RESUMEN

As specialized intracellular parasite, viruses have no ability to metabolize independently, so they completely depend on the metabolic mechanism of host cells. Viruses use the energy and precursors provided by the metabolic network of the host cells to drive their replication, assembly and release. Namely, viruses hijack the host cells metabolism to achieve their own replication and proliferation. In addition, viruses can also affect host cell metabolism by the expression of auxiliary metabolic genes (AMGs), affecting carbon, nitrogen, phosphorus, and sulfur cycles, and participate in microbial-driven biogeochemical cycling. This review summarizes the effect of viral infection on the host's core metabolic pathway from four aspects: cellular glucose metabolism, glutamine metabolism, fatty acid metabolism, and viral AMGs on host metabolism. It may facilitate in-depth understanding of virus-host interactions, and provide a theoretical basis for the treatment of viral diseases through metabolic intervention.

RESUMEN

OBJECTIVE To investigate the inhibitory effect of natural compound XCQ-9 of Cynanchum paniculatum on the proliferation and apoptosis of Jurkat cell line of human T-cell acute lymphoblastic leukemia and its possible mechanism. METHODS Jurkat cell was used as the leukemia cell model, and MTT assay was adopted to detect the inhibitory effects of 0 (blank control), 2.5, 5, 10, 20 and 40 μmol/L XCQ-9 on the proliferation of Jurkat cell after treated for 24, 48, 72 h. After treated with 0 (blank control), 2.5, 5, 10 μmol/L XCQ-9 for 24 h and 48 h, the cell cycle and apoptosis were analyzed by flow cytometry. The expressions of Caspase-9, Cleaved Caspase-9, Caspase-3, Cleaved Caspase-3, poly ADP-ribose poly-merase (PARP), Cleaved-PARP, cyclin-dependent kinase 1 (CDK1) and Cyclin B1 were detected by Western blot after treated for 24 h. RESULTS Compared with blank control group, XCQ-9 at different concentrations could significantly decrease the survival rate of Jurkat cells (P<0.01), and showed a dose and time-dependent manner. After 48 h treatment of 5, 10 μmol/L XCQ-9, Jurkat cell apoptosis was induced significantly (P<0.05 or P<0.01), and the cell was arrested in G2 phase (P<0.01). After 24 h treatment of 10 μmol/L XCQ-9, the protein expressions of CDK1 and Caspase-9 were remarkably down-regulated (P<0.01), while the protein expressions of Cyclin B1, Cleaved Caspase-9, Cleaved Caspase-3 and Cleaved PARP were significantly up-regulated (P<0.05 or P<0.01). CONCLUSIONS XCQ-9 plays anti-tumor effect through inducing G2 phase arrest to inhibit proliferation and 5008) activating Caspase pathway to increase apoptosis.

Objective:To evaluate the additional value of chest thin layer CT over 99Tc m-hydrazinonicotinyl-(polyethylene glycol) 4-E[(polyethylene glycol) 4-c(RGDfK)] 2(HYNIC-PEG 4-E[PEG 4-c(RGDfK)] 2; 3PRGD 2) SPECT/CT in detecting isolated pulmonary space. Methods:This was a prospective study conducted in General Hospital of Ningxia Medical University. There were 87 patients with solitary pulmonary space occupying between July 2015 and December 2016, and 74 of those patients (49 males, 25 females, age range: 37-80 (58.4±9.6) years) who had pathological results were enrolled. 99Tc m-3PRGD 2 SPECT/CT imaging was performed routinely, and then the chest thin layer CT images were acquired. The maximum radioactive counts ratio of tumor to non-tumor (T/N)≥1.5 was the standard for positive planer 99Tc m-3PRGD 2 imaging, and that ≥2.0 was the standard for positive SPECT/CT imaging. According to the pathological results as gold standard, the diagnostic efficiencies of 99Tc m-3PRGD 2 planer and SPECT/CT imaging, chest thin layer CT and chest thin layer CT+ 99Tc m-3PRGD 2 SPECT/CT imaging for malignant pulmonary lesions were calculated. Kappa test was used to compare the consistency of the imaging methods and pathological results. Results:The post-surgery histopathology confirmed that 51 patients were with malignancy and 23 had benign lesions. The sensitivity, specificity, accuracy, positive predictive value and negative predictive value of 99Tc m-3PRGD 2 planer imaging, SPECT/CT imaging and chest thin layer CT in the diagnosis of malignant pulmonary lesions were 47.1%(24/51), 65.2%(15/23), 52.7%(39/74), 75.0%(24/32), 35.7%(15/42); 86.3%(44/51), 47.8%(11/23), 74.3%(55/74), 78.6%(44/56), 11/18 and 84.3%(43/51), 52.2%(12/23), 74.3%(55/74), 79.6%(43/54), 12/20, respectively. Those of the chest thin layer CT+ SPECT/CT were 98.0%(50/51), 73.9%(17/23), 90.5%(67/74), 89.3%(50/56) and 17/18 respectively. The Kappa values between the imaging methods ( 99Tc m-3PRGD 2 planer imaging, SPECT/CT imaging, chest thin layer CT and the chest thin layer CT+ SPECT/CT) and pathological examination were 0.100, 0.250, 0.354 and 0.765 (all P<0.001). Conclusion:Chest thin layer CT has an incremental value over 99Tc m-3PRGD 2 SPECT/CT imaging in the differential diagnosis of benign and malignant pulmonary lesions.

