RESUMEN

To observe the effects of hypothermia on the repolarization duration and the expression of Kir2.1 protein of ventricular myocytes in isolated rat heart and explore the role of Kir2.1 protein. Eighteen healthy adult male Sprague-Dawley rats were randomly divided into three groups (n=6 per group): Control group (C group), 35℃ group (H group), 32℃ group (H group). Langendorff isolated heart models were established. After 15 min 37℃ K-H fluid banlanced perfusion, C group continued to perfuse the K-H solution at 37℃ for 30 minutes, H group continued to perfuse the K-H solution at 35℃ for 30 minutes, H group continued to perfuse the K-H solution at 32℃ for 30 minutes. At 15 min of balanced perfusion (T), and 30 min of continuous perfusion (T), the heart rate,and the MAP in the three layers of the left ventricular anterior wall were recorded, the action potential duration at 50% repolarization (MAPD), the action potential duration at 90% repolarization (MAPD) and transmural dispersion of repolarization(TDR) were calculated. At the same time, the occurrence of arrhythmia was recorded. The expression of Kir2.1 protein was measured by Western blot. The average optical density (AOD) and the distribution of Kir2.1 protein were measured by immunohistochemistry in the ventricular tissue measured by electrophysiology. Compared with T, the heart rate was decreased, MAPD and MAPD were prolonged significantly (P<0.05), and TDR was increased significantly (P<0.05) in H group, H group at T. Compared with C group, the HR was decreased, the MAPD was prolonged significantly (P<0.05), TDR was increased significantly (P<0.05),the expression and the AOD of Kir2.1 protein were decreased significantly (P<0.05) in Hgroup, Hgroup at T. Compared with H group, the heart rate of H group was decreased significantly (P<0.05), MAPD and MAPD were prolonged significantly (P<0.05), and TDR was increased significantly (P<0.05) at T. The distribution of Kir2.1 protein in group C was normal, while the distribution of Kir2.1 in H group and H group was disordered. Hypothermia prolonged the ventricular duration of repolarization and increased the dispersion of repolarization. The mechanism is related to the down-regulation the expression of Kir2.1 protein and the disorder of the distribution of Kir2.1 protein.

Objective To investigate the effects of vaspin on insulin resistants of 3T3-L1 adipocyte through the insulin receptor substrates (IRS) /phosphatidylinositol 3-kinase (PI3K) /protein kinase B (Akt) /glucose transporter (Glut) signaling pathway.Methods 3T3-L1 cells cultured by palmitic acid (PA) were used to establish insulin resistance models,which were divided into PA group,PA + 100 ng/ml vaspin group,PA+200 ng/ml vaspin group,PA+400 ng/ml vaspin group and PA+400 ng/ml vaspin+wortmannin (PI3K inhibitor) group.Glucose uptake and consumption were assessed by 2-deoxy H3-D-glucose incorporation and glucose oxidase-peroxidase respectively.IRS/PI3K/Akt/Glut signaling pathway was evaluated using reverse transcription polymerase chain reaction and Western blot analysis.Results Compared with PA group,glucose uptake and consumption increased gradually with the increasing of vaspin concentration in other groups (P < 0.05).mRNA levels of IRS-1,Akt and Glut 4 increased gradually as vaspin concentration increasing (P<0.05),and the ratios of p-IRS-1 to IRS-1,p-Akt to Akt and Glut 4 protein level also showed the same trends (P<0.05).However,glucose uptake and consumption in PA+400 ng/ml vaspin+wortmannin group were less than that of PA +400 ng/ml vaspin group (P<0.05).PA+400 ng/ml vaspin+wortmannin group showed lower mRNA and protein phosphorylation levels of IRS-1,Akt and Glut 4 (P<0.05),and that the ratios of p-IRS-1 to IRS-1,p-Akt to Akt and Glut 4 protein levels decreased (P<0.05).Conclusions Vaspin can improve the insulin sensitivity of 3T3-L1 adipocyte by activating IRS/PI3K/Akt/Glut signaling pathway.

