RESUMEN

Objective::To study the correlation between endogenous hormone content, related enzyme activity and embryogenesis during the stratification of Acanthopanax senticosus seeds, and to provide theoretical basis for application of endogenous hormones and related physiological indicators in regulating germination of A. senticosus seeds in production and scientific research. Method::Endophytic fungi as well as different concentrations of nitric oxide and hydrogen peroxide solution were used for soaking the seeds of A. senticosus, and then thermophilic stratification was conducted for the seeds. The content of endogenous hormones such as gibberellin (GA3), abscisic acid (ABA), indole acetic acid (IAA), indole butyric acid (IBA) and salicylic acid (SA) in seeds of A. senticosus were tested by high performance liquid chromatography (HPLC), and the activity change of enzymes in vivo such as catalase (CAT), superoxide dismutase (SOD), peroxidase (POD), and malondialdehyde (MDA) were tested. The correlation between the embryo rate and the hormones and their enzyme activities in the seeds of A. senticosus was studied by the grey correlation method. Result::The embryo rate was significantly positively correlated with IAA, GA3, and CAT, with correlation coefficient of 1.086, 0.935 and 1.067, respectively. There was a significant positive correlation between IAA, GA3, CAT, IBA, and SA, but with a significant negative correlation with ABA, MDA, and POD. The correlation degree was as follows: IAA>CAT>GA3>IBA>SA>SOD>ABA>POD>MDA. Conclusion::IAA, GA3 and CAT have significant promoting effects on embryo rate, and there is a significant correlation between hormone content and enzyme activity, which provides a basis for exploring the seed germination mechanism of A. senticosus.

Objective: To study the quality of Acanthopanax senticosus seedlings,develop grading standards and optimize the best analytical method. Method: Observing and measuring the plant height,diameter,leaf area,root length,chlorophyll content and other main agronomic traits of A. senticosus seedlings from different habitats(Baoqing,Qitaihe,Dongfanghong,Yilan,Acheng,Raohe Linkou and Yabuli) in each year. K-cluster grading method,principal component evaluation factor K-cluster analysis method,standard deviation method for grading,three classification methods were evaluated with different levels of seedling survival rate as indicators. Result: The direct K-cluster analysis method was used to determine the quality of A. senticosus seedlings as the best method. The seedlings of A. senticosus were divided into 3 grades,among in the first level the seedling height is ≥ 13 cm,the stem diameter is ≥ 0.37 cm,the root length is ≥ 8 cm,the leaf area is ≥ 28 cm2,the chlorophyll content is ≤ 31,the main origin is Baoqing,Qitaihe area. In the second level the seedling height is 8-13 cm,the stem diameter is 0.30-0.37 cm,the root length is 6-8 cm,the leaf area is 13-28 cm2,the chlorophyll content is 31-32,and the main origin is Acheng,Dongfanghong,Raohe area. In the third level the seedling height is 5-8 cm,the stem diameter is 0.26-0.30 cm,the root length is 5-6 cm,the leaf area is 5-13 cm2,the chlorophyll content is 32-38,and the main origin is Yabuli,Yilan and Linkou. Conclusion: In this experiment,the quality grading standards of A. senticosus seedlings were preliminarily established,Baoqing and Qitaihe can be used as a high-quality production area for breeding seeds in the Heilongjiang province.which provided the basis for quality evaluation of A. senticosus seedling planting and artificial cultivation.

Objective: To investigate the effect of exogenous nitric oxide (NO) on breaking the dormancy of Acanthopanax senticosus seeds and the changes in endogenous hormones and enzymes,and provide a basis for breaking the dormancy as well as artificial cultivation of A. senticosus seeds.Method: Different concentrations (1,5,10,20 mmol·L-1) of sodium nitroprusside (NO donor) were used to treat the A. senticosus seeds, and then thermophilic stratification was conducted. The content changes of endogenous hormones such as gibberellin (GA3), abscisic acid (ABA),indolo acetic acid (IAA),indolo butyric acid (IBA) and salicylic acid (SA) at different stratification time (0, 30, 50, 80, 100,130 d) were tested by high performance liquid chromatography (HPLC). The activity change of its in vivo enzymes[catalase (CAT), superoxide dismutase (SOD), peroxidase (POD), and malondialdehyde (MDA)] were tested by enzyme-labeled instrument.Result: In the seed germination process of A. senticosus,the contents of GA3,IAA,IBA,and SA were increased gradually,while the content of ABA was reduced gradually. The enzyme activities of POD and MDA were significantly reduced,and the enzyme activities of CAT and SOD were increased obviously. Exogenous NO could increase the seed germination rate and shorten the seed germination time. The effect of 20 mmol·L-1 sodium nitroprusside showed the most obvious effect and 10 mmol·L-1 SNP showed the weakest effect in promoting seed germination,showing an obvious "V" shape for changes.Conclusion: Sodium nitroprusside could promote the seed germination effect of A. senticosus, probably by increasing the content of hormones and enzyme in the stage of seed germination and improving the contents of endogenous NO during germination.

