] and liquid chromatography-photodiode array detection [LC-PDA] techniques, a dereplication strategy for the analysis of the secondary metabolites constituents of the genus Hypericum has been developed. From the crude methanolic extract of the aerial parts of H. triquetrifolium [leaves, stems, and flowers] and on the basis of their UV- profiles, chromatographic retention times and [+,-]-ESI-MS [TIC and SIM] mass spectral data, seven known [1-7] compounds were dereplicated fairly rapidly. The compounds were classified into three structural classes: phloroglucinols: hyperfirin and adhyperfirin; naphthodianthrones: hypericin, pseudo-hypericin, proto-hypericin, and protopseudo-hypericin; and the flavonoid rutin

ABSTRACT

The potent photodynamic properties of hypericin [HY] elicit a range of light-dependant virucidal and tumoricidal activities. This work highlights the interaction of liposomes loaded with HY with the aqueous humor and crystalline lens of the rabbits' eye in comparison with those treated with HY only. Six group of New Zealand rabbits of six rabbits each were used in this study. The rabbits received 30 micro l of HY or liposomes loaded with hypericin topically once a day for one week as an eye drop of HY concentration 1-9 micro M. The eyes of rabbits were exposed to He-Ne laser for 5 min after 30 min from administration of HY or liposomes loaded with HY for the studied groups. Total soluble protein, molecular weight distribution, electrophoretic mobility and refractive index for both aqueous humor and soluble lens crystallins were investigated for the studied samples. The obtained results showed an increase in the solubility of lens protein and aqueous humor, an increase in the molecular weights of beta and gamma proteins accompanied by fluctuations in he electrophoretic mobility and slight decrease in the refractive index at high concentration of HY. The changes were higher in the samples treated with HY more than those treated with liposomes loaded with HY. It is concluded from the obtained data that hypericin embedded in liposomes may be used as a hotosensitizer for photodynamic therapy at low fractionated doses with minimal side effects
