RESUMEN

RESUMEN El CCU es la segunda causa de muerte en mujeres de nuestro país. Dentro de los primeros mecanismos de defensa del hospedero se encuentra la respuesta inmune de las células NK y su función lítica a expensas de su receptor activador NKG2D, el cual posee como ligandos mica, micb y ulbp (1-6), los cuales se expresan en células transformadas y/o infectadas por virus. Uno de los mecanismos de evasión por parte de la célula tumoral es el clivaje de estas proteínas a través de metaloproteinasas como adam10, adam17 y mmp14. Se analizó la expresión de estos ligandos y metaloproteinasas mediante PCR tiempo real, en lineas celulares de referencia para cáncer cervical como HeLa (positiva para VPH-18) y C33A (negativa para VPH). Se obtuvieron valores representativos de expresion relativa genica con diferencias significativas asi: mmp14 en linea HeLa (p= 0.006); y mica y ulbp-3 en la linea C33A (p= 0.020 y p=0.003 respectivamente). Por lo tanto, se podría sugerir que la expresión de mmp14 se encuentra posiblemente involucrados con la presencia de VPH causante del cancer cervical y la respuesta inmunne innata desarrollada.


ABSTRACT Cervical cancer is the second leading cause of death in women in our country. Within the first host defense mechanisms is the immune response of NK cells and their lytic function at the expense of its NKG2D receptor activator which has as ligands mica, micb and ulbp (1-6), which are expressed in transformed cells and / or virally infected. One of the mechanisms of evasion by the tumor cell is the cleavage of these proteins through metalloproteinases as adam10, adam17 and mmp14. We analyzed the expression of these ligands and metalloproteinases by real time PCR, in reference to cell lines HeLa cervical cancer (positive for HPV-18) and C33A (negative for HPV). We obtained representing relative gene expression with significant differences from the other lines of study as follows: mmp14 in HeLa (p = 0.006); and mica and ulbp-3 in C33A (p = 0.020 and p = 0.003 respectively). Thus one might suggest that the expression of mmp14 is possible involved with HPV presence causing high risk of cervical cancer and innate inmunne response developed.

RESUMEN

Aim To observe whether matrix metallo-proteinase-14 ( MMP-14) and tissue matrix metallopro-teinase inhibitor-4 ( TIMP-4) were involved in the car-diac-protection of mitochondrial aldehyde dehydrogen-ase 2 ( ALDH2) against high glucose induced rat pri-mary cardiomyocyte injury. Methods Rat primary cardiomyocytes were cultured. The cardiomyocyte via-bility was detected by MTT assay at different concentra-tion of glucose at different time point. After established high glucose-induced cardiomyocytes injury model, cardiomyocytes were randomly divided into 4 groups:normal control group ( NG, glucose at 5.5 mmol· L-1) , NG + Alda-1 group ( Alda-1 at 20 μmol·L-1) , high glucose group ( HG, glucose at 30 mmol·L-1) and HG+Alda-1 group. The cell viability at 48 h and oxidative stress level were detected by MTT and DHE staining methods. The protein expressions of ALDH2, MMP-14 and TIMP-4 were determined by Western blot. Results The cardiomyocytes injury model was established according to the cell activity result. Com-pared with NG group, the cell viability, the protein ex-pressions of ALDH2, MMP-14, the ratio of MMP-14/TIMP-4 were decreased, TIMP-4 protein expression and the level of oxidative stress were increased in HG group. Compared with HG group, in HG + Alda-1 group, the cell viability, the protein expressions of AL-DH2, MMP-14, the ratio of MMP-14/TIMP-4 were in-creased, the levels of oxidative stress and TIMP-4 pro-tein expression were decreased. Conclusion Activa-tion of mitochondrial ALDH2 may relieve high glucose induced cardiomyocytes injury. The protective effect was likely related to the inhibition of oxidative damage, down-regulation of MMP-14 and up-regulation of TIMP-4 proteins.

