Abstract The locust bean gum (LBG) is a polysaccharide with thickening, stabilizing and gelling properties and it has been used in the preparation of pharmaceutical formulations. Hydrogels (HGs) are obtained from natural or synthetic materials that present interesting properties for skin application. This study aimed to develop HGs from LBG using indole-3-carbinol (I3C) as an asset model for cutaneous application. HGs were prepared by dispersing LBG (2%, 3% and 4% w/v) directly in cold water. The formulations showed content close to 0.5 mg/g (HPLC) and pH ranging from 7.25 to 7.41 (potentiometry). The spreadability factor (parallel plate method) was inversely proportional to LBG concentration. The rheological evaluation (rotational viscometer) demonstrated a non-Newtonian pseudoplastic flow behavior (Ostwald De Weale model), which is interesting for cutaneous application. The HET-CAM evaluation showed the non-irritating characteristic of the formulations. The bioadhesive potential demonstrated bioadhesion in a concentration-dependent manner. Permeation in human skin using Franz cells showed that the highest LBG concentration improved the skin distribution profile with greater I3C amounts in the viable skin layers. The present study demonstrated the feasibility of preparing HGs with LBG and the formulation with the highest polymer concentration was the most promising to transport active ingredients through the skin.

In order to reveal the main nutrients and functional ingredients in the shoots of Polygonatum cyrtonema, the polysaccharides, proteins, amino acids, and total phenols were determined. The tested samples cultured in Ma'nijiaonong, Hengtang village, Tianmushan town, Lin'an, Zhejiang, which were collected from three provenances(Pan'an and Longquan in Zhejiang and Qingyang in Anhui). The results showed that the polysaccharide content of the shoots varied from 2.34% to 12.73%, roughly one-third of rhizomes. The protein content varied from 107.75 to 192.49 mg·g~(-1), nearly 5.50 times more than rhizomes. Moreover, the average of total amino acid content was 193.13-248.74 mg·g~(-1), approximately 4.16 times of rhizomes. And the essential amino acids account for 35.57%-39.44% of the total amino acids content, which was close to the standard of the ideal protein proposed by FAO/WHO(the essential amino acid/total amino acid is about 40%). In addition, the taste amino acids(TaAA) changed from 160.12 to 208.29 mg·g~(-1), revealing the material basis of "shoots were extremely delicious" in Chinese ancient herbal medicine. Additionally, the total phenols varied from 51.21-58.76 mg·g~(-1), about 2.96 times of rhizomes. The DPPH free radical scavenging rate of tested shoots was over 95%, which obviously superior to rhizomes. Therefore, the shoots of P. cyrtonema is a very high-quality vegetable and functional food with good development potential. Furthermore, the main nutrients and functional substances in P. cyrtonema shoots are closely related to the provenances and harvesting seasons. It is important to improve the quality and yield of the shoots by strengthening the variety of breeding and cultivation techniques.

Based on the ancient method of nine-steaming and nine-sun-curing,the chemical composition changes and quality profiles in different processes of Polygoni Multiflori Radix were studied. Their contents of stilbene glycoside,anthraquinones and polysaccharides were determined by nine-steaming and nine-sun-curing with black bean juice and pharmacopoeia method. HPLC chemical fingerprints were established,and orthogonal partial least squares-discriminant analysis( OPLS-DA) was performed on different processed products using SIMCA 14. 1 software to evaluate the quality difference between samples. The results of content determination show that,with the increase of the number of processing and steaming times,the stilbene glycoside and the combined anthraquinone showed a decreasing trend,and the free anthraquinone,total anthraquinone and polysaccharide showed an upward trend in the different preparations of Polygoni Multiflori Radix and Pharmacopoeia. Six-steamed and six-sun-cured products can be used as the finishing point for the classic steaming. Fingerprint results showed that there were significant differences in chemical composition in Polygoni Multiflori Radix at different processing processes. It can be identified stilbene glycoside( peak 13),emodin( peak 21),and physcion( peak 24). By comparing the relative peak areas of the 26 chromatographic peaks in the sample after normalization( the reference is peak 7),it was found that the relative peak areas of 12 peaks in the processed products were higher than the raw products,13 peaks were reduced; according to statistical analysis of OPLS-DA,Polygoni Multiflori Radix at different processing degrees was further divided into three categories,sample S1 was class I,S2-S5 were class Ⅱ,and S6-S11 were class Ⅲ. And 8 peaks with the VIP value higher than 1. 0 were peak 13,21,4,3,11,14,5,and 24 in order. The eight chemical components were the main components to distinguish the difference between Polygoni Multiflori Radix in the process of nine-steaming and nine-sun-curing,suggesting that it was rational to use stilbene glycoside,emodin and emodin methyl ether as quality control indicators of Polygoni Multiflori Radix. The method established in this experiment conformed to the methodological verification requirements,established a method of multi-component content determination combined with fingerprint,and clarified that six-steaming and six-sun-curing was used as an improved classical processing technology,and more clearly defined the whole dynamic change of chemical composition in Polygoni Multiflori Radix by nine-steaming and ninesun-curing process. It provides a basis for the chemical quality evaluation model about different processed products of Polygoni Multiflori Radix.

