Abstract In soybean breeding program, continuous selection pressure on traits response to yield created a genetic bottleneck for improvements of soybean through hybridization breeding technique. Therefore an initiative was taken to developed high yielding soybean variety applying mutation breeding techniques at Plant Breeding Division, Bangladesh Institute of Nuclear Agriculture (BINA), Bangladesh. Locally available popular cultivar BARI Soybean-5 was used as a parent material and subjected to five different doses of Gamma ray using Co60. In respect to seed yield and yield attributing characters, twelve true breed mutants were selected from M4 generation. High values of heritability and genetic advance with high genotypic coefficient of variance (GCV) for plant height, branch number and pod number were considered as favorable attributes for soybean improvement that ensure expected yield. The mutant SBM-18 obtained from 250Gy provided stable yield performance at diversified environments. It provided maximum seed yield of 3056 kg ha-1 with highest number of pods plant-1 (56). The National Seed Board of Bangladesh (NSB) eventually approved SBM-18 and registered it as a new soybean variety named Binasoybean-5 for large-scale planting because of its superior stability in various agro-ecological zones and consistent yield performance.


Resumo No programa de melhoramento da soja, a pressão pela seleção contínua para a resposta das características de rendimento criou um gargalo genético para melhorias da soja por meio da técnica de melhoramento por hibridação. Portanto, foi desenvolvida uma variedade de soja de alto rendimento, aplicando técnicas de reprodução por mutação, na Divisão de Melhoramento de Plantas, no Instituto de Agricultura Nuclear de Bangladesh (BINA), em Bangladesh. A cultivar popular BARI Soybean-5, disponível localmente, foi usada como material original e submetida a cinco doses diferentes de raios gama usando Co60. Em relação ao rendimento de sementes e às características de atribuição de rendimento, 12 mutantes genuínos foram selecionados a partir da geração M4. Altos valores de herdabilidade e avanço genético com alto coeficiente de variância genotípico (GCV) para altura da planta, número de ramos e número de vagens foram considerados atributos favoráveis ao melhoramento da soja, garantindo, assim, a produtividade esperada. O mutante SBM-18, obtido a partir de 250Gy, proporcionou desempenho de rendimento estável em ambientes diversificados e produtividade máxima de sementes de 3.056 kg ha-1 com o maior número de vagens planta-1 (56). O Conselho Nacional de Sementes de Bangladesh (NSB) finalmente aprovou o SBM-18 e o registrou como uma nova variedade de soja, chamada Binasoybean-5, para plantio em larga escala por causa de sua estabilidade superior em várias zonas agroecológicas e desempenho de rendimento consistente.

OBJECTIVE To investigate the effects of soybean isoflavones (SI) on the reproductive development of young mice. METHODS C57BL/6 young mice were randomly divided into control group, SI low-dose and high-dose groups (10, 100 mg/kg), with 10 mice in each group (half male and half female). The young mice in each group were given corresponding liquid intragastrically, once a day, for 2 consecutive weeks. After the last administration, the percentage of body weight increase was calculated; serum estradiol and testosterone levels, malondialdehyde (MDA) content, total antioxidant capacity (T-AOC), superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) activities in the reproductive organs of the young mice were determined. The histopathological changes in the reproductive organs were observed. The cell apoptosis of reproductive organs was detected. RESULTS Compared with the control group, the percentage of body weight increase in female mice was increased significantly in the SI high-dose group, while that of male mice was decreased significantly (P<0.05 or P<0.01). Cystic follicles could be seen in the ovarian tissue in SI groups, a loose arrangement of spermatocytes could be seen in the testicular tissue, and partial epithelial cell shedding could be seen in epididymal tissue. The serum level of testosterone in female young mice and the serum levels of testosterone and estradiol in male young mice in SI groups, GSH-Px activity in the ovarian tissue of female young mice in the SI low-dose group, T-AOC activities in the ovarian tissue of female young mice in SI groups as well as the apoptotic rates of cells in testicular and epididymal tissue of male young mice in SI groups were increased significantly (P<0.05 or P< 0.01); the serum level of estradiol in female young mice in SI groups, SOD activity in the ovarian tissue of female young mice in the SI high-dose group, and MDA contents in the ovarian tissue of female young mice in SI groups as well as the apoptotic rates of cells in ovarian tissue of female mice in SI groups were decreased significantly (P<0.05 or P<0.01). CONCLUSIONS SI can enhance the antioxidant stress capacity of ovarian tissue in female young mice and reduce their oxidative stress damage, but it has certain toxicity to reproductive organs in male mice.