Surfactin has great potential applications in enhancing oil recovery, agriculture, pharmaceuticals, foods and beverages, and cosmetics due to its extraordinary surface activity, biodegradability, anti-bacterial activity and biocompatibility. Enhancing surfactin production by engineering surfactin-producer and optimizing culture conditions is the key of its industrial production and subsequent applications. In this study, the effect of fatty acid synthesis pathway on surfactin synthesis was investigated, and Bacillus subtilis THBS-2 and THBS-8 with high surfactin titer were constructed by overexpressing key genes involved in the fatty acid synthesis pathway. To optimize culture condition, the amount and adding time of isopropyl-beta-D-thiogalactopyranoside (IPTG) and amino acids were studied, and a two-stage culture method was obtained: IPTG (final concentration: 1.25 mmol/L) and leucine (final concentration: 5 g/L) were added at 3 h, leucine (final concentration 5 g/L) and condensed culture medium (5 mL) were added at 24 h. Applying this strategy, the surfactin titer of B. subtilis THBS-2 reached to 24 g/L in shake flask at 48 h and up to 34 g/L after 68 h fermentation in a 30-L fermentor. The results provide basis for large-scale production and broad application of surfactin.

Objective To analyze the image characteristics of whole-body bone planar scan and SPECT/CT imaging in patients with polyostotic bone fibrous dysplasia ( PFD) . Methods Twenty-three pa-tients with PFD (12 males, 11 females, age: 10-77(31.4±3.4) years) between June 2007 and March 2017 were enrolled. Twenty-one patients were confirmed pathologically and 2 was diagnosed by follow-up re-sults. The images of 99Tcm-methylene diphosphonate (MDP) whole-body bone scan and SPECT/CT imaging were retrospectively analyzed. Results Bone involvement in the extremities was the most common and lesions in the lower and right limbs were more than those in the upper and left limbs. Lesions were frequently unilat-erally on whole-body bone planar images in 18 of the other 23 patients ( 78. 3%) . Among them, 15/16 with limb lesions showed no bone deformation in limbs, while the enlargement and deformity were more common in the skull, ribs and pelvis. Vertebral involvement was found in 7 of 23 patients (30.4%), and the hand and foot bone involvement was found in 6 of 23 patients (26.1%). Most lesions (96.9%, 248/256) presented high or mod-erate abnormal uptake, which distributed in a stripe shape in the extremities, ribs and skull. On SPECT/CT ima-ges, the ground glass, vegetable sponge and mixed lesions showed higher uptake compared with the cystic le-sions. Conclusions The PFD has certain characteristics on whole-body bone scan. SPECT/CT imaging can reflect pathological, blood flow and metabolic changes of PFD.