ABSTRACT Soybeans contain about 30% carbohydrate, mainly consisting of non-starch polysaccharides (NSP) and oligosaccharides. NSP are not hydrolyzed in the gastrointestinal tract of monogastric animals. These NSP negatively affect the development of these animals, especially the soluble fraction. This work aimed to establish a method to quantify NSP in soybeans, using high performance liquid chromatography (HPLC), and to estimate correlations between NSP, oligosaccharides, protein and oil. Sucrose, raffinose + stachyose, soluble and insoluble NSP contents were determined by HPLC. Oil and protein contents were determined by near-infrared spectroscopy (NIRS). The soluble PNAs content showed no significant correlation with protein, oil, sucrose and raffinose + stachyose contents, but oligosaccharides showed a negative correlation with protein content. These findings open up the possibility of developing cultivars with low soluble NSP content, aiming to develop feed for monogastric animals.

A bactéria gram-positiva Streptococcus agalactiae do grupo B (GBS) faz parte da microbiota normal do trato geniturinário humano, sendo um organismo comensal do corpo da mulher. No entanto, em mulheres grávidas, quando há alterações na composição microbiana do canal vaginal, pode ocorrer a proliferação e a infecção pelo GBS. Este microrganismo, em sua forma patogênica oportunista, pode infectar o neonato durante o parto natural, assim como contribuir para infecções urinárias e uterinas durante a gestação. O GBS já foi identificado como um dos responsáveis pela alta taxa de mortalidade neonatal, sendo um dos principais agentes de infecção em recém-nascidos no mundo. Ele também pode ser a causa de infecções nas gestantes, levando a várias complicações, como corioamnionite, endometrite e infecções do trato urinário e do sítio cirúrgico. Pode haver comprometimento da gestação e do feto, com abortamento, morte fetal intrauterina e ruptura da membrana coriônica, levando a parto prematuro - que pode resultar em outras consequências graves. Este trabalho foi desenvolver um modelo vacinal para mucosa sublingual, utilizando o polissacarídeo capsular do Streptococcus agalactiae como antígeno, encapsulado em Nanopartículas de quitosana. Para o estudo de otimização dos parâmetros de fermentação, para aumentar a produtividade de cápsula polissacarídica (PS) presente na superfície celular, utilizou-se o Banco de Dados Kegg (Kyoto Encyclopedia of Genes and Genomes). A adição do suplemento L-Prolina foi o que propiciou a principio, maior relação entre crescimento bacteriano e formação de cápsula polissacarídica. A purificação e extração da cápsula polissacarídica foi realizada com etapas sucessivas de ultra filtração tangencial e precipitação alcóolica dos contaminantes. As caracterizações físico-químicas: difração de raios-X (DRX), cromatografia gasosa (CGMS), ressonância magnética (NMR) e determinação de açúcares pelo método fenol-sulfúrico, foram realizadas para identificação da composição e estrutura monossacarídica de açucares. O PS isolado apresenta ramificações de fucose, manose, glicose, galactose e N-acetil-glucosamina, apresentando estrutura amorfa. A liofilização do polissacarídeo foi realizada para fins de concentração e conservação. A encapsulação do polissacarídeo acoplada quimicamente com OVA, em uma Nanopartícula de quitosana, teve como finalidade aumentar a mucoadesividade e possibilitar maior absorção do antígeno entre as células da junção epitelial das mucosas sublinguais. A partir da análise de DLS (Espalhamento dinâmico de luz), as Nanopartículas apresentaram dimensões entre 200 a 400 nm e o Potencial Zeta acima de 20. O índice de polidispersão (PDI) está dentro do esperado (abaixo de 0.3). A capacidade de encapsulamento em relação à OVA foi de 92,8% dos grupos que continham PS. O teste IgG sérica total mostrou que o grupo G2 (Nanopartícula com Polissacarídeo e Proteína acoplados) foi o que teve maior reatividade no teste de ELISA, pela Análise de Variância (ANOVA) com ferramenta de Bonferrone. O teste sIgA mostrou que o grupo G2 (Nanopartícula com Polissacarídeo e Proteína acoplados) foi o que teve maior concentração de anticorpo sIgA total. Como resultado e conclusão, o polissacarídeo capsular do Streptococcus agalactiae é um bom candidato a antígeno vacinal