Objective To study the distribution of connexin 43 (Cx43) in cervical cancer HeLa cells, and to verify the localization of Cx43 in mitochondria. Methods HeLa cells were segregated into cytoplasm, cell nucleus, mitochondria and supernatant after segregation by using the method of homogenate and centrifuge. Immunoelectron microscope was used to observe the morphology of mitochondria and the localization as well as the distribution of Cx43 in HeLa cells. Voltage-dependent anion channel 1 (VDAC1) was used to confirm the localization of mitochondria. Immunofluorescence was used to costain HeLa cells with Cx43 and mitochondrial marker VDAC1 to verify mitochondria localization of Cx43 in cervical cancer HeLa cells. Then Western blot was used to quantify the expression of Cx43 in fractions (cytoplasmic fraction,nuclear, mitochondria and post mitochondrial supernatant). Mitochondrial markers including VDAC1 and cytochrome c oxidaseⅣ(COXⅣ) were used to confirm mitochondria. Plasma membrane marker (LHR) was used to confirm plasma membrane. Results Immunoelectron microscope confirmed that the normal mitochondria or cystic swollen one could be seen in the complete HeLa cells and the detached HeLa cells mitochondria, with the presence of Cx43 and VDAC1 in detached mitochondria. Immunofluorescence showed Cx43 colocalized with VDAC1. There was a significant difference in the Cx43 expressions of the subcellular structure in the HeLa cells [cytoplasm (1.23±0.11), cell nucleus (0.39±0.09), mitochondria (3.67±0.59), supernatant after segregation (0.16±0.06); F =84.17, P <0.05]. It also showed that the relative amount of Cx43 in mitochondria was enriched. Conclusions Cx43 is enriched in mitochondria in cervical cancer HeLa cells. Therefore, Cx43 in mitochondria might be a potential target in diagnosis, therapy and prognosis of cervical cancer.

Objective To explore the changes in serum concentrations of irisin,vaspin and reactive oxygen species (ROS) in patients with type 2 diabetes mellitus (T2DM) and metabolic syndrome (MS),and to investigated the correlation of irisin and vaspin with clinical parameters of MS.Methods A total of 260 T2DM patients were enrolled.Age and gender were recorded,anthropometrics,biochemical parameters,and levels of irisin,vaspin and ROS in fasting serum were measured,and homeostatic model assessment of insulin resistance (HOMA-IR) calculated.Wilcoxon rank sum test,correlation analysis,Logistic regression analysis,multiple linear regression analysis and receiver operating characteristic (ROC) curve analysis were performed.Results Compared to T2DM patients without MS,T2DM patients with MS had lower serum level ofirisin [male:112.81 (86.96-191.84) μg/Lvs.156.23 (110.61-225.97) μg/L,female:141.09 (77.52-175.55) μg/L vs.172.15 (95.69-240.37) μg/L,P <0.01],higher levels of vaspin and ROS [male:1.13 (0.95-1.38) μg/Lvs.0.36 (0.21-0.82) μg/L,1 540 (1 250-1 860) kU/Lvs.1 020 (920-1 350) kU/L;female:1.52 (1.13-1.80) μg/Lvs.0.51 (0.47-1.08) μg/L,1 650 (1 320-1 940) kU/Lvs.1 120 (980-1 420) kU/L,P <0.01].In the T2DM patients,serum irisin level was negatively correlated with vaspin (r =-0.382,P < 0.01) and ROS (r =-0.410,P < 0.01),while vaspin was positively correlated with ROS (r =0.400,P < 0.01).Multiple linear regression analyses showed that irisin was significantly correlated with body mass index (BMI),waist circumference and triglyceride,while vaspin was correlated with gender,BMI,and waist circumference (all P < 0.05).Logistic regression analysis revealed that irisin,vaspin and ROS were all associated with MS (OR =0.77,95 % CI 0.608-0.978;OR=1.39,95% CI 1.252-1.539;OR=1.38,95% CI1.112-1.718,all P<0.05).ROC analysis demonstrated that irisin and vaspin had significant area under the curve (AUC =0.931,P <0.01;AUC =0.777,P < 0.01) for the prediction of MS.Conclusions Serum irisin level was significantly decreased,while vaspin and ROS were significantly increased in T2DM patients with MS.Irisin and vaspin were associated with clinical presentations of MS,suggesting that irisin and vaspin might be valuable predictors of MS.

