RESUMEN

Background and Aim: Despite the economic importance of buffalo as a main source of milk and meat, only little attention has been directed to its immune and reproductive performance. The early diagnosis of subclinical endometritis may reduce the economic loss of buffalo’s production. The difference in expression profiles of immunity-related genes has an important role in the early detection of subclinical endometritis. This study aimed to assess the expression of five immunity-related genes: TGFBR1, PTGER2, PTGER4, HP and CXCL5 in endometritis-infected buffaloes. Materials and Methods: Total RNA was extracted from 120 buffalo uterine samples; 60 infected with endometritis and 60 healthy ones. Qt-PCR was performed on cDNA synthesized from extracted RNA using Sybr green and GAPDH as a house-keeping gene. Results: The results showed the up-regulation of two tested genes; TGFBR1 and CXCL5 in endometritis-infected buffalo compared to healthy animals by 7.9 and 4.3 folds, respectively at a significance level of p<0.05. The other three tested genes; PTGER2, PTGER4 and HP were down-regulated in buffalo during endometritis infection at different levels; PTGER2 and HP (0.6 folds, p<0.05) and PTGER4 (0.4 fold, p= 0.2). Conclusions: It is to be concluded that the assessment of expression of inflammation-related immunity genes may have an effective role on the detection of endometritis infection in buffalo during its early stages and this early diagnosis can reduce the economic loss of buffalo production and reproduction.

Objective To analyze the infection situation and risk factors of Helicobacter pylori (Hp) in children in Yulin, Shaanxi province. Methods A total of 374 preschool children with digestive tract symptoms in Yulin area from December 2019 to December 2020 were selected. All the research object of personal data by using self-designed questionnaire (gender, age), personal experiences and health habits (common tableware, common koubei brushing your teeth, wash your hands before the rice then empress), the life habits (whether diet, koubei utilities, brushing your teeth regularly to suck finger), socioeconomic status, place of residence, with the number of living, family economic income), caregivers has a history of stomach problems (parents, nanny) And so on. The investigation method is the combination of telephone inquiry and on-site investigation. 3ml fasting venous blood was collected, and the supernatant was centrifuged. Serum IL-17 and IL-10 levels were determined by ENZYme-linked immunosorbent assay, and Treg and Th17 cell levels were determined by flow cytometry. Hp infection was detected by 14C urea breath test. Univariate analysis and logistic regression were used to analyze the independent risk factors of Hp infection in children. Results Eighty-three of the 374 children were infected with Hp, and the infection rate was 22.19%. The levels of Treg and IL-10 in peripheral blood of Hp infected children were significantly lower than those of non-hp infected children (P<0.05). The levels of Th17 and IL-17 in peripheral blood of children with Hp infection were significantly higher than those without Hp infection (P<0.05). In univariate analysis, there were statistically significant differences between the two groups in age, parental education level, per capita monthly income, sharing tableware, sharing toothbrushing cups, washing hands before and after meals, sucking fingers, parents' knowledge of Hp, and caregivers' history of stomach disease (P<0.05). Logistic regression analysis showed that sharing tableware, sharing toothbrushing cups, sucking fingers, parents' lack of knowledge about Hp, and caregivers' history of gastric diseases were independent risk factors for Hp infection in children (P<0.05). Conclusion The infection rate of Hp in children is high, and Hp infection can cause immune system dysfunction in children. It is necessary to take targeted and effective clinical measures to prevent Hp infection.

