Abstract This in situ study aimed to evaluate the antibacterial and anti-demineralization effects of an experimental orthodontic adhesive containing triazine and niobium phosphate bioglass (TAT) around brackets bonded to enamel surfaces. Sixteen volunteers were selected to use intra-oral devices with six metallic brackets bonded to enamel blocks. The experimental orthodontic adhesives were composed by 75% BisGMA and 25% TEGDMA containing 0% TAT and 20% TAT. Transbond XT adhesive (TXT) was used as a control group. Ten volunteers, mean age of 29 years, were included in the study. The six blocks of each volunteer were detached from the appliance after 7 and 14 days to evaluate mineral loss and bacterial growth including total bacteria, total Streptococci, Streptococci mutans, and Lactobacilli. Statistical analysis was performed using GLM model - univariate analysis of variance for microhardness and 2-way ANOVA for bacterial growth (p<0.05). The 20% TAT adhesive caused no difference between distances from bracket and the sound zone at 10-µm deep after 7 and 14 days. After 14 days, higher mineral loss was shown around brackets at 10- to 30-µm deep for TXT and 0% TAT adhesives compared to 20% TAT. S. mutans growth was inhibited by 20% TAT adhesive at 14 days. Adhesive with 20% TAT showed lower S. mutans and total Streptococci growth than 0% TAT and TXT adhesives. The findings of this study show that the adhesive incorporated by triazine and niobium phosphate bioglass had an anti-demineralization effect while inhibiting S. mutans and total Streptococci growth. The use of this product may inhibit mineral loss of enamel, preventing the formation of white spot lesions.

PURPOSE: This study was performed to evaluate the osteogenic potential of 3mol% yttria-stabilized tetragonal zirconia polycrystals (3Y-TZP) and niobium oxide containing Y-TZPs with specific ratios, new (Y,Nb)-TZPs, namely YN4533 and YN4533/Al20 discs. MATERIALS AND METHODS: 3Y-TZP, YN4533 and YN4533/Al20 discs (15 mm diameter and 1 mm thickness) were prepared and their average surface roughness (Ra) and surface topography were analyzed using 3-D confocal laser microscope (CLSM) and scanning electron microscope (SEM). Mouse pre-osteoblast MC3T3-E1 cells were seeded onto all zirconia discs and evaluated with regard to cell attachment and morphology by (CLSM), cell proliferation by PicoGreen assay, and cell differentiation by Reverse-Transcription PCR and Quantitative Real-Time PCR, and alkaline phosphatase (Alp) staining. RESULTS: The cellular morphology of MC3T3-E1 pre-osteoblasts was more stretched on a smooth surface than on a rough surface, regardless of the material. Cellular proliferation was higher on smooth surfaces, but there were no significant differences between 3Y-TZP, YN4533, and YN4533/Al20. Osteoblast differentiation patterns on YN4533 and YN4533/Al20 were similar to or slightly higher than seen in 3Y-TZP. Although there were no significant differences in bone marker gene expression (alkaline phosphatase and osteocalcin), Alp staining indicated better osteoblast differentiation on YN4533 and YN4533/Al20 compared to 3Y-TZP. CONCLUSION: Based on these results, niobium oxide containing Y-TZPs have comparable osteogenic potential to 3Y-TZP and are expected to be suitable alternative ceramics dental implant materials to titanium for aesthetically important areas.

Nanopartículas (NPs) tem ganhado notoriedade crescente em aplicações biomédicas. Podendo ser constituídas de diversos materiais, NPs tem sido empregadas como agentes de contraste, na liberação direcionada e controlada de fármacos, em terapia para tratamento de câncer, em catálise heterogênea, entre outras aplicações. As nanopartículas magnéticas de óxido de ferro (MNP) destacam-se pela multiplicidade de aplicações, apesar de serem pouco caracterizadas quanto à toxicidade celular. Outras nanopartículas com excelente potencial são as constituídas de óxido de nióbio (NbONPs), as quais merecem atenção especial, pois o Brasil é detentor de 98% das reservas comercialmente viáveis deste elemento. Neste trabalho NPs destes dois metais de transição (ferro e nióbio) foram sintetizadas, almejando entender suas interações com materiais, biomoléculas e meios biológicos. Diversas metodologias foram desenvolvidas e testadas com intuito de otimizar a morfologia e o rendimento da preparação, resultando na escolha de decomposição térmica para MNP e, para NbONPs, escolheu-se a impregnação do óxido de nióbio sobre uma matriz de MNPs recobertas com sílica. No caso das MNPs, procedeu-se ao recobrimento das mesmas com lipídeos zwitteriônicos (Dioleilfosfatidilcolina (DOPC)) e carregados positivamente (Brometo de dioctadecildimetilamônio (DODAB)). Foram inicialmente caracterizadas as suas propriedades em diversos ambientes biológicos para posteriormente realizarmos ensaios de citoxicidade em queratinócitos humanos (HaCaT). Avaliamos também a degradação das NPs em diferentes pH, bem como, a interação das mesmas com membranas miméticas de vesículas gigantes unilamelares (GUVs - Giant Unilamellar Vesicles), com visualização microscópica. As MNPs recobertas com DODAB mostraram-se mais tóxicas para os queratinócitos em cultura e também causaram lise das GUVs. No caso das NbONPs, avaliou-se a acidez proveniente do Nb2O5 e o seu potencial em catálise heterogênea, bem como a avaliação da citotoxicidade em HaCaT revelou um potencial uso biomédico