Recent studies have shown that statins can inhibit the apoptosis of vascular endothelial cells. Many Pharmacological actions of statins have nothing to do with thier cholesterol lowering effects. These effects are called non lipid lowering cardiovascular protective effects. In this study, by establishing a high glucose induced vascular endorhelial cell apoptosis model, we explored the mechanism of the non lipid - related cardiovascular protective effect of atorvastatin. The results showed that atorvastatin could protect human umbilical vein endothelial cells from damage induced by new high glucose and inhibit apoptosis. High concentration atorvastatin can up regulate the expression of Bcl-2 and down regulate the expression of Baxprotein [P<0.05]. This suggests that statins may play a role in cardiovascular protection by inhibiting endothelial cell apoptosis. This results is confirmed by experiments, which can provide clues for clinical treatment, and combine drug therapy and lifestyle intervention to reduce blood sugar

Purpose To compare the diagnostic efficacy of 99Tcm-MIBI SPECT/CT tomography and SPECT imaging in evaluating neoadjuvant chemotherapy (NAC) in breast cancer,and to explore the clinical gain value of SPECT/CT tomography.Materials and Methods Forty-seven patients with biopsy pathologically confirmed breast cancer in General Hospital of Ningxia Medical University from 2010 to 2015 were retrospectively analyzed.99Tcm-MIBI SPECT/CT test was performed before NAC and 4-6 courses after.The patients were assigned into effective group and ineffective group based on the pathological response after NAC,the MIBI intake rate of tumors in the two groups was compared,and the efficiency of SPECT,CT and SPECT/CT in evaluating NAC were respectively calculated and compared.Results According to the postoperative pathological response,47 patients were divided into effective group (n=30) and ineffective group (n=17).In early phase,the T/N value of effective group and ineffective group was 1.46±0.58 and 1.10+0.48,respectively,with a statistically significant difference (t=2.185,P<0.05).There was no significant difference between SPECT,CT and SPECT/CT in evaluating NAC efficacy and pathological response (P>0.05).The sensitivity,specificity and accuracy of these three methods in evaluating NAC efficacy were respectively:80.0%,52.9% and 70.2% (SPECT);86.7%,70.6% and 80.9% (CT);92.3%,88.2% and 89.4% (SPECT/CT).There was a statistically significant difference between the specificity and accuracy of SPECT/CT tomography and SPECT imaging in evaluating NAC efficacy (x2=5.10 and 8.16,P<0.05).Conclusion The intake rate of tumors can predict the sensitivity of NAC.The diagnostic efficacy of 99Tcm-MIBl SPECT/CT tomography is more effective than that of SPECT imaging in evaluating the efficacy of NAC in breast cancer.

Objective To investigate th grade of CT in colonic diverticulitis and the value in predicting clinical treatment.Methods CT data of totally 41 patients with colonic diverticulitis were analyzed retrospectively.According to the imaging signs,the grading of diverticulitis cases was performed.Patients with level 1 to 3 were considered as only need non-operative treatment,those with level 4 or 5 were considered as need surgical treatment.The consistency of predictive treatment plans were contrasted with clinical practical treatment results.Results The CT features of colonic diverticulitis were saclike protrusions bulged outside of the bowel wall with colon wall thickening in 40 cases (40/41,97.56%) and fuzzy pericolonal fat space in 39 cases (39/41,97.12%).The diagnostic accuracy of preoperative CT grading in predictive treatment plan was 85.37 % (36/41).Conclusion Colonic diverticulitis have certain characteristic CT findings.CT grading of diverticulitis is helpful to formulate appropriate clinical treatment.

Objective To explore the imaging characteristics and diagnostic value of 18 F-FDG PET/CT in patients with hepatic epithelioid hemangioendothelioma ( HEH) . Methods From January 2014 to December 2015, 5 HEH patients (2 males, 3 females; average age (49.2±13.7) years) confirmed by pathology and underwent 18 F-FDG PET/CT imaging were enrolled in this retrospective study. The character-istics of PET/CT images were analyzed. Two-sample t test and Pearson linear correlation analysis were used for data analysis. Results A total of 26 lesions were detected in 4 patients with multiple lesions, of which 88.5%( 23/26) distributed at the liver edge, and 42. 3%( 11/26) had hepatic capsule retraction sign. SUVmax of the 26 lesions was 5.1±2.6 and it was not correlated with tumor size (r=0.054, P>0.05). Lesion FDG uptake in patients with distant metastases was higher than that in patients without distant metastases (6.08±1.80 vs 3.84±1.14;t=4.15, P<0.05). Two patients underwent the delayed imaging, and FDG up-take in 11/14 lesions was higher compared with that of early imaging, and 3/14 lesions were newly detected by the delayed imaging. One patient had diffuse low-density lesions with diameters between 6.1 and 35.5 mm and SUVmax between 2.6 and 8.1. Conclusion Liver lesions of HEH patients show some features on 18F-FDG PET/CT images which can find distant metastases, and the delayed imaging can improve the detection rate of lesions.

