RESUMEN

OBJECTIVE: To study the immunomodulatory effects of Astragalus polysaccharide (APS) in type 1 diabetic mice. METHODS: A mouse model of type 1 diabetes mellitus was established by intraperitoneal injection of multiple low dose streptozotocin (MLD-STZ). The diabetic mice were intraperitoneally administered 100, 200, 400 mg/kg APS or 1 ml normal saline (NS) every day respectively, then the diabetic mice were sacrificed after 15 or 30 days of treatment. The effect of APS on insulitis was determined via pancreatic histological analysis. Serum insulin autoantibody (IAA) levels were measured by radio-immunoassay (RIA). Proliferation ability of splenocytes to concanavalin A was tested by using [(3)H] thymidine incorporation assay. The levels of cytokine interferon-gamma (IFN-gamma) and interleukin-4 (IL-4) secreted by splenocytes were determined by enzyme linked immunosorbent assay (ELISA) method, and the expression of peroxisome proliferator-activated receptor gamma (PPARgamma) in spleens was characterized using Western-blot analysis. RESULTS: Attenuated insulitis, down-regulation of the serum IAA levels and Th1/Th2 cytokine ratio, decrease of the proliferation ability of splenocytes to concanavalin A, and up-regulation of the PPARgamma levels in spleens showed a significant time- and dose-dependent response to APS treatment as compared with the NS-treated group. CONCLUSION: APS possesses immunotherapeutic effects on mice with type 1 diabetes mellitus through improving the cell- and humoral-mediated immunity.

Objective To study the protective effects of vitamin E(VE) on injury to male reproductive function in tail-suspended rats. Method Thirty adult male Spraque-Dawley(SD) rats were randomly divided into three groups:control, TS (tail-suspension) and VE+TS groups. TS and VE+TS groups were tail-suspended for 14 d,and VE was given orally to VE+TS group during tail-suspension. After the treatment, weight and morphology of testes, quality and amount of sperm, as well as serum hormones were observed. Objective To study the effects of simulated weightlessness induced by tail-suspension on reproduction in adult male rats and its mechanisms. Methods Twenty Spraque-Dawley adult male rats were randomly divided into two groups: the experimental group and control group. Rats in the experimental group were tail-suspended for 14 days. After the treatment, the weight and morphology of testis, the quantity and quality of sperm, the level of serum testosterone and the apoptosis rate of testicular cells were observed. Results Compared with the control, the testicular weight, the quantity and quality of sperm, the level of serum testosterone significantly decreased (P<0.05) while the apoptosis rate of testicular cells significantly increased (P<0.05) in tail-suspended rats. In addition, hematoxylin and eosin (HE) staining showed that seminiferous tubules atrophied, spermatogenic cells decreased, seminiferous epithelial cells disintegrated and shed, and the lumina were azoospermic. Conclusion Simulated weightlessness induced by tail-suspension has a harmful effect on male reproduction of adult rats by increasing the apoptosis of testicular cells.

Objective To investigate the proliferative response and time course of endogenous neural stem/progenitor cells after cerebral cortical concis in the adult rats. Methods Eighty adult male Sprague-Dawley rats were used in this study. Cumulative BrdU labeling was employed to detect the proliferating cells. At 1 d, 3 d, 7 d, 14 d, and 21 d after cerebral cortical concis, the rats were killed for BrdU immunohistochemical staining and cell counting in the injured ipsilateral SVZ. Results Little BrdU immunoreactivity cells was present in SVZ of the control rats from day 7 to day 21 after sham operation. The number of BrdU immunoreactivity cells in the injured ipsilateral SVZ increased at day 1 and peaked at day 7 after cerebral cortical concis. Conclusion After cerebral cortical concis of the adult rats, neural stem/progenitor cells in the injured ipsilateral SVZ markedly proliferated with a peak at day 7. This finding may be important for manipulating SVZ cells to promote the recovery from cerebral cortical concis.