Nanoparticles (NPs) have received increasing attention in biomedical applications. NPs can be constituted by different materials and have been used as contrast agents, in drug delivery, in cancer therapy, in heterogeneous catalysis, among other applications. Magnetic iron oxide nanoparticles (MNP) are notable for their multiplicity of applications, although they are poorly characterized for cellular toxicity. Other nanoparticles with excellent potential are made of niobium oxide (NbONPs), which deserve special attention, since Brazil holds 98% of the commercially viable reserves of this element. In this Thesis, NPs of these two transition metals (iron and niobium) were synthesized, aiming to understand their interactions with materials, biomolecules and media biological. Several methodologies were developed and tested to optimize the morphology and yield of the preparation, resulting in the choice of thermal decomposition for MNPs and, for NbONPs, the impregnation of niobium oxide on a matrix of silica-coated MNPs. In the case of MNPs, they were also coated with lipid zwitterionics (Dioleoyl phosphocholine (DOPC)) and positively charged (Dimethyldioctadecylammonium bromide (DODAB)) lipids. Its properties were initially characterized in several biological environments for later cytotoxicity assays in human keratinocytes (HaCaT). It evaluated the degradation of the NPs in different pH, as well as their interaction with giant unilamellar vesicle (GUVs) mimetic membranes, with microscopic visualization. MNPs coated with DODAB were more toxic to keratinocytes in culture and caused lysis of GUVs. In the case of NbONPs, acidity from Nb2O5 was evaluated in heterogeneous catalysis, as well as the evaluation of HaCaT cytotoxicity revealed a potential biomedical use

Abstract The aim of this study was to evaluate the cytotoxicity and bioactivity of calcium silicate-based cements combined with niobium oxide (Nb2O5) micro and nanoparticles, comparing the response in different cell lines. This evaluation used four cell lines: two primary cultures (human dental pulp cells - hDPCs and human dental follicle cells - hDFCs) and two immortalized cultures (human osteoblast-like cells - Saos-2 and mouse periodontal ligament cells - mPDL). The tested materials were: White Portland Cement (PC), mineral trioxide aggregate (MTA), white Portland cement combined with microparticles (PC/Nb2O5µ) or nanoparticles (PC/Nb2O5n) of niobium oxide (Nb2O5). Cytotoxicity was evaluated by the methylthiazolyldiphenyl-tetrazolium bromide (MTT) and trypan blue exclusion assays and bioactivity by alkaline phosphatase (ALP) enzyme activity. Results were analyzed by ANOVA and Tukey test (a=0.05). PC/Nb2O5n presented similar or higher cell viability than PC/Nb2O5µ in all cell lines. Moreover, the materials presented similar or higher cell viability than MTA. Saos-2 exhibited high ALP activity, highlighting PC/Nb2O5µ material at 7 days of exposure. In conclusion, calcium silicate cements combined with micro and nanoparticles of Nb2O5 presented cytocompatibility and bioactivity, demonstrating the potential of Nb2O5 as an alternative radiopacifier agent for these cements. The different cell lines had similar response to cytotoxicity evaluation of calcium silicate cements. However, bioactivity was more accurately detected in human osteoblast-like cell line, Saos-2.


Resumo O objetivo deste estudo foi avaliar a citotoxicidade e bioatividade de cimentos à base de silicato de cálcio associados com óxido de nióbio (Nb2O5) micro e nanoparticulados, e comparar a resposta em diferentes linhagens celulares. Foram utilizadas quatro linhagens celulares: duas culturas primárias (células da polpa dentária humana - hDPCs e células do folículo dentário humano - hDFCs) e duas culturas imortalizadas (células osteoblásticas humanas - Saos-2 e células do ligamento periodontal de ratos - mPDL). Os materiais analisados foram: Cimento Portland branco (PC); Agregado trióxido mineral (MTA); PC associado com micropartículas (PC/Nb2O5µ) ou nanopartículas (PC/Nb2O5n) de óxido de nióbio (Nb2O5). A citotoxicidade foi avaliada pelos ensaios de brometo de metil-tiazolil-difeniltetrazólio (MTT) e azul de tripan, e a bioatividade pela atividade da enzima fosfatase alcalina (ALP). Os resultados foram analisados por ANOVA e teste de Tukey (a=0,05). O grupo do PC/Nb2O5n apresentou viabilidade celular semelhante ou maior do que o grupo do PC/Nb2O5μ em todas as linhagens celulares. Além disso, ambos os grupos apresentaram viabilidade celular semelhante ou maior do que o MTA. Saos-2 apresentaram maior atividade de ALP, com destaque para o material PC/Nb2O5μ aos 7 dias de exposição. Concluiu-se que cimentos de silicato de cálcio associados com Nb2O5 micro ou nanoparticulado apresentaram citocompatibilidade e bioatividade, demonstrando potencial do Nb2O5 como agente radiopacificador alternativo para estes cimentos. As linhagens celulares estudadas apresentaram resposta semelhante na avaliação da citotoxicidade de cimentos de silicato de cálcio. No entanto, a bioatividade é melhor detectada na linhagem de células osteoblásticas humanas, Saos-2.