Objective To investigate the effect of continuous quality improvement on subjective well-being quality of life and satisfaction of esophageal cancer patients. Methods A total of 120 esophageal cancer patients in our hospital were randomly divided into two groups, 60 cases in the observation group treated with continuous quality improvement while the other 60 cases in the control group treated with routine care. The two groups were compared in terms of subjective well-being,quality of life and nursing satisfaction. Results Before the intervention, there were no significant differences between the two groups in view of the quality of life scale score and subjective well-being score (P>0.05). After the intervention, in view of symptoms, the scores on all the items in the boservation group were significantly lower than those in the control group (P<0.05), except constipation, economic difficulty and diarrhea and the function items were higher. The subjective satisfaction of the observation group was significantly better than that of the control group (P<0.05). Conclusion Continuous quality improvement care can promote the improvement of subjective well-being, improve the quality of life and satisfaction of patients with esophageal cancer.

By formulating the standard operating procedures for clinical skills, and standardizing the operation steps, the Affiliated Hospital of Inner Mongolia Medical University further refines and quantifies the scoring criteria, and this is also applied in the teaching practice of medical students' clinical skills. The two operations of clean wound dressing and diagnostic peritoneal puncture are taken as an example for description. The assessment results shows that the level of medical students' clinical skills has improved significantly. Questionnaire survey also shows that teachers' training efficiency and students' learning effi-ciency have been improved , indicating that clinical skills standard operation procedures have achieved good teaching results in clinical skills teaching practice.

Objective To establishthe sensitivity indexes system for estimating nursing quality which conforms to China's national conditions and have the certain representative among the whole country. Methods After usingStructure-process-outcome as the theoretical framework and surveying the literatures domestic and abroad,a total of 42 nursing management experts from 30 provinces were consulted by the Delphi method,including symposium and two rounds of questionnaire consultation. Results The authority coefficient of the two rounds consultation were 0.89 and 0.91,familiarity coefficient were 0.86 and 0.88 respectively,and the coordination coefficient of expert opinion were 0.20 and 0.25. The sensitivity indexes system for estimating nursing qualityconsists of two level index systems,which were divided into three first level indexes and seven second level indexes. The first level index in-cluded nursing manpower allocation,nursing safety and nursing events,while the second level index includes nurse-inpatient ratio,accuracy rate of identification,the number of medication error events,the number of blood transfusion error events,the number of specimen error events,the number of drug extravasation events and high-risk patient falls (falling bed) incidence. Conclusion The results of this study are reliable. There is a clear definition and interpreta-tion of sensitivity indexes for estimating nursing quality and index monitoring is operable.

RESUMEN

Primary biliary cholangitis (PBC) is an autoimmune liver disease mainly involving intrahepatic interlobular bile ducts and can progress to liver fibrosis, liver cirrhosis, and even liver failure. Ursodeoxycholic acid (UDCA) is the first-line therapeutic drug for PBC and can delay disease progression, but as high as 40% of patients have suboptimal response to UDCA. Obeticholic acid, a farnesoid X receptor agonist, has been approved by FDA in May 2016 for patients who have no response to UDCA treatment or cannot tolerate such treatment. Other drugs such as fibrates, glucocorticoids, immunosuppressants, biological agents, and mesenchymal stem cells are gradually used in clinical practice and bring new hope to patients with refractory PBC.

Objective To compare the composition of γδ T cells in the peripheral blood of gestational diabetes mellitus(GDM) patients with those of normal pregnant women,so as to explore the association of γδ T cells with the onset and progression of GDM.Methods The peripheral blood mononuclear cells(PBMC) of GDM patients(case group) and normal pregnant women(con-trol group) were separated by Ficoll-Hypaque density gradient centrifugation.γδ T cells were labeled with FITC conjugated anti TCRγδ monoclonal antibody(McAb) and PE conjugated anti-CD3 McAb and analyzed by flow cytometry.Results γδ T cells in GDM patients' peripheral blood were about(6.89 ± 3.22) %,which was higher than normal control group(4.26 ± 1.64) %.The percentage of γδ T cells in non-treated GDM group was significantly increased to(8.79 ± 2.33)%,while in treated GDM group,the percentage of γδ T cells was drop to normal(3.76±1.62)%.Conclusion γδ T cells in GDM patients' peripheral blood and CD3+ T Cells were slightly increased.The alteration may be related to the onset of GDM.

