RESUMEN

Allergic asthma is a complex, polygenic disease characterized by chronic inflammation and airway hyperresponsiveness. Fungus and dust mites are the most important inhaled allergens of allergic asthma, and often exist in the form of mixed allergens. In recent years, genetic studies have shown that several genes are associated with allergic asthma attacks. This article reviews the studies on the genes related to allergic asthma caused by dust mites and fungus, such as a disintegrin and metalloproteinase 33(ADAM33),interleukin-4(IL-4),glycoprotein A repetitions predominant(GARP),toll like receptor 3(TLR3),mannose-binding lectin 2(MBL2),chemokine(C-C motif)ligand 17(CCL17)and other genes .

RESUMEN

Abstract Southern corn leaf blight (SCLB) is one of the most important corn leaf diseases. Appropriate management strategies and the use of resistant cultivars allow disease control. Therefore, knowing the aspects related to the pathogen and the response of hosts makes it possible to design efficient strategies for selecting genotypes resistant to this disease. In this sense, the objective was to carry out the Bipolaris maydis isolate characterization, evaluating the pathogenicity in different popcorn lines and the symptoms generated in the host after inoculation. The isolate characterization consisted of the macromorphological evaluation of the colonies and the micromorphological evaluation of the conidia in the PDA medium. An experiment was carried out in a greenhouse to evaluate the pathogenicity of the isolate, using 20 inbred lines of popcorn in a randomized block design with four replicates. Inoculation was carried out by spraying leaves, with a suspension containing 1.0 x 104 conidia.ml-1 of the CF/UENF 501 isolate of B. maydis. An incidence assessment and three assessments of disease symptom severity were performed, with seven days intervals between evaluations. The morphological characterization data of the isolate were analyzed using descriptive statistics, and for disease severity, the linear regression model was applied the first-degree model. The variance analysis was performed for the linear and angular coefficients obtained for each treatment. When a difference was found, the Scott-Knott clustering algorithm at 5% significance was applied. The isolate had gray-green colonies, a cottony appearance, and an irregular shape. The lines L353, L354, and L624 show more resistance at the beginning and throughout the evaluations. The high virulence of the CF/UENF 501 isolate made it possible to differentiate the lines in terms of disease intensity and the pattern of symptoms presented.

Abstract Introduction Coronavirus disease 2019 (COVID-19) caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has cast a gloom spell on healthcare worldwide, infecting millions of people. Objective The aim of the present study is to determine the prevalence and review the contributing comorbidities and the precipitating factors leading to the emergence of the fungal infections in COVID-19-affected patients. To assess the utility of different laboratory techniques for confirmation of fungal infections. To assess the strengths and limitations of the diagnostic methods. Methods We have studied 252 clinical samples obtained from 121 COVID-positive patients. Results Among the 121 patients clinically diagnosed with fungal infections, 88 had diabetes and were given steroids for treatment (p-value = 0.001). Ninety-five patients (78.5%) had a positive laboratory diagnosis (either culture positive, potassium hydroxide [KOH]-positive or positive histopathology report). Fungal culture was positive in 75 (61.9%) patients and histopathology report was positive in 62 (51.2%). Histopathology was positive in 7 (5.8%) patients in whom culture and KOH were negative. Conclusion Aggressive treatment methods, administration of immune suppressants, and antibiotics, with an intention to salvage, have made patients susceptible to the benign fungus, causing it to evade the host immunity, thus leading to invasive infections. Applying different laboratory modalities would not only aid in providing fast and valuable information but also help in understanding the pathology which would assist the clinician in selecting the correct treatment for the patient.

Invasive fungal infections are common in immunosuppressed patients and in those admitted to critical care areas; these infections cause high morbidity and mortality. Fungi are a heterogeneous group of eukaryotic microorganisms that interact with humans. As a result, a wide spectrum of situations can occur, ranging from eradication of the fungus to death of the person. The most common species is Candida albicans. Most grow in the form of yeast, but some species can form pseudohyphae and produce different virulence factors, such as adhesins and secreted aspartic proteinases. They allow the microorganism to adhere to both human tissues and prosthetic devices in addition to causing tissue injury, thus favoring invasion at the site of infection respectively. Many factors are necessary such as the use of corticosteroids, antibiotics, disruption of natural barriers in addition to various comorbidities such as chronic kidney failure, liver disease, chronic and acute necrohemorrhagic pancreatitis that contribute to infection by this fungus. In addition to conventional culture media that can take 48 hours or more to obtain results, there are currently new diagnostic tools that can reduce the diagnosis time to just a few hours in the patients in whom Candida sp infection is suspected, among the most studied are the detection of fungal antigens in blood, nucleic acid amplification methods and clinical prediction indices. Early diagnosis, timely and appropriate antifungal treatment determine the survival of patients.