RESUMEN

BACKGROUND: Chronic lymphocytic leukaemia (CLL) is a neoplasm of B-cells characterized by variable prognosis. Exploring the proteome of CLL cells may provide insights into the disease. Therefore, eleven proteomics experiments were conducted on eleven primary CLL samples. RESULTS: We reported a CLL proteome consisting of 919 proteins (false discovery rate (FDR) 1%) whose identification was based on the sequencing of two or more distinct peptides (FDR of peptide sequencing 1%). Mass spectrometry-based protein identification was validated for four different proteins using Western blotting and specific antibodies in different CLL samples. Small sizes of nucleolin (~57 kDa and ~68 kDa) showed a potential association with good prognosis CLL cells (n = 8, p < 0.01). Compared with normal B-cells, CLL cells over-expressed thyroid hormone receptor-associated protein 3 (THRAP3; n = 9; p = 0.00007), which is implicated in cell proliferation; and heterochromatin protein 1-binding protein 3 (HP1BP3; n = 10; p = 0.0002), which promotes cell survival and tumourogenesis. A smaller form of HP1BP3, which may correspond to HP1BP3 isoform-2, was specifically identified in normal B-cells (n = 10; p = 0.0001). HP1BP3 and THRAP3 predicted poor prognosis of CLL (p 0.05). Consistently, THRAP3 and HP1BP3 were found to be associated with cancer-related pathways (p 0.05). CONCLUSIONS: Our findings add to the known proteome of CLL and confirm the prognostic importance of two novel cancer-associated proteins in this disease.

RESUMEN

Objective: To reduce or avoid the volatilization loss of camphor (CA) in the industrialized preparation of cataplasm, using the inclusion technology of HP-β-CD. Methods: The HP-β-CD inclusion compound of CA (CA-HP-β-CD) was prepared by the constant temperature stirring method. The preparation processing of CA-HP-β-CD was optimized with inclusion efficiency as an index, and the pharmaceutical properties of CA-HP-β-CD were characterized by DSC, UV and SEM. The CA and CA-HP-β-CD were prepared to cataplasm at the industrial-scale production respectively, then the content of CA was determined and the statistical analysis was carried out. Results: The inclusion efficiency of CA-HP-β-CD for CA was (68.57 ± 1.26)% when the CA-HP-β-CD was prepared with the mass ratio of CA to HP-β-CD 1:3, the working temperature at 35 ℃ and stirring at 300 r/min for 4.0 h. The DSC, UV and SEM scans showed that there was a good compatibility between CA and HP-β-CD, and CA could be fully encapsulated by HP-β-CD and formed a stable inclusion complex of CA-HP-β-CD. Results of industrial-scale production of cataplasm for CA and CA-HP-β-CD revealed that the retention rate (actual content/feeding quantity) were (80.13 ± 1.05)% and (39.45 ± 1.38)%, respectively. And there was a statistically significant difference between them (t-test, P < 0.01), indicating that the HP-β-CD inclusion complex effectively reduce the volatilization loss of camphor CA. Conclusion: The volatilization loss of CA at the industrial-scale production of cataplasm was effectively reduced by the CA-HP-β-CD, which improves the utilization rate of CA and reduces the cost, and provides ideas and reference for the research and development of cataplasm of volatile drugs.

RESUMEN

D-Glycero-D-mannno-heptose 1β, 7-bisphosphate (HBPβ) is an important intermediate for constructing the core structure of Gram-negative bacterial lipopolysaccharides and was reported as a pathogen-associated molecular pattern (PAMP) that regulates immune responses. HBPβ with 3-O-amyl amine linker and its monophosphate derivative D-glycero-D-mannno-heptose 7-phosphate (HP) with 1α-amyl amine linker have been synthesized as candidates for immunity study of HBPβ. The O3-amyl amine linker of heptose was installed by dibutyltin oxide-mediated regioselective alkylation under fine-tuned protecting condition. The stereoselective installation of 1β-phosphate ester was achieved by NIS-mediated phosphorylation at low temperature. The strategy for installation of 3-O-amyl amine linker onto HBP derivative can be expanded to the syntheses of other conjugation-ready carbohydrates bearing anomeric phosphoester.