<p><b>OBJECTIVE</b>To establish objectively biomechanical criteria for temporomandibular joint disc tissue engineerings.</p><p><b>METHODS</b>Eight temporomandibular joint discs from a four-month-old goat were separated completely. A cylindrical sample with diameter of 3 mm in the anterior, intermediate, and posterior bands was obtained, and the samples underwent pre-compression test with three cycles under a speed of 0.025 mm · s⁻¹. With the end condition for the maximum load of less than 4.9 N and 1.5 mm compression displacement, the formal compression test was performed with the same speed. Moreover, this test obtained the stress-strain relationship and elastic modulus of each disc band. Ultrastructure and collagen fiber orientation of the district zone were observed by scanning electron microscope and polarized light microscope.</p><p><b>RESULTS</b>1) The physiological strain range of an articular disc was within 10%, and the quasilinear constitutive equation of articular disc tissues can be fitted with the polynomial function: y = ax + bx² + cx³. 2) The elastic modulus of the intermediate disc zone was the largest (P < 0.05). No significant difference existed between the anterior and posterior bands (P = 0.361). 3) Scanning electron microscope and polarized light microscope showed an annular disc collagen network structure, which was the internal part of the goat temporomandibular joint disc. The collagen arrangement of intermediate bands was tensely arranged anterior-posteriorly. The collagen of anterior and posterior bands went through mediolaterally with intersection of anterior-posterior branch fibersd.</p><p><b>CONCLUSION</b>Annular collagen network structure is the basis for goat temporomandibular joint disc compression resistance properties. The intermediate band demonstrated higher compression resistance performance than the anterior and posterior bands.</p>

Purpose To evaluate the role of 18F-FDG PET/CT tumor metabolic imaging in the detection of lung cancer lesions,using receiver operating characteristic (ROC) curves analysis and investigate the diagnostic value of TNR and TLR in lung lesions.Materials and Methods A total of 82 patients who were confirmed with lung space-occupying lesionsby 18F-FDG PET/CT tumor metabolic imaging were retrospectively analyzed.Based on the pathological gold standard,evaluate the diagnostic value of 18F-FDG PET/CT tumor metabolic imaging in lung space-occupying lesions by t-test and ROC curve.Results There was a significant difference of TNR and TLR between benign and malignant lesion (P<0.01).The AUC on ROC curve of TLR ratio was 0.943 in the lung lesions while the TNR was 0.927.The sensitivity,specificity and the accuracy were 86.36%,62.50% and 81.71%,respectively with the TNR cutoff of 6.87.The sensitivity,specificity and the accuracy were 90.91%,68.75% and 86.58%,respectively with the TLR cutoff of 2.86.Conclusion 18F-FDG PET/CT tumor metabolic imaging is useful for differentiating lung space-occupying lesions.But for cases of atypical 18F-FDG uptake,it is suggested to combine with clinical information.