Objective Through the detection of the levels of serum betatrophin,anthropometric and biochemical indices,to determine the alteration of betatrophin levels in patients with polycystic ovary syndrome (PCOS) and the relationship between betatrophin levels and metabolic indexes such as 25-hydroxy vitamin D3 [25 (OH) D3].Methods The study group was composed of 24 women with PCOS and 20 age-natched healthy women as controls.Following general physical examination of the subjects,anthropometrie measurements were performed (height,weight,waist circumference,hip circumference).Body mass index (BMI) and waist-hip ratio were calculated;25 (OH) D3,Ca2+,fasting blood glucose,fasting insulin,triglycerides,high-density lipoprotein cholesterol (HDL-C),low-density lipoprotein cholesterol (LDL-C) were also measured and homeostasis model assessment of insulin resistance (HOMA-IR) were evaluated in all subjects.Serum betatrophin levels were examined with an enzyme-linked immunosorbent assay (ELISA).Using independent sample T test to compare the differences between groups,the linear correlation analysis was performed to study the correlation of betatrophin with anthropometric and biochemical indices,the influencing factors of betatrophin were analyzed by multiple linear regression.Results Circulating betatrophin levels [(0.341±0.034) ng/ml vs.(0.810±0.162) ng/ml,t=4.271,P<0.001] and HDL-C [(1.014±0.321) mmol/L vs.(1.419±0.287) mmol/L,t=2.218,P<0.05] significantly decreased in women with PCOS compared with controls,and BMI [(27.691±4.392) kg/m2 vs.(23.310±4.781) kg/m2,t=-2.073,P<0.05],fasting blood glucose [(5.950±0.411) mmol/L vs.(4.883±0.314) mmol/L,t=-2.142,P< 0.05],HOMA-IR [(4.946 ± 0.741) vs.(3.387± 0.397),t=-2.493,P<0.05],triglycerides [(1.510± 0.848) mmol/L vs.(1.037±0.402) mmol/L,t=-2.223,P<0.05],LDL-C [(3.431±0.479) mmol/L vs.(2.396±0.435) mmol/L,t=-2.433,P<0.05] were obviously increased.Moreover,there was a postive correlation between betatrophin and HOMA-IR (r=0.425,P<0.05) as well as 25(OH)D3 (r=0.577,P<0.05) and Ca2+ (r=0.448,P<0.05),while the betatrophin was negatively related to BMI (r=-0.451,P<0.05),triglycerides (r=-0.454,P<0.05),LDL-C (r=-0.551,P<0.05).Circulating betatrophin levels were higher when 25 (OH) D3 levels were beyond 35 nmol/L in PCOS patients [(0.539±0.092) ng/ml vs.(0.199±0.031) ng/ml,t=3.072,P<0.001],and the multiple linear regression analysis showed that the main factors affecting the levels of betatrophin were BMI (OR=-0.260),HOMA-IR (OR=0.218),25(OH)D3 (OR=0.238),and glycerol (OR=-0.162).Conclusion It is speculated that betatrophin may be a valuable predictive factor of PCOS because betatrophin is closely associated with insulin resistance,lipid metabolism disorders and the lack of 25-hydroxy vitamin D3 in PCOS patients.

<p><b>OBJECTIVE</b>To compare between MLO-Y4 osteocyte and osteoblast to support osteoclast formation in co-culture system.</p><p><b>METHODS</b>MLO-Y4 cells and murine osteoblast cells were co-cultured with bone marrow cells with or without vitamin D₃ presence.Bone marrow cells were as control group. Tartrat resistant acid phosphatase (TRAP)+ giant cells with three or more nuclei were counted and compared under a microscope at day 9.</p><p><b>RESULTS</b>In the absence of vitamin D₃, (1963.3 ± 93.1)/plate osteoclasts were observed when MLO-Y4 cells co-cultured with bone marrow cells in 24-well plate.While only (12.7 ± 5.5)/plate osteoclasts were found in the osteoblast group, and (6.0 ± 1.0)/plate in control group. The statistical difference occurs for any two groups (P < 0.05). Vitamin D₃ could significantly increase osteoclast formation in the three groups.</p><p><b>CONCLUSIONS</b>Osteocytes could induce osteoclastogenesis without the presence of vitamin D₃ and vitamin D₃ could enhance the induction effects of MLO-Y4 and osteoblast cells.</p>

