RESUMEN

This study assessed and compared different methods for vegetative propagation of a miniature ornamental pineapple hybrid (ORN-MUT), seeking to determine the best method for production of plantlets, as well as for removal of the PMWaV viral complex from plants cultured in vitro, for production of healthy parent plants. Pineapple wilt is a disease that can cause large economic and is caused by a viral complex called Pineapple mealybug wilt-associated virus (PMWaV). For this, four propagation methods were evaluated (conventional, stem sectioning, micropropagation and etiolation of nodal segments). The time necessary for each method and the number of plants formed were assessed. Stem tips (0.5 mm) were cultured and indexed for three PMWaV types. Conventional propagation produced 17 plantlets per plant in 566 days, stem sectioning produced 2.3 plantlets per stem in 591 days, while the conventional micropropagation technique produced 1,284 plants after four subcultures in 778 days. Stems etiolated for 60 days showed peak production in the second subculture, with 1,224 plants. This method required 883 days to obtain plants with ideal size for transplantation to the field. In turn, stems etiolated for 120 days produced 935 plants at the end of four subcultures, with peak output in the third subculture, in which the plants could be cultivated in the field after 943 days. Conventional micropropagation and etiolation for 60 days were the best methods for production of plantlets of the ORN-MUT hybrid. The results of this work showed that the cultivation of shoot tips is an efficient strategy to remove the PMWaV complex and obtain healthy mother plants and can be a useful tool for other varieties of pineapple.

RESUMEN

We aimed to evaluate the effects of different irrigation depths with saline water on growth, yield, water-use efficiency, and fruit quality of 'Pérola' pineapple plant grown in the semi-arid of Bahia state. The experiment was carried out in a randomized block design with five treatments which represented the irrigation depths: 100% of crop evapotranspiration (ETc) with irrigation water of 0.75 dS m-1 in electrical conductivity (ECiw); and 50, 75,100, and 125% of ETc with water of 3.6 dS m-1 in ECiw. Pineapples were grown under field conditions watered by drip irrigation in which pressure compensating emitters had 8 L h-1 flow rate. We observed that the irrigation depth 100% of ETc with water of 0.75 and 3.6 dS m-1 in ECiwprovides higher pineapple yields under the semi-arid conditions of this study, and the chemical quality of the fruits are up to commercial standards, except when applying 125% of ETc with water of 3.6 dS m-1 in ECiw.

Pineapple peels has several beneficial properties including antioxidant activity. We investigated the antioxidant effect of five different peels of pineapple lyophilized extracts, not adsorbed and adsorbed onto Amberlite. They were examined using total phenolic contents (TPC), antioxidant effect by 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical scavenging and ferric reducing antioxidant power (FRAP). In addition, we analyzed the chemical composition by HPLC-ESI-QTOF-MS/MS. The main constituents of pineapple peels were tentatively identified as quercetin glycosides and N,N'-diferuloylspermidine. We conclude that the antioxidant activity in pineapple peels from District of Poroto, Province of Trujillo, Region of La Libertad, can be associated with the presence of flavonoid and spermidines.

Ananas comosus, popularly known as pineapple, has well defined plant anatomy and numerous pharmacologically active phytochemicals. Some of these are responsible for the species antimicrobial potential, which has been widely studied given the bacterial resistance to the currently used antibiotics. Considering this, it is defined that the main characteristic of a drug is the absence of toxic effects, thus, there is a need to seek data regarding the toxicity of A. comosus, collaborating for its possible use as a drug. Comparing hydroalcoholic extracts from the crown leaves, skin and pulp of the infructescence, it was possible to determine that the three tested extracts are non-toxic, being the one with the lowest toxicity to Artemia salina Leach (leaves extract with LC50 equal to 994 μg/mL) and to lamb's blood (pulp extract with percentage of haemolysis equal to 0.83%). Studies concerning principles and methodologies similar to those used here have found comparable results stating the low toxicity of the plant. These results contribute to the research that promotes the use of pineapple as a phytotherapeutic agent and reaffirms its presence in the Brazilian Unified Health System.

