RESUMEN

@#A new method based on rat liver microsomes and chromatographic fingerprint comparison was established to investigate the possible herb-drug interactions between Liuwei Dihuang Pills(LDP)and nifedipine(NIF). LDP and NIF were incubated simultaneously with rat liver microsomes at 37 °C for 60 min in a shaking water bath. The separation was achieved on a C18 column using 0. 1% formic acid solution and acetonitrile as mobile phase with a liner gradient program. The flow rate was set at 1. 0 mL/min. Detection was achieved by UV light at 240 nm. To evaluate the interactions between LDP and NIF, the similarity of the fingerprinting chromatograms before and after co-incubation was calculated by similarity evaluation system for chromatographic fingerprint of TCM. Furthermore, pharmacokinetic experiments in rat suggested that there was no significant difference in the pharmacokinetic parameters of the typical components in LDP.

RESUMEN

Objective The High-performance thin-layer(HPTLC) chromatographic fingerprint of Saikosaponin from Chaihu(roots of Bupleurum chinense DC.) was established and fingerprints similarity of different species of Bupleurum spp.were evaluated.Methods High-performance thin-layer chromatography(HPTLC)were carried out.The chromatographic conditions were as follows:pre-coated HPTLC silica-gel plate as stationary phase,dichloromethane-methanol-ethyl acetate-water(30:40:15:3) as mobile phase(solvent system) and 2 %p-DMBA/10 %Sulfuric acid alcoholic solution as derivatization reagent.The common pattern of HPTLC fingerprints were obtained through’Chromafinger’solution software,and authentication and quality assessment were analyzed by similarity and Principle Component Analysis.Results The common pattern of the roots of Bupleurum chinense DC.consists of 19 characteristic peaks,and higher similarities existed between the roots of Bupleurum chinense DC(Bei Chaihu),B.falcatum(San-dao Chaihu) and B.scorzonerifolium Willd.(Nan Chaihu),but Nan Chaihu contains much lower total saponins than that in Bei Chaihu.The other species of Bupleurum demonstrated their different chemical distribution.The toxic species of B.longiradiatum can be easily differentiated from other spp.by comparison with the HPTLC images.Conclusion The survey showed that the main commodities of’Chaihu’in the domestic market can be attributed to’Bei Chaihu fingerprint-pattern’.The toxic species of B.longiradiatum can be easily differentiated from other spp.by comparison with the HPTLC images.

RESUMEN

Objective To study the standardization of similarity evaluation method of chromatographic fingerprints. Methods HPLC and computer simulation method were used to analyze the content variation pattern of the characteristic compounds, the construction method of standard fingerprints and the statistical meanings of similarity evaluation respectively. Results The content distribution of characteristic compounds should obey normal probability. It is recommended that content information and median algorism should be used to generate reference fingerprints. The confidence factor of similarity coefficient also should be tested. Conclusion The process of compound sampling, establishment of criteria and result test are described and standardized from statistical aspect.

RESUMEN

Objective To study the standardization of similarity evaluation method of chromatographic fingerprints. Methods Computer simulation and HPLC method were used to investigate the characteristics of different similarity evaluation methods and the criteria of characteristic variables selection in chromatographic fingerprints. Results Cosine ratio and correlation coefficient should be the first choice for similarity calculation based on chromatographic peak area. Peak area is recommend to be used as the characteristic variable for chromatographic fingerprints. Conclusion The features of the commonly used chromatographic fingerprint evaluation methods are described and their range of application are defined.

RESUMEN

AIM: To study and evaluate release rules of Zuojin Sustained-Release Tablet(ZJSRT). METHODS: The total quantum statistical moment similarity(TQSMS)was used to evaluate similarity of the chromatographic fingerprints of ZJSRT in vitro dissolution; RESULTS: Compared with the chromatographic fingerprint of the complete dissolution sample,the averages of area under curve of total quantum,mean chromatographic retention time of total quantum and variance of mean chromatographic retention time of total quantum of the ZJSRT's release rules in vitro,from 1 to 12 h,were 2.620?10~8 ?v?sec,74.599 min and 252.481 min~2 respectively,meanwhile their RSD were 62.445%、1.268% and 2.777% coincidently,the similarities range from 0.947 to(0.991,)as average of 0.977 and RSD of 1.491%. CONCLUSION: It is suggested that the TQSMS can evaluate release rules of multicompon and control its quality.

RESUMEN

AIM: To develop and perfect the similarity algorithm on chromatographic fingerprints of traditional Chinese medicine(TCM). METHODS: The experimental and simulated data were used to investigate and verify the characteristics on the new and former methods on similarity evaluation for chromatographic fingerprints method. RESULTS: There are many strong points for new similarity algorithm in practice. CONCLUSION: The new method provides more rationality and practicability in TCM quality control and analysis compare to existing methods.

RESUMEN

AIM:To establish the normalized chromatographic fingerprints(NCFP) of Shegan Antiviral Injectionl(Rhizoma Belamcandae,Flos Lonicerae japonicae,Herba Artemisiae scopariae,Radix Bupleuri,etc.)(SGKBDI) and to evaluate them by using the 25 relatively normalized parameters.METHODS:The chromatographic fingerprints of the eight herbal drugs,two intermediates and preparation of SGI were determined by RP-HPLC.The 11 chromatographic fingerprints were evaluated by 25 relatively normalized index calculated by the software of Digitalized Evaluation System of Super-information Characteristic for Traditional Chinese Medicine Fingerprints 3.0.RESULTS:The best fingerprints were S4 Herba Artemislae Scopariae and S7 Radix Isatidis,the next were S2 Flos Lonicerae、 S5 Radix Bupleuri、 S6 Herba Taraxaci、 S8 Folium Isatidis,S10 intermediat Ⅱ and S11 Sgkbdi.CONCLUSION:The 25 characteristic parameters can be used to objectively,authentically and thoroughly reveal the separation of chromatographic fingerprints and its normalized characteristics of the NCFP of SGKBDI.