<p><b>OBJECTIVE</b>To assess the quality of orthodontic clinical trials published in 4 major dental journals in the past 10 years and establish the reference standard for orthodontic clinical trials and quality control of dental journals.</p><p><b>METHODS</b>All the clinical trials published in Chinese Journal of Stomatology, West China Journal of Stomatology, Journal of Practice Stomatology and Chinese Journal of Orthodontics from 1999 to 2008 were searched. The demographic information of the papers was extracted and the quality of the clinical trials according to the consolidated standards of reporting trials (CONSORT) was assessed.</p><p><b>RESULTS</b>Four hundred and ninety-four clinical trials were retrieved, and 21.3% (105/494) of them were supported by grants. For the study design, only 26.1% (129/494) were prospective studies, and 3.8% (19/494) were randomized clinical trials. It was hard to evaluate precisely due to the lack of information about the details of the study designs. For the randomized clinical trials, the lack of details for randomization, allocation concealment, blinding and intention to treat compromised the quality.</p><p><b>CONCLUSIONS</b>The general quality of clinical trials in orthodontics is poor. It needs to be improved both in the clinical study design and the paper writing.</p>

RESUMEN

Objective To explore molecular fluorescence imaging features of the growth and metastasis of DsRed-marked mouse bladder carcinoma. Methods The study used lipofectamine 2000 transfection method,transferred on the BTT739 cells with plasmid chickenβ-actin-DsRed-Neo vector.The stably expressing BTT739-DsRed monoelonal cells were got with G418 selection.It randomly divided the 615 mouse of 24 into three groups,injected cell suspension on the hindlimb,the first and second group with BTT739-DsRed cell and the third group with BTT739 cell to found xenograft roodel.MAESTRO imager recorded fluorescence images of the growth and metastasis of the tumors in vivo and the fluorescence intensity was measured.The excitation wavelength was 560-580 nm,emission wavelength was 590-610 nm and exposure time was 5000 ms.After continuous observation of 4 weeks,every week killed the mouse of the second group and cut into image,made records of the red fluorescent mouse bladder cancer xenograft model,measured the tumor size and fluorescence sighal values; analyzed the relations between the tumor size and fluorescence signal values as well as between the whole image and cut image. Results DsRed tumor could be observed at the first week. Central local fluorescence loss could be detected at the second week, pathologically confirmed necrotic tumor tissue and a little connective tissue. At the fourth week, a local lymph node metastasis could be observed with no distant metastasis. The measured values of fluorescent signal were as follows: (89±18), (122±55), (133±69), (715±343)counts; the tumor size were as follows: (13±4), (45±22), (83±29), (253±67)mm2. The whole body image of tumor size were as follows: (12± 3),(50±23), (90±29), (290±74)mm2. The cut image of tumor size were as follows: (12±5), (72±30), (141±43), (524±237)mm2. The tumor size and fluorescent signal values reflect positive linear correlation with 0. 74 coefficient (t= 3. 97, P<0.05), whole body imaging and cut image reflect positive linear correlation with 0. 97 coefficient (t=10, P<0.05). The whole body image of tumor size was (70. 85±17.13) % of cut image. Conclusions Red fluorescent mouse bladder cancer xenograft model could observe the growth and metastasis of the tumor intuitively, continuously, and sensitively.As the tumor increased, the fluorescence range also increased, the fluorescence disappeared after tumor necrosis, the expression of the red fluorescent transferred after the metastasis of the tumor.

Objective To establish an indicator system for performance assessment of women and children health care institutions,and provide basis for performance management of such institutions.Methods In this study,literature searching,expert discussion,Delphi method,coefficient of variation method,correlation analysis,principal components analysis,factor analysis and cluster analysis were used to establish the indicator system.In addition,expert scoring and CRITIC methods were called into play to calculate the weight coefficient of each index,and to test the validity and reliability of such an indicator system.Results Thanks to the framework of this indicator system,we set up six class 1 indicators,including heahhcare quality,healthcare efficiency,quality and efficiency of public health service,social efficiency,economic benefit,and development potential.Conclusions Scientific studies can help us build a scientific and practical general performance assessment indicator system applicable to women and children care hospitals at all levels.