Abstract To evaluate the influence of rewetting solutions on bond strength to root dentin of conventional gutta-percha (GP) or niobium phosphate glass-based gutta-percha (GNb) associated with a bioceramic sealer. The root canals of 80 human mandibular premolars were prepared using nickel-titanium instruments and irrigation with sodium hypochlorite and EDTA. The teeth were randomly divided into four groups according to the gutta-percha used: GNb or GP associated with EndoSequence BC Sealer (BC) and the solution for rewetting dentin before filling (distilled water; phosphate buffer saline solution - PBS; simulated body fluid - SBF; or no solution). The root canals were filled with a single cone using warm vertical condensation. Micropush-out bond strengths associated with the filling materials in slices from middle root thirds was determined 30 days after root filling. The failure mode was analyzed with stereoscopic lens. The data were statistically analyzed by two-way ANOVA and Holm-Sidak test (p < 0.05). There was significant difference in the types of gutta-percha (p < 0.001) and in the different rewetting solutions (p = 0.003). The interaction between gutta-percha and rewetting solutions was not significant (p = 0.53). The SBF solution provided an increase in bond strength for both gutta-percha solutions. The GNb+BC (3.42 MPa) association increased bond strength when compared with GP+BC (2.0 MPa). The use of SBF as a dentin rewetting solution increased bond strength in the groups studied. Association of GNb with bioceramic sealer was beneficial, increasing the bond strength to dentin when compared with the association with GP.

PURPOSE: The purpose of this study was to investigate the electrochemical characteristics of nanotubular Ti-25Nb-xZr ternary alloys for dental implant materials. MATERIALS AND METHODS: Ti-25Nb-xZr alloys with different Zr contents (0, 3, 7, and 15 wt.%) were manufactured using commercially pure titanium (CP-Ti), niobium (Nb), and zirconium (Zr) (99.95 wt.% purity). The alloys were prepared by arc melting in argon (Ar) atmosphere. The Ti-25Nb-xZr alloys were homogenized in Ar atmosphere at 1,000℃ for 12 hours followed by quenching into ice water. The microstructure of the Ti-25Nb-xZr alloys was examined by a field emission scanning electron microscope. The phases in the alloys were identified by an X-ray diffractometer. The chemical composition of the nanotube-formed surfaces was determined by energy-dispersive X-ray spectroscopy. Selforganized TiO₂ was prepared by electrochemical oxidation of the samples in a 1.0 M H₃PO₄+0.8 wt.% NaF electrolyte. The anodization potential was 30 V and time was 1 hour by DC supplier. Surface wettability was evaluated for both the metallographically polished and nanotube-formed surfaces using a contact-angle goniometer. The corrosion properties of the specimens were investigated using a 0.9 wt.% aqueous solution of NaCl at 36℃±5℃ using a potentiodynamic polarization test. RESULT: Needle-like structure of Ti-25Nb-xZr alloys was transform to equiaxed structure as Zr content increased. Nanotube formed on Ti-25Nb-xZr alloys show two sizes of nanotube structure. The diameters of the large tubes decreased and small tubes increased as Zr content increased. The lower contact angles for nanotube formed Ti-25NbxZr alloys surfaces showed compare to non-nanotube formed surface. The corrosion resistance of alloy increased as Zr content increased, and nanotube formed surface showed longer the passive regions compared to non-treatment surface. CONCLUSION: It is confirmed that corrosion resistance of alloy increased as Zr content increased, and nanotube formed surface has longer passive region compared to without treatment surface.

Abstract To evaluate the effect of incorporating niobium phosphate bioactive glass (NbG) into commercial etch-and-rinse adhesive systems, with and without silane, on their degree of conversion (DC) (%) and microtensile bond strength (μTBS). The NbG micro-filler was added to two etch-and-rinse adhesive systems: One Step (OS) and Prime & Bond (PB) at 40% concentration. The following groups were formed: control without glass addition OS; addition of unsilanized NbG (OSNbG); addition of silanized NbG (OSNbGS); control without glass PB; addition of unsilanized NbG (PBNbG); addition of silanized NbG (PBNbGS). The DC was determined using total Fourier spectroscopy reflection (FTIR/ATR). For μTBS testing, 48 human third molars (n=8) were restored and sliced to obtain specimens (0.8 mm2) and they were tested at two different time intervals: immediately and after 6 months. The fracture mode was evaluated with a stereoscopic loupe (40×) and by scanning electron microscopy (SEM). The data were subjected to ANOVA and Tukey tests (a=0.05). NbG addition did not compromise the adhesive system DC values (p>0.05). Furthermore, the NbG added to the adhesive systems did not affect μTBS values (p>0.05). Fracture occurred predominantly at the dentin-adhesive interface. NbG bioactive glass did not affect the DC or microtensile bond strength results.