The present study was conducted to explore the effect of endophytic fungi fraction on growth and anti-oxidative activity of Eleutherococcus senticosus. The growth,yield,contents of MDA,and antioxidant activities were assessed in E. senticosus under five fungi fractions,namely BZ,MH,DT,JS,and XFZ. The results showed that fungi fractions and component significantly affected the growth,low concentration of DT fungi fraction significantly increased the biomass of E. senticosus,reduced the MDA content in cells,and the antioxidant activities of the aqueous extracts were superior to the others. The results indicated that low concentration of DT fungi fraction was the optimum fraction to achieve high yield and quality of E. senticosus.

Objective: To study the Methods: of endophytic fungi to break the dormancy of Acanthopanax senticosus seeds and the changes of endogenous hormones and enzymes. Methods: The seeds of A. senticosus were stimulated with endophytic fungi, then treated under thermophilic stratification. The content of endogenous hormones such as GA3, ABA, IAA, IBA, and SA, in the seeds of A. senticosus were tested by HPLC. And the changes of its in vivo enzymes (CAT, SOD, POD, and MDA) activity were tested. Results: Five strains of endophytic fungi from A. senticosus apparently promoted the germination of seeds. In the process of fluctuating temperature stratification, the content of GA3, IAA, IBA, and SA were increased, at the same time, the content of ABA was reduced. The activity of POD and MDA enzymes was significantly reduced, and the enzyme activities of CAT and SOD increased obviously. Conclusion: It is suggested that endophytes have significant effect on the content of seed hormones and enzymes, in addition endophytes could promote the initiation of seed germination.

Resequencing Pathogen Microarray (RPM) is a new pathogen detection and identification technology based on DNA microarray. In order to apply RPM in the detection of unexplained infection and as a result, to improve the emergency response capacity, a new RPM-based respiratory pathogens detection assay was developed to simultaneously detect 19 common respiratory viruses, 9 influenza A viruses (Flu A),11 human rhinoviruses(HRV), 28 enteroviruses and 18 rare respiratory viruses. The specificity of multiplex system was examined by confirmed positive specimens for 16 common respiratory virus. The sensi-tivity was evaluated by serial ten-fold dilutions of plasmids or in vitro-transcribed RNA. RPM could detect and differentiate 16 virus types/subtypes at 10 - 1 000 copies/reaction level. Nucleic acids of 8 throat swabs with unexplained respiratory tract infections were pooled and detected by the new assay. The RPM result was verified by common PCR followed by sequencing as well as PLEX-ID (Abbott). Except for a false-positive of PIV1, no difference among the three assays was found. These results indicate the assay based on the new RPM is a highly sensitive, high throughput test for the detection of respiratory virus infections, which is significant for the management of emergent and epidemic infectious disease.

Acute respiratory tract infections (ARTIs) are widely distributed among the population, mainly caused by respiratory viruses. ARTIs are responsible for significant morbidity and mortality among the elderly and infants or young children, causing a serious economic burden. The rapid and accurate identifi cation of a pathogen will provide a guideline for the clinical diagnosis and therapy. Multiplex polymerase chain reaction (PCR) technologies combine the rapidness and high sensitivity of PCR with high through put, thus achieving the capability of detecting multiple pathogens simultaneously. The commercial kits based on these multiplex PCR methods allow to detect more than twelve respiratory viruses simultaneous ly, reaching the comparable sensitivities and specificities to those of real-time PCR. The recent progress of novel multiplex PCR assays and their principles as well as applications in respiratory virus diagnosis were reviewed in this paper.

<p><b>OBJECTIVE</b>A reverse transcription loop-mediated isothermal amplification (RT-LAMP) assay was developed for rapid visual detection of HIV-1.</p><p><b>METHODS</b>RT-LAMP primers were designed according to conservative sequences of HIV-1 gag gene, and their sensitivity and specificity were evaluated by the established RT-LAMP protocol with the addition of the hydroxynaphthol blue (HNB) dye prior to amplification. The performance of RT-LAMP on clinical samples was compared with real-time reverse transcription PCR(qRT-PCR).</p><p><b>RESULTS</b>The RT-LAMP assay showed a high specificity, and its detection limit was 1000 copies RNA per tube. The sensitivity and specificity of this method using 43 clinical samples were 94.6% and 100%, respectively,in comparison with those of qRT-PCR.</p><p><b>CONCLUSION</b>RT-LAMP assay using hydroxynaphthol blue dye does not need expensive instruments, and offer an alternative for the rapid detection of HIV-1 with the potential to be applied in field diagnosis.</p>

In order to investigate the clinical manifestations, diagnosis, therapy and prognosis of lymphoplasmacytic lymphoma/Waldenström macroglobulinemia (LPL/WM), 16 patients with LPL/WM were analyzed retrospectively. The results showed that the average age of 16 patients with LPL/WM was 65.1 years old, the most common syndromes were anemia and hyperviscosity syndrome, bone marrows were composed of small lymphocyte, admixed with variable numbers of plasma cells and plasmacytoid lymphocytes. And lymph node biopsy revealed that most cells expressed B-cell-associated antigen. Among the 16 cases, complete remission was 25%, overall response rate (ORR) was 81.3%, overall survival time was 6 to 108 months. 3 patients died and survival rate was 81.3 %. It is concluded that the clinical course of LPL/WM is typically indolent. These patients can acquire remission in clinic, but can not be cured, some of them can transform into patients with more malignant lymphoma.