<p><b>OBJECTIVE</b>To explore the effect of focal-adhesion micromanipulation on the biological behavior of fibroblast.</p><p><b>METHODS</b>Micro-pot was made by microcontact printing. The molecules of constitutive protein was adhered on micro-pot by self-assemble of peptides. Skin fibroblasts were cultured on the membrane by self-made biomechanical cell culture for 2 weeks. Morphology observation and cell immunohistochemistry analysis was performed.</p><p><b>RESULTS</b>After 2 weeks, the morphology of the fibroblasts was diverse and more compliant. Cell immunohistochemistry analysis found that the expression of integrinbeta1, alpha5 and tensin was dramatically reduced.</p><p><b>CONCLUSIONS</b>The morphology and the biological behaviour of the fibroblasts in hypertrophic scar can be changed after micromanipulation of focal adhesion.</p>

<p><b>OBJECTIVE</b>To assess the effectiveness of vertical control with high-pull headgear in the treatment of high angle cases.</p><p><b>METHODS</b>Forty high angle cases (MP-SN >40 degrees) were included in this study and all the patients were treated with extraction. Twenty patients in the study group and control group, respectively. Totally 14 cephalometric measurements were selected for the assessment Paired t-test and independent t-test were used in the analysis.</p><p><b>RESULTS</b>No significant differences were found between the two groups (P > 0.05). In the study group, after treatment Y angle, MP-SN angle, MP-FH angle and U6-PP distance increased 1.9 degrees, 1.5 degrees, 2.3 degrees and 1.1 mm, respectively (P < 0.01).</p><p><b>CONCLUSIONS</b>High-pull headgear could not provide effective vertical control in high angle cases.</p>

Objective: To use in vivo fluorescence image analysis system for evaluating the efficacies of pVAX1-Ag85A and pVAX1-Ag85B DNA vaccines in treatment of bladder cancer cell-implanted tumors in mice. Methods: Discosomasp red fluorescent protein (DsRed) stably transfected bladder cancer BTT cell line (BTT-DsRed) was established and BTT-DsRed cell-implanted mouse model was constructed. Six days later, 24 BTT-DsRed-bearing mice were randomly divided into pVAX1-Ag85A DNA vaccine group, pVAX1-Ag85B DNA vaccine group, and saline group through injecting the pVAX1-Ag85A, pVAX1-Ag85B, and saline into the right hind limbs of mice, respectively. The growth and metastasis of implanted BTT-DsRed tumors were examined by in vivo fluorescence image analysis system. Results: BTT cell line stably transfected with DsRed (BTT-DsRed) was successfully established. Fluorescence visible mouse model was successfully es-tablished by inoculating BTT-DsRed cells into hind limbs of mice. After treatment with pVAX1-Ag85A or pVAX1-Ag85B for 2 weeks, the in vivo tumor fluorescence intensity in pVAX1-Ag85B group was significantly lower than that in the saline group (P <0.05). After 3 weeks, tumor fluorescence intensities in both pVAX1-Ag85A and pVAX1-Ag85B groups were significantly lower than that in the saline group (P < 0.01). But the efficacies of pVAX1-Ag85A and pVAX1-Ag85B groups were similar (P > 0.05). The distant lymphatic metastasis rate in pVAX1-Ag85B group was significantly lower than those in the saline (25.0% vs 87.5%) and pVAX1-Ag85A groups (25.0% vs 62.5%) (P <0.05). Conclusion: In vivo fluorescence image analysis system can dynamically, sensitively and visually evaluate the anti-tumor effects of DNA vaccines against bladder cancer cell-implanted tumors. Both pVAX1-Ag85A and pVAX1-Ag85B DNA vaccines have anti-tumor effects for bladder cancers.