Since the Amsterdam declaration in 2011, the taxonomy of two names (sexual and asexual) for each species has been abandoned, and the first name described has also been prioritized. Since it is recommended to start using the updated names of the genera and species of yeasts and filamentous fungi, a transition is proposed using the current name, and the old one in parentheses written with lower case, in order to avoid confusion in clinician physicians already accustomed to the previous nomenclature.

RESUMEN

Despite the evolution of modern medicine, traditional medicine remains widespread in developing countries and its use continues to increase in industrialized countries. It is the same way that the effectiveness of the hydroalcoholic extract of Terminalia ivorensis was tested on the feet fungus disease of volunteers. Objective: The present work is oriented in the preparation of an antimicrobial hydroalcoholic extract of Terminalia ivorensis, a medicinal plant in order to enhance it. Materials and Methods: One hundred (100) grams of powder from trunk bark’s Terminalia ivorensis were extracted by homogenisation in a solvent mixture of 70% ethanol and 30% distilled water in a blender. After six grinding cycles, the homogenate obtained in each case was first wrung out in a clean white cloth square and then successively filtered twice on cotton wool and on Whatman 3 mm filter paper. The filtrate obtained was dried in a Venticell oven. The powder obtained constitutes the hydroalcoholic extract (or The 70% hydroethanolic extract). The 70% hydroethanolic extract of Terminalia ivorensis obtained was mixed with water to obtain a pasty liquid form before being tested on feet fungus disease using a cotton ball. Results: The extract had activity on these different shapes of feet fungus disease with a marked improvement. The volunteers who finished their treatment have been cured of feet fungus disease. Conclusion: The treatment results obtained revealed that the hydroalcoholic extract has good antimicrobial activity. The hydroalcoholic extract can be an undeniable source for the development of Improved Traditional Medicines (ITM) against feet fungus disease.

RESUMEN

Background: Mucormycosis, once a rare fungal disease, has now shown a surge in cases, and its epidemiology in India is intriguing. Rising incidence confers the necessity of defining the demography of the population at risk, early diagnosis, and recommendations for the management of mucormycosis in patients with coronavirus disease (COVID-19) infection. Aims: To study the clinico-epidemological profile, risk factors, and histopathological features of COVID-19-associated Mucormycosis (CAM). Setting and Design: This is a retrospective observational study. Material and Methods: Fifty-six biopsy specimens taken from patients of COVID-19 with clinical suspicion of mucormycosis were included. Results: Diabetes mellitus, corticosteroids, and oxygen therapy were the most common risk factors associated with CAM. The mean duration between the onset of symptoms of COVID-19 and onset of CAM was 25.2 ± 15.3 days. Rhino-orbito-cerebral mucormycosis (ROCM) was present in 24.5% of the cases, with a survival rate of 58.3%. Histopathology showed tissue invasion by branching broad-based, pale, fungal hyphae consistent with mucor in 49 (87.5%) cases. Inflammation was acute neutrophilic type in 61.2% of the patients with a survival rate of 63.33%. The survival rate with chronic non-granulomatous and chronic granulomatous was 100% and 81.8%, respectively. Conclusion: Keeping a high index of suspicion for mucormycosis in COVID-19 by all medical fraternities of the health community in the existence of risk factors is the need of the hour for prompt diagnosis. Biopsy of antral necrotic tissue should be performed immediately after suspicion for histopathological study to confirm the diagnosis of mucormycosis and predict prognosis depending on the type of inflammation incited, fungal morphology, load, and necrosis.

RESUMEN

Purpose: To evaluate the knowledge and skills of clinicians in differentiating cases of Pythium from fungal keratitis based on their clinical appearance. Methods: Thirty slit?lamp photographs from culture?proven cases of Pythium and fungal keratitis were selected for conducting a prospective online?based photographic survey. The participants were asked to identify the causative organism and enlist the specific clinical features which helped in reaching the diagnosis. Both cornea specialists and clinical fellows/trainees participated in the survey. Sensitivity, specificity, and predictive values were estimated, and the factors associated with these, including the identification rates of clinical signs, were evaluated using bivariate and multivariate linear regression analyses. The identification rates between the consultants and fellows were compared. Results: A total of 42 cornea specialists (28 consultants and 14 clinical fellows) participated in the survey. Clinicians were able to differentiate Pythium from fungal etiology in only 56% of cases. The differentiating ability between the consultants and fellows was statistically insignificant. Average sensitivity, specificity, and positive and negative predictive values for differentiating Pythium from fungus were 56%, 65%, 56%, and 66%, respectively. The specific clinical features enlisted by the participants to differentiate Pythium from fungus were the presence of tentacles, peripheral guttering, dot?like infiltrates, and elevated plaques. Conclusion: Clinical knowledge regarding the signs and symptoms and the microbiological identification of features for Pythium are still lacking among clinicians. As the treatment protocols are distinctly different for both, it is imperative to enhance the knowledge and diagnosing ability to tackle this emerging microorganism which causes high morbidity

