RESUMEN

【Objective】 To explore the efficiency of hierarchical teaching model in advanced training of therapeutic blood component apheresis. 【Methods】 A total of 76 students who participated in the advanced training(≥3 months) of therapeutic blood component apheresis in Blood Transfusion Department of our hospital from January 2016 to December 2021 were taken as subjects. They were divided into observation group(hierarchical teaching mode, n=46) and control group (traditional teaching mode, n=30) using random number table method. The assessment scores of the two groups in terms of theoretical knowledge and experimental operation ability before and after the advanced traning were compared, and the satisfaction of students for the teaching models were collected by questionnaires. 【Results】 After training, the average score of the observation group and control group in terms of theoretical knowledge and experimental operation ability were 52.57±2.17 vs 51.00±2.73, 34.74±1.99 vs 33.40±2.42, respectively (P<0.05), and the training satisfaction was 95.35% (41/43) vs 78.57% (22/28) (P<0.05). 【Conclusion】 Compared to traditional teaching mode, hierarchical teaching mode in advanced training of therapeutic blood component apheresis has better effect on the trainees to master relevant theoretical knowledge and operational skills.

RESUMEN

Autologous ozonized blood transfusion(AOBT) is a therapy of re-transfusion of 100-200 mL of autologous blood after shaking and agitation with appropriate amount of oxygen-ozone in vitro. The oxidation of blood through the strong oxidation of ozone can enhance the non-specific immune response of the body, regulate the internal environment and promote health. This therapy has been increasingly applied in clinical practice, while no unified standard for the operation process in terms of ozone concentration, treatment frequency and treatment course had been established. This operation process of AOBT is primarily explored in order to standardize the operation process and ensure its safety and efficacy.

Dopamine D3 receptor (D3R) is implicated in multiple psychotic symptoms. Increasing the D3R selectivity over dopamine D2 receptor (D2R) would facilitate the antipsychotic treatments. Herein, novel carbazole and tetrahydro-carboline derivatives were reported as D3R selective ligands. Through a structure-based virtual screen, ZLG-25 (D3R Ki = 685 nmol/L; D2R Ki > 10,000 nmol/L) was identified as a novel D3R selective bitopic ligand with a carbazole scaffold. Scaffolds hopping led to the discovery of novel D3R-selective analogs with tetrahydro-β-carboline or tetrahydro-γ-carboline core. Further functional studies showed that most derivatives acted as hD3R-selective antagonists. Several lead compounds could dose-dependently inhibit the MK-801-induced hyperactivity. Additional investigation revealed that 23j and 36b could decrease the apomorphine-induced climbing without cataleptic reaction. Furthermore, 36b demonstrated unusual antidepressant-like activity in the forced swimming tests and the tail suspension tests, and alleviated the MK-801-induced disruption of novel object recognition in mice. Additionally, preliminary studies confirmed the favorable PK/PD profiles, no weight gain and limited serum prolactin levels in mice. These results revealed that 36b provided potential opportunities to new antipsychotic drugs with the multiple antipsychotic-like properties.

Objective To evaluate the relationship between perioperative peripheral blood Type 17 helper (Th17) cells and Th17-related cytokines and postoperative cognitive dysfunction (POCD) in elderly patients undergoing general anesthesia.Methods Ninety-six patients of both sexes,aged 65-86 yr,of American Society of Anesthesiologists physical status Ⅱ or m,scheduled for elective hip replacement under general anesthesia,were selected.At 3 days before operation and 1,2,3 and 7 days after operation,Montreal Cognitive Assessment (MoCA) was used to evaluate the cognitive function,and fasting venous blood samples were taken for determination of the percentage of Th17 cells and serum interleukin-17 (IL-17) and IL-22 concentrations.The patients were divided into POCD group and non-POCD group according to whether the patients developed POCD at day 7 after operation or not.Pearson linear correlation of the percentage of Th17 cells and serum IL-17 and IL-22 concentrations with MoCA scores was analyzed.Results Twenty-six patients developed POCD (27.1%).The percentage of peripheral blood Th17 cells and serum IL-17 and IL-22 concentrations were significantly higher at each time point after operation than before operation in POCD group and at 1 and 2 days after operation than before operation in non-POCD group (P<0.05).The percentage of peripheral blood Th17 cells and serum IL-17 and IL-22 concentrations were significantly higher at each time point after operation in POCD group than in non-POCD group (P<0.05).The percentage of peripheral blood Thl7 cells was negatively correlated with MoCA scores (r =-0.867,P<0.01) and serum IL-17 and IL-22 concentrations were negatively correlated with MoCA scores (r=-0.662 and-0.638,P<0.01) in group POCD.Conclusion The development of POCD is related to the increase in the percentage of peripheral blood Th17 cells and concentrations of Th17-related cytokines IL-17 and IL-22 in elderly patients undergoing general anesthesia.

RESUMEN

We developed phase-switch microfluidic devices for molecular profiling of a large number of single cells. Whole genome microarrays and RNA-sequencing are commonly used to determine the expression levels of genes in cell lysates (a physical mix of millions of cells) for inferring gene functions. However, cellular heterogeneity becomes an inherent noise in the measurement of gene expression. The unique molecular characteristics of individual cells, as well as the temporal and quantitative information of gene expression in cells, are lost when averaged among all cells in cell lysates. Our single-cell technology overcomes this limitation and enables us to obtain a large number of single-cell transcriptomes from a population of cells. A collection of single-cell molecular profiles allows us to study carcinogenesis from an evolutionary perspective by treating cancer as a diverse population of cells with abnormal molecular characteristics. Because a cancer cell population contains cells at various stages of development toward drug resistance, clustering similar single-cell molecular profiles could reveal how drug-resistant sub-clones evolve during cancer treatment. Here, we discuss how single-cell transcriptome analysis technology could enable the study of carcinogenesis from an evolutionary perspective and the development of drug-resistance in leukemia. The single-cell transcriptome analysis reported here could have a direct and significant impact on current cancer treatments and future personalized cancer therapies.

