BACKGROUND: LXYL-P1-2 is the first reported glycoside hydrolase that can catalyze the transformation of 7-b-xylosyl-10-deacetyltaxol (XDT) to 10-deacetyltaxol (DT) by removing the D-xylosyl group at the C7 position. Successful synthesis of paclitaxel by one-pot method combining the LXYL-P1-2 and 10- deacetylbaccatin III-10-b-O-acetyltransferase (DBAT) using XDT as a precursor, making LXYL-P1-2 a highly promising enzyme for the industrial production of paclitaxel. The aim of this study was to investigate the catalytic potential of LXYL-P1-2 stabilized on magnetic nanoparticles, the surface of which was modified by Ni2+-immobilized cross-linked Fe3O4@Histidine. RESULTS: The diameter of matrix was 20­40 nm. The Km value of the immobilized LXYL-P1-2 catalyzing XDT (0.145 mM) was lower than that of the free enzyme (0.452 mM), and the kcat/Km value of immobilized enzyme (12.952 mM s 1 ) was higher than the free form (8.622 mM s 1 ). The immobilized form maintained 50% of its original activity after 15 cycles of reuse. In addition, the stability of immobilized LXYL-P1-2, maintained 84.67% of its initial activity, improved in comparison with free form after 30 d storage at 4 C. CONCLUSIONS: This investigation not only provides an effective procedure for biocatalytic production of DT, but also gives an insight into the application of magnetic material immobilization technology.

RESUMEN

Abstract In this work, four isolates of endophytic fungi (Alternaria alternata, Colletotrichum gloesporioides, Glomerella cingulata and Nigrospora sphaerica), deposited in the culture collection 'University Recife Mycologia' (URM) at the Universidade Federal de Pernambuco, were characterized for the genes ITS 1 and 4 (region 5.8 S) and evaluated for taxol production.

RESUMEN

Of the initial six cell lines originating from explants of Taxus globosa, or Mexican yew (stem internode, leaves and meristematic tissue), three were selected for their microbial and oxidation resistance, two from leaves and the other from stem internode. A study of their behavior, both in terms of cell growth, and of baccatin III and paclitaxel production, was developed in suspension cultures with an initially standardized biomass (fresh weight 0.23 g/L) using modified Gamborg's B5 medium, and an elicitor (methyl jasmonate), on either the first or seventh day of culture, at several levels (0, 0.1, 1, 10, 100 microM). In most of the conditions used, the three cell lines showed growth associated baccatin III production. The cell line from stem internode was the highest producer of baccatin III using 1 microM elicit or, sampling at 10 days.

RESUMEN

A fungus was isolated from the stem cuttings of Taxus celebica, which produced paclitaxel in liquid-grown cultures. The fungus was identified as Fusarium solani based on colony characteristics, morphology of conidia and the 26S rDNA sequence. Paclitaxel was identified by chromatographic and spectroscopic comparison with authentic paclitaxel and its cytotoxic activity towards Jurkat cells in vitro.

Taxol is a diterpeniod with complex structure, which is extracted frequently from the genus Taxus. At present, this drug is one of the most important anticancer natural agents with different mechanism from other similar drugs, used for remedy of different cancers such a melanoma, lung cancer, non-hodgkins lymphoma, meatus urinarius cancer, esophagus cancer, etc. At present, many scientists are trying total synthesis and semi-synthesis of tax and also its extraction from cell culture. However, these methods have characteristic problems. The increasing demand of taxol for clinical use as an anticancer drug combined with limited supply from plant sources require a thorough understanding of its biosynthesis and extraction methods

Importance of yew tree [Taxus spp.] originates from the entering of Taxol [Paclitaxel] in the treatment of breast cancer since 1982. Taxus spp. is one of the Taxaceae family, distributed in North America, Europe, meditranean region, Asia and North forests of Iran. Efficacy of Taxol in the treament of some cancers and low concentration of this compound in tree, stimulated semi synthetic preparation of taxol from other more aboundant compounds such as baccation-III which is found with higher concentrations in all tree tissues. Despite entering some other routs for preparation of taxol e.g. cell culture, cultivation and studies on yew tree is in high importance, therefore review on this plant was made by authors
