RESUMEN

【Objective】 To summarize the clinical features, serological features, blood transfusion protocols and treatment of 3 cases of passenger lymphocyte syndrome(PLS) after ABO-incompatibility liver and renal transplantation in our hospital, in order to provide guidance for comprehensive clinical understanding and recognition of this disease, especially early recognition and treatment. 【Methods】 By collecting the basic information of the patients and the time of cross-matching incompatibility of homologous blood after transplantation, observing the skin yellow staining, detecting hemoglobin value and other hemolysis indexes, and blood group serological detection results before and after transfusion, the diagnosis and analysis were performed. The diagnosis and treatment effect of PLS were analyzed by collecting the clinical outcome information after immunization and blood transfusion. 【Results】 Three cases of ABO-incompatible liver transplantation showed decreased hemoglobin and hemolysis, incompatible cross-matching of homologous blood, and anti-A and anti-B IgG antibodies were confirmed by serological test. After treatment such as immunosuppression and plasma exchange, blood transfusion was effective, hemolysis was improved, and antibodies gradually disappeared. 【Conclusion】 ABO blood group antibody screening, unexpected antibody screening and direct antiglobulin test(DAT)should be performed regularly for ABO-incompatible liver and renal transplantation cases, in order to detect the PLS early. A series of laboratory tests related to PLS should be performed in time to diagnose and adjust the treatment plan, including transfusion strategy, when homologous cross-matching is incompatible.

ObjectiveTo establish a high performance liquid chromatography(HPLC) fingerprint of Yanghetang benchmark sample, and evaluate its quality with chemometric methods, so as to provide a reference for the quality control of this benchmark sample. MethodHPLC was used to establish the fingerprint of Yanghetang benchmark sample with ZORBAX SB-C18 column(4.6 mm×250 mm, 5 μm), the mobile phase was consisted of acetonitrile(A) -0.05% phosphoric acid aqueous solution (containing 0.05% triethylamine solution)(B) for gradient elution(0-5 min, 2%-3%A; 5-15 min, 3%-5%A; 15-65 min, 5%-30%A; 65-90 min, 30%-70%A), the flow rate was 1.0 mL·min-1, the column temperature was 35 ℃, and the detection wavelength was 210, 260 nm. Traditional Chinese Medicine(TCM) Chromatographic Fingerprint Similarity Evaluation System (2012 edition) combined with cluster analysis, principal component analysis(PCA) and partial least squares-discriminant analysis(PLS-DA) were used to evaluate the quality differences between different batches of Yanghetang benchmark samples, and to find the main chemical components responsible for the quality differences. ResultHPLC fingerprint of Yanghetang benchmark sample was established, 13 common peaks were identified and attributed to each common peak, including peaks 2 and 8 from Rehmanniae Radix Praeparata, peaks 10 and 11 from Cinnamomi Cortex, peaks 1, 3-6 from fried Sinapis Semen, peak 13 from Ephedrae Herba, and peaks 7, 9, 12 from Glycyrrhizae Radix et Rhizoma. Eight of them were identified by comparing with control substance, which were 5-hydroxymethylfurfural(peak 2), sinapine thiocyanate(peak 4), glycyrrhizin(peak 7), verbascoside(peak 8), cinnamic acid(peak 10), cinnamaldehyde(peak 11), glycyrrhizic acid(peak 12) and ephedrine hydrochloride(peak 13). The similarities of the HPLC fingerprints of 15 batches of Yanghetang benchmark samples with the control fingerprint were all greater than 0.80. The three chemometric methods could classify the samples into two categories. Eight differential components were screened out, among which 5-hydroxymethylfurfural, sinapine thiocyanate, verbascoside and ephedrine hydrochloride were identified. ConclusionThe established fingerprint analysis method is accurate, stable and reproducible, which basically reflects the overall chemical composition of Yanghetang benchmark sample, and can provide a basis for establishment of quality standards for compound preparations of this famous classical formula.

Objective To identify the retention time of semen(stains)inside and outside the cadaver using Fourier Transform Infrared(FTIR)spectroscopy and chemometrics.Methods A simulated environment inside the porcine vagina was used to retain fresh semen for 0~168 hours inside and outside the body.Infrared spectroscopy data were collected,and chemometric methods including Principal Components Analysis(PCA)and Partial Least Squares(PLS)regression models were established.Results The infrared spectrum peak positions of semen(stains)inside and outside the body did not show significant changes over time.The PCA results showed good clustering relationships,and the predicted deviation value in the PLS regression model was 15.786 3 hours,with an R2 value of 0.922 4.The characteristic wavenumbers selected by the CARS algorithm were in the wavelength ranges of 1 000~1 180 cm-1,1 430~1 480 cm-1,and 1 700~1 760 cm-1.Conclusion Fourier Transform Infrared spectroscopy has the potential to identify the retention time of semen(stains)inside and outside the body,and there are differences in the degradation trends of semen(stains)inside and outside the body.