ABSTRACT Calcium silicate-based materials have been shown to be bioactive due to their ability to produce biologically compatible carbonated apatite. The objective of this study was to analyze the bioactivity of Biodentine ™ and MTA Repair HP® in contact with human dentine discs, which were sealed and divided randomly to form four groups: group 1 Biodentine™, group 2 MTA Repair HP®, positive control group MTA Angelus® and negative control group IRM®, which were incubated in PBS solution for 10 days, for a subsequent analysis by means of MEB-EDS and Raman spectroscopy. The three calcium-based materials analyzed in this study proved to be bioactive because upon contact with a phosphate-based solution they were triggered at the onset of amorphous calcium phosphate, as the precursor during the formation of carbonated apatite.

RESUMEN

The study aims to use CRISPR/Cas9 introducing foreign gene targeted knock-in into chicken EAV-HP genome. First, specific primers were designed for amplification of EAV-HP left, right homologous arms and enhanced green fluorescent protein (eGFP) expression cassette. PCR products of homologous arms were ligated to both sides of eGFP by overlap extension PCR, resulting in full-length donor DNA fragment designated as LER. Then LER fragments were cloned into pMD19-T to obtain donor vector pMDT-LER. Subsequently, the donor vector pMDT-LER was transfected into HEK293T cells to verify the expression of eGFP gene. Furthermore, co-transfection of CRISPR/Cas9 expression vector and pMDT-LER into chicken DF-1 cells was performed to achieve eGFP transgenic cells. Meanwhile, eGFP expression was observed in cells, and the event of eGFP integration into EAV-HP genome was detectable by amplification of target DNA. Finally, the transgenic DF-1 cells were passaged seven times, and the stable integration and expression of eGFP was checked by PCR and Western blotting. These results demonstrated that eGFP gene was knocked into the EAV-HP genome successfully, which provides a new integration site for research of transgenic chicken.

RESUMEN

Objective To investigate the effects and the law of anti bitterness efficiency of hydroxypropyl-β-cyclodextrin (HP-β-CD) and other different types bitterness masking at different concentrations on the bitterness decoction of Chinese materia medica (BDCMM) by using the traditional human taste panel method (THTPM) and Electronic tongue (E-tongue). Methods Based on THTPM, the bitterness reduction value (ΔI), the correction rate of bitterness suppression (CRBS), and the correction rate of bitterness suppression potency index (PI50) were used as indicators to evaluate the bitterness suppression effect of four bitterness masking at different concentrations on BDCMM such as Sophorae Flavescentis Radix (SFR), and the relationship model between the concentrations of ΔI and bitterness masking was established to explore the bitterness suppression rule of bitterness masking on different BDCMM. Based on the E-tongue method, the E-tongue taste response information values of HP-β-CD and Acesulfame K to BDCMM such as SFR were measured at different concentrations, and the electronic tongue bitterness reduction value (ΔIe) was used as an index to explore the change rule of ΔIe with bitterness masking concentration. THTPM and E-tongue was combined to establish a prediction model of the effect of bitterness suppression. Results The ΔI, CRBS, and PI50 of four kinds of BDCMM with different concentrations of bitterness masking were measured, which can be used to compare the bitterness suppression effect of bitterness masking; The relation between ΔI and bitterness masking concentration accorded with Weibull curve model; Based on E-tongue, ΔIe was obtained, and the model of the relationship between ΔIe and bitterness masking concentration was established. The relationship between ΔIe and ΔI was established. The model determination coefficients of HP-β-CD for the two types of ΔIe of the two BDCMM were 0.986 1, 0.977 9, 0.989 0, and 0.982 0 respectively (P < 0.01, n = 6). Acesulfame had no response to the sensor and did not establish a model of bitterness suppression law. Conclusion Based on THTPM and E-tongue, a method for evaluating the bitterness suppression effect of bitterness masking on BDCMM was established. The bitterness suppression effect of bitterness masking on BDCMM such as SFR was studied by molecular inclusion, high-efficiency sweetening, and other bitterness suppression mechanisms were inverstigated.