RESUMEN

Plant fiber is a renewable and biodegradable material that can be used effectively to reinforce various composites. Pineapple hybrids selected for their fiber quality are in the phase of agronomic validation in Brazil by the Embrapa Cassava and Fruits research unit. The selection of a hybrid for large-scale fiber production depends on obtaining a large number of seedlings. This study evaluated the morphogenetic response and propagation potential of eight hybrids of Ananas comosus var. erectifolius, for the purpose of producing high-quality seedlings on a large scale. Stem and crown buds were reduced and placed in MS nutritive medium supplemented with BAP at 0.5 mg L-1, NAA at 0.01 mg L-1 and Phytagel® at 2.5 g L-1. After 45 days, the number of oxidized, contaminated and surviving buds was determined. Swollen buds and plantlets were transferred to a multiplication medium containing MS sucrose, salts and vitamins. The propagation potential was evaluated based on the geometric growth rate among sub-cultures. The FIB-NEG hybrid presented the best results for the establishment phase (40.28%). The best propagative potential was obtained from crown buds with the highest values for FIB-EST (3.93), FIB-MIN (3.91) and FIB-BOY (3.91) hybrids.

ABSTRACT Pollen morphology characterization is important in taxonomy, conservation and plant breeding, and pollen viability studies can support breeding programs. This study investigated pollen morphology and male fertility in 18 species of Bromeliaceae with ornamental potential. For morphological characterization, pollen grains were acetolyzed and characterization of exine was done using scanning and transmission electron microscopy. Pollen viability was investigated by in vitro germination and histochemical tests. Species belonging to Aechmea and Ananas genera presented medium size pollen, except for Ae. fasciata, with large pollen. Al. nahoumii, P. sagenarius and the Vriesea species analyzed showed large pollen, except for V. carinata, with very large pollen. Pollen of Aechmea, Ananas and P. sagenarius presented bilateral symmetry, diporate, exine varying from tectate to semitectate. Al. nahoumii and Vriesea species presented pollen with bilateral symmetry, monocolpate; exine was semitectate, reticulate and heterobrochate. Germination percentage and tube growth were greater in SM and BKM media. Histochemical tests showed pollen viability above 70% for all species, except for Ananas sp. (40%). Pollen morphology is important for the identification of species, especially in this family, which contains a large number of species. High rates of viability favor fertilization and seed production, essential for efficient hybrid production and conservation.

RESUMEN

ABSTRACT: This study examines the effect of inoculation with diazotrophic bacteria and nitrogen fertilization on the growth of micropropagated pineapple cv. 'Vitória' plantlets during the acclimatization period. The experiment was carried out in a greenhouse in Campos dos Goytacazes, in randomized blocks, using a 2x5x5 factorial scheme, with the factors being two types of inocula (absence or presence of a mixture of diazotrophic bacteria that contained Burkholderia sp. UENF 114111, Burkholderia silvatlantica UENF 11711, and Herbaspirillum seropedicae HRC 54), five levels of urea (0, 2, 5, 7, and 10g L-1), and five evaluation periods (30, 60, 90, 120, and 150 days following the planting of plantlets in trays), and with four replications of each treatment. An increase in the dried weight of shoots and roots of micropropagated pineapple cv. 'Vitória' plants depended on inoculation with diazotrophic bacteria, and plantlets acclimatization could be reduced by up to 57 days, depending on the concentration of urea fertilizer.