The soybean looper (SBL), Chrysodeixis includens (Walker, [1858]) (Lepidoptera: Noctuidae), is a soybean and cotton pest in South America countries. Field-evolved resistance of SBL to inhibitors of chitin biosynthesis has been reported in Brazil; however, this mode of action is still widely used against SBL. On this basis, we conducted laboratory bioassays to investigate if adjuvants (Nimbus®, TA 35®, Break-Thru® S 240, and Rizospray Extremo®) added to the teflubenzuron spray increase the mortality of SBL strains (resistant, heterozygous, and susceptible to chitin biosynthesis inhibitors). Using chromatography analysis, we also evaluated the amount of teflubenzuron on soybean leaves when applied alone or in combination with adjuvants. In laboratory bioassays, the biological activity of teflubenzuron increased against the susceptible SBL strain when adjuvants were added. In contrast, no relevant effects of adjuvants added to the teflubenzuron spray against heterozygous and resistant SBL larvae were detected. In leaf bioassays, even leaves from the upper third part of the plants containing a significantly higher amount of teflubenzuron (3.4 mg/kg vs 1.7 and 0.6 mg/kg); the mortality of SBL strains was similar when teflubenzuron was applied alone or in mixture with adjuvants. Our findings indicated that adjuvants added to teflubenzuron spray do not provide a substantial increase in the mortality of SBL strains resistant to chitin biosynthesis inhibitors. Therefore, it is necessary to reduce the use of this mode-ofaction insecticide against SBL and to give preference to other insecticides or control tactic.


A lagarta falsa-medideira, Chrysodeixis includens (Walker, [1858]) (Lepidoptera: Noctuidae), é uma praga da soja e do algodão nos países da América do Sul. A resistência de C. includens a inibidores da biossíntese de quitina tem sido relatada no Brasil. Entretanto, esse modo de ação ainda é amplamente utilizado para controle de C. includens. Com base nisso, conduzimos bioensaios em laboratório para investigar se adjuvantes (Nimbus®, TA 35®, Break-Thru® S 240 e Rizospray Extremo®) adicionados à calda inseticida de teflubenzuron aumentam a mortalidade de linhagens de C. includens (resistentes, heterozigotos e suscetíveis a inibidores da biossíntese de quitina). Usando análise cromatográfica, também avaliamos a quantidade de teflubenzuron em folhas de soja quando aplicado isolado ou em combinação com adjuvantes. Em bioensaios de laboratório, a atividade biológica do teflubenzuron aumentou para a linhagem suscetível quando os adjuvantes foram adicionados à calda inseticida. Em contraste, nenhum efeito relevante de adjuvantes adicionados ao teflubenzuron foi detectado para os heterozigotos e resistentes. Em bioensaios de folhas, mesmo naquelas do terço superior das plantas, as quais apresentaram uma maior deposição de teflubenzuron (3,4 mg/kg vs 1,7 e 0,6 mg/kg); a mortalidade das linhagens de C. includens foi semelhante quando o teflubenzuron foi aplicado isolado ou com adjuvantes. Nossos resultados indicam que os adjuvantes adicionados ao teflubenzuron não fornecem um aumento substancial na mortalidade de linhagens de C. includens resistentes aos inibidores da biossíntese de quitina. Portanto, é necessário reduzir o uso desse modo de ação para o manejo de C. includens e dar preferência a outros inseticidas ou tática de controle.

Objective To explore the nutritional efficacy of compound protein powder formulations from different sources. Methods Three groups of compound protein powder formulations were obtained through scientific blending using soy protein, whey protein and yeast protein as raw materials. The effects of the compound protein powders on nitrogen metabolism, serum biochemical indicators, and pathological changes of liver tissue and epididymal fat in rats were evaluated. Results Compared with the control (casein), the net protein utilization, biological evaluation, and protein efficacy ratio of the compound protein powders in rats were significantly improved, and the changes in these indicators in the formula with the highest whey protein content were most significant among all three formulas. The compound protein powders effectively increased the levels of albumin and globulin, while decreased the content of total cholesterol, indicating beneficial effects on improving immunity and controlling lipid metabolism, with the formula group 2 being the most effective among all three groups. The pathological examination showed that the three groups of protein powder did not have adverse effects on liver tissue and epididymal fat. Conclusion The present study demonstrates that the compound protein powder formulation has nutritional value, which suggests a potential of the application of the compound protein powder formulation in the elderly, and people with special nutritional needs, such as sports people.