<p><b>OBJECTIVE</b>To compare the dental change and the mechanism after non-extraction treatment between passive self-ligating and conventional bracket systems.</p><p><b>METHODS</b>Twenty-six non-extraction crowding cases were selected. Half of them were treated with Damon3 self-ligating brackets and the others were treated with conventional brackets. The results were compared. Stepwise regression analysis was performed for the two groups.</p><p><b>RESULTS</b>There were no statistically significant differences in the changes of the intercanine and interpremolar widths and L1-MP angle (P > 0.1). In the self-ligating bracket group, the intermolar width increased by 1.42 mm and the L1-APo increased by 2.66 mm. In the conventional appliance group, the intermolar width increased by 0.65 mm and L1-APo increased by 1.57 mm. The differences were statistically significant between the two groups (P < 0.1). Stepwise regression analysis showed that in self-ligating bracket group, the coefficient of determination for the changes of L1-MP was 96.6%.</p><p><b>CONCLUSIONS</b>In crowding cases treated with non-extraction, there were an overall increase in the proclination of the mandibular incisors and the arch width in both bracket groups. Compared with cases treated with conventional appliances, the cases treated with Damon3 brackets resulted in greater intermolar width increases; In Damon3 bracket group, the change of the lower incisor inclination was influenced not only by the mandibular crowding and the intermolar width before treatment but also by the patient's skeletal pattern and the changes of the arch width during treatment.</p>

RESUMEN

<p><b>OBJECTIVE</b>To evaluate the dynamic changes of Streptococcus mutans concentration of plaque during fixed appliance treatment and the effects of two materials of ligation on Streptococcus mutans concentration.</p><p><b>METHODS</b>Twenty-eight patients undergoing fixed appliance treatment were observed. Ligature wire and elastomeric rings were applied on one side of arches, stochastically. The dynamic changes on the quantity and percentage of Streptococcus mutans were observed before and after fixed appliance bonding.</p><p><b>RESULTS</b>Statistically significant increase of the quantity and percentage of Streptococcus mutans was found after fixed appliance bonding, and the percentage of Streptococcus mutans in the plaque around the brackets ligated with elastomeric rings was more than that of ligature wire at the beginning of fixed appliance bonding, statistically.</p><p><b>CONCLUSIONS</b>The finding suggested that the caries-associated capability of the plaque increased after bonding and there was greater caries-associated capability of the plague on the teeth when elastomeric rings was used than that of the plague when ligature wire was used at treatment beginning. The ligature wire is recommended in the fixed appliance treatment.</p>

RESUMEN

<p><b>OBJECTIVE</b>To investigate the stability of upper molars with the application of micro-screw implant anchorage during orthodontic treatment.</p><p><b>METHODS</b>Fifteen adult patients with severe maxillary protrusion were included. Upper first premolars were extracted and upper posterior anchorage was reinforced with micro-screw implant in all patients. The average treatment period to close the extraction space was 10.5 months. Cephalometric and cast analysis were carried out.</p><p><b>RESULTS</b>During the treatment, the micro-screw implants kept stable in sagittal plane; neither the mesial-distal movement nor the rotation or tipping of the upper molars during the treatment was of statistic significance (P > 0.05); the edge of upper incisors was retracted by 6.99 mm on average, and no significant vertical change was observed (P > 0.05).</p><p><b>CONCLUSIONS</b>Micro-screw implant could provide good anchorage control in the orthodontic treatment.</p>

<p><b>OBJECTIVE</b>To record condylar movements of patients with mandibular deviation and analyze their characteristics.</p><p><b>METHODS</b>Computer aided diagnosis axiography (CADIAX III) was used to record and analyze condylar lateral movements in 31 patients (9 males, 22 females and the age range 12 - 26, mean age 18 years old) with mandibular deviation. Paired-t test and rank sum test were used to compare condylar movements. Multinomial logistic analysis was used to clarify the relationship between condylar movement and mandibular deviation.</p><p><b>RESULTS</b>The condylar lateral movements in patients with mandibular deviation were asymmetric. The tracing of the shifted side was longer than the contralateral side (P < 0.01). The transverse inclination of the shifted side was smaller than the contralateral side (P < 0.01). With the increase of deviation, the movement length difference between the two sides increased and the length of contralateral side decreased (P < 0.01).</p><p><b>CONCLUSIONS</b>The patients with mandibular deviation not only have morphological asymmetry but asymmetric condylar movement length and inclination in lateral movements. There is specific relationship between condylar movement and mandibular deviation.</p>