Resumo Avaliar o efeito da incorporação de vidro niobofosfato bioativo (NbG, 40% em peso) em dois sistemas adesivos simplificados convencionais (One Step [OS] e Prime & Bond {PB}) com e sem silano, no grau de conversão (%) e resistência de união (RU) após 6 meses. Os seguintes grupos foram testados: Controle OS: sem adição de partículas; OS e adição de NbG sem silano (OSNbG); OS e adição de partículas silanizadas NbG (OSNbGS); Controle PB: sem adição de partículas; PB e adição de partículas de NbG sem silano (PBNbG); PB e adição de partículas silanizadas NbG (PBNbGS). O grau de conversão (GC) foi determinado utilizando espectroscopia de Fourier (FTIR/ATR). Para RU, 48 terceiros molares humanos (n=8) foram restaurados e cortados para obter corpos-de-prova (0,8 mm2) e, em seguida, testados em dois momentos: imediato e após seis meses. O padrão de fratura foi avaliado com lupa estereoscópica (40×) e microscópio eletrônico de varredura (MEV). Os dados foram submetidos à análise de variância e Tukey (a=0,05). NbG não comprometeu valores de GC dos sistemas adesivos (p>0,05). Além disso, a adição de NbG aos sistemas adesivos não afetou os valores de RU (p>0,05). O padrão de fratura ocorreu predominantemente na interface dentina-adesivo. NbG não afetou os resultados de GC e RU.

ABSTRACT Objective: To identify respiratory symptoms and evaluate lung function in mine workers. Methods: This was a cross-sectional observational study involving production sector workers of a pyrochlore mining company. The subjects completed the British Medical Research Council questionnaire, which is designed to evaluate respiratory symptoms, occupational exposure factors, and smoking status. In addition, they underwent pulmonary function tests with a portable spirometer. Results: The study involved 147 workers (all male). The mean age was 41.37 ± 8.71 years, and the mean duration of occupational exposure was 12.26 ± 7.09 years. We found that 33 (22.44%) of the workers had respiratory symptoms and that 26 (17.69%) showed abnormalities in the spirometry results. However, we found that the spirometry results did not correlate significantly with the presence of respiratory symptoms or with the duration of occupational exposure. Conclusions: The frequencies of respiratory symptoms and spirometric changes were low when compared with those reported in other studies involving occupational exposure to dust. No significant associations were observed between respiratory symptoms and spirometry results.


RESUMO Objetivo: Determinar a presença de sintomas respiratórios e avaliar a função pulmonar de trabalhadores da mineração. Métodos: Estudo observacional de caráter transversal realizado com trabalhadores do setor de produção de uma mineradora de pirocloro. Para avaliar os sintomas respiratórios, fatores de exposição ocupacional e tabagismo, foi aplicado o questionário de sintomas respiratórios British Medical Research Council, e a função pulmonar foi avaliada utilizando-se um espirômetro portátil. Resultados: Participaram do estudo 147 trabalhadores, todos do sexo masculino, com média de idade de 41,37 ± 8,71 anos e com tempo de exposição ocupacional de 12,26 ± 7,09 anos. Foi observado que 33 (22,44%) dos trabalhadores apresentaram sintomas respiratórios e que 26 (17,69%) dos trabalhadores apresentaram alguma alteração nos resultados espirométricos. Entretanto, os resultados espirométricos não se correlacionaram significativamente com sintomas respiratórios e tempo de exposição. Conclusões: As frequências de sintomas respiratórios e de alterações espirométricas apresentaram-se baixas quando comparadas às de estudos que envolviam exposição à poeira ocupacional. Não foram observadas associações significativas dos sintomas respiratórios com os valores espirométricos.

Objective Mineral Trioxide Aggregate (MTA) is composed of Portland Cement (PC) and bismuth oxide (BO). Replacing BO for niobium oxide (NbO) microparticles (Nbµ) or nanoparticles (Nbη) may improve radiopacity and bioactivity. The aim of this study was to evaluate the radiopacity and cytotoxicity of the materials: 1) PC; 2) White MTA; 3) PC+30% Nbµ; 4) PC+30% Nbη. Material and Methods For the radiopacity test, specimens of the different materials were radiographed along an aluminum step-wedge. For cell culture assays, Saos-2 osteoblastic-cells (ATCC HTB-85) were used. Cell viability was evaluated through MTT assay, and bioactivity was assessed by alkaline phosphatase activity assay. Results The results demonstrated higher radiopacity for MTA, followed by Nbµ and Nbη, which had similar values. Cell culture analysis showed that PC and PC+NbO associations promoted greater cell viability than MTA. Conclusions It was concluded that the combination of PC+NbO is a potential alternative for composition of MTA. .

