RESUMEN

In order to improve the effectiveness of private class inquiry with the development of information teaching, the smart teaching platform has been established, with instructional management, curriculum setting, teacher preparation, classroom application, supervision and monitoring modules. Taking the platform as the medium, the small class inquiry learning community of entity curriculum is constructed between students and the teachers. In the eight-year medical teaching, the content of learning cycle is designed according to the entity curriculum, which is issued on cloud platform before class, in class and after class. Students learn basic concepts by themselves in the learning community, explore the application of knowledge under the guidance of teachers, and expand knowledge in class or after class. After having test in teaching procedure, the small class learning community based on smart teaching cloud platform has a submission rate, interaction rate and score rate of more than 90%. It can not only make full use of the advantages of information-based teaching resources, but also build face-to-face learning community in the course teaching, reflecting the emotional interaction of personalized teaching. It's suggested that new approaches to teaching should be student-centered and activity-based, engaging students actively in the learning process, which can promote students' autonomous learning ability and innovative thinking ability.

OBJECTIVE:To study the effects of Modified liuwei dihuang decoction on kidney/bone injury of chronic kidney disease-mineral and bone disorder(CKD-MBD)model rats. METHODS :The male SD rats were randomly divided into normal group(n=10),high phosphorus group (n=30),model group (n=30),calcitriol group (positive control ,0.09 μg/kg,n=30), Modified liuwei dihuang decoction group (10 g/kg by crude drug ,n=30). CKD-MBD model was established by high phosphorus and adenine diet for 6 weeks. After modeling ,normal group and model group were given normal diet/high phosphorus diet and intragastric administration of water. Administration groups were fed with normal diet and given corresponding solution intragastrically(water as solvent ),0.1 mL/kg,once a day ,for consecutive 6 weeks. Blood sample of rats in the normal group were collected ,and they were sacrificed after the last administration. Blood sample of 10 rats in each other group were collected , and they were sacrificed at 2,4 and 6 weeks after administration. The contents of blood urea nitrogen (BUN),serum creatinine (Scr),calcium,phosphorus,iPTH,FGF-23,RANKL and osteocalcin in serum were detected in each group. The bone mineral density(BMD)of femoral was measured ,the morphological changes of renal tissue and bone tissue were observed ,and the percentage of renal tubular injury and the score of renal interstitial fibrosis were calculated. RESULTS :Compared with normal group, above indexes in high phosphorus group had no significant change at different time points (P>0.05). There was no abnormal change in renal/bone tissue. Compared with high phosphorus group at the same time point ,the contents of BUN ,Scr, phosphorus,iPTH,FGF-23,RANKL and osteocalcin in serum ,the percentage of renal tubular injury and the score of renal interstitial fibrosis in the model group were significantly increased ,while the contents of calcium in serum and the BMD of femoral were significantly decreased (P<0.05 or P<0.01). The renal tissue showed diffuse fibrosis. The width of trabecular bone was increased and the number of osteoblasts was decreased. Compared with the model group at the same time point ,the contents of BUN(except for Modified liuwei dihuang decoction group after 2 weeks of administration ),Scr,serum phosphorus ,iPTH, FGF-23,RANKL and osteocalcin ,the percentage of renal tubular injury and the score of renal interstitial fibrosis in Modified liuwei dihuang decoction group and calcitriol group were decreased significantly at each time point ;serum calcium content and BMD(except for 2 weeks of administration )were significantly increased (P<0.05 or P<0.01),and the pathological changes of renal/bone tissue were significantly improved ;there was no statistical significance in above indexes between Modified liuwei dihuang decoction group and calcitriol group (P>0.05). RESUMEN

Objective·To establish a cell-based screening system for identification of compounds with activity in regulating retinoic acid receptor (RARα) stability. Methods·The modified pMSCV plasmid constructs, named as RARα-EGFP-IRES-DsRed, consists of enhanced green fluorescent protein (EGFP) fusing to RARα and red fluorescent protein (DsRed) as internal references incorporating the internal ribosome entry site (IRES) as interval sequence. The RARα-EGFP-IRES-DsRed plasmid was stably transfected into NB4 cells which were named as NB4-pMGIR-RARα. Fluorescence signals of EGFP and DsRed indirectly reflecting the expression of RARα, were detected by flow cytometry in cells that were treated with all-trans retinoic acid, sodium valproate, cytarabine, lenalidomide, etoposide, montelukast and gambogic acid, respectively. Effects of these compounds on the expression of RARα protein were further examined by Western blotting. Results·A double fluorescence reporter system for screening compounds that can increase the stability of RARα protein was successfully established, and sodium valproate was identified as a potent compound to promote the stability of RARα. Conclusion·The double fluorescence reporter system can be used to screen compounds regulating the stability of RARα protein, which can be further used to identify compounds regulating the stability of other proteins.

RESUMEN

It is necessary to monitor intraoperative spinal function in order to prevent spinal neurological deficit during spine surgery. This study aims to extract characteristic electrophysiological monitoring parameters during surgical treatment of scoliosis. The problem, "the monitoring parameters in time domain are of great variability and are sensitive to noise", may also be solved in this study. By use of continuous wavelet transform to analyze the intraoperative cortical somatosensory evoked potential (CSEP), three new characteristic monitoring parameters in time-frequency domain (TFD) are extracted. The results indicate that the variability of CSEP characteristic parameters in TFD is lower than the variability of those in time domain. Therefore, the TFD characteristic monitoring parameters are more stable and reliable parameters of latency and amplitude in time domain. The application of TFD monitoring parameters during spine surgery may avoid spinal injury effectively.