OBJECTIVES@#To explore the potential biomarkers for the diagnosis of primary brain stem injury (PBSI) by using metabonomics method to observe the changes of metabolites in rats with PBSI caused death.@*METHODS@#PBSI, non-brain stem brain injury and decapitation rat models were established, and metabolic maps of brain stem were obtained by LC-MS metabonomics method and annotated to the HMDB database. Partial least square-discriminant analysis (PLS-DA) and random forest methods were used to screen potential biomarkers associated with PBSI diagnosis.@*RESULTS@#Eighty-six potential metabolic markers associated with PBSI were screened by PLS-DA. They were modeled and predicted by random forest algorithm with an accuracy rate of 83.3%. The 818 metabolic markers annotated to HMDB database were used for random forest modeling and prediction, and the accuracy rate was 88.9%. According to the importance in the identification of cause of death, the most important metabolic markers that were significantly up-regulated in PBSI group were HMDB0038126 (genipinic acid, GA), HMDB0013272 (N-lauroylglycine), HMDB0005199 [(R)-salsolinol] and HMDB0013645 (N,N-dimethylsphingosine).@*CONCLUSIONS@#GA, N-lauroylglycine, (R)-salsolinol and N,N-dimethylsphingosine are expected to be important metabolite indicators in the diagnosis of PBSI caused death, thus providing clues for forensic medicine practice.

【Objective】 To investigate the clinical characteristics and diagnosis and treatment of passenger lymphocyte syndrome (PLS) in patients following allogeneic hematopoietic stem cell transplantation (allo-HSCT). 【Methods】 A total of 489 patients who underwent allo-HSCT in Suzhou Hongci Hematology Hospital were retrospectively enrolled. The clinical process, diagnosis and treatment measures and prognosis of four patients complicated with PLS after transplantation were analyzed. 【Results】 Among the 489 patients, 4 were diagnosed with PLS. The blood types of donor/recipient ABO were all secondary incompatible (The blood type of donors were O and the recipients were A or B). The overall incidence of PLS in allo-HSCT was 0.82%(4/489)and 2.2%(4/179)in transplants with donor/recipient secondary incompatible ABO-blood types. PLS occured in 6-13 days after donor stem cell infusion. Clinical manifestations were dizziness and fatigue, low back pain, jaundice, deepening urine, rapid decrease in hemoglobin on laboratory tests, elevated indirect bilirubin and lactate dehydrogenase, positive urobilinogen, positive direct anti-human globulin test (DAT), and anti-A or anti-B antibodies against recipient red blood cells were detected in plasma. After the treatment of O-type washed red blood cells, methylprednisolone, gamma globulin, rituximab and other treatments, the hemolysis was improved. All patients achieved engraftment of neutrophil and platelet. Red blood cell transfusion was halted in 3 weeks. 【Conclusion】 PLS is a rare complication of allo-HSCT, which mainly occurs in allo-HSCT patients with secondary incompatibility of ABO blood group of donor/recipient. The clinical prognosis is good after properly treatment.

Resumo As cidades inteligentes vêm crescendo ao redor do mundo, impulsionadas por inovações tecnológicas. Com elas surgem diversas oportunidades, mas também novas ameaças à segurança e privacidade do usuário nessa realidade interconectada. Este estudo tem como objetivo investigar a percepção de segurança e confiança na tecnologia por parte dos cidadãos e como esta afeta a propensão ao seu uso e, consequentemente, à vida na cidade inteligente. Para tanto, conduziu-se um survey (n = 601), por meio do método PLS-SEM, para testar as hipóteses formuladas. Os resultados obtidos confirmam que o modelo proposto demonstra ser consistente. As relações "confiança e segurança subjetiva" e "segurança objetiva e privacidade de dados" obtiveram relações mais consistentes, assegurando a forte influência das barreiras "tangíveis" e "intangíveis" da percepção de segurança. Dessa forma, para obter e manter a confiança dos usuários, as instituições por trás da tecnologia precisam estar atentas à opinião deles e da sociedade, de forma a manter uma boa reputação para que possam, assim, perpetuar uma percepção positiva de segurança. Conclui-se, assim, que o conceito de segurança adquire uma nova dimensão no contexto da cidade inteligente por ser um componente crucial para toda a sua base e estar intimamente ligado à tecnologia, além de se apresentar como uma preocupação fundamental para os governos e as entidades que buscam implementar soluções e aplicações do conceito.