BACKGROUND: Although we have gained much information about leadinduced organ damage, the effect of blood lead level on T cell subgroup is yet to be determined.OBJECTIVE: To assess the effect of blood lead level on T cell subgroup of children, and the association of T cell subgroup with threshold limit value of blood lead level.DESIGN: Comparative observation.SETTING: The Institute of Molecular Microorganism, Maternity and Child Care Hospital, Shanxi Provincial Children Hospital.PARTICIPANTS: Sixty children, (32 boys and 28 girls), aged 3-6 years with a mean of (4.5±1.5) years old, were selected from the suburbs of Taiyuan city between May 2003 and October 2003. Informed consents were obtained from all their guardians. The enrolled children according to their blood lead levels were assigned into three groups, 13 in Group Ⅰ with blood lead level ≥ 0.48 μmol/L, 20 in Group 2 with lead level ≥ 0.24 μmol/L but < 0.48 μmoL/L and 27 in Group 3 with lead level < 0.24 μmol/L.METHODS: Blood lead level and expression of T cell subgroup were measured with graphite furnace atomic absorption spectroscopy and immunofluorescence methods respectively. Student t test was used in data analysis, and linear correlation analysis was used to assess the correlation between blood lead level and expression of T cell subgroup.level and expression of T cell subgroup.pression of CD3 and ratio of CD4 to CD8 were lower in Group 1 (lead level ≥0.48 μmol/L) than Group 3 (lead level < 0.24 μmol/L) (t=3.27,blood lead level showed had significant inverse correlation with CD3 expression and the ratio of CD4 to CD8(r=-0.689,-0.674,P < 0.01).CONCLUSION: A blood lead level ≥ 0.48 μmol/L, is shown to significantly decrease the expression of CD3 and ratio of CD4 to CD8 in peripheral blood, which may impair the children's immunological function.

<p><b>OBJECTIVE</b>To introduce a new modified twin-block advancement appliance and investigate the effects on respiratory variables in patients with OSAS.</p><p><b>METHODS</b>29 patients with OSAS participated in the study and were fitted with modified twin-block appliances to hold the mandible in an anterior and inferior position. Polysomnography was performed with and without appliance insertion. And questionnaires were used for registration of patients subjective symptoms. Pair-t analysis was used to evaluate the effects of appliances in patients with OSAS.</p><p><b>RESULTS</b>26 patients responded to the appliance therapy. Apnea-hypopnea index, apnea index and hypopnea index were reduced significantly (P < 0.01). Lowest arterial oxygen saturation improved significantly (P < 0.01). Discomfort with mandibular advancement disappeared within one week.</p><p><b>CONCLUSIONS</b>Modified twin-block advancement appliance is a conservative, successful treatment alternative that could benefit patients suffered from OSAS.</p>

<p><b>OBJECTIVE</b>The research tended to approach applying of adjustable appliance in the treatment of obstructive sleep apnea-hypopnea syndrome (OSAHS).</p><p><b>METHODS</b>30 OSAHS patients (24 males and 6 females) participated in the adjustable group, with a mean age of (49.9 +/- 9.9) years old. AHI was (33.1 +/- 22.7) per hour. The control group consisted of 30 OSAHS patients wearing ordinary mandibular advancing appliance in the corresponding period, with age, weight and AHI at the same level. Monoblind way was designed to obtain and analyze the therapy differences. Differences in changes of upper airway, mandible and hyoid bone were also analyzed among the doctor-experience position, final adjusted position and original position.</p><p><b>RESULTS</b>AHI decreased by 85.5% in the adjustable appliance group. The change in AHI was greater significantly (P = 0.025) in the adjustable group than in the control group. In the final adjusted position, the amount of mandibular advancement was (5.8 +/- 1.4) mm [(71 +/- 26)% of the maximum range of protrusion] and that of bite opening (the distance between upper and lower incisor edges) was (4.6 +/- 1.1) mm.</p><p><b>CONCLUSIONS</b>The adjustable appliance had shown better therapy effect in OSAHS patients. The final adjusted position provided useful information on determining mandibular position using other appliances.</p>