Objective To summarize the SPECT/CT manifestation of spondylitis caused by Brucells infection and to evaluate the diagnostic value.Methods From June 2012 to October 2015,a total of 28 patients (14 males,14 females,average age 46.4 years) with Brucellosis spondylitis confirmed by laboratory test and pathology were included.The images of whole-body bone scan and SPECT/CT fusion imaging were retrospectively analyzed.According to the pathological and serologic test results,the diagnostic efficacy of imaging was calculated.x2 test was used.Results Most of the Brucellosis spondylitis happened in the lumbar(76.7%,43/56),and the most common locations were L3,L4,L5 (72.1%,31/43).Two or more involved consecutive vertebra were found in 71.4% (20/28) of the patients.Moderate radioactive distribution was showed in 89.2% (50/56) of lesions,high radioactive distribution was showed in 5.4% (3/56) of lesions,and mild radioactive distribution was showed in the rest 3 lesions.Thirty-three lesions(58.9%,33/56) had diffuse increased radioactivity uptake in the affected vertebra,and 32.1%(18/56) showed diffuse increased radioactivity at the superior and inferior margin of the vertebra;only 8.9% (5/56) of lesions were on one side of the vertebral bodies.The SPECT/CT results were as follows:(1) Bone destruction was showed in 80.4% (45/56) of lesions,and the edge of the lesion was clear.(2) For 66.7% (30/45) of lesions,bone hyperplasia was seen along with bone destruction and moderate radioactivity concentration on the edge of destruction area.(3) The damage of the intervertebral disc was mild,and the vertebral abscess was relatively rare (5.4%,3/56).The diagnostic accuracy of SPECT/CT was statistically higher than that of whole-body bone scan:67.8%(38/56) vs 96.2%(54/56);x2=13.1,P<0.05.Conclusion SPECT/CT imaging has a higher diagnostic efficiency than whole-body bone scan in Brucellosis spondylitis.

RESUMEN

Objective:To explore the clinical appearance,diagnosis method and curative efficacy of one patient with esophageal teratoma complicated with rupture of esophagus in order to raise the awareness of the clinicians for the disease. Methods:The patient with chest pain after eating was admitted to hospital,and upper gastrointestinal radiography,chest CT and endoscopy were taken,which showed rupture of esophagus.Then an emergency thoracic exploration was performed.A too long cleft and much necrosis were identified,so the repair of esophagus was changed into excision and anastomosis. Results: The operation was successful,and the pathological findings showed the rupture of esophageal teratoma.Follow-up showed no complication till now.Conclusion:Esophageal teratoma complicated with esophageal rupture is rare.Clinicians should improve the understanding and reduce the misdiagnosis rate.

RESUMEN

Objective To investigate the functions of microRNA-1 43 (miR-1 43)in esophageal cancer cell line ECA1 09.Methods ECA1 09 cells were transfected with negative control (NC),miR-1 43 mimics or miR-1 43 inhibitors.3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide (MTT)assay was per-formed to evaluate the growth of ECA1 09 cells after transfection.Annexin V-FITC /PI apoptosis test kit was used to detect early apoptosis rate in ECA1 09 cells.Transwell migration and invasion assays were conducted to compare the migration and invasion capacity of ECA1 09 among different groups.Real-time PCR and Western blotting were used to analyze the mRNA and protein alteration after transfection.Results Three and four days after transfection,compared with NC (absorbance value:0.90 ±0.02 and 1 .09 ±0.07),miR-1 43 mimics inhibited ECA1 09 cell proliferation (absorbance value:0.66 ±0.05 and 0.80 ±0.04),while miR-1 43 inhibi-tors promoted cell proliferation (absorbance value:1 .1 3 ±0.09 and 1 .51 ±0.08),with statistical signifi-cances (F =49.1 6,P =0.000;F =1 00.34,P =0.000).Early-stage apoptosis rates of ECA1 09 transfected with NC,miR-1 43 mimics and miR-1 43 inhibitors were 3.42% ±0.72%,1 1 .63% ±1 .1 5% and 0.94% ± 0.1 0%,respectively,with statistical significance (F =1 51 .61 ,P =0.000).Meanwhile,compared with NC (migration cell number:336 ±1 3,invasion cell number:1 47 ±1 6),miR-1 43 mimics inhibited cell migration (1 48 ±1 6)and invasion (75 ±1 0),while miR-1 43 inhibitors promoted cell migration (51 0 ±1 4)and inva-sion (238 ±1 6),with statistical significances (F =470.99,P =0.000;F =90.04,P =0.000).Compared with NC (1 .00 ±0.00),miR-1 43 mimics down-regulated mRNA (relative expression level 0.22 ±0.08)and protein expression (relative expression level 0.46 ±0.08)of K-ras,whereas miR-1 43 inhibitors up-regulated mRNA (1 .55 ±0.1 2)and protein expression (1 .33 ±0.05)of K-ras (F =1 31 .36,P =0.000;F =88.1 7, P =0.000).Conclusion miR-1 43 functions as a tumor suppressor in esophageal cancer cell line ECA1 09, probably by down-regulating K-ras expression.