Resumen Las ciudades inteligentes están creciendo en todo el mundo, impulsadas por las innovaciones tecnológicas. Con ellas surgen diversas oportunidades, pero también nuevas amenazas a nuestra seguridad y privacidad en esta realidad interconectada. Este estudio tiene como objetivo investigar la percepción de los ciudadanos sobre la seguridad y la confianza en las tecnologías y cómo afectan la propensión a usarlas y, en consecuencia, la vida en la ciudad inteligente. Para ello, se realizó una encuesta (n = 601) utilizando el método PLS-SEM para contrastar las hipótesis formuladas. Los resultados obtenidos confirman que el modelo propuesto resulta ser consistente. Las relaciones 'confianza y seguridad subjetiva' y 'seguridad objetiva y privacidad de datos' obtuvieron relaciones más consistentes, confirmando la fuerte influencia de las barreras 'tangibles' e 'intangibles' de la percepción de seguridad. De esta forma, para obtener y mantener la confianza de los usuarios, las instituciones que están detrás de las tecnologías deben estar atentas a su opinión y a la de la sociedad a los efectos de mantener una buena reputación, para que puedan, así, mantener una percepción positiva de la seguridad. Se concluye que el concepto de seguridad adquiere una nueva dimensión en el contexto de la ciudad inteligente, ya que es un componente crucial de toda su base, estrechamente vinculado a la tecnología además de presentarse como una preocupación fundamental para los gobiernos y entidades que buscan implementar soluciones conceptuales y aplicaciones.


Abstract Smart cities are growing around the world, driven by technological innovations. With them come several opportunities and new threats to our security and privacy in this interconnected reality. This study investigates citizens' perception of security and trust in technologies and how they affect the propensity to use them and, consequently, life in the smart city. Therefore, a survey was conducted (n = 601) using the PLS-SEM method to test the formulated hypotheses. The results obtained confirm that the proposed model proves to be consistent. The relationships between 'trust and subjective security' and 'objective security and data privacy' obtained stronger relationships, confirming the strong influence of the 'tangible' and 'intangible' barriers of the perception of security. Thus, to obtain and maintain users' trust, the institutions behind the technologies need to be attentive to the opinion of their users and society to keep a good reputation and a positive perception of security. The users' opinion is a crucial component of smart cities' entire base, closely linked to technology, and presents as a fundamental concern for governments and entities that seek to implement concept solutions and applications.

RESUMEN

The present study developed an ultra-fast liquid chromatography coupled with triple quadrupole-linear ion trap composite mass spectrometry(UHPLC-QTRAP-MS) to simultaneously determine the content of potential active components in Scutellariae Barbatae Herba and also to provide a reference approach for screening out the differential quality control components among different batches of Scutellariae Barbatae Herba. Chromatographic separations were conducted on a Thermo Acclaim~(TM) RSLC 120 C_(18) column(3.0 mm×100 mm, 2.2 μm) in a gradient program. The mobile phase consisted of 0.1% aqueous formic acid and acetonitrile, and the column temperature was maintained at 40 ℃. The flow rate was 0.4 mL·min~(-1) and the injection volume was 2 μL. The targeted compounds were monitored in the multiple reaction monitoring(MRM) mode. The acquired data were processed by hierarchical cluster analysis(HCA) and partial least square discriminant analysis(PLS-DA). Sixteen compounds all showed good linear relationship within the corresponding linear ranges and the R~2 values were all higher than 0.993 2. The RSDs of precision, repeatability, and stability were less than or equal to 3.7%. Mean recovery rates were in the range of 95.67% and 104.8% with RSDs≤3.2%. According to HCA and PLS-DA, all samples were clustered into four categories. Scutellarin, acteoside, scutellarein, and scutebarbatine X(VIP>1) were considered as differential chemical markers in the four categories. In conclusion, the developed method can be used for the simulta-neous determination of the multiple components and quality control of Scutellariae Barbatae Herba.