Galactinol synthase (GolS) genes play important roles in plant response to abiotic stress. In this research, the plant expression vector of soybean GmGolS2-2 gene was constructed and transformed into tobacco to study the drought tolerance of transgenic tobacco. A GmGolS2-2 gene with 975 bp coding sequence was cloned from soybean leaves by reverse transcription-polymerase chain reaction (RT-PCR). GmGolS2-2 was linked to the plant expression vector pRI101 by restriction enzyme sites Nde Ⅰ and EcoR Ⅰ, and transformed into tobacco by leaf disc method. Genomic DNA PCR and real-time PCR showed that three GmGolS2-2 transgenic tobacco plants were obtained. The growth status of GmGolS2-2 transgenic tobacco under drought stress was better than that of wild-type tobacco. After drought stress treatment, the electrolyte leakage and malondialdehyde content of transgenic tobacco were lower than those of wild-type tobacco, but the proline content and soluble sugar content were higher than those of wild-type tobacco. The results of real-time PCR showed that the heterologous expression of GmGolS2-2 increased the expression of stress-related genes NtERD10C and NtAQP1 in transgenic tobacco. The above results indicated that GmGolS2-2 improved drought resistance of transgenic tobacco.

OBJECTIVE@#To explore the mechanism that mediates the effect of soybean isoflavones (SI) against cerebral ischemia/reperfusion (I/R) injury in light of the regulation of regional cerebral blood flow (rCBF), ferroptosis, inflammatory response and blood-brain barrier (BBB) permeability.@*METHODS@#A total of 120 male SD rats were equally randomized into sham-operated group (Sham group), cerebral I/R injury group and SI pretreatment group (SI group). Focal cerebral I/R injury was induced in the latter two groups using a modified monofilament occlusion technique, and the intraoperative changes of real-time cerebral cortex blood flow were monitored using a laser Doppler flowmeter (LDF). The postoperative changes of cerebral pathological morphology and the ultrastructure of the neurons and the BBB were observed with optical and transmission electron microscopy. The neurological deficits of the rats was assessed, and the severities of cerebral infarction, brain edema and BBB disruption were quantified. The contents of Fe2+, GSH, MDA and MPO in the ischemic penumbra were determined with spectrophotometric tests. Serum levels of TNF-α and IL-1βwere analyzed using ELISA, and the expressions of GPX4, MMP-9 and occludin around the lesion were detected with Western blotting and immunohistochemistry.@*RESULTS@#The rCBF was sharply reduced in the rats in I/R group and SI group after successful insertion of the monofilament. Compared with those in Sham group, the rats in I/R group showed significantly increased neurological deficit scores, cerebral infarction volume, brain water content and Evans blue permeability (P < 0.01), decreased Fe2+ level, increased MDA level, decreased GSH content and GPX4 expression (P < 0.01), increased MPO content and serum levels of TNF-α and IL-1β (P < 0.01), increased MMP-9 expression and lowered occludin expression (P < 0.01). All these changes were significantly ameliorated in rats pretreated with IS prior to I/R injury (P < 0.05 or 0.01).@*CONCLUSION@#SI preconditioning reduces cerebral I/R injury in rats possibly by improving rCBF, inhibiting ferroptosis and inflammatory response and protecting the BBB.

Organic fertilizers increase soil aeration, water and nutrient holding capacity, improve seed germination and encourage the production of thicker roots, increase the yield and quality of the crop. This study was aimed to determine the effects of organic fertilizer (vermicompost, sheep manure, cattle manure, poultry manure and inorganic fertilizer as control) applications on the root and plant development of soybean during two growth stages, namely third trifoliate stage (V3) and beginning of flowering of the plants (R1) in the pot condition. The experiment was established with a two-factorial randomized plot design with four replications. The fertilizer applications had a significant effect (p<0.01) on the V3 and R1 growth stages of soybean. The highest plant height (36.78 cm) and root length (41.18 cm) were achieved with vermicompost, and the lowest plant height (21.73 cm) was recorded with inorganic fertilizer. The number of nodules was highest for vermicompost and cattle manure applications (17.16 plant-1 and 16.76 plant-1, respectively), and lowest for poultry manure (0.22 plant-1). In the R1 growth stage, poultry manure produced the highest biological fresh and dry weights, root dry weight at 25.08 g plant-1 and 5.67 g plant-1, 3.99 g plant-1 respectively. As a result, it was demonstrated that vermicompost and poultry manure applications promote strong root and plant development due to high organic matter and phosphorus and can be used successfully without inorganic fertilize application in soybean farming.