RESUMEN

Os objetivos deste estudo foram o isolamento e diferenciação de células da polpa (hDPCs) e do folículo (hDFCs) dentário de terceiros molares humanos e utilização para avaliação da citotoxicidade e bioatividade de novos cimentos endodônticos. (Capítulo 1) - Após a obtenção das culturas primárias foram realizados os ensaios vermelho de Alizarina, atividade da enzima fosfatase alcalina (ALP). A expressão gênica de proteína morfogenética óssea (bmp7), sialofosfoproteína dentinária (dspp) e alp foi avaliada. Os ensaios de bioatividade demonstraram maior atividade para hDPCs, além de maior expressão de dspp e alp; a expressão de bmp7 foi semelhante nas duas linhagens. Conclui-se que hDPCs demonstram maior capacidade de diferenciação em células com potencial de mineralização, quando comparadas com hDFCs. (Capítulo 2) – Foram utilizadas linhagens primárias (hDPCs e hDFCs), em comparação com linhagens imortalizadas (células osteoblásticas humanas Saos-2, e do ligamento periodontal de ratos mPDL) para análise dos materiais: 1) Cimento Portland branco (CP); 2) MTA; 3) CP/ óxido de nióbio microparticulado (Nb2O5?); 4) CP/ óxido de nióbio nanoparticulado (Nb2O5n). A citotoxicidade foi avaliada pelos ensaios MTT e Azul de Trypan e a bioatividade pela atividade da ALP. Viabilidade celular foi observada em todas as linhagens; e a atividade de ALP apresentou maior resposta na linhagem Saos-2. Conclui-se que as diferentes linhagens celulares são similares para avaliação da citotoxicidade; entretanto, para avaliação da bioatividade o uso de uma linhagem celular osteoblástica é mais indicado. Além disso, observa-se que materiais à base de Nb2O5? e Nb2O5n mostram potencial como radiopacificador alternativo no MTA


The aims of this study were to isolate and differentiate pulp (hDPCs) and follicle (hDFCs) cells obtained from human third molars and to use them in cytotoxicity and bioactivity assays of new dental materials. (Chapter 1) - After obtaining the stem cultures, Alizarin Red, alkaline phosphatase (ALP) activity and gene expression by Real Time PCR of bone morphogenetic protein (BMP7), dentin sialophosphoprotein (DSPP) and ALP were performed. The bioactivity asssays showed greater activity for hDPCs, and increased expression of dspp and alp; the expression of bmp7 was similar in both cell lines. We conclude that hDPCs demonstrate greater ability to differentiate into cells with mineralization potential when compared with hDFCs. (Chapter 2) - stem lineages (hDPCs and hDFCs) were used, compared to immortalized lineages (human osteoblastic cells Saos-2, and periodontal ligament of mice cells mPDL) for analysis of the following materials: 1) white Portland cement (PC); 2) MTA; 3) C/ microparticulated of niobium oxide (Nb2O5µ); 4) CP/nanoparticulated niobium oxide (Nb2O5n). Cytotoxicity was performed by MTT and Trypan Blue assays and bioactivity by ALP activity assay. Cell viability was observed in all cell lines, and ALP activity showed greater response in Saos-2. We conclude that the different cell lines are similar in cytotoxicity, however, to evaluate the bioactivity, using an osteoblastic cell line is more appropriate. Moreover, it is observed that materials based on Nb2O5µ and Nb2O5n show potential for alternative use to MTA

Os objetivos deste estudo foram desenvolver e caracterizar um compósito para obturação endodôntica à base de guta-percha e vidro niobofosfato. Foi avaliado a resistência de união à dentina radicular do compósito experimental, da guta EndoSequence BC, ambos sem o uso de cimento além do cimento AH Plus associado a guta-percha através do ensaio de push-out. A análise dos padrões de fratura foi realizada com auxílio de MEV. Análises usando EDX and MEV-EDS foram realizadas para verificar a composição e distribuição das partículas de vidro na superfície e no interior da matriz de guta-percha dos materiais testados. Os resultados foram analisados estatisticamente pelos testes ANOVA e Tukey, sendo considerado significante quando p < 0,05. Foi avaliado também a influência do compósito experimental, do vidro niobofosfatos bioativo, diferentes tipos de gutapercha e hidroxiapatita na adesão e formação de biofilme por bactérias orais com coloração Live/dead usando um microscópio confocal à laser. O biofilme multiespécies foi formado na superfície de discos de hidroxiapatita - HA, vidro niobofosfato - VNB, guta-percha Obtura - OBT, guta-percha Protaper - PTP, EndoSequence BC guta-percha - GBC e o compósito experimental guta-percha associado com o vidro niobofosfato - GNB. O biovolume total (mm3), biovolume de bacterias viáveis (mm3), e porcentagem de bactérias viáveis (%) foram quantificados...