ABSTRACT: Exporting the organic fresh fruit (FF) to Europe and America could be regarded as great opportunity for smallholders and suppliers of Gilgit Baltistan, Pakistan. Yet, the global organic farmland and the number of organic producers are continued to grow. To date, little is known about the behavior of Pakistani farmers towards organic fresh fruit production. This study examined the factors that influence the decision-making of fresh fruit smallholders concerning the conversion to organic farming (COF). This study uses Ajzen's theory of planned behavior (TPB) to predict the behavior of Pakistani smallholders of fresh fruit. Path analysis examined the utility of the extended version of the TPB model to anticipate 277 small-scale farmers' behavioral intentions towards the adoption of organic farming (AOF) and their prospect of converting their farm to organic produce within the next three years. The present study reported that the smallholders' attitude (ATT), subjective norms (SN), perceived behavior control (PBC), and farmer's group participation (FGP) were positively correlated with farmer's intention to adopt organic farming, while group participation of farmers has significant correlation with their behavior regarding production of organic fruits. The present study justifies using the model of farmers' group participation in policies that deliberately cheer up smallholders of fresh fruit to adopt organic farming, to raise farm incomes and to alleviate poverty.


RESUMO: Exportar a fruta fresca orgânica (FF) para a Europa e América pode ser considerada uma grande oportunidade para pequenos proprietários e fornecedores de Gilgit Baltistan, Paquistão. No entanto, as terras agrícolas orgânicas globais e o número de produtores orgânicos continuam a crescer. Até o momento, pouco se sabe sobre o comportamento dos agricultores paquistaneses em relação à produção de frutas frescas orgânicas. O objetivo deste estudo, portanto, foi examinar e determinar os fatores que influenciam a tomada de decisão dos pequenos produtores de frutas frescas sobre a conversão para a agricultura orgânica (COF). Este estudo usa a teoria do comportamento planejado de Ajzen (TPB) para prever o comportamento dos pequenos produtores de frutas frescas do Paquistão. A análise de caminho examinou a utilidade da versão estendida do modelo TPB para antecipar as intenções comportamentais de 277 agricultores de pequena escala em relação à adoção da agricultura orgânica (AOF) e sua perspectiva de converter sua fazenda em produtos orgânicos nos próximos três anos. O presente estudo descobriu que a atitude dos pequenos proprietários (ATT), as normas subjetivas (SN), o controle do comportamento percebido (PBC) e a participação do grupo de agricultores (FGP) foram positivamente correlacionadas com a intenção do agricultor de adotar a agricultura orgânica, enquanto a participação do grupo de agricultores correlação significativa com seu comportamento em relação à produção de frutas orgânicas. O presente estudo justifica o uso do modelo de participação de grupos de agricultores em políticas que encorajam deliberadamente os pequenos produtores de frutas frescas a adotar a agricultura orgânica, para aumentar a renda agrícola e diminuir a pobreza.

The present study evaluates different processing and drying methods and investigates their effects on the chemical components in Paeoniae Radix Alba via content determination. The fresh medicinal materials of Paeoniae Radix Alba collected from Bozhou of Anhui province were processed(boiled and peeled) and dried(hot air-dried, infrared-dried, and microwave-dried) at different temperatures(40, 50, 60 and 70 ℃), and the 11 components(monoterpene glycosides, polyphenols, tannin, and benzoic acid) in Paeoniae Radix Alba were determined by ultra-performance liquid chromatography coupled to triple quadrupole with electrospray tandem mass spectrometry(UPLC-TQ-MS). Then the compounds in processed and dried samples were analyzed by partial least squares discriminant analysis(PLS-DA) and orthogonal partial least squares discriminant analysis(OPLS-DA), and the contribution rates of differential components were evaluated by variable important in projection(VIP). The results indicated that the samples obtained by different processing and drying methods could be distinguished. Albiflorin, gallic acid, 1,2,3,4,6-pentakis-O-galloyl-β-D-glucose, and benzoic acid were the common differential components in boiled Paeoniae Radix Alba. Benzoic acid was the common differential component in peeled Paeoniae Radix Alba. Gallic acid was the common differential component in Paeoniae Radix Alba dried by different methods. The samples could not be distinguished after drying at different temperatures due to the lack of common differential components. This study is expected to provide a reference for the selection of processing and drying methods and the optimization of processing parameters.