This research was aimed at obtaining varieties of soybean adaptive to acid soils and to obtain Plant Growth-Promoting Rhizobacteria (PGPR) isolates that can improve the agronomic characteristics of soybean and increase the ultisols fertility. This research was conducted in two-stages research on Sampali Village, Percut Sei Tuan sub-District, Deli Serdang District, North Sumatra Province, Indonesia from August 2019 until March 2020. The first stage (adaptive varieties screening) using the non-factorial Randomized Block Design (RBD) with the varieties of Argomulyo, Wilis, Kaba, Dena-1, Devon-1, Dega-1, Demas-1, Burangrang, Detam-1, and Kipas Merah. The second stage (application of PGPR isolates singly and in combinations) using the factorial RBD, the first factor of applicative single and the combination of PGPR isolates, the second factor of adaptive varieties including Detam-1 and Wilis. Data were analyzed with ANOVA and followed by DMRT at P<0.05. The results showed that the Detam-1 and Wilis varieties had significantly higher yield per plant of 14.73 g and 14.54 g, respectively, than other varieties. The applications of a single and combination of PGPR isolates significantly increased the number of branches, stem diameter, plant height, yield per plant, soil pH, organic-C, available-P, and total-N and decreased the soil C/N. The Detam-1 variety had the higher in yield per plant compared to Wilis variety. The isolates combination of Rhizobium leguminosarum+Rhizobium sp2+Bacillus sp+Burkholderia sp for Detam-1 and Wilis varieties can be recommended to support the growth and yield of soybean on ultisols.

A four-week feeding trial on the simultaneous replacement of 0 to 50% of soybean meal and 0 to 100% of soybean oil on the growth performance, carcass composition and profitability was conducted using 45 randomly chosen one-week-old growing Japanese quail. The five experimental diets were; D0 = Diet containing 20% soybean meal and 4% soybean oil (control diet), D1 = Diet, where 12.5% of soybean meal and 25% of soybean oil has been replaced by RSC; T2 = Diet, where 25% of soybean meal and 50% of soybean oil has been replaced by RSC; T3 = Diet, where 37.5% of soybean meal and 75% of soybean oil has been replaced by RSC; T4 = Diet, where 50% of soybean meal and 100% of soybean oil has been replaced by RSC. Results demonstrated that in raw rubber seed the proportion of kernel to hull is 64: 36. Proximate components of the boiled and sundried full fat rubber seed kernel (RS) were, moisture = 96.6%, ME = 5305 kcal / kg DM, CP = 17.6%, EE = 51.05%, CF = 8.5%, NFE = 18.25% and Ash = 4.6%. The body weights of the birds at 35 days of age in all dietary treatments were statistically similar. The feed intake of the quail among different dietary groups was varied significantly (p < 0.05) and there was a tendency of decreasing the feed intake at 25% and above inclusion level of RSC. The body weight gain and FCR were also statistically similar in all dietary groups. The feed cost and total production cost /kg BW of quail was also found to become steadily higher (p> 0.05) at higher RSC inclusion level. The muscle development was found to better at lower (%) RSC level, whereas the organs were enlarged at higher RSC inclusion. Results on the majority of the production parameters, and economics and carcass yield parameters suggested that the simultaneous replacement of maximum 50% soybean oil 25% soybean meal by processed RSC might be suggested, particularly, there will be a crisis in the availability of these two ingredients.


Um ensaio de alimentação de quatro semanas na substituição simultânea de 0 a 50% de farelo de soja e 0 a 100% de óleo de soja no desempenho de crescimento, composição de carcaça e lucratividade foi conduzido usando 45 codornas japonesas em crescimento com uma semana de idade escolhidas aleatoriamente. As cinco dietas experimentais foram: D0 = Dieta contendo 20% de farelo de soja e 4% de óleo de soja (dieta controle); D1 = Dieta, onde 12,5% de farelo de soja e 25% de óleo de soja foram substituídos por RSC; T2 = Dieta, onde 25% do farelo de soja e 50% do óleo de soja foram substituídos por RSC; T3 = Dieta, onde 37,5% do farelo de soja e 75% do óleo de soja foram substituídos por RSC; e T4 = Dieta, onde 50% do farelo de soja e 100% do óleo de soja foram substituídos por RSC. Os resultados demonstraram que em sementes de borracha crua a proporção de kernel para casca é 64: 36. Os componentes próximos do caroço de semente de borracha gordurosa fervida e seca ao sol foram: umidade = 96,6%; EM = 5305 kcal / kg MS; CP = 17,6%; EE = 51,05%; CF = 8,5%; NFE = 18,25%; e Ash = 4,6%. Os pesos corporais das aves aos 35 dias de idade em todos os tratamentos dietéticos foram estatisticamente semelhantes. O consumo de ração das codornas entre os diferentes grupos dietéticos variou significativamente (p < 0,05) e houve uma tendência de diminuição do consumo de ração em 25% e acima do nível de inclusão do RSC. O ganho de peso corporal e a FCR também foram estatisticamente semelhantes em todos os grupos dietéticos. O custo da ração e o custo total de produção / kg de PC de codornas também se tornaram cada vez mais altos (p > 0,05) no nível de inclusão de RSC mais alto. Constatou-se que o desenvolvimento muscular melhorou com o nível de RSC mais baixo (%), enquanto os órgãos aumentaram com a inclusão de RSC mais alta. Os resultados sobre a maioria dos parâmetros de produção e parâmetros econômicos e de rendimento de carcaça sugeriram que a substituição simultânea de no máximo 50% de óleo de soja e 25% de farelo de soja por RSC processado pode ser sugerida, particularmente, haverá uma crise na disponibilidade desses dois ingredientes.