RESUMEN

The present paper introduced the management status of wild medicinal resources (WMR) including the law system, the government system and the rule system, and analyzed the main problems and their reasons for WMR management. It pointed out that the old management system for WMR was not fit the need of conversation and management of WMR. It suggested to revise the "the Law of Conversation and Manage System of Wild Medicinal Resources", and discussed the law type, the objective and the principle he government system, management field and rule system for the revised law in details.

RESUMEN

Objective To observe the clinical effect of blood transfusion on earthquake victims from Wenchuan earthquake area, Sichuan province.Methods The blood routine, electrolyte and coagulation function of earthquake victims were detected.Results A total of 41(27.5%) victims received blood transfusion. The average volume of blood transfusion was 1 233 ml, and the average volume of plasma was 1 550 ml. No patient was found to have adverse transfusion reaction. There was no significant difference between the electrolytes before and after the transfusion.Conclusion Blood transfusion is important in the care of earthquake victims.

One major problem to successful treatment of cancer is the development of resistance by tumor cells to multiple chemotherapeutic drugs, a phenomenon named multidrug resistance (MDR). Searching for the novel chemotherapeutical agents is one of the important strategies for overcoming MDR. By using a cytotoxicity assay, flow cytometry analysis, Western-blotting and RT-PCR, a drug (Taxol, TAX) resistant human nasopharyngeal carcinoma KB cell line (KB/TAX) was established by addition of the drug to the cell cultures gradually, then a novel N-sugar substituted thalidomide analogue (STA-35) was investigated for its reversal effect on MDR of KB/TAX cells and possible mechanism. The results showed that KB/TAX cells were resistant to several chemotherapeutical agents, and the relative resistance to TAX was 73.1. Compared with parental KB cells, the function and protein expression of P-glycoprotein (P-gp), as well as mdr1 gene in the KB/TAX cells were remarkable reduced. Moreover, both KB and KB/TAX cells were sensitive to STA-35, the relative resistance to TAX on KB/TAX cells was decreased by the addition of STA-35. Furthermore, STA-35 (5 ~20 ?mol/L)was capable to reduced the activity of P-gp by increasing the accumulation of rhodamine 123, decreasing P-gp expression in KB/TAX cells in a dose dependent manner , but had no effect on the mdr1 gene expression. These results suggest a potential action of STA-35 as MDR reversing agent, and one of the possible mechanisms could be the suppression of P-gp function and protein expression.

Objective To screen and analyze the differentially expressed genes of Jurkat cells cocultured with bone marrow stromal cells(BMSCs) from leukemia patients.Methods Suppression subtractive hybridization(SSH) was employed to establish subtracted cDNA library of differentially expressed genes in Jurkat cells cocultured with BMSCs from leukemia patients in vitro.The cDNA fragments were sequenced and analyzed.Results The differentially expressed gene cDNA library of Jurkat cells cocultured with BMSCs from leukemia patients in vitro was developed by SSH.Primary screening was done by reverse northern hybridization.Thirty up-regulated and 22 down-regulated cDNA fragments were isolated and sequenced,and sequence homology was performed by BLAST in GenBank.These genes were related to cell cycle regulation,cellular apoptosis and energy metabolism.The information of differentially expressed genes in Jurkat cells implied that their secretion function,cell cycle and apoptosis were affected by BMSCs from leukemia patients.Conclusion BMSCs from leukemia patients might influence gene expression of Jurkat cells.The differentially expressed genes might be significantly associated with the protection of BMSCs from leukemia patients on Jurkat cells during Daunorubicin exposure.

Objective To observe the effects of all-tram retinoic acid(ATRA)on intercellular communication function and connexin 43 expression in acute leukemia bone marrow stromal cells(ALBMSCs).Methods ALBMSCs were cultured in vitro,and the ATRA(1?10~(-5)mol/L)was added after serial subcultivation.The expression level of Cx43 was detected by immunochemistry and flow cytometry, and the gap junction intercellular communication(GJIC)was examined by using cell scarification with dye transfer(CSDT)technique.Re- sults Before and after addition of ATRA,the positive rates of Cx43 expression as detected by immunochemistry in ALBMSCs were(47.2?2.04)% and(54.5?5.86)%,respectively.The positive rates of Cx43 expression in ALBMSCs as detected by flow cytometry before and after the addition of ATRA were(38.75?23.95)% and(49.5?5.46)%,respectively.The expression of Cx43 in ALBMSCs after adding ATRA was significantly higher than that before the addition of ATRA(P

Objective To investigate the expression level of CRIF1 in human leukemic Jurkat T-cell line cocultured with primary leukemic bone marrow stromal cells.Methods RT-PCR was used to study the expression level of CRIF1 in human leukemic Jurkat T-cell line cocultured with primary leukemic bone marrow stromal cells in vitro. Results After coculture with leukemic bone marrow stromal cells, the expressions of CRIF1 and Gadd45 mRNA in Jurkat cells were significantly higher than the control.Conclusion High mRNA levels of CRIF1 might be associated with the G0/G1 phage arrest in human leukemic Jurkat T-cell line cocultured with primary leukemic bone marrow stromal cells.