Objective:To explore the learning guidance model and its influencing factors of undergraduates in medical schools.Methods:Based on the developmental learning guidance theory, 1 117 undergraduates from two undergraduate medical universities were selected as the research objects, through questionnaire surveys to collect their knowledge learning, curriculum learning and other core variables, and balanced guidance, compound guidance and other outcome variables, using Smart PLS 3.0 to test the validity of the samples.Results:There were statistical significance in the path relationship affecting the core variables and the outcome variables of the learning guidance model, P<0.05, indicating that the influencing factors of the learning guidance model had a significantly positive effect on the learning guidance model construction. Conclusion:Constructing a "balanced-compound-special" learning guidance model suitable for undergraduates' learning activities in medical universities provides a reference for the application and promotion of learning guidance model in medical undergraduate stage.

OBJECTIVE:To study spectrum-effect relationship of 11 different solvent extracts from Trollius chinensis against hypoxia/reoxygenation injury of cardiomyocyte . METHODS :HPLC-MS/MS method was used to establish the fingerprints of 11 different solvent extracts from T. chinensis ,the compounds corresponding to the common peaks were identified by comparing with the substance control and literature information. MTT assay was used to detect the effects of 11 different solvent extracts from T. chinensis on the survival rate of rat myocardial H 9c2 cells injured by hypoxia/reoxygenation. The MDA content ,ROS level in cells and LDH content in the supernatant were detected by ELISA. GRA and PLS method were used to analyze the spectrum-effect relationship between the compounds corresponding to common peak and anti-hypoxia/reoxygenation injury of cardiomyocyte (drug effect). RESULTS :There were 22 common peaks in 11 different solvent extracts from T. chinensis ,and 22 compounds were identified. Compared with hypoxia/reoxygenation injury group ,survival rate of hypoxia/reoxygenation injury+S 1-S6,S9 and S 10 groups were increased significantly ,while MDA content ,ROS level and LDH content were decreased significantly (P<0.05); ROS level and LDH content of hypoxia/reoxygenation injury+S 8 group w ere decreased significantly (P<0.05). The r of GRA analysis of 22 compounds with drug effects were all higher than 0.8. OBJECTIVE: To discriminate and quantify of Gleditsia japonica Miq. thorn (SZJ) and Gleditsia microphylla Gordon ex Y. T. Lee thorn (YZJ) in the Gleditsia sinensis Lam thorn (GST). METHODS: Fourier transform near-infrared spectroscopy (FT-NIR) combined with linear discriminate analysis (LDA), support vector machine (SVM), as while as back propagation neural network (BPNN) algorithms were applied to construct the identification models. The SZJ and YZJ content in adulterated GST were determined by partial least squares regression (PLSR). RESULTS: The SVM models performance best compared with LDA and BP-NN models for it could reach 100% accuracy in training and validation set for identifying authentic GST and GST adulterated with SZJ and YZJ based on the spectral region of 5 000-4 200 cm-1 combined with SG+VN processing. The rp, RMSEP (the root mean standard error of prediction) and bias for the prediction by PLS regression model were 0.993, 2.919% and -0.330 3 for SZJ, 0.995, 2.57% and 0.364 9 for YZJ, respectively. CONCLUSION: Our results suggest that the combination of NIR spectroscopy and chemometric methods offers a simple, fast and reliable method for classifification and quantifification of SZJ and YZJ adulterants in the GST.