Sumatran fleabane (Conyza sumatrensis [Retz.] E. Walker) can be found in many different agricultural environments and impact different crops, such as soybeans and corn. It is believed that the application of burndown and preemergence herbicides in the off-season are effective in controlling Sumatran fleabane in soybean crops. The objective was to evaluate the effectiveness of burndown and preemergence herbicides in the off-season, with one or two applications, in the control of Sumatran fleabane in soybean cultivation. Five field experiments were conducted in Maripá, state of Paraná (PR), Brazil. The treatments consisted of the application of burndown herbicides in combinations with preemergence ones, with one or two applications. Control of Sumatran fleabane and soybean yield were evaluated. With the set of experiments, it is highlighted that the strategy combining more applications, with different herbicides, burndown and preemergence, is more promising in the control of Sumatran fleabane. When comparing synthetic auxins, dicamba and triclopyr stand out. For sequential application, worse performance was observed for diquat. Combinations between burndown and preemergence herbicides were effective in controlling Sumatran fleabane, for pre sowing application in soybean. With emphasis on managements with sequential applications of saflufenacil with glufosinate or glyphosate. The strategy combining more applications, with different herbicides, burndown and preemergence herbicides, is more promising in the control of Sumatran fleabane.

A four-week feeding trial on the simultaneous replacement of 0 to 50% of soybean meal and 0 to 100% of soybean oil on the growth performance, carcass composition and profitability was conducted using 45 randomly chosen one-week-old growing Japanese quail. Results demonstrated that in raw rubber seed the proportion of kernel to hull is 64: 36. Proximate components of the boiled and sundried full fat rubber seed kernel (RS) were, moisture = 96.6%, ME = 5305 kcal / kg DM, CP = 17.6%, EE = 51.05%, CF = 8.5%, NFE = 18.25% and Ash = 4.6%. The body weights of the birds at 35 days of age in all dietary treatments were statistically similar. The feed intake of the quail among different dietary groups was varied significantly (p 0.05) at higher RSC inclusion level. The muscle development was found to better at lower (%) RSC level, whereas the organs were enlarged at higher RSC inclusion. Results on the majority of the production parameters, and economics and carcass yield parameters suggested that the simultaneous replacement of maximum 50% soybean oil 25% soybean meal by processed RSC might be suggested, particularly, there will be a crisis in the availability of these two ingredients.


Um ensaio de alimentação de quatro semanas na substituição simultânea de 0 a 50% de farelo de soja e 0 a 100% de óleo de soja no desempenho de crescimento, composição de carcaça e lucratividade foi conduzido usando 45 codornas japonesas em crescimento com uma semana de idade escolhidas aleatoriamente. As cinco dietas experimentais foram: D0 = Dieta contendo 20% de farelo de soja e 4% de óleo de soja (dieta controle); D1 = Dieta, onde 12,5% de farelo de soja e 25% de óleo de soja foram substituídos por RSC; T2 = Dieta, onde 25% do farelo de soja e 50% do óleo de soja foram substituídos por RSC; T3 = Dieta, onde 37,5% do farelo de soja e 75% do óleo de soja foram substituídos por RSC; e T4 = Dieta, onde 50% do farelo de soja e 100% do óleo de soja foram substituídos por RSC. Soybean mosaic virus (SMV) is a critical pathogen that reduces the soybean yield and seed quality worldwide, and SMV has a restricted natural host range. In this study, we used sequence-independent amplification (SIA) methods to identify the viruses that may cause the Atractylodes macrocephala Koidz disease. Results revealed that there is SMV in diseased Atractylodes macrocephala leaves, and we named this isolate as SMV-Am. To further characterize the genomic structural and phylogenetic relationships of SMV-Am, genomic dsRNA was extracted, and the genomic sequence was amplified by RT-PCR and RACE. The results of bioinformatics analysis showed that the genomic RNA of SMV-Am is 9587 nucleotides in length, which conforms to the typical characteristics of Potyvirus. According to the sequence alignment of complete nucleotide sequences, SMV-Am showed the highest level of nucleotide homology and amino acid sequences to SMV-Liaoning, 96. 57% and 98. 86%, respectively. In addition, phylogenetic analysis based on the nucleotide sequences of other SMV isolates of SMV-Am revealed that SMV-Am was most closely related to SMV-Liaoning. Then, the SMV-Am protein was further analyzed by I-TASSER and PyMOL software, revealing that the key amino acid mutations led to the structural changes of P1, HC-Pro, P3, 6K2, NIa-pro and NIb proteins, with P1 the most obvious. Finally, recombination has been detected at the position of 6560-8950 nucleotides, the main parent is the isolate SMV-XFQ012 (accession number KP710875. 1), and the secondary parent is isolate SMV-pCB301-SC15 (accession number MH919386. 1). This study is the first report of SMV in Atractylodes macrocephala Koidz, and these results are expected to provide a scientific basis for the prevention and control of SMV infection on Atractylodes macrocephala Koidz.