Gram-positive bacteria Streptococcus agalactiae group B (GBS) is part of the normal microbiota of the human genitourinary tract, being a commensal organism of the female body. However, in pregnant women, when there are changes in the microbial composition of the vaginal canal, GBS proliferation and infection may occur. This microorganism, in its opportunistic pathogenic form, can infect the neonate during natural childbirth, as well as contribute to urinary and uterine infections during pregnancy. The GBS has already been identified as one of the responsible for the high neonatal mortality rate, being one of the main agents of infection in newborns in the world. It can also be the cause of infections in pregnant women, leading to various complications such as chorioamnionitis, endometritis, and urinary tract and surgical site infections. There may be pregnancy and fetal impairment, with abortion, fetal intrauterine death, and rupture of the chorionic membrane, leading to premature labor - which can result in other serious consequences. This work was to develop a vaccine model for sublingual mucosa using the capsular polysaccharide of Streptococcus agalactiae as antigen, encapsulated in chitosan nano particles. For the study of optimization of the fermentation parameters, the Kegg (Kyoto Encyclopedia of Genes and Genomes) database was used to increase the productivity of polysaccharide capsule (PS) present on the cell surface. The addition of the L-Proline supplement gave rise to a higher ratio between bacterial growth and polysaccharide capsule formation. The purification and extraction of the polysaccharide capsule was performed with successive stages of tangential ultrafiltration and alcoholic precipitation of the contaminants. The physicochemical characterization of X-ray diffraction (XRD), gas chromatography (CGMS), magnetic resonance (NMR) and determination of sugars by the phenol-sulfuric method were performed to identify the composition and monosaccharide structure of sugars. The isolated PS presents branches of fucose, mannose, glucose, galactose and N-acetyl-glucosamine, presenting amorphous structure. Lyophilization of the polysaccharide was performed for concentration and conservation purposes. The encapsulation of the polysaccharide coupled chemically with OVA in a chitosan nano particle was aimed at increasing mucoadhesiveness and allowing greater absorption of the antigen between the cells of the sublingual mucosal epithelial junction. From the analysis of DLS (dynamic light scattering), the nanoparticles presented dimensions between 200 to 400 nm and the Zeta potential above 20. The polydispersity index (PDI) is within the expected range (below 0.3). The encapsulation capacity for OVA was 92.8% of the groups containing PS. The total serum IgG test showed that the G2 group (Nano particle with Polysaccharide and Protein coupled) was the one that had the highest reactivity in the ELISA test, by Analysis of Variance (ANOVA) with Bonferrone tool. The sIgA test showed that the G2 group (Nanoparticle with Polysaccharide and Protein coupled) had the highest concentration of total sIgA antibody. As a result and conclusion, the capsular polysaccharide of Streptococcus agalactiae is a good candidate for vaccine antigen