RESUMEN

@#Abstract:Objective To investigate the clinical characteristics and early diagnostic methods of patients with Talaromyces marneffei infection, so as to reduce the mortality of patients. Methods The clinical characteristics and microbiological analysis data including fungal culture, smear examination and mass spectrometry were collected from 18 patients with Talaromyces marneffei infection in the Department of Respiratory Medicine, Department of Tuberculosis, and Department of Critical Respiratory Medicine in Fuzhou Pulmonary Hospital from January 2017 to December 2021, and descriptive analysis was conducted. Results All the 18 patients were confirmed to be infected with Talaromyces marneffei by conventional culture and matrix-assisted laser desorption/ionization-time of flight (MALDI-TOF) mass spectrometry (MS). The main infection sites of 18 patients with Talaromyces marneffei infection were lungs and lymph nodes, and the patients were accompanied by clinical manifestations such as cough, sputum and fever. The imaging features such as patchy shadows, mediastinal lymph node shadows and nodular shadows were common. Microbiological testing showed a statistically significant difference between smear and culture with a higher positive culture rate (χ2=13.74, P<0.05). The positive rate of blood culture in microbiological test was 60.0% (9/15), the positive rate of bronchial lavage fluid culture was 26.7% (4/15), the positive rate of sputum culture was 5.6% (1/18), one case each of pus, bone marrow, pleural fluid and cerebrospinal fluid was positive for culture and the other cases were negative, one case of sputum and one case of pus were positive for smear and the rest were negative. Colony characteristics showed that the colony morphology was mycelial phase at 25 ℃, producing red pigment, and the branching pattern of the penicillus was seen microscopically as monoverticillate or biverticillate; At 35 ℃, the yeast phase appeared at the initial stage, and then the mycelium phase changed after 5-6 days; the yeast phase was observed at 37 ℃, and yeast-like cells were seen under the microscope. All 18 patients with Talaromyces marneffei infection got better after using antifungal drugs. Compared with non-HIV patients with Talaromyces marneffei infection, leukopenia and anemia were common in HIV patients with Talaromyces marneffei infection, and the differences were statistically significant (P<0.05). Conclusions The infection of Talaromyces marneffei can be divided into localized type and disseminated type, which usually invade the lungs, skin, lymph nodes and other places. The main manifestations of patients are fever, cough, phlegm and other atypical symptoms. At present, the diagnosis of Talaromyces marneffei infection is mostly based on the fungal culture test, and the application of MALDI-TOF MS method can effectively shorten the diagnosis time of Talaromycosis marneffei. Clinical characteristics combined with microbiological analysis provide an objective basis for early diagnosis of patients with Talaromyces marneffei infection, and timely use of antifungal therapy can improve the prognosis of patients.

RESUMEN

Aims@#Providing safe drinking water is an ongoing global concern. Coagulation is an essential process in water treatment. However, most of the coagulants are chemical in nature and have negative impacts on human health and the environment. This study investigated the production of myco-coagulant in solid-state fermentation using a fungal strain. @*Methodology and results@#A scale-up was performed using the tray method to investigate the influence of substrate thickness (from 2-30 mm) on myco-coagulant production. The results revealed that the turbidity removal efficiency of myco-coagulant in kaolin suspension was found to be increasing with the increase in thickness of the coco peat substrate. However, the myco-coagulant extracted from the media with a thickness of 30 mm was able to remove the highest turbidity by 96%. Three different subculturing methods for mycelium inoculation were evaluated. The surface inoculation approach produced better results than other inoculation processes. The effect of initial turbidity values (50-300 NTU) on turbidity removal was studied too. The myco-coagulant was found to be the most suitable for high-turbidity water (300 NTU) with turbidity removal of 52%. Subculturing of fungus from solid-state to solid-state was also studied, which showed that the strategy was just as effective as an inoculum-based subculture. @*Conclusion, significance and impact of study@#Excellent bio-coagulation activity has been shown for the myco-coagulant that was isolated from the fungus strain. Subculturing using existing substrates will be more economical than subculturing using fresh inoculum. This strategy saves time, labour and cost of the coagulant production.