Abstract: It has learned that soybean is being affected by a floral disorder known as floral malady where plants fail to develop pod and do notattendfull maturity. For this floral disorder, we present a new methylation-sensitive amplified polymorphism (MSAP) approach for the evaluation of relative quantitative characteristics of non-methylation, hyper-methylation, hemi-methylation, and full methylation status of CCGG sequences, which are recognized by the isoschizomers HpaII and MspI. We applied a technique to analyze alterations in the cytosine methylation a popular Indian soybean (Glycine max L.) genotype, JS-335.The result revealed that in the symptomatic plant, out of 392 MSAP sites, 281 (71.68%), 33 (8.41%),38 (9.69%), and 40(10.20%) found to beun-methylated, hemi-methylated, fully methylated and hyper-methylated, respectively. Whereas, the MSAP profile of asymptomatic plants revealedout of 402MSAP sites, 330 (81.28%) was un-methylated, 22(5.41%) hemi-methylated,29(7.14%) fully methylatedand 25 (6.15%) hypermethylated. In comparison with asymptomatic(18.71%) plant, approximately 10% increased methylation was noted in symptomatic(28.31%) plantprofiles. The increased levels of methylation was recorded in the symptomatic plants about 28.31%and18.71% in asymptomatic. The study showed a higher epigenetic influence on JS-335 genotype of floral malady symptomatic than same genotype of asymptomatic plant. No pod formation in symptomatic plant induce genome wide changes either in promoter or coding region of gene(s) and DNA fragments showing polymorphism related to differences in pattern and extent of methylation associated with floral malady.

Abstract The mortality of the Alabama argillacea and Chrysodeixis includens (Lepidoptera: Noctuidae) larvae caused by the kaolin inert powder and the entomopathogenic fungus Beauveria bassiana were determined under laboratory conditions. Using the caterpillar submersion method, the CG 138 B. bassiana isolate was more pathogenic to A. argillacea than the CG 70, GC 82, ESALQ 634, and ESALQ 645. All five tested isolates caused similar mortality of C. includens. The mortality of first-instar larvae of A. argillacea and C. includens by feeding on leaf-disc impregnated with B. bassiana (CG 138) and kaolin was also determined. Higher A. argillacea mortalities were observed in the B. bassiana (CG 138) treatments, regardless of the presence of kaolin. However, the activity of kaolin + B. bassiana (CG 138) against C. includens was higher than each ingredient alone, indicating an additive action against C. includes larvae. The mortality of A. argillacea and C. includens larvae treated with kaolin + B. bassiana (CG 138) was similar, and the A. argillacea mortality was higher than that of C. includens with kaolin and B. bassiana (GC 138) separated. The treatment kaolin + B. bassiana (CG 138) is promising for the simultaneous management of these two defoliator pests, mainly A. includes. In addition, the monophagous A. argillacea is more susceptible to both kaolin and B. bassiana (GC 138) than the polyphagous C. includens, suggesting that the nutritional ecology plays an important role in the susceptibility of these defoliator species to alternative insecticides.


Resumo A mortalidade de larvas de Alabama argillacea e Chrysodeixis includens (Lepidoptera: Noctuidae), causada pelo pó inerte do caulim e pelo fungo entomopatogênico Beauveria bassiana, foi determinada em condições de laboratório. Usando o método de submersão da lagarta, o isolado CG 138 de B. bassiana foi mais patogênico para A. argillacea que os CG 70, GC 82, ESALQ 634 e ESALQ 645 desse fungo. Os cinco isolados testados causaram mortalidade semelhante de C. includens. A mortalidade de larvas de primeiro ínstar de A. argillacea e C. includens pelo método de alimentação em discos foliares impregnados com B. bassiana (CG 138) e caulim foi, também, determinada. A mortalidade de A. argillacea foi maior nos tratamentos com B. bassiana (CG 138), independentemente da presença do caulim. A atividade do caulim + B. bassiana (CG 138) contra C. includens foi maior que cada ingrediente isolado, indicando ação aditiva contra larvas desse Lepidoptera. A mortalidade de larvas de A. argillacea e C. includens, com caulim + B. bassiana (CG 138), foi semelhante e as de A. argillacea maiores que as de C. includens com caulim e B. bassiana (GC 138) isoladamente. O tratamento caulin + B. bassiana (CG 138) é promissor para o manejo simultâneo dessas duas pragas desfolhadoras, principalmente A. includens. Além disso, a monófaga, A. argilacea, é mais suscetível ao caulim e à B. bassiana (GC 138) que a polífaga, C. includens, sugerindo que a ecologia nutricional desempenha papel importante na suscetibilidade dessas espécies desfolhadoras a inseticidas alternativos.