Objective To study the expression of interleukin-2 (IL-2), soluble interleukin-2 receptor (sIL-2R),determine the alteration of erythrocytic immunity and T cell subgroup in the blood of outer circulation in patients with hypertensive cerebral hemorrhage so and to probe into the relationship between them, and to explore the clinical significance. Methods Enzyme linked immnunosorbent assay (ELISA) was used to determine the content of IL-2 and sIL-2R. The immunoadsorption was employed to examine the erythrocytic immune activity and its regulating function.Streptavidin-peroxidase(S-P) was used to determine the cell number of CD3 (cluster of differentiation3), CD4 and CD8. Results The content of IL-2 in the group with hypertensive cerebral hemorrhage was significantly lower than that in the control group (P<0.01), and the content of sIL-2R increased. Red blood cell C3b receptor (RBC. C3b R)and RBC immune adherence enhancing factor (RFEB) dropped greatly (P<0.01), while RBC immune complex rosette (RBC. ICR) and RBC immune adherence inhibiting factor (RFIR) increased greatly. The cell number of CD3 and CD4decreased (P<0.01) and there was no obvious change in CD8 (P<0.05). Conclusion The decrease of immune function was observed in patients with hypertensive cerebral hemorrhage. The determination of the content of IL-2, sIL-2R, erythrocytic immunity and the activity of T subgroup has an important clinical significance in the occurrence,development, treatment, and prognosis of hypertensive cerebral hemorrhage.

RESUMEN

The regulation of vascular and trabecular smooth muscle relaxation or contraction in the penis, that is, the physiology of corporal smooth muscle tone, determines penile erection or flaccidity. There is considerable evidence that the potassium channel and calcium channel, like many other vascular tissues, are the major modulators of smooth muscle tone in the corpora. Moreover, data on cultured corporal smooth muscle cells and isolated corporal tissue strips have demonstrated that the neurotransmitters participating in erection modulate corporal smooth muscle tone largely through their effects on ionic channels and transmembrane ionic flux.

RESUMEN

The signal transduction pathways within corporal smooth muscle cells are the intracellular molecular mechanisms of corporal smooth muscle tone regulation. Various neurotransmitters activate the membrane receptor proteins or intracellular enzyme pathways and result in the production of extracellular chemical signals. Second messenger molecules and ions transmit and amplify the signals, and subsequently induce the relaxation of smooth muscle cells and penile erection. Therefore, the study of signal transduction in cavernous smooth muscle play an important role in understanding the physiology of erection and the pathophysiology of erectile dysfunction as well as in developing new selective drugs for the treatment of erectile dysfunction.

Objective To study the content of interleukin- 6 (IL-6) and the altering pattern of erythrocyte immunity and T cell subgroup in the blood of outer circulation in the patients with cerebral glioma, and to expl ore their relationship and the clinical significance. Methods Enzyme linked immunosorbent assay (ELISA) was used to determine the content o f IL-6. The immunoabsorption was employed to examine the erythrocytic immune ac tivity and its regulatory function. Streptavidin-Peroxidase (S-P) was used to determine the cell number of CD3 (cluster of differentiation 3), CD4, CD8. Results The content of IL-6 in the group with cerebral glioma i s significantly lower than that in the control group (P0.05). Conclusion The weakening of immune function is obse rved in the patients with cerebral glioma. Testing of the content of IL-6, eryt hrocyte immunity and the activity of T subgroup has important clinical significa nce in the occurrence, development, treatment, outcome and prognosis of the cere bral glioma.

RESUMEN

Objective To assess the efficacy of radiotherapy and synthetic therapy for vertebral hemangiomas.Methods Clinical symptoms,imaging findings,therapeutic methods and following up after treatment in 22 cases with vertebal hemangiomas were retrospectively analyzed.Results Complete relief of symptoms(CR)were occurred in 19 of 22 patients(86.4%) and partial relief(PR)were achieved in 3 of 22 patients(13.6%).There was no obvious complication of radiotherapy and radiation-induced neoplasms.There was no relapse.Conclusion For vertebral hemangioma,the synthetic therapy which includes intenventional therapy before operation as well as radiotherapy after operation is of obvious superiority in relieving compression rapidly,reducing hemorrhage during operation and obtaining good efficacy.

ObjectiveTo investigate the influence of aging and castration on the density of andro gen receptor(AR) in the male rat heart. MethodsAR in the heart of rats aged 40d,90d(intact and castrated) and 2 years were demonstrated by means of immunohistochemical technique and the den sity of the positive staining was measured by using image analys is. The optical density of AR in the septum of the rats were analyzed by anova and doncan process of the SAS data analysis system. Re sultsOD(AR) of the group aged 2 years was higher than that of the groups aged 40d and 90d (0. 3550±0. 0077,0. 31080. 0264 and 0. 3149±0. 0200 respectively), thetwolattergroups showed similarly intensive staining, while the value of castrated group(0. 4136±0. 0320) was signif icantly greater than that of intact same aged group. Conclusion The development of sexual gland has little influence in the density of AR in the male rat heart during prepuberty to adult, both old age and castration increase the density of AR in the rat heart.