OBJECTIVE:To improve the willin gness of chinese pharmaceutical enterprises to participate in drug patient assistance programs ,improve the level of drug use and access to innovative drugs in China and fully evaporate the supplementary role of the medical security system. METHODS :Through grounded theoretical analysis of the interview results ,the theoretical framework of the factors affecting the implementation of drug patient assistance programs by pharmaceutical companies was proposed. The above theoretical framework was empirically studied through questionnaire survey on the personnel who understood or participated in drug patient assistance program in Chinese pharmaceutical enterprises. Based on partial least squares structural equation model ,Smart PLS 3.0 software was used for data processing to analyze the influence of different factors on the willingness of pharmaceutical companies to participate in drug patient assistance programs ,and the relevant suggestions were put forward. RESULTS & CONCLUSIONS :Economic benefits ,market competition ,social benefits and policy environment have significants impacts on the willingness of pharmaceutical enterprises to implement drug patient assistance programs. The path coefficients were 0.244 (P<0.01),0.236 (P<0.01),0.235 (P<0.05),0.228 (P<0.01). Human resource efficiency and cooperation unit have no significant effects on the willingness of pharmaceutical enterprises to implement drug patient assistance programs(P>0.05). Economic benefits are in line with the nature of enterprises ’profit-making and have the most close influence. Therefore,the pricing strategy of drugs and the tax policies of government departments are very important for enterprises. Participation in market competition is also an important factor for enterprises to participate in patient assistance programs. The earlier the implementation of patient assistance ,the more opportunities of relevant drugs will be to seize more markets. The social benefits manifest as reducing the economic burden of patients ,improving the quality of life ,and promoting the social and economic development. At the same time ,enterprises have set up good social image and forms a word-of-mouth effect. The policy environment generally includes the legal environment and medical insurance policies. The legal provisions restrict the donations of pharmaceutical enterprises to a certain extent ,but corresponding regulatory measures are still essential. Medical insurance policies are closely related to the establishment and adjustment of patient assistance programs ,and the connection between medical insurance access and patient assistance programs needs to be further improved. Therefore ,from the perspective of enterprises ,it is recommended to establish a cost and benefit calculation system for drug patient assistance programs ,and broaden the publicity channels and increase the publicity intensity of patient assistance programs. From the perspective of the government ,it is recommended that tax incentives for donations should be increased,the patient assistance supervision system should be improved ,the access and adjustment rules for the drugs of patient assistance programs in medical insurance list should be clarified ,so as to promote pharmaceutical enterprises to actively participate in patient assistance programs and improve the drug use level and drug accessibility of patients.

Objective: To establish the quality evaluation system based on spectrum-antibacterial effect correlation of Yinhuang Granules in order to detect its raw medicinal materials, extracts, and preparation simultaneously. Methods: Firstly, the spectrum- antibacterial effect correlation quality evaluation system of Yinhuang Granules was established by using Least Squares Support Vector Machine (LS-SVM). Then, by using supervised partial least square-discriminant analysis models (PLS-DA), the antibacterial effect evaluation was judged based on the spectrum-antibacterial effect data of Scutellariae Radix, Lonicerae Japonicae Flos, Scutellaria extract, Lonicerae japonica extract, and Yinhuang Granules. Results: The mathematical model of Yinhuang Granules based on spectrum-antibacterial effect correlation was established; The average relative error of the prediction results was less than 5%, and the antibacterial rate of Scutellariae Radix, Scutellaria Radix extract, Lonicerae Japonicae Flos, Lonicerae japonica extract was greater than 43%, 5.5%, 11%, 37% calculated by their mathematical model, which can ensure the antibacterial effect was greater than 11% (correct rate was 87%). Conclusion: The quality evaluation system can realize the quality control of the key link of the production of traditional Chinese medicine, the evaluation results are more scientific, comprehensive, and accurate.

Objective: To establish a method for the identification of the Periplaneta americana and other insectivorous herbs (Scorpio and Hirudo) based on infrared spectroscopy combined with chemometrics, and to provide a basis for the identification of the P. americana. Methods: Fourier transform infrared spectroscopy was used to collect the infrared spectrum data of three kinds of insect medicine powders. After the second order derivation of the obtained spectral data, the ordinate verticalization method and standardization method were used to optimize the spectrum. The spectral data was further analyzed by chemometrics, such as hierar chicalcluster analysis (HCA), principal component analysis (PCA) and partial least squares discriminant analysis (PLS-DA). Results: There were differences in the infrared fingerprints of the P. americanas, Scorpios, Leeches. The absorption peaks of the P. americanas at 1 711, 1 410, and 712 cm-1 were obvious. The absorption peaks of the Scorpios at 1 753, 1 400, 1 168, and 717 cm-1 were obvious. The absorption peaks of the Leeches at 1 558, 1 457, 1 400, and 669 cm-1 were obvious, And the leeches have no absorption peak at 1 753-1 711 cm-1. The peak shape of the three insectivorous herbs was significantly different at 1 800-1 700 cm-1. In the second derivative spectrum, the positions of the main peaks are the same, but the intensity of the common peaks is different. Using the HCA analysis method, it was found that the three insectivorous herbs could be quickly distinguished. The PCA and PLS-DA analysis methods were used to find that the three insectivorous herbs were distributed in different regions. Conclusion: Infrared spectroscopy combined with chemometrics can easily and quickly identify the P. americanas and other insectivorous herbs (Scorpios and Leeches), and provide reference for the quality control and evaluation of P. americanas.