The objectives of this study were to develop and characterize a composite for endodontic obturation based gutta-percha and niobium phosphate glass. The study evaluated the composites micropush-out bond strength to root dentine of the experimental gutta-percha and niobium phosphate glass composite applied with thermoplastic technique to the root canals without sealer in a moist environment and compare to the conventional root canal obturation Ah Plus and gutta-percha, and a commercial bioceramic gutta without sealer. Additionally, the cores materials were characterized using Scanning Electron Microscopic/Energy-dispersive X-ray (SEM/EDS) and Energy Dispersive X-ray fluorescence spectrometry (EDX) analysis. The failure mode was analyzed with SEM. Analysis using EDX and SEMEDS was carried out to verify the composition and distribution of the particles of the tested materials. Data were statistically analyzed by one-way ANOVA and Tukeys test (p < 0,05). Also was analysed the influence of different types of gutta-percha and bioactive niobium phosphate glass on the adherence and biofilm formation by oral bacteria from human dental plaque with Live/dead staining assay using confocal laser scanning microscopy (CLSM). The multispecies biofilm was grown from plaque bacteria on discs of hydroxyapatite - HA, niobium phosphate bioactive glass- VNB, Obtura pellets - OBT, Protaper gutta-percha - PTP, EndoSequence BC gutta-percha - GBC and gutta-percha associated with niobium phosphate glass - GNB in brain-heart infusion broth for 3,14 and 30 days. OBJECTIVE@#To evaluate the cytotoxicity of a new type of titanium alloy Ti-25Nb-10Ta-1Zr-0.2Fe by studying the induced proliferation of L929 cells in contrast with other titania widely used in clinical practice.@*METHODS@#The cell line was treated with extracting liquid containing different concentrations of titanium alloys. The number and morphology of cells was observed under an inverted phase contrast microscope. MTT was used to measure the relative growth rate (RGR) and judge the cytotoxicity grade. Flow cytometry was used to observe cell cycle progression.@*RESULTS@#The RGR of TNTZ group cells at the 3 time points was (93.7±0.8), (100.6±0.4), and (106.4±0.3); the cytotoxicity grade was 1, 0 and 0 after treating for 1, 3 and 5 days; with influence on neither the cell morphology nor the cell cycle. The flow cytometry showed that the sequence of S phase cells was Ti>TNTZ>TC4>blank control >TC4ELI, with no significant difference (P>0.05). None of the 4 materials inhibited the cell proliferation.@*CONCLUSION@#The cell morphology and proliferation are not affected by TNTZ. The new titaniu alloys shows good cyto-compatibility. The cytotoxicity is grade 0, meeting the clinical application standard.

O objetivo desta tese foi caracterizar o pentóxido de nióbio (Nb2O5), produzir materiais compósitos de matriz polimérica de uso odontológico e avalia-los. As partículas de Nb2O5 foram caracterizadas com difração de raios-X, microscopia eletrônica de varredura, granulometria a laser, método de Brunauer–Emmett–Teller e espectroscopia micro-Raman. Um adesivo experimental foi desenvolvido com a incorporação de diferentes concentrações de Nb2O5 (0, 5, 10 e 20 %, em peso). Este adesivo foi avaliado quanto à radiopacidade, grau e cinética de conversão, microdureza e a sua interface com a dentina. Um cimento endodôntico experimental de cura dual também foi desenvolvido com a incorporação de diferentes concentrações de Nb2O5 (0, 80, 100 e 120 %, em peso). Este cimento foi avaliado quanto à radiopacidade, escoamento, espessura de película, microdureza e grau de conversão (imediatamente, 7 dias e 14 dias após a fotoativação). A avaliação do Nb2O5 mostrou que as partículas utilizadas apresentavam estrutura monoclínica, com formato irregular, com tamanho médio de 38,16 μm, área de superfície de 3,86 m2/g e estrutura química compatível com pentóxido de nióbio. A radiopacidade e dureza do adesivo aumentaram com a incorporação do Nb2O5 e ele pode ser detectado no interior da interface dentina/adesivo. No cimento endodôntico a incorporação do Nb2O5 também aumentou a radiopacidade e a dureza e reduziu o escoamento apenas no grupo com 120 %. O grau de conversão aumentou até 14 dias após a fotoativação, com exceção do grupo com 80%. Após a caracterização do Nb2O5 e da produção dos materiais experimentais, foi feito o depósito de pedido de patente de invenção. The aim of this thesis was to characterize the niobium pentoxide (Nb2O5), to produce resin-based composite for dental applications and evaluate them. Niobium pentoxide was characterised by X-ray diffraction, surface area, particle size, micro-Raman and scanning electron microscopy. An experimental adhesive resin was formulated with 0, 5, 10 and 20 wt% Nb2O5. The formulated adhesive resins were evaluated based on microhardness, degree of conversion, radiopacity and interface (resin/dentin) characterisation by micro-Raman. An experimental dual-cured root canal sealer was produced with a methacrylate-based comonomer blend. Nb2O5 was added at four different concentrations: 0, 80, 100 and 120wt%. Radio-opacity, flow, film thickness, microhardness and degree of conversion (after 1, 7 and 14 days) were evaluated. The particles used in this study presented a monoclinic crystalline phase with typical chemical groups, mean size of 38.16 μm and 3.86 m2/g of surface area. Microhardness and radiopacity increased with higher amounts of Nb2O5, and the particles were able to penetrate into the hybrid layers. Microhardness and radiopacity increased with higher amounts of Nb2O5 in the adhesive resin, and the particles were able to penetrate into the hybrid layers. For the endodontic cement, the radiopacity and microhardness increased with the Nb2O5 addition. The flow was significantly smaller for the 120% group. All groups except those with 80wt% had higher values for degree of conversion after 14 days than immediately after photocuring. <p><b>OBJECTIVES</b>To investigate the effect of magnetron sputtered niobium nitride (NbN) on the bonding strength of commercially pure cast titanium (Ti) and low-fusing porcelain (Ti/Vita titankeramik system).</p><p><b>METHODS</b>Sixty Ti specimens were randomly divided into four groups, group T1, T2, T3 and T4. All specimens of group T1 and T2 were first treated with 120 microm blasted Al2O3 particles, and then only specimens of group T2 were treated with magnetron sputtered NbN film. All specimens of group T3 and T4 were first treated with magnetron sputtered NbN film and then only specimens of group T4 were treated with 120 microm blasted Al2O3 particles. The composition of the deposits were analyzed by X-ray diffraction (XRD). A universal testing machine was used to perform the three-point bending test to evaluate the bonding strength of Ti and porcelain. The microstructure of NbN, the interface of Ti-porcelain and the fractured Ti surface were observed with scanning electron microscopy (SEM) and energy depressive spectrum (EDS), and the results were compared.</p><p><b>RESULTS</b>The XRD results showed that the NbN deposits were cubic crystalline phases. The bonding strength of Ti and porcelain in T1 to T4 group were (27.2+/-0.8), (43.1+/-0.6), (31.4+/-1.0) and (44.9+/-0.6) MPa. These results were analyzed by one-way analysis of variance and differences between groups were compared using least significant difference test. Significant inter-group differences were found among all groups (P<0.05). The results of SEM showed that with treatment of Al2O3 or NbN, alone, pre-cracks were found in the interface of Ti-porcelain, while samples treated with both Al2O3 and NbN had better bond. EDS of Ti-porcelain interface showed oxidation occurred in T1, T2 and T3, but was well controlled in T4.</p><p><b>CONCLUSIONS</b>Magnetron sputtered NbN can prevent Ti from being oxidized, and can improve the bonding strength of Ti/Vita titankeramik system. Al2O3 blast can also improve the bonding strength of Ti/Vita titankeramik system.</p>