RESUMEN

The efficacy of standard therapeutic strategies for Helicobacter pylori (H. pylori) infection is decreasing over time due to the emergence of drug-resistant strains. As an alternative, the present study investigated the capacity of Lactobacilllus paracasei (L. paracasei) HP7, isolated from kimchi, to inhibit H. pylori growth. The effects of L. paracasei HP7 on H. pylori adhesion and H. pylori-induced inflammation were examined in AGS human gastric adenocarcinoma epithelial cells and a mouse model of H. pylori SS1 infection. L. paracasei HP7 reduced H. pylori adhesion to AGS cells and suppressed the inflammatory response in infected cells by downregulating interleukin-8. H. pylori colonization in the stomach of C57BL/6 mice was demonstrated by rapid urease test, and results showed significant decrease in mice post-treated with L. paracasei HP7. Additionally, L. paracasei HP7 decreased gastric inflammation and epithelial lesions in the stomach of H. pylori-infected mice. These results demonstrate that L. paracasei HP7 treatment can inhibit H. pylori growth and is thus a promising treatment for patients with gastric symptoms such as gastritis that are caused by H. pylori infection.

Objective:To investigate the effect of Helicobacter pylori(Hp) infection in patients with chronic atrophic gastritis (CAG) and the expression of transforming growth factor-β receptorⅡ,interleukin 6 and tumor necrosis factor-α in gastric mucosa.Methods:76 cases of gastroscope biopsy specimens were collected in the CAG patients, the infection of Hp was detected by PCR fluorescence, and immunohistochemical staining was used to determine the expression of TGF-βRⅡ,IL-6 and TNF-α in gastric foveolar epithelium and stromal inflammatory cells. Results:There were significant differences in chronic inflammation between Hp-positive and Hp-negative group (P<0.05).Expression of IL-6 in stromal inflammatory cells was significantly different between Hp-positive and Hp-negative group (P<0.05).Expression of TGF-βRⅡ and TNF-α was not significantly different in two groups(P>0.05).IL-6 expressed instromal inflammatory cells were correlated with chronic inflammation (r=0.249,P=0.03).The degree of intestinal metaplasia and dysplasia were correlated with the atrophic severity respectively(r=0.697,0.366).Conclusion:IL-6 is related to the chronic inflammation in patients with CAG who has Hp infection. The chronic atroplic severity promotes the development of intestinal metaplasia and dysplasia.

RESUMEN

Objective To observe the Hp infection effect on blood lipid metabolism and oxidative stress through detecting acute cerebral infarction (ACI) with Helicobacter pylori (Hp) infection serum lipid and malondialdehyde (MDA) level and superoxide dismutase (SOD) activity. Methods Chose ACI 350 cases of ACI group,80 cases of control group and detected serum Hp-IgG antibody (by colloidal gold) and 14C urea breath test (by14 C-UBT).Used the American beckman coulter AU680 fullautomatic biochemical analyzer to detect the serum lipid and SOD activity (colorimetry),and used Shanghai now 752 visible spectrophotometcr (532 nm) to detect the serum MDA level (TBA).Results The Hp infection rate of ACI group (69.5 %) was significantly higher than that in the control group (33.8%),the differences was statistically significant (x2 =18.882,P<0.01).The differences in serum TC,TG,LDL-C and HDL-C levels were statistically significant (t=4.167 ~5.521,all P<0.01) compared with Hp non-infection group. The serum MDA level and SOD activity of the control group and Hp infection group were statistically significant (F=34.891~46.613,all P<0.01).Hp infection with Hp infection group to compare serum MDA level and SOD activity differences statistically significant (t=5.197,9.713,all P<0.01),the Hp infection group and control group in comparative differences were statistically significant (t=3.173,8.228,all P<0.01).Conclusion Acute cerebral infarction merger of HP infection, blood lipid metabolic disturbance in body increased and enhanced oxidative stress reaction, this may be intensified HP infection is one of the reasons for acute brain infarction dis ease.