OBJECTIVE@#To explore the effect of arsenic trioxide on the apoptosis of retinoblastoma cell line HXO-RB(44) and the possible mechanism.@*METHODS@#The effect of arsenic trioxide on the proliferation of retinoblastoma cell line HXO-RB(44) was observed by MTT colorimetric assay; the apoptosis of the HXO-RB(44) was examined by AO/EB staining and flow cytometry analysis (Annexin V+ PI staining); caspase-3 activity and bcl-2/bax expression in the HXO-RB(44) were detected by cpp32 colorimetric assay kit and Western blot.@*RESULTS@#Arsenic trioxide inhibited the proliferation of HXO-RB(44) cell in dose and duration-dependent manner in vitro; arsenic trioxide significantly increased the apoptosis in HXO-RB(44) cells. The activation of caspase-3 was increased, and the rate of bcl-2/bax was down-regulated in the HXO-RB(44) cells processed with arsenic trioxide.@*CONCLUSION@#Arsenic trioxide can inhibit the proliferation of retinoblastoma cell HXO-RB(44) in vitro by apoptosis induction. The apoptosis induction is possibly related to the caspase-3 activation and bcl-2/bax down-regulation.

RESUMEN

<p><b>OBJECTIVE</b>To explore the significance of surgical treatment of HCC originating from caudate lobe.</p><p><b>METHODS</b>From 1995 to 2003, caudate lobectomy, including 19 cases of isolated lobectomy and 20 cases of combined lobectomy, were performed in 39 patients with HCC originating from caudate lobe, and the factors that might influence postoperative liver function were compared between the two groups.</p><p><b>RESULTS</b>All tumors were resected successfully. One patient died of postoperative renal failure. Hydrothorax occurred in 3 patients, ascites in 4 patients, and bile leakage in 1 patient. The survival rates of 1, 3, 5 year were 53%, 50%, 39% respectively.</p><p><b>CONCLUSIONS</b>Caudate lobectomy is a effective method for HCC originating from caudate lobe.</p>

<p><b>OBJECTIVE</b>To establish a preoperative scoring system to predict the lymph node metastases (N) in gastric cancers.</p><p><b>METHODS</b>The clinicopathologic data of 291 cases with gastric cancer were analyzed retrospectively. The factors influencing significantly actual lymph node status (pN) were selected through the univariate and the multivariate analysis, and the score of each factor was identified. Scores predicting different N stages were identified using receiver operating characteristic curves. The N stages defined by the score system were compared with the actual pN status using kappa statistics and diagnostic test.</p><p><b>RESULTS</b>Tumor size, depth of invasion and histopathological types were selected to establish the scoring system. According to this score system, scores 0-4 predict N0, scores 5-7 predict N1, scores 8-9 predict N2 and scores 10-13 predict N3. There was a good agreement between N stages predicted by the scoring system and the actual pN status (weighted kappa = 0.605, u = 14.548, P < 0.0001). The crude agreement, positive predictive value and negative predictive value of the scoring system were 82.8%, 65.6% and 88.5%, respectively.</p><p><b>CONCLUSION</b>The scoring system can provide accurate and reliable information to predict the lymph node metastases of gastric cancers preoperatively. It is simple and practical to use in clinical work and can help surgeons to choose an optimal extent of lymph node dissection for gastric cancer.</p>

<p><b>OBJECTIVE</b>To evaluate the value of serum TR(6) for the diagnosis and TNM classification in patients with gastric carcinoma.</p><p><b>METHODS</b>Serum TR(6) levels were measured using ELISA method in 31 gastric cancer patients, 19 patients with nonmalignant conditions and 29 healthy individuals. TR(6) expression in tumor mass was studied with immunohistochemistry. TR(6) gene copy number in tumor tissues was evaluated by real time PCR.</p><p><b>RESULTS</b>Ninety-seven point nine percent (47 of 48 cases) of healthy individuals and patients with nonmalignant conditions were serum TR(6)-negative. In contrast, 71% (22 of 31 cases) of gastric cancer patients were serum TR(6)-positive. Serum TR(6) positiveness was closely correlated with tumor differentiation status and TNM classification. TR(6) gene amplification did not occur in gastric carcinoma.</p><p><b>CONCLUSIONS</b>Serum TR(6) levels were correlated significantly with TNM stage and histopathological type of tumor. This can help to determine the pre-operative TNM classification and to choose the optimal extent of lymph node dissection for gastric cancer.</p>