Objectives: This study aims to evaluate and compare the mechanical and metallurgical properties of stainless steel and titanium molybdenum alloy (TMA) archwires, with recently introduced timolium and titanium niobium arch wires. Materials and Methods: Archwires were categorized into four groups (group I to IV) with 10 samples in each group. They were evaluated for tensile strength, yield strength, modulus of elasticity, load deflection, frictional properties and weld characteristics. Results: The results were statistically analyzed using ANOVA test and it indicated that stainless steel has high strength, high stiffness and low friction compared to other arch wires, thereby proving that it is the best choice for both sliding as well as frictionless retraction mechanics. TMA with its high formability, low stiffness and low load deflection property is suited to apply consistent force in malaligned teeth but, high friction limits its use in retraction only with loop mechanics. Conclusion: Timolium possesses comparatively low stiffness, better strength and behaves as an intermediate between stainless steel and TMA and hence can be tried for almost all clinical situations. Low springback and high formability of titanium-niobium archwire allows creation of finishing bends and thus it can be used as finishing archwire.

STATEMENT OF PROBLEM: The success of titanium implants is due to osseointegration or the direct contact of the implant surface and bone without a fibrous connective tissue interface. PURPOSE: The purpose of this study was to evaluate the osteoblast precursor response to titanium - 10 tantalum - 10 niobium (Ti-Ta-Nb) alloy and its sputtered coating. MATERIAL AND METHODS: Ti-Ta-Nb coatings were sputtered onto the Ti-Ta-Nb disks. Ti6-Al-4V alloy disks were used as controls. An osteoblast precursor cell line, were used to evaluate the cell responses to the 3 groups. Cell attachment was measured using coulter counter and the cell morphology during attachment period was observed using fluorescent microscopy. Cell culture was performed at 4, 8, 12 and 16 days. RESULTS: The sputtered Ti-Ta-Nb coatings consisted of dense nanoscale grains in the range of 30 to 100 nm with alpha-Ti crystal structure. The Ti-Ta-Nb disks and its sputtered nanoscale coatings exhibited greater hydrophilicity and rougher surfaces compared to the Ti-6Al-4V disks. The sputtered nanoscale Ti-Ta-Nb coatings exhibited significantly greater cell attachment compared to Ti-6Al-4V and Ti-Ta-Nb disks. Nanoscale Ti-Ta-Nb coatings exhibited significantly greater ALP specific activity and total protein production compared to the other 2 groups. CONCLUSIONS: It was concluded that nanoscale Ti-Ta-Nb coatings enhance cell adhesion. In addition, Ti-Ta-Nb alloy and its nanoscale coatings enhanced osteoblast differentiation, but did not support osteoblast precursor proliferation compared to Ti-6Al-4V. These results indicate that the new developed Ti-Ta-Nb alloy and its nanoscale Ti-Ta-Nb coatings may be useful as an implant material.