A ingestão regular de fibras alimentares pode auxiliar na redução do risco de certas doenças crônicas, como doenças cardiovasculares, síndromes metabólicas, diabetes tipo 2 e alguns tipos de câncer. Dentre as principais fontes de fibras alimentares, destaca-se o grão de trigo, que é rico em fibras solúveis e insolúveis. O grão de trigo pode ser identificado por três macrorregiões distintas, sendo elas o pericarpo, o gérmen e o endosperma. O principal alimento oriundo do trigo é a farinha produzida através da moagem seriada do endosperma dos grãos, sendo o principal subproduto a casca do grão conhecido como farelo de trigo, cujo pericarpo é o principal constituinte. As fibras alimentares estão concentradas no pericarpo, ou seja, no farelo do trigo, e estão associadas através de uma estrutura tridimensional não uniforme e compactada, o que prejudica a solubilidade de polissacarídeos complexos diminuindo suas biodisponibilidades caso ingeridos na alimentação humana. Apesar do farelo de trigo ser um resíduo com razoável valor biológico e com alto teor de fibras, o principal destino do farelo é a alimentação animal. Devido ao fato do farelo de trigo possuir baixo custo, elevado teor de fibras e grande volume disponível no mercado, o objetivo desse trabalho é estudar três métodos físico-químicos que modifiquem a estrutura do farelo de trigo e permitam que uma maior quantidade de polissacarídeos se tornem mais solúveis em água. Uma vez diluídas, essas frações de polissacarídeos consideradas como fibras alimentares solúveis (FAS) poderão ser extraídas, isoladas e aplicadas como ingredientes na produção de alimentos funcionais, como margarinas e sucos. Assim, o projeto permitirá que a indústria de alimentos desenvolva uma fonte de fibras solúveis a partir de uma matéria prima de baixo custo e alta produção. As modificações químicas foram feitas em meio alcalino (solução de NaOH nas concentrações 0,025, 0,05 e 0,1M), meio ácido (solução de H2SO4 nas concentrações 0,25, 0,5e 1,0M) e com agentes oxidantes (solução de hipoclorito de sódio a 10%).Com relação ao rendimento da extração das frações solúveis, as extrações em meio ácido foram as melhores. Todos os polissacarídeos não amido extraídos do farelo de trigo apresentaram baixo peso molecular e composição de açúcares diferentes. Os polissacarídeos oriundos das extrações em meio ácido demonstraram mais glicose e um menor percentual de arabinose e xilose ao serem comparadas com a água, enquanto os oriundos das extrações alcalinas apresentaram um teor menor de glicose e maior de arabinose e xilose (em comparação com as frações da água). A partir da análise de açúcares (foi avaliado maiores quantidades dos monossacarídeos arabinose, xilose e glicose) e do rendimento das extrações, foram escolhidas 5 amostras, sendo elas a fração solúvel extraída em água, H2SO4 0,25 e 0,5M e NaOH0,25 e 0,1M para as análises do teor de ß-glicanos, compostos fenólicos, propriedades funcionais (propriedades espumantes, absorção de água, solubilidade e umidade) e efeito prebiótico. Como resultado, todas os polissacarídeos não amido apresentaram elevada solubilidade e não possuíram propriedades espumantes e de absorção de água (WAC) relevantes. O teor de compostos fenólicos foi reduzido em todas as frações solúveis, quando comparado com o teor de compostos fenólicos do farelo de trigo e o pequeno percentual remanescente de compostos fenólicos possui baixo poder redutor. Foi encontrado ß-glicano em todas as frações dos polissacarídeos não amido extraídas, sendo 0,82% nas frações extraídas em água, 0,97 e 2,79% nas frações extraídas com H2SO4 e 5,42 e 4,62% nas frações extraídas com NaOH 0,025 e 0,1M. Embora tenha sido encontrado ß-glicano em todas as amostras extraídas, somente as extrações em água e com NaOH 0,1 M apresentaram possível efeito prebiótico in vitro


Regular intake of dietary fiber can help to reduce the risk of certain chronic diseases, such as cardiovascular disease, metabolic syndromes, type 2 diabetes and some types of cancer. Among the main sources of dietary fiber is the wheat grain, which is rich in soluble and insoluble fibers. The wheat grain can be identified by three distinct macroregions, they are the pericarp, the germ and the endosperm. The main food made by wheat is its flour produced by serial grinding of the grain endosperm, and the main by-product is the husk of the grain known as wheat bran, whose pericarp is the main constituent. Food fibers are concentrated in the pericarp and are associated through a non-uniform and compacted three-dimensional structure, which impairs the solubility of complex polysaccharides, decreasing their bioavailability when ingested in human food. Although wheat bran is a residue with reasonable biological value and high fiber content, the main destination of the bran is animal feed. Due to the fact that wheat bran has a low cost, high fiber content and a great volume available in the market, the objective of this work was to study three physic-chemical methods that modify the structure of wheat bran and allow a greater amount of polysaccharides to become more soluble in water. Once diluted, these fractions of polysaccharides considered as soluble dietary fiber (FAS) can be extracted, isolated and applied as ingredients in the production of functional foods such as margarines and juices. Thus, the project will enable the food industry to develop a source of soluble fiber from a low-cost, high-yield raw material. The chemical modifications were made in alkaline medium (NaOH solution at concentrations of 0.025, 0.05 and 0.1M), acid medium (H2SO4 solution at concentrations of 0.25, 0.5 and 1.0M) and with oxidizing agents (10% sodium hypochlorite solution). In relation to the extraction yield of the soluble fractions, acid extraction was the best. All non-starch polysaccharides extracted from wheat bran had low molecular weight and different sugar composition. The polysaccharides from acid extraction showed more glucose and a lower percentage of arabinose and xylose when compared to water, while those from the alkaline extractions had a lower glucose content and higher arabinose and xylose (compared to the fractions from water). From the analysis of sugars (higher amounts of monosaccharides arabinose, xylose and glucose) and extraction yields, 5 samples were chosen, being the soluble fraction extracted in water, 0.25 and 0.5M H2SO4 and NaOH 0.25 and 0.1M for analyzes of ß-glycan content, phenolic compounds, functional properties (foaming properties, water absorption, solubility and moisture) and prebiotic effect. As a result, all non-starch polysaccharides exhibited high solubility and did not possess relevant foaming and water absorption properties (WAC). The content of phenolic compounds was reduced in all soluble fractions when compared to the phenolic compounds content of wheat bran and the small remaining percentage of phenolic compounds had low reducing power. ß-glucan was found in all fractions of the extracted non-starch polysaccharides, 0.82% in fractions extracted in water, 0.97 and 2.79% in fractions extracted with H2SO4 and 5.42 and 4.62% in fractions extracted with 0.025 and 0.1M NaOH. Although ß-glycan was found in all the extracted samples, only extractions in water and with 0.1M NaOH showed possible prebiotic effect in vitro