ObjectiveTo study the distribution, developmental patterns, and hormonal control of androgen re- ceptors(AR) in the developing and ethane dimethane sulfonate(EDS) treated male SD rat epididymis. MethodsThe ABC technique of immunohistochemistry and image analysis were used to assess optodensity means (OPTDM) of AR, providing a measure of relative nuclear AR concentration. ResultsThe specific AR immunostaining was observed in the nuclei of epididymal epithelium, peritubular smooth muscle cells and intertubular connective tissue cells. The rela- tive AR concentrations varied with the different segments of the epididymis in the adult rat(P<0. 05 or P<0.01). AR protein was highest in the caput (0. 763--0. 026),lowest in the corpus (0. 712±0. 025) and intermediate in the cauda (0. 736±0. 008). Levels of epididymal AR changed with development. In the cauda, AR level was highest on day 21 (0. 773±0. 028),intermediate on day 35(0. 762±0. 022),and lowest on day 90~120(0. 736±0. 008). The 90~120d group was significantly different from the 21d group (P<0. 01)and 35d group (P<0. 05). After the adult rats were treated with EDS to eradicate Leydig cells and endogenous testosterone, it was observed that the OPTDM of AR in the epididymal cauda epithelium was significantly reduced (P<0. 001), and was restored to the control level by using ex- ogenous testosterone replacement (P<0. 001). Conclusion These results suggest that the epididymal corpus depends least on androgens and the AR expression in the epididymis decreases with age and is dependent on circulating andro- gens.

Objective:To construct a recombinant adenovirus vector harboring fusion gene NT4-p53(N15)-Ant,laying a foundation for gene therapy research of malignant tumors.Methods:The p53(N15)-Ant gene was obtained by T-vector method and was inserted in pBV220/NT4 vector after digested with restriction enzyme.The fusion gene of NT4-p53(N15)-Ant was subcloned into the shuttle plasmid of adenovirus;the products were cotransfered into HEK-293 cell line with helper plasmid PJM17.The recombinant adenovirus was produced by homologous recombination of above 2 plasmids in HEK-293 cells and its titer was measured by plaque-forming.The expression of Ad.NT4p53Ant in transfected 293 cells was confirmed by reverse transcription polymerase chain reaction(RT-PCR)procedure.The effect of Ad.NT4p53Ant on HepG2 cell line was measured by a colorimetric 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide(MTT)assay.Results:The p53(N15)-Ant gene was confirmed by restriction enzyme digestion and DNA sequencing.High titer of recombinant adenovirus was obtained by homologous recombination in HEK-293 cells(1?10 11pfu/ml).The expression of NT4-p53(N15)-Ant gene in 293 cells was confirmed by RT-PCR.Ad.NT4p53Ant had strong killing effect on HepG2 cells.Compared with Ad.GFP,Ad.NT4p53Ant significantly decreased the survival rate of HepG2 cells.Conclusion:The recombinant adenovirus vector encoding gene NT4-p53(N15)-Ant has been successfully constructed in this experiment by molecular cloning and in vitro recombination techniques,laying a foundation for further research of gene therapy of cancer.

Objective: To investigate the influence of transfection of TCRVJ37.1 gene in cytotoxicity of PBMC of healthy donor to breast cancer cell line MCF-7/S. Methods :pcDN*31vp'1 packaged by lipofectamine were transfected to healthy donor's PBMC,Flow Cytometer Analysis and MTT Colorimetric Assay were used respectively to test the expression of pcDNA3.1VB7.1 gene and the cytotoxicity of PBMC before and after gene transfection of healthy donor to breast cancer cell line MCF-7/S. Results: The expression of TCRVJ37.1 gene after transfection was obviously higher.There was distinguish differences of cytotoxicity before and after gene transfection. Conclusion:The modified healthy donor's PBMC by TCR gene could have stronger cytotoxicity to breast cancer cell line.