<p><b>AIM</b>The piezoelectric properties and cytotoxicity of a porous lead-free piezoelectric ceramic for use as a direct bone substitute were investigated.</p><p><b>METHODOLOGY</b>Cold isostatic pressing (CIP) was applied to fabricate porous lithium sodium potassium niobate (Li0.06Na0.5K0.44) NbO3 specimens using a pore-forming method. The morphologies of the CIP-processed specimens were characterized and compared to those of specimens made by from conventional pressing procedures. The effects of the ceramic on the attachment and proliferation of osteoblasts isolated from the cranium of 1-day-old Sprague-Dawley rats were examined by a scanning electron microscopy (SEM) and methylthiazol tetrazolium (MTT) assay.</p><p><b>RESULTS</b>The results showed that CIP enhanced piezoelectricity and biological performance of the niobate specimen, and also promoted an extracellular matrix-like topography of it. In vitro studies showed that the CIP-enhanced material had positive effects on the attachment and proliferation of osteoblasts.</p><p><b>CONCLUSION</b>Niobate ceramic generated by CIP shows a promise for being a piezoelectric composite bone substitute.</p>

<p><b>OBJECTIVE</b>A novel biological piezoelectric ceramic was made by beta-tricalcium phosphate (beta-TCP) and lithium sodium potassium niobate (LNK) piezoelectric ceramics. To study its biocompatibility to osteoblast isolated from the cranium of 1-day-old Sprague-Dawley mice.</p><p><b>METHODS</b>The biological piezoelectric ceramic TCPLNK1/10, TCPLNK5/5 respectively mixed by beta-TCP and LNK piezoelectric ceramic at the ratio of 1/10 and 5/5. Then osteoblasts were used and seeded respectively on the negative and positive surfaces of TCPLNK1/10 and TCPLNK5/5. Growth and proliferation of the osteoblasts on TCPLNK1/10 and TCPLNK5/5 surfaces were evaluated in vitro by means of scanning electron microscopy (SEM) examination, fluorescence dyeing of osteoblast skeleton protein and MTT assay.</p><p><b>RESULTS</b>Cell morphology of osteoblast on positive and negative surfaces of TCPLNK1/10 and TCPLNK5/5 was normal, and both adhesion and growth characteristics showed better than control group. The growing osteoblasts on the TCPLNK1/10 negative surface were significantly higher than others. The negative surface of TCPLNK1/10 possessed better osteogenesis potential than others in vitro.</p><p><b>CONCLUSION</b>The surface of TCPLNK may permit the imitation piezoelectric effect of natural bone for bone regeneration.</p>

Visando comparar a relação carga/deflexão de fios ortodônticos retangulares de aço inoxidável e titânio-nióbio, foram selecionados fios 0,019" x 0,025" pré-contornados das seguintes marcas: Abzil Lancer, 3M Unitek e Ormco, em aço inoxidável, e da marca Ormco em titânio-nióbio. A metodologia consistiu em posicionar os fios pré-contornados em um manequim ortodôntico, com aparelho fixo colado e ausência do incisivo central superior direito. O conjunto foi fixado a uma máquina universal de ensaios, Emic 10000, provida de uma ponteira de aço, que transmitiu cargas no centro do segmento do fio selecionado, correspondente ao incisivo extraído. A aplicação de forças era mensurada por um programa de computador, gerando automaticamente gráficos e tabelas da relação carga/deflexão. Os resultados mostraram uma menor relação carga/deflexão nos fios de aço inoxidável da marca 3M Unitek, seguidos pela Ormco. No entanto, os fios de titânio-nióbio da Ormco apresentaram valores médios de carga/deflexão estatisticamente menores em relação a todos os fios de aço inoxidável avaliados, independentemente de suas marcas comerciais.


Aiming to compare load/deflection relation of rectangular arch wires of stainless steel(SS) and titanium-niobium, were selected pre-conformed 0.019''x 0.025" arch wires of next marks: Abzil Lancer; 3M Unitek and Ormco, in stainless steel, and Titanium-niobium from Ormco. The methodology consisted in installation the arch wires in orthodontic simulator, with bond fixd orthodontics appliances and absence of right maxillary central incisive. The simulator was fixed in a Universal Tests Machine, Emic 10000, where a stainless steel ferrule transmitted load in the middle of arch wires segment of incisive absence. According to transmitting of loads, the software measured the deflection of arch wire and automatically created graphics and tables of load/deflection relation. The results showed a lower load/deflection relation in 3M Unitek stainless steel arch wires, followed by Ormco samples. The Ormco titanium-niobium presented the statistically lowest load/deflection relation, when compared with all stainless steel brands tested.

The recovery force of Ti-Nb coated and uncoated TiNi shape memory alloy rods was investigated. The rods were 6.0 mm, 6.5 mm and 7.0 mm in diameter respectively. The mean transition temperature was 33.0 degrees C. The rods were stored at -18 degrees C and pre-bent with a three-point bending fixture, the span was 20. 0 centimeters and the deflections were 5.0 mm, 10.0 mm, 15.0 mm and 20.0 mm, respectively. The rods were then heated in a constant temperature saline solution chamber. The experimental temperature was 37.0 C and 50.0 C respectively. The recovery force was measured in a constant displacement mode on biomaterial test machine. The results showed that the recovery force of the memory alloy rod increased with increasing recovery temperature, rod diameter and deformation of both Ti-Nb coated and uncoated surface. The recovery force of Ti-Nb coated rods of 6.0 and 6.5 millimeter in diameter was lower than the uncoated rods in the same diameter. However, the recovery force of 7.0-mm-diameter rods showed no significant difference between coated and uncoated surface.