Abstract Background: Soybean milk by-product (SMBP) is a potential alternative feed ingredient in swine diets due to its high protein content. However, information on energy and nutritional values of SMBP used as swine feed ingredient is limited. Objective: To estimate energy values and protein digestibility of SMBP in pigs based on in vitro assays. Methods: Four SMBP samples were obtained from 3 soybean milk-producing facilities. In vitro total tract disappearance (IVTTD) and in vitro ileal disappearance (IVID) of dry matter (DM) in the SMBP samples were determined. In vitro ileal disappearance of crude protein was determined by analyzing crude protein content in undigested residues after determining IVID of DM. Digestible and metabolizable energy of SMBP were estimated using gross energy, IVTTD of DM, and prediction equations. Results: Sample 4 had greater IVTTD of DM than that of sample 3 (97.7 vs. 94.4%, p<0.05), whereas IVID of DM in sample 4 was lower compared with sample 1 (53.5 vs. 65.0%, p<0.05). In vitro ileal disappearance of crude protein in sample 2 was greater than that in sample 1 and 3 (92.6 vs. 90.6 and 90.1%; p<0.05). The estimated metabolizable energy of SMBP ranged from 4,311 to 4,619 kcal/kg as-is basis and the value of sample 3 was the least (p<0.05) among SMBP samples. Conclusion: Energy values and protein digestibility should be determined before using SMBP in swine diets.


Resumen Antecedentes: El subproducto de la leche de soja (SMBP) es un ingrediente alimenticio alternativo con uso potencial en dietas porcinas dado su alto contenido de proteína. Sin embargo, la información sobre sus valores energéticos y nutricionales para alimentación de cerdos es muy limitada. Objetivo: Estimar los valores de energía y la digestibilidad de la proteína del SMBP en cerdos con base en ensayos in vitro. Métodos: Se obtuvieron cuatro muestras de SMBP de tres empresas productoras de leche de soja. Se determinaron la desaparición de tracto total in vitro (IVTTD) y la desaparición ileal in vitro (IVID) de la materia seca (DM) en las muestras de SMBP. La desaparición ileal in vitro de proteína cruda se determinó analizando el contenido de proteína cruda en residuos no digeridos después de determinar la IVID de la DM. La energía digestible y metabolizable de SMBP se estimó utilizando la energía bruta, IVTTD de la DM y ecuaciones de predicción. Resultados: La muestra 4 tuvo una mayor IVTTD de la DM que la muestra 3 (97,7 vs. 94,4%, p<0,05), mientras que la IVID de la DM en la muestra 4 fue menor en comparación con la muestra 1 (53,5 vs. 65,0%, p<0,05). La desaparición ileal in vitro de la proteína cruda en la muestra 2 fue mayor que la de las muestras 1 y 3 (92,6 vs. 90,6 y 90,1%; p<0,05). La energía metabolizable estimada de SMBP varió de 4.311 a 4.619 kcal/kg (en base húmeda) y el valor de la muestra 3 fue el menor (p<0.05) entre las muestras de SMBP. No entanto, as informações sobre os valores energéticos e nutricionais do SMBP usado como ingrediente alimentar para suínos são limitadas. Objetivo: Estimar valores energéticos e digestibilidade protéica do SMBP em suínos com base em ensaios in vitro. Métodos: Foram obtidas quatro amostras de SMBP de três instalações produtores de leite de soja. Foram determinados o desaparecimento total do trato in vitro (IVTTD) e o desaparecimento ileal in vitro (IVID) da matéria seca (DM) nas amostras de SMBP. O desaparecimento ileal in vitro da proteína bruta foi determinado pela análise do conteúdo de proteína bruta em resíduos não digeridos após a determinação da IVID do DM. A energia digerível e metabolizável do SMBP foi estimada usando energia bruta, IVTTD do DM e equações de predição. Resultados: a amostra 4 apresentou maior IVTTD de DM do que a amostra 3 (97,7 vs. 94,4%, p<0,05) enquanto a IVID do DM na amostra 4 foi menor em comparação com a amostra 1 (53,5 vs. 65,0%, p<0,05). O desaparecimento ileal in vitro da proteína bruta na amostra 2 foi superior ao da amostra 1 e 3 (92,6 vs. 90,6 e 90,1%; p<0,05). Objective:To explore the feasibility of replacing wood (or wood chips) with crop residues for culturing <italic>Armillaria gallica</italic> targeting the problems of forest resource destruction and increased cultivation cost caused by the extensive use of wood in <italic>Gastrodia elata</italic> cultivation, so as to reduce the cultivation cost of <italic>G. elata</italic>, promote the effective use of crop residues, and protect forest resources. Method:The growth situation of <italic>A. gallica</italic> in different media was observed, followed by the measurement of its growth rate using streaking method and the determination of total polysaccharide content of <italic>A. gallica</italic> by phenol-concentrated sulfuric acid colorimetric method. In order to further optimize the soybean straw cultivation medium, we carried out a four-factor three-level L<sub>9</sub>(3<sup>4</sup>) orthogonal assay on the ratio of main ingredients, sucrose content, inorganic salt content, and water content. Result:The comparison of growing states of <italic>A. gallica</italic> cultured in different media revealed that <italic>A. gallica</italic> in soybean straw medium began to grow since the fourth day of inoculation, and the mycelium grew well, with the growth rate being 0.352 cm·d<sup>-1</sup>, which was 1.48 times that in birch wood medium. The total polysaccharide content of <italic>A. gallica</italic> cultured in soybean straw medium was the highest, which was 39.260 mg·g<sup>-1</sup>, much higher than 17.028 mg·g<sup>-1</sup> of that cultured in birch wood medium. This demonstrated the obvious advantage of soybean straw medium, whose main ingredients were soybean straw and wheat bran at the ratio of 8:2, with the sucrose and inorganic salt content accounting for 1% and 0.5% of the main ingredients, respectively. When the water content reached 50%, the growth rate of <italic>A. gallica</italic> was maintained at 0.392 cm·d<sup>-1</sup>. Conclusion:This study has provided a basis for utilizing soybean straw instead of wood (or wood chips) as cultivation medium for <italic>A. gallica</italic>, thus better reducing the waste of forest resources and protecting the natural environment in the cultivation of <italic>G. elata</italic>.

