ABSTRACT

Purpose: This study aimed to compare the degree of maturation and development of fetal pig segmental intestinal tissue with that of spheroids created by in-vitro reaggregation of dissociated fetal intestinal cells after transplantation into immunodeficient mice. Methods: Fetal pig small intestines were transplanted as segmental grafts into the omentum and subrenal capsules of immunodeficient mice or enzymatically treated to generate single cells. Spheroids made by in-vitro reaggregation of these cells were transplanted into the subrenal capsules of immunodeficient mice. The segmental grafts and spheroids were harvested four and eight weeks after transplantation, and the structural maturity and in-vivo development of these specimens were histologically evaluated. Results: The spheroids were engrafted and supplied blood vessels from the host mice, but an intestinal layered structure was not clearly observed, and there was almost no change in size. On the other hand, the segmental grafts formed deep crypts in the mucus membrane, the inner circular layer, and outer longitudinal muscles. The crypts of the transplanted grafts harvested at eight weeks were much deeper, and the smooth muscle layer and the enteric nervous system were more mature than those of grafts harvested at the fourth week, although the intestinal peristaltic wave was not observed. Conclusions: Spheroids created from fetal small intestinal cells could not form layered structures or mature sufficiently. Conversely, segmental tissues structurally matured and developed after in-vivo transplantation and are therefore potential grafts for transplantation.

ABSTRACT

This study aimed to evaluate the characteristics of two different murine models of hormone-treated renal-encapsulated bovine ovarian tissue xenotransplantation. Two immunodeficient mouse models (BALB/c Nude and C57BL6 SCID) were xenografted with ovarian pieces from heifers and each group was subjected to two hormonal treatments of eCG or a combination of FSH+LH. Donor ovaries and recipients were evaluated by histology and infrared thermography at different times. At the time of xenograft collection, animals were evaluated for alterations in hepatorenal biochemistry. The statistical test used in the study was ANOVA, followed by Tukey's test. Among the strains, 80% of C57BL6 SCID and 77% of BALB/c Nude mice showed development and vascularization of the transplanted tissue, which acquired cyclicity at 19 and 9 days post-transplant, respectively. Hemorrhagic follicles in xenografts induced with FSH+LH were found in the C57BL6 SCID strain. Infrared thermography was insufficient to distinguish the tissue donor recipient. In conclusion, the C57BL6 SCID strain appears to be the best host for ovarian xenografts, since the transplants in these mice were viable and showed robust follicular development. This work will aid future choices of immunodeficient strains for xenografting procedures.(AU)


Este estudo teve como objetivo avaliar as características dos dois diferentes modelos de murinas tratadas hormonalmente após xenotransplante de tecido ovariano bovino sob a cápsula renal. Dois modelos de camundongos imunodeficientes (BALB/c NUDE e C57BL6 SCID) receberam fragmentos de ovário de novilhas e cada grupo foi submetido a dois tratamentos hormonais de eCG ou uma combinação de FSH+LH. Ovários doadores e receptores foram avaliados por histologia e termografia infravermelha em diferentes momentos. No momento da retirada do xenotranplante, os animais foram avaliados quanto a alterações na bioquímica hepatorrenal. O teste estatístico utilizado no estudo foi ANOVA, seguido do teste de Tukey. Entre as linhagens, 80% de C57BL6 SCID e 77% das BALB/c NUDE mostraram desenvolvimento e vascularização do tecido transplantado, que adquiriu a ciclicidade 19 e 9 dias após o transplante, respectivamente. Corpos hemorrágicos foram encontrados após o xenotransplante induzidos com FSH+LH na linhagem C57BL6 SCID. A termografia infravermelha foi insuficiente para distinguir o tecido doador do receptor. Em conclusão, a linhagem C57BL6 SCID demonstrou ser o melhor hospedeiro para xenotransplante de ovário, uma vez que os transplantes nestes camundongos foram viáveis e mostraram desenvolvimento folicular. Este trabalho ajudará futuras escolhas de linhagens imunodeficientes para procedimentos de xenotranplante.(AU)

ABSTRACT

Foi realizada falha segmentar de 6mm na região metafisária da tíbia de 12 coelhos,sendo utilizado enxerto ósseo cotical heterólogo fragmentado conservado em glicerina (98 por cento), para a sua reconstrução. O enxerto foi depositado no leito receptor e a integração deste foi avaliada radiologicamente por 20, 40 e 60 dias. Houve incorporação gradativa, em relação ao tempo, do enxerto no leito receptor, em 100 por cento dos casos. O uso de heteroenxerto ósseo cortical fragmentado e conservado em glicerina a 98 por cento promove a reparação das falhas ósseas, sem sinais de infecção ou rejeição, podendo ser utilizado como substituto ósseo na medicina veterinária...