The aims of this study were to evaluate the effects of shade in pineapple plant with cassava, on the yields of pineapple fruit sand cassava roots, on the morph-anatomical variation, damage by solar radiation and chlorophyll content of pineapple leaves. Pineapple plants were cultivated in plots comprising three parallel row sunder different shade conditions provided by cassava plants grown at various spacings within single rows located on either side of the plots. The experiment was of a randomized block design with five treatments (cassava spacings of 0.50, 0.75, 1.00 and 1.25 m and a pineapple monoculture) and four replications. Yields of fruits and roots, together with the morphological, chemical and histological characteristics of the "D" leaves of pineapple, were determined14 months after planting the pineapples plant. Data were submitted to analysis of variance with the Scott-Knott test or Friedman test (P< 0.05) and to regression analysis. Pineapple plants grown under the majority of shade conditions presented higher fruit weight and overall yield per hectare in comparison with plants grown in direct sunlight. The maximum productivities of pineapple fruit were achieved when cassava plants were spaced 0.75 m apart and, under these shade conditions, fruits were fully protected against burning by the sun. The "D" leaves of shaded plants were longer and thicker, with higher content of chlorophyll a and total chlorophyll, reduced stomatal density and dimensions of stomatal pores, narrower guard cells, thinner aquiferous hypodermis, and reduced abaxial and adaxial epidermis. The yield of cassava roots per plant increased linearly with increasing distance between the plants, but the yield per hectare decreased with decreasing plant density.

This research describes the drying kinetics and compares the convective drying rates of in natura and osmodehydrated pineapple slices in inverted sugar. The effective moisture diffusivity during air drying was estimated using Fick's second law of diffusion. The suitability of a theoretical liquid-diffusion model and seven semi-theoretical mathematical models for use in describing the experimental drying curves was also evaluated. Goodness of fit between experimental and predicted values was based on the root mean square error, mean absolute percentage error, mean bias error, agreement index, residual plot analysis and the principle of parsimony. Osmotic dehydration was conducted in 155, 310, and 465 mL L-1 osmotic solutions, at 40 and 50 ºC for 2 h at 60 rpm. Convective drying was performed in a tray cabinet dryer using heated ambient air at 60 ºC and 1.15 m s-1. Osmotic pretreatment facilitated water removal during the first hours of drying, a trend that was reversed towards the end of the process for samples osmodehydrated at the highest solution concentration. The effect of the osmotic pretreatments on drying rate was negligible at 40 C, but at 50 C the rate of moisture removal was more intense for samples in natura and osmodehydrated at the lowest solution concentration. Effective moisture diffusivity increased with temperature and solution concentration. The single-exponential, three-parameter semi-theoretical drying model gave the best predictions of the drying curves of pineapple slices both in natura and pre-osmodehydrated in inverted sugar.

Objective: To investigate the chemical constituents from the leaves of Ananas comosus and their biological activities. Methods: The chemical constituents from the air-dried leaves of A. comosus were isolated and purified by the chromatography on silica gel and Sephadex LH-20 columns as well as recrystallization. Their structures were identified on the basis of physiochemical properties and spectroscopic data analyses, and the antibacterial activity and artemia lethal activity of the compounds were determined. Results: Eight compounds were isolated from the 95% ethanol extract in the leaves of A. comosus and identified as tricin-4′-O-[10″-O-(8″-hydroxyl) feruloyl-(9‴-O-p-coumaroyl) glyceryl] ether (1), 2, 4-dichlorobenzoic acid (2), tricin (3), chrysoeriol (4), 1-O-p-coumaroylglycerol (5), 1-O-feruloylglycerol (6), 1-O-feruloyl-3-O-p-coumaroyl-glycerol (7), and 1, 3-O-diferuloylglycerol (8). Compound 1 exhibited as well inhibitory activities as positive control Ciprofloxacin (CPFX) against Staphylococcus aureus and Escherichia coli, with MIC values of 0.156 μg/mL. The results of artemia lethal activity showed that the IC50 of the compounds 1 and 4 were 21.4 and 25.0 μg/mL, respectively. Conclusion: All the compounds except 5 are isolated from this plant for the first time. Among them, compound 1 is a new one and named ananasin A, which has the good antibacterial and artemia lethal activity.

