ABSTRACT

Celiac is a hereditary disease presented with chronic inflammation of small intestine. Several studies supposed a relationship between Celiac disease and Inflammatory Bowel Disease [IBD]. Tissue trans-glutaminase antibody is one of the best serological markers in Celiac disease. This study was designed to evaluate the association of Celiac and IBD using tissue trans-glutaminase antibody. In this descriptive study 127 confirmed IBD patients who were referred by gastroentrologists 2005-08 enrolled into the study. A questionnaire was completed and tissue trans-glutaminase antibody was evaluated with ELISA method with a Cut-off=12 U/ml. Among 127 referred patients, serum samples of 102 patients were collected. Mean +/- SD of age was 36.17 +/- 15.2 years and 48% were males. Ulcerative colitis, Crohn's disease and other colitis were observed in 76 [74.5%], 7 [6.9%] and 19 [18.6%] of patients, respectively. Tissue trans-glutaminase antibody was positive [19.8 U/L] in one 46-years-old male patient with ulcerative colitis. Tissue trans-glutaminase antibody titer was not significantly different between IBD patients and controls, thus it seems not appropriate to suggest as one of the routine tests in IBD patients

ABSTRACT

La transglutaminasa tisular (tTG) se conoce como el autoantígeno más específico en pacientes con enfermedad celíaca. Por ello, nuestro objetivo fue estandarizar un ensayo ELISA para detectar anticuerpos específicos anti-tTG en pacientes con dicha patología. Se purificó la tTG a partir de hígado de cobayo y compararla con la tTG de una casa comercial, comprobando su eficacia en la inducción de anticuerpos en conejos inmunizados. La tTG procesada en el laboratorio se purificó unas 100 veces respecto al extracto original, conservando una elevada actividad tTG; además se demostró que la tTG purificada estaba en condiciones más puras que la de la casa comercial. El método ELISA para detección de anticuerpos IgA anti tTG se probó con ambas tTG (purificada y comercial) en 5 pacientes celíacos, 15 controles de diagnóstico diferencial y 8 controles sanos. A pesar de lo bajo de la muestra se observó una diferencia significativa (p=0.019) en el grupo de celíacos respecto a los sanos empleando la tTG purificada, no así en los otros grupos ni con la tTG comercial. La tTG purificada es mejor con relación a los resultados obtenidos con la tTG comercial, a pesar de la ausencia de estandarización del ensayo por la baja muestra de pacientes con enfermedad celíaca activa

Para verificar a participação dos dendrócitos dérmicos fator XIIIa positivos (DDFXIIIa+) na patogenia da leishmaniose tegumentar americana (LTA) fez-se a sua demonstração por técnica imuno-histoquímica em lesões de pele de 47 doentes e comparação de sua população com a de grupo controle normal. Verificou-se a interação dos DDFXIIIa+ com os parasitas por técnica imuno-histoquímica de dupla marcação com os anticorpos anti-fator XIIIa e anti-Leishmania. Os DDFXIIIa+ eram hipertróficos e numerosos nas lesões de LTA quando comparados a pele normal. Evidenciou-se parasitismo de DDFXIIIa+ em 11 de 22 casos. Os resultados sugerem a participação dos DDFXIIIa+ no mecanismos patogenéticos da LTA como célula - alvo, efetora e/ou apresentadora de antígenos / In order to verify factor XIIIa positive dermal dendrocyte (FXIIIa+DD) involvement in american tegumentary leishmaniasis pathogenesis, 47 patients skin lesions were subjected to immunohistochemistry technique, quantified and compared to a normal skin group sample. Interaction of FXIIIa+DD and parasites was disclosed by double-staining technique with anti-factor XIIIa and anti- Leishmania antibodies. FXIIIa+DD were hypertrofic and numerous in leishmaniasis skin lesions. FXIIIa+DD harboring Leishmania were observed in 11 of 22 biopsies.