It was realized a 6mm of segmental defect at the metaphyseal region of the tibia of 12 rabbits and fragmented heterolog cortical bone graft conserved in glycerin (98 percent) was used to the reconstruction. The graft was placed in the receptor bed and its integration was evaluated by radiological exam after 20, 40, and 60 days. There was gradative graft incorporation, by the time, in the receptor bed, in 100 percent of the cases. The use of fragmented cortical bone heterograf conserved in glycerin 98 percent promotes bone defect reparation without signs of infection and can be used like osseous substitute in the veterinary medicine...

ABSTRACT

Procurou-se verificar a viabilidade da colopexia laparoscópica com o emprego de pericárdio bovino conservado em glicerina e testar seus resultados em relação ao procedimento incisional por celiotomia em 20 cães, separados em dois grupos com igual número de animais. A técnica laparoscópica envolveu a realização de duas incisões paralelas no músculo transverso abdominal, sob o qual foi posicionado um retalho da membrana conservada, que envolveu completamente o cólon descendente. Com a aplicação de dois clipes que uniram as bordas do pericárdio e com a sua fixação às camadas serosa e muscular do cólon, promoveu-se a aproximação do órgão à parede abdominal. Vinte e oito dias após, por ocasião do exame laparoscópico, verificou-se que todos os pacientes do grupo da celiotomia mantiveram a aderência do cólon, situação que não foi observada nos animais submetidos à cirurgia laparoscópica. Conclui-se que a técnica proposta não possibilita colopexia permanente, sendo contra-indicada para cães


The laparoscopic colopexy with bovine pericardium preserved in glycerol was compared to incisional approach by celiotomy. Twenty mongrel dogs were used after being separated in two groups. In the laparoscopic surgery, two incisions were made on the transverse abdominal muscles, and the implant was positioned through the muscle flap, involving the bowel. Finally, the extremities of the mesh were clipped. Twenty-eight days after this surgery, the experimental animals were submitted to a new laparoscopic surgery, now aiming at observing the results of both techniques. All patients of the celiotomy group maintained the colopexy. However, none of the animals submitted to the laparoscopic surgery maintained the bowel fixation. The proposed technique is not indicated to promote colopexy in dogs

ABSTRACT

Avaliou-se o uso de biomaterial de origem bovina na regeneração de defeitos ósseos segmentares empregando-se 12 coelhos, fêmeas, da raça Norfolk, com idade de seis meses e pesos entre 3 e 4,5kg. Realizou-se falha segmentar bilateral de um centímetro de comprimento na diáfise do rádio, com inclusão do periósteo. No membro direito, o defeito foi delimitado por membrana de pericárdio liofilizada, contendo em seu interior mistura de proteínas morfogenéticas ósseas adsorvidas a hidroxiapatita, colágeno liofilizado e osso inorgânico. No membro esquerdo, o defeito não recebeu tratamento. Radiografias foram obtidas ao término do procedimento cirúrgico e aos sete, 30, 60, 90, 120 e 150 dias de pós-operatório. Após eutanásia de seis coelhos aos 60 dias e seis aos 150 dias de pós-cirúrgico, os resultados radiográficos e histológicos mostraram que a regeneração óssea foi inibida nos defeitos segmentares tratados com o biomaterial.


Biomaterials of bovine origin in regenerating segmental bone defects were evaluated. Twelve six-month old Norfolk rabbits, weighting 3 to 4.5kg were used. A 1cm long segmental defect was created in the radial diaphysis, including the periosteum, of both forelimbs. In the right forelimb, the defect was filled using a mixture of bone morphogenic proteins adsorbed to hydroxyapatite, agglutinant of lyophilized collagen in granules and anorganic cortical bone in granules delimited by a pericardial membrane. In the left forelimb, the defect did not receive treatment and served as a control. Radiographies were taken immediately after surgery and at seven, 30, 60, 90, 120 and 150 days post-operatively. Six rabbits were euthanized at 60 days and the other six at 150 days post-surgery for histological evaluation. Radiographic and histological results revealed that bone regeneration was inhibited in the segmental defects receiving biomaterials.