RESUMEN

This study was aimed to observe the effect ofpomegranate leaf tennis and Ananas coumosus leaf phenols on the growth of lactating mice through breast milk. Intragastric administration of different doses of drug was given to maternal mice. The general condition of newborn mice, body weight, tail length and organ coefficient of liver-related factors regulating the expression of lipid metabolism were observed. The results showed that both pomegranate leaf tennis and A. coumosus leaf phenols can obviously affect the growth and development of newborn mice through breast milk. And the effect of pomegranate leaf tennis was stronger than A. coumosus leaf phenols. Meanwhile, it obviously downregulated the expression of liver-related lipid metabolism enzymes in newborn mice. It was concluded that pomegranate leaf tennis and A. coumosus leaf phenols can affect the growth and development of newborn mice through breast milk. Its effect was related to the influence of lipid metabolism enzyme in the liver. Attention should be paid on taking this medication during lactation. Its clinical significance still required in-depth study.

The natural light application on natural ventilation micropropagation systems makes a bigger irradiance and gaseous exchanges on in vitro culture, occasioning foliar anatomy alterations and possibiliting the culture more close of phothoautrofic conditions. The present study had the purpose to evaluate growth in vitro and foliar anatomy of pineapple plants (Ananas comosus L.) incubated in natural or artificial light, using two micropropagation systems (conventional and natural ventilation) with different sucrose concentrations. The treatments were composed of different sucrose concentrations (0; 15; 30 e 45 g. L-1) combined with different micropropagation systems (conventional and natural ventilation), incubated under natural light. Two additional treatments were made with conventional and natural ventilation micropropagation system incubated on growth room with control of irradiance and temperature. The biggest in vitro growth were observed on natural ventilation under natural light with 30 g. L-1 of sucrose on culture media, as well as the foliar anatomy alterations that made bigger chlorophyll parenquimata during in vitro culture. Natural ventilation micropropagation system in combination with natural light produces the greatest biomass production and the better foliar anatomy.

The world market for flowers is showing great economic prominence in recent years. Despite the establishment of micropropagation techniques, improvements are still needed in the protocol. In this sense, the aim of the present study was to evaluate the in vitro multiplication of species of ornamental pineapple Ananas comosus var. bracteatus and A. comosus var. erectifolius at different evaluation periods and concentrations of BAP. Were used as explants axillary buds of ornamental pineapple. We used a completely randomized design, consisting of 16 treatments with 5 replicates and 2x4x2 factorial design, with one explant per tube. For leaf number, variety erectifolius was more efficient than bracteatus; evaluation performed 60 days, the number of leaves was two times higher than at 30 days at the concentration of 1 mg l-1 BAP. The variety erectifolius presented leaves longer in the concentration of 1 mg L-1 BAP. The length of the leaves decreased with increasing concentration of BAP. The number of roots produced was not influenced by the variety, or the evaluation period. The variety erectifolius showed greater root length main bracteatus that in the absence of BAP at 60 days. The concentration of 3 mg L-1 BAP was more effective in the formation of shoots. The variety bracteatus at 60 days showed the highest shoot formation in ornamental pineapple.

RESUMEN

Introduction: there are natural products from different fruits and plants that are effective as spermicides, but it is important that they should have little or no cytotoxic effect on epithelial cells. Currently available spermicides with nonoxynol-9 cause vaginal irritation and damage to the vaginal mucosa, the uterine epithelium, and the microbial flora of the vagina. Objective: to elucidate the effect on cell viability, cytotoxicity and apoptosis of spermicidal extracts of Ananas comosus (L.) Merr. and Sapindus saponaria L. over HeLa cell line. Methods: both extracts were evaluated on HeLa cell line using the novel ApoTox-Glo™ Triplex Assay to determine whether cell viability, cytotoxicity and apoptosis were affected. Results: it was determined that treatment with Sapindus saponaria and Ananas comosus extracts initially affected cell viability, but the latter tended to be restored. There was a sign of cell apoptosis that also tended to decrease over time. Conclusions: extracts of Sapindus saponaria and Ananas comosus may affect the survival of cells at the beginning, but these can continue replicating over time. There was a sign of cell apoptosis that also tended to decrease over time. Something similar happened to cell cytotoxicity, indicating that although the extracts may affect the survival of cells at the beginning (6 hours of treatment), these can continue dividing over time.