Objective To study the effects of vascular endothelial growth factor(VEGF) in early stage diabetic retinopathy and their mechanisms,so as to guide the clinical work theoretically.Methods Thirty-six SD rats were randomly divided into four groups.Nine rats were used as normal controls(M Group).Streptozocin were injected intraperitoneally to induce diabetes for 1 month(M1 Group),2 months(M2 Group),and 3 months(M3 group).At the experimental ends of each group,the rats were over anesthetized and their eyeballs were extracted to make digest preparation.Immunofluorescence were used to investigate the expression of VEGF on retinal blood vessels.Transmission electromicroscopy was used to observe the histology of the retinal vessels.Results VEGF immunofluorescence showed an expression rate of 56% in M2 group and 89% in M3 group.Transmission electromicroscopy showed no change in M Group.However,it showed swelling of endothelial cells,finger like process into the capillary cavity,uneven distribution of heterochromatin in pericytes in M1 Group.Obvious fragmental thickening and splitting of basement membrane,swelling and deformation,finger like process to the capillary cavity,and concentration and margination of heterochromatin in endothelial cells,and swelling and deformation of rough surfaced endoplasmic reticulum and mitochondrion in pericytes were observed in M2 and M3 groups,especially in the latter.Conclusion The expression of VEGF proteins in the retinas of diabetic rats increases with the progression of diabetes.It proves that VEGF plays an important role in the occurrence and development of early diabetic retinopathy.

Objective To construct the expression box of fusion gene NT4-p53(N15)-Ant.Methods The gene of p53(N15)-Ant was obtained by PCR of two primers templating with each other and T-vector cloning method.The positive clone was identified and analyzed by the restriction enzymes and sequencing respectively.After digested with restriction enzyme,the interest gene of p53(N15)-Ant was subcloned into the plasmid pBV220/NT4.Results The gene of p53(N15)-Ant was confirmed by the digestion of restriction enzyme and sequencing.The recombinant plasmid pBV220/NT4p53(N15)Ant was identified by the digestion of restriction enzyme and agarose gel electrophoresis and the results conformed theoretical values.Conclusion The plasmid pBV220 containing the expression box of NT4-p53(N15)-Ant was successfully constructed by molecular cloning and recombination techniques in vitro,which will guide further study on gene therapy of cancer.

In recent half century,the incidence of male infertility is in an increasing tendency;however,the etiological factors and its pathogenesis are not fully clarified yet.By associating with the some results of our research,some hot spots on the pathogenesis of male infertility are briefly approached here,and the effects of varicocele,formaldehyde and infertility-related genes on male infertility are especially focused.

In ultrastructural level, this paper shows that the pig bile extracts and its effective constituent, the sodium deoxycholate, could fragment the Trichomonas vaginalis in vitro, The flagellum and caudal projection were splitted up; the plasma membrane, the intracellular membrane system, such as the limiting membranes of hydrogcnosome, phagosome and nuclear envelope were injured too; and cytoplasm was condensed, even the whole body was fragmented. The mechanism of the effeces is probably due to the fact that the surface activity of the drugs accelerated degradation of lipids which are made of the biomembrane system of human trichomonas vaginalis.

Objective To study the microstructural and ultrastructural changes of the epididymis of experimental varicocele in adolescent rats and it's role in infertility resulting from varicocele. Methods A varicocele model was performed in adolescent Sprague Dawley rats by partially ligating the left renal vein,the different segments of the epididymides of the rats were prepared for light and electron microscopy,the microstructure and ultrastructure of the epididymis were studied. Results There were lesions of different degree and segment specific changes in the epididymis with varicocele.Light microscopically,the main changes were interstitial vascular hyperemia,lymphocytes infiltration,sperm granuloma developed in the interstitial;The structure of the columnar epithelia was anomalies,epithelial cells degeneration,even the vacuoles appeared in the epithelial cells.The number of halo and clear cells increased.Inside the cavity of the duct,there were shedding cells,macrophage,deformed sperms and residual bodies.Electron microscopically,numerous large lysosomes,the residual bodies,the defected main cellular organelles(e.g. the endoplasmic reticulum,the mitochondrion and the Golgi complex etc.)and even large clear vacuoles were presented in the cytoplasm of principal cells.Clear cells were filled with lysosomes that made them frequently bulging into the lumen.The microvilli of the columnar epithelia were sparse and showed local defects.The thickness of the basal membrane increased.Conclusion\ The experimental varicocele in adolescent rats lead to microstructure and ultrastructure lesions in the epididymis,which may be another important reason of infertility resulting from varicocele.\;[