Objective To investigate the expression and significance of MMP-14 and EMMPRIN and the correlation between them in oral squamous cell carcinoma ( OSCC) . Methods The expression of MMP-14 and EMMPRIN mR-NA in 43 cases of OSCC and 11 cases of normal oral tissue were detected by RT-PCR. Results The expression of MMP-14 and EMMPRIN mRNA in OSCC was significantly higher than those in normal oral tissue ( P <0.05 ) . Compared with those without lymph node metastasis in OSCC, the expression of MMP-14 and EMMPRIN mRNA with lymph node metastasis was significantly higher ( P <0.05 ) . Furthermore, the expression of MMP-14 and EMMPRIN was significantly associated with pathological stage and the lymph node metastasis,but neither of them was correlated with sex and age. Correlation analysis showed that MMP-14 expression level was positively related to EMMPRIN level(r=0.801,P<0.01). Conclusion The expression of MMP-14 and EMMPRIN in OSCC is signif-icantly correlated with occurrence, progress and metastasis of OSCC. Therefore, the combination of detecting the expression of MMP-14 and EMMPRIN mRNA may evaluate the degree of malignancy and predict the prognosis of patients with OSCC.

RESUMEN

PURPOSE: The purposes of this study were to compare and quantify the expression of PGE2, MMP-14 and TIMP-1 in the gingival tissues of patients with type 2 diabetes mellitus and healthy adults of chronic periodontitis with alveolar bone resorption. MATERIALS AND METHODS: Gingival tissue samples were obtained during periodontal surgery or tooth extraction. According to the patient's systemic condition & clinical criteria of gingiva, each gingival sample was devided into three groups. Group 1 (n=8) is clinically healthy gingiva without bleeding and no evidence of bone resorption or periodontal pockets, obtained from systemically healthy 8 patients. Group 2 (n=8) is inflammed gingiva from patients of chronic periodontitis with alveolar bone resorption. Group 3 (n=8) is inflammed gingiva from patients of chronic periodontitis with alveolar bone resorption associated with type 2 diabetes. Tissue samples were prepared and analyzed by Western blotting. The quantification of PGE2, MMP-14 and TIMP-1 were performed using a densitometer and statistically analyzed by one-way ANOVA followed by Tukey test. RESULTS: The expressions of MMP-14 and TIMP-1 were showed increasing tendency in group 2 & 3 compared to group 1. The expressions of PGE2, MMP-14 were showed increasing tendency in group 3 compared to group 1 and group 2. According to MMP-14 levels were increasing, PGE2 showed increasing tendency in group 3, and although PGE2, MMP-14 levels were increasing, TIMP-1 levels were similar expressed comparing to group 2. CONCLUSION: In conclusion, this study demonstrated that the expression levels of MMP-14 and TIMP-1 had increasing tendency in inflammed tissue. It can be assumed that PGE2 and MMP-14 may be partly involved in alveolar bone resorptive process and the progression of periodontal inflammation associated to type 2 DM.

RESUMEN

Objective:To detect the expression of MMP-14,MMP-2 and TIMP-2 in gastric ulcer tissues, gastric cancer and tissues around gastric cancer. Methods:The expression of MMP-14,MMP-2,and TIMP-2 were detected by S-P immunohistochemistry in gastric ulcer tissues,gastric cancer tissues and around tissues from operated patients of gastric cancer (n=30) and gastric ulcer patients(n=20)who were diagnosed by stomach lens. Results:(1) The expression rates of TIMP-2, MMP-2 and MMP-14was 43.3%,76.7% and 53.3% respectively in gastric cancer tissues and 56.7%,46.7% and 40%in tissues around it,but 70%,45% and 30% in gastric ulcer tissues. (2) The expression of TIMP-2 in gastric cancer was lower than that in tissues or that in gastric ulcer tissues(P0.05).The expression of TIMP-2 was negatively correlated with the expression of MMP-2 or MMP-14(rs=-1.0,P