RESUMEN

Objective: To compare pharmacodynamic difference of Ribes diacanthum (RDP) and Ribes mandshuricum (RMK) treatment on renal fibrosis in vivo and in vitro. Methods: Both of TGFβ1-induced HK-2 cell fibrosis model and UUO-induced kidney fibrosis mice model were used in the present study. The cell morphology, ratio of cell length to width, renal histopathology, protein expressions of α-SMA and E-cadherin in kidney tissues were evaluated through biological and pharmacological methods and technologies, including Western blot, immunohistochemistry, HE staining, Masson staining and so on. In addition, partial least squares-discriminant analysis (PLS-DA) was applied to analyze the renal histopathological score as well. Results: RDP (1.5, 5, 15 μg/mL) and RMK (3, 10 μg/mL) effectively improved morphological changes and reduced the ratio of cell length to width in TGFβ1-induced HK-2 cell fibrosis; Moreover, RDP (40 mg/kg) and RMK (80 mg/kg) remarkably decreased the expression of α-SMA and increased the expression of E-cadherin in UUO mice model. The degree of pathological damage and fibrosis were also alleviated in both groups. PLS-DA analysis showed no significant difference in anti-fibrotic effects between RDP and RMK treatment. Conclusion: Both RDP and RMK have anti-fibrosis effects on TGFβ1-induced HK-2 cell fibrosis model and UUO-induced kidney fibrosis mice model, and there is no significant difference between these two herbs.

Objective:To compare the effects of different processing techniques on the chemical constituents of Aurantii Fructus for screening the dominant decoction pieces. Method:UPLC-Q/TOF-MS was used to detect the chemical constituents of Aurantii Fructus, chromatography separation was achieved on an ACQUITY UPLC BEH C18 column (2.1 mm×100 mm, 1.7 μm), and gradient elution was performed with 0.1% formic acid aqueous solution (A)-0.1% formic acid acetonitrile solution (B) as mobile phase (0-10 min, 5%-35%B; 10-18 min, 35%-75%B; 18-21 min, 75%-100%B; 21-24 min, 100%B; 24-24.1 min, 100%-5%B; 24.1-28 min, 5%B). Data acquisition was carried out in electrospray ionization (ESI) under the positive ion mode, the scanning range was m/z 50-1 200. The chemical constituents in methanol extract of Aurantii Fructus were identified according to reference substance, relative molecular weight, mass spectrometric cleavage rule and literature information. SIMCA-P 13.0 software was used to establish principal component analysis (PCA) model and partial least squares-discriminant analysis (PLS-DA) model of Aurantii Fructus processed products, PCA score plot, PLS-DA loading plot and variable importance in the protection (VIP) values were obtained to screen the material basis for the main differences before and after processing of Aurantii Fructus. Result:A total of 54 chemical components were identified by UPLC-Q/TOF-MS. PCA indicated that there were significant differences among different groups of Aurantii Fructus processed by different methods. A total of 14 chemical components with VIP value >1 were screened by PLS-DA as the main chemical markers for the differences before and after processing, including hesperidin, poncirin, narirutin, etc. The comprehensive weighted score showed that the content of effective components in Aurantii Fructus processed with honey bran was the highest. Conclusion:The contents of chemical constituents in Aurantii Fructus before and after processing are significantly changed. Flavonoids are the most important compounds to distinguish different processed products of Aurantii Fructus. Aurantii Fructus processed with honey bran is the dominant variety.

@#Women are likely to leave the job sector as a result of the crisis between their commitments to the career with the household interest. In response to this issue, this study aims to build a career challenge model that caters to the demand among women in this century. Hence, this study has identified the key factors to the challenges faced by female engineers in pursuing their career as an engineer through the Delphi Modified Technique. The result shows this study looking into the relationship between four independent constructs namely, life balance, childcare, leaves and gender discrimination. Meanwhile, the dependent construct of this study is career challenges faced by women. The scope of the study comprises female engineers with families and 211 respondents were selected to answer the questionnaire. The data obtained were analysed using the PLS-SEM 2.0 software via the algorithm, bootstrapping and blindfolding method. The construction process of this model involves two tests including the construction of the measurement model and the structure model. Testing the measurement model involves internal consistency namely (a) convergent validity and (b) discriminant validity in which these two validities have six analyses; (i) external loading, (ii) composite reliability, (iii) average variance extracted (AVE), (iv) Fornell-Larcker, (v) cross loading, and (vi) Heterotrait-Monotrait Ratio (HTMT). Meanwhile, the structural model testing involves the analysis of (i) Multicollinearity (Inner VIF), (ii) Path Coefficient, (iii) R square (R2), (iv) size effect (f2), and (v) Predictive Relevance (Q2). The findings indicate that gender discrimination and life balance have significant relationships in influencing career challenges. Hence, this model is expected to contribute to the literature of Human Resource Management.