RESUMEN

Objective To investigate the modulatory effect of orexin A on glutamate receptor-mediated current in the freshly isolated pyramidal neurons from the rat prefrontal cortex (PFC). Methods Deep layer (Ⅴ-Ⅵ) prefrontal cortical pyramidal neurons from postnatal 10 to 14 day-old Wistar rats were acutely dissociated by a combination of mechanical and enzymatic method. Subsequently, the effect of orexin A on the current induced by glutamate was studied by the technique of whole cell patch clamp. Results Both orexin A and glutamate dose-dependently evoked the inward transmembrane current. The current was evoked by 1 mmol/L glutamate as a control group(100%). After treatment with 1 mmol/L orexin A for 4-10 s, 1 mmol/L glutamate induced-current was increased by (46.59?15.19)% (n=8, P

RESUMEN

Objective To observe the postnatal development and perinatal electrophysiological characteristics of oligodendrocyte precursor cell (OPC) in rats. Methods Immunohistochemistry and Western blot were applied to determine the expression of NG2 OPC in cerebral cortex and hippocampus at various developmental stages of SD rats. Electrophysiological characteristics of OPC were also recorded in slices of 7-day rats. Results The majority of hippocampal and cerebral OPC exhibited stellate shape,a small cell body with a few processus. Total population of the NG2 immunopositive OPC was numerous at P7d in cerebral cortex and hippocampus. OPC expressed in adult rats with slightly more quantity. Moreover,OPC in hippocampus of P7d rats typically exhibited small inward sodium current and weak active responses, whereas only outward potassium current and inactive responses were recorded in white matter OPC of P7d rats. Conclusion Total population of OPC and relative optical density of NG2 are the highest in P7d rats at the postnatal developmental stages. OPC in cerebrum and hippocampus of P7d rats displays electrophysiological heterogeneity.

RESUMEN

Objective To study the effects of hypoxia and glutamic acid on the kinetic properties of NMDA receptor channel of the hypothalamic neurons in rats. Methods Cell-attached mode patch clamp technique was employed to record the single channel current of the NMDA receptor. Results The open probability of NMDA receptor channel was increased after acute hypoxia compared with that of normal state, the open time τ1,τ2 was changed from (0.33±0.10)ms,(4.36±0.26)ms to (0.93±0.22)ms,(7.64±0.72)ms, and the close time τ1,τ2 was from (18.03±3.50)ms,(171.50±19.10)ms to (3.42±1.02)ms,(19.39±3.07)ms. The mean open probability was changed from 0.12±0.05 in normal state to 0.66±0.36 in hypoxia state. Furthermore, glutamic acid can increase open time and open probability of NMDA receptor channel, decrease close time. Conclusion The excitability and the open probability of NMDA receptor channel of hypothalamic neurons increased by hypoxia is related to glutamate.

RESUMEN

Objective To observe the electrophysiological characteristics of the inward current of cardiomyocyte-like cells derived from rat bone marrow mesenchymal stem cells (MSCs). Methods Bone marrow specimens was extracted from SD rats aged 1 month. MSCs were isolated by gradient centrifugation and obtained by adherence culture. The second-passage MSCs were treated with 10 ?mol/L 5-azacytidine and incubated for 24 h. After 4 weeks, cardiomyocyte-like cells were identified by immunocytochemical staining technique, then used to detect the inward current with the whole-cell patch-clamp technique, which results were compared with the normal cardiomyocytes and the MSCs without induction. Results Some cardiomyocyte-like cells were stained positive for troponin T, and the quickly activated and inactivated inward currents were elicited 4 weeks after 5-aza cytidine treatment. The inward currents of cardiomyocyte-like cells recorded in the same square pulse were weaker than that of the normal cardiomyocytes, and the I-V curve of cardiomyocyte-like cells was right-shifted. Conclusion After treated with 5-azacytidine, MSCs differentiate into cardiomyocyte-like cells which express the excitable inward current.

RESUMEN

Objective To develop new methods to cultivate, retrieve and purify oligodendrocyte progenitor cells (OPCs). Methods Primary OPCs were isolated from mixed glial cultures of newborn Sprague Dawley rat forebrains by using a selective detachment procedure, further purified by differential adhesion, and maintained in a chemical conditioned medium. The characteristic of OPCs was determined by both electrophysiological recording and immunostaining. Results Immunocytochemical analysis revealed that more than 90% of the isolated cells expressed the OPCs-specific marker NG2. OPCs exhibited a moderate input resistance, delayed rectifier K+ currents (IK) that could be antagonized partly by TEA-Cl and CsCl, small tetrodotoxin (TTX) sensitive Na+ currents that failed to generate typical action potentials. Conclusion Isolation and purification by selective detachment and differential adhesion procedure are effective to obtain highly purified OPCs. OPCs, distinct from neuron, astrocyte, oligodendrocyte or microglia, possess unique physiological properties.