In this study, artemether (ARM)-loaded mixed micelles (MM) composed of the sodium glycocholate (SGC) and soybean lecithin (SL) were prepared by film dispersion method. The effects of hydration medium, SL mass ratio and total concentration of excipients on the solubilization of ARM were investigated and the stability of MM was evaluated. Results showed that the particle size distribution of SGC-SL-MM prepared by phosphate buffer solution (PBS, pH 7.4, 0.05 mol·L-1) was uniform, with an average size of 3.58 ± 0.14 nm and the polydispersity index (PDI) value was 0.16 ± 0.04. The solubility of ARM increased significantly from 0.64 ± 0.04 mg·mL-1 to 13.7 ± 0.13 mg·mL-1 along with the concentration of total excipient increasing from 1.0% to 30.0% (w/w). The calculated results of Arrhenius parameter and storage stability showed that the degradation rate constant of ARM in MM was smaller than that in acetonitrile-PBS (pH 7.4) at either 37 ℃ or 60 ℃. The experimental ARM-MM was clear after storing for two months at 25 ℃ and the degradation of ARM was less than 7.0%. In conclusion, the SGC-SL-MM can not only improve the solubility of ARM in aqueous solution, but also improve its chemical stability in aqueous solution at low temperature.

Recently we witness the rising number of genetically modified (GM) soybean (Glycine max) events approved for importing from abroad and developed domestically, so it is urgent to establish a rapid screening protocol that can cover more events with less detection targets and fit the national condition. Additionally, in order to control the detection workload, it is also necessary to construct a multi-targets plasmid (MTP) molecule that can be used as the positive material. In this study, the information of the transgenic elements in 29 GM soybean events was collected and the combinations and frequencies of these elements were analyzed, to establish a novel screening protocol. It includes eight detecting targets, CaMV 35S promoter (P-35S), NOS terminator (T-nos), herbicide tolerance gene pat, E9 terminator (T-E9), insecticidal gene cry1Ac, AHAS promoter (P-AHAS), pin Ⅱ terminator (T-pin Ⅱ), and the event-specific sequence of the transgenic event DP305423, and an endogenous reference gene of soybean Lectin. After validation, the 29 GM soybean events described above can be screened by detection of the nine targets. This is referred to as the “8+1” protocol for GM soybean screening. Then these targeted sequences described in the protocol were simultaneously inserted into a cloning vector to construct the corresponding MTP pDDSC-1910. Finally, we tested whether it could be a positive plasmid. As expected, PCR analysis using pDDSC-1910 as a template showed that specific amplicons were observed with high sensitivity. Therefore, the “8+1” screening protocol for GM soybean was established, and the positive plasmid molecule pDDSC-1910 containing corresponding targets was successfully constructed. These results would facilitate the efficient screening and detection of transgenic soybeans.
