RESUMEN

Objective To optimize the best macroporous resin through static adsorption and desorption and establish enrichment process of juglone-constituents from Qinglongyi with the transfer rate of juglone constituents as the index. Methods The static adsorption and desorption of four different resins with different polarity to juglone-constituents from Qinglongyi were compared. AB8 resin was selected as the best resin. Results The craft parameters of the best resin were optimized, and the results were as follows: initial concentration was 0.0760 g/ml, the pH was 2.0, the flow rate of adsorption was 3BV/h, the elution concentration of ethanol was 70%, the elution volume was 3BV,the desorption flow rate was 2BV/h, and the ratio of diameter to height was 1:10. After the process of optimization, the content of juglone- constituents from Qinglongyi increased from 1.026% to 11.08% and the transfer rate was 72.08%. Conclusion The AB8 macroporous resin could be used to enrich and purify juglone-constituents from the Qinglongyi.

OBJECTIVE:To study the effects of different drying methods on the quality of North Qinglongyi,thus optimize the best drying way of North Qinglongyi. METHODS:North Qinglongyi was processed using five kinds of drying methods includ-ing drying in the shade,drying in the sun,oven drying,microwave drying,freeze drying and oven drying under different tempera-ture (40,50,60 ℃). The indicators were investigated,such as drying time,recovery rate of water,water content,alcohol ex-tract,content of active ingredients walnut quinone and juglone,in vitro anti-tumor activity of methanol extract to human gastric ade-nocarcinoma BGC823 cells and human lung cancer A549 cells. The effects of different drying methods on the quality of North Qinglongyi were analyzed comparatively. RESULTS:Among 5 kinds of drying method,microwave drying time was the shortest, and the time of drying in the shade was the longest;water content of sample with freeze drying was the highest,and that with mi-crowave drying was the lowest;the content of alcohol extract has little difference;the content of walnut quinone of sample with freeze drying was the highest,and that with microwave drying was the lowest;the content of juglone of sample dried at 40 ℃ was the highest,and that of freeze drying was the lowest;anti-tumor experiment in vitro showed that inhibition rate of sample with dif-ferent drying methods on the growth of 2 kinds of cells was in descending order of freeze drying>40 ℃ drying>drying in the shade,and the inhibition rate decreased with the increase of drying temperature. CONCLUSIONS:Different drying methods have obvious influence on the quality of North Qinglongyi. 40 ℃ drying method is the best for North Qinglongyi drying from the com-prehensive analysis including cost,the contents of effective composition,anti-tumor activity and practice.

Objective: To study the effect of Qinglongyi polysaccharides (QP) in the exocarp of Juglans mandshurica on the complex mobility of erythrocytes in S180 mice. Methods: Erythrocytes were collected and prepared into suspensions, and the complex mobility of cells was measured using high performance capillary electrophoresis (HPCE). Optimized experimental conditions were as follows: 50 cm × 75 μm capillary, buffer for electrophoresis; phosphate solution containing hydroxypropylmethyl cellulose (0.1 mol/L, pH 7.4), injection pressure 3.448 kPa, injection time 10 s, separation voltage 20 kV, and column temperature 25 °C. Results: The migration time of erythrocytes in S180 mice was longer than that in normal mice, which was 18.09 min for the model group and 12.11 min for the control group, and the complex mobility of erythrocytes in S180 mice was lower than that in normal mice, which was 0.92 × 10-4 cm2/(Vs) for the model group and 1.38 × 10-4 cm2/(Vs) for the control group. It was also found that S180 mice treated by QP could shorten the migration time and increase the complex mobility of erythrocytes. Conclusion: QP could improve the complex mobility of erythrocytes in S180 mice, and HPCE could be used as a powerful tool for determining the physiological state and functions of erythrocytes. © 2013 Tianjin Press of Chinese Herbal Medicines.

RESUMEN

Object To study the effects of the different Qinglongyi extracts (QE) on cell membrane fluidity of S 180 and H 22 tumor-bearing mice. Methods The tumor models of S 180 and H 22 were used. The oxter of the normal mice were injected S 180 tumor cell liquid and the abdomen of those were injected H 22 tumor cell liquid. Tumor-bearing mice were given QE by ip cold alcohol extract (191.37, 95.68, 47.84 mg/kg), hot alcohol extract (121.17, 60.58, 30.29 mg/kg), and cytoxan (20 mg/kg as positive control), normal saline (as negative control) for 7 d then the mice were killed, and the tumor cell membrane liquids were made according to the methods. The membrane fluidity was studied with fluorescence polarization (P) and microviscosity (?) with 1,6-dipheyl-1,3,5-hexatriene (DPH) as a probe. Results The QE by both cold and hot alcohol can make the membrane fluidity of S 180 tumor cell improve, the membrane fluidity of H 22 tumor cell decline. RESUMEN

Object To observe antitumor effects of different extract fractions from Qinglongyi (DEQ). Methods S180-mice and H22-mice were given by ip as model. Antitumor effects of DEQ on S180-mice were observed, and also the effect on indexes of thymus and spleen, sialic acid (SA) content in cancer cell membrane and erythrocyte membrane in S180-mice and H22-mice. Results The two extract fractions by cold and hot alcohol have obvious antitumor effects in a dose-effect manner to S180-mice, and can prolong H22-mice life. The two extract fractions from Qinlongyi can significantly decrease SA in cancer cell membrane and increase SA in erythrocyte membrane. Conclusion The two extract fractions from Qinglongyi have antitumor effects.