OBJECTIVE:To establish the method for content dete rmination of polyphyllin Ⅱ,Ⅵ,Ⅶ in Ypsilandra thibetica , and to compare the differences of 3 saponins in different parts of Y. thibetica from different producing areas. METHODS :HPLC method was adopted to determine the contents of polyphyllin Ⅱ,Ⅵ,Ⅶ in whole grass part and underground part of Y. thibetica from 10 producing areas. The determination was performed on Kromasil C 18column with mobile phase consisted of acetonirile-water (gradient elution )at the flow rate of 1.0 mL/min. The column temperature was 35 ℃,and detection wavelength was set at 203 nm;sample size was 10 μL. With the contents of 3 saponins as the index ,20 batches of Y. thibetica were analyzed by cluster analysis and PLS-DA analysis ;the aggregation of samples was analyzed and determined the primary difference components. RESULTS:The linear range of polyphyllin Ⅱ,polyphyllin Ⅵ and polyphyllin Ⅶ were 0.051-2.04,0.007-0.28,0.168-6.72 μg, respectively(r≥0.999 5);the detection limits were 1.92,1.75,1.87 ng,respectively;and the quantitative limits were 6.40,5.87, 6.23 ng,respectively;RSD of precision ,reproducibility and stability tests (24 h)were all lower than 2%(n=6);the average recovery rates were 99.29%,101.38% and 99.64%,with RSDs of 1.17%,2.64%,0.75%(n=6),respectively. The content of polyphyllin Ⅱ was 0.615-1.875 mg/g,that of polyphyllin Ⅵ was 0-0.095 mg/g,and that of polyphyllin Ⅶ was 3.158-12.354 mg/g. Cluster analysis showed that 20 batches of samples were clustered into two groups ,batch S 9-S12 were clustered in to one group,and the other 16 batches of samples were clustered into another group. PLS-DA analysis showed that 20 batches of samples were divided into 3 areas,batch S 1,S2,S8,S14,S16,S20 were included in area Ⅰ;batch S 9-S12 included in area Ⅱ;and the rest of the samples included in area Ⅲ. The quality of Y. thibetica from different habitats was different ,and there was no difference in the saponin composition between the whole grass and the underground part. Weight ranking found that mail:cscdtcm@126.com polyphyllin Ⅶ was the main difference component in Y. thibetica,and the content of polyphyllin Ⅶ in Y. thibetica from Pengzhou city and Dayi county was the highest. CONCLUSIONS :The established method is simple ,convenient and sensitive. It can be used for the content determination of 3 saponins in Y. thibetica . The content of active components is higher and the quality is better in Y. thibetica from Pengzhou city and Dayi county.

The grading and quality analysis methods for different commercial Glycyrrhiza Polygalae Radix slices were established. The qualities of different grade samples were analyzed and compared, in order to provide useful information for the formulation of the grading standards of Glycyrrhiza Polygalae Radix slices. A total of 34 batches of Glycyrrhiza Polygalae Radix slice samples collected from 12 companies were divided into two grades: first-grade (diameter ≥ 3.0 mm) and second-grade (diameter < 3.0 mm). Thin-layer chromatography (TLC), multi-component content determination and fingerprint analysis were used to analyze the qualities of different grades of Glycyrrhiza Polygalae Radix slices, and the fingerprints were statistically analyzed using partial least squares-discriminant analysis (PLS-DA) and orthogonal partial least squares-discriminant analysis (OPLS-DA). The results showed that the established TLC method can simultaneously identify three major types of components, including sugar esters, xanthones, and saponins in Glycyrrhiza Polygalae Radix slices, and has obvious advantage compared to the existing methods for its rich information, low cost, and easy or safe operation. The multi-component determination showed that the contents of three index components (polygalaxanthone Ⅲ, 3,6'-disinapoyl sucrose and tenuifolin) in the first-grade products of Glycyrrhiza Polygalae Radix slices were lower than those in the second-grade products. The results of PLS-DA and OPLS-DA indicated significant differences were observed between the first-grade and second-grade products, with sibiricose A5, sibiricose A6, polygalaxanthone Ⅲ, 3,6'-disinapoyl sucrose and tenuifoliside A being identifies as the major differentiate markers.