The aim of this in vitro study was to assess the root dentin demineralization caused by a microcosm biofilm model that has been exposed to sucrose in different ways. Materials and Methods: Saliva of two volunteers was inoculated into an artificial medium for biofilm growth and dentin blocks were immersed into these media. Dentin specimens were randomly exposed to one of the five experimental conditions: C (control group - no saliva inoculum or sucrose), 0S (saliva inoculum without sucrose, negative control), 3S (three daily one-minute immersions in 20 % sucrose), 6S (six daily one-minute immersions in 20 % sucrose), and CS (continuously immersed in 5 % sucrose). After five days, biofilm was collected to determine the concentration of intracellular and extracellular polysaccharides and the dentin surface hardness loss (SHL) was measured. The experiment was carried out in triplicate. Results: The dentin SHL was higher in groups that were exposed to sucrose (3S, 6S and CS) and there was a statistically significant difference between all groups (p<0.001). CS had higher concentrations of polysaccharides (p>0.001) and there was no statistically significant difference between the other groups (0S, 3S and 6S) (p>0.005). Conclusion: The microcosm biofilm model developed has the potential to produce root dentin demineralization at different exposures to sucrose.


El objetivo de esta investigación in vitro fue evaluar la desmineralización de la dentina radicular causada por un modelo de biofilm microcosmo que fue expuesto de diferentes maneras a la sacarosa. La saliva de dos voluntarios fue colocada en un medio artificial para crecimiento del biofilm y los bloques de dentina fueron sumergidos en estos medios. Al aza rlos bloques fueron expuestos a una de las cinco condiciones experimentales: C (grupo control ­ sin inoculación de saliva o sacarosa), 0S (inoculación de saliva sin sacarosa, control negativo), 3S (tres inmersiones diarias de un minuto en sacarosa a 20 %), 6S (seis inmersiones diarias de un minuto en sacarosa a 20 %), y CS (sumergidos continuamente en 5 % de sacarosa). Después de cinco días, el biofilm fue recogido para determinar la concentración de polisacáridos intracelulares y extracelulares y fue medida la pérdida de dureza superficial de la dentina (SHL). El experimento se repitió en tres ocasiones. La dentina SHL era mayor en los grupos que fueron expuestos a la sacarosa (3S, 6S E CS) y hubo diferencia estadísticamente significativa entre todos los grupos (P<0,001). CS presentó mayor concentración de polisacáridos (p<0,001) y no hubo diferencia estadísticamente significativa entre los demás grupos (0S, 3S E 6S) (p>0,005). El modelo del biofilm desarrollado tiene potencial para producir desmineralización de la dentina radicular en diferentes exposiciones a la sacarosa.

A serosurvey for Mayaro virus (MAYV), eastern (EEEV), western (WEEV) and Venezuelan (VEEV) equine encephalitis viruses was conducted with 237 sheep, 87 free-ranging caimans and 748 equids, including 37 collected from a ranch where a neurologic disorder outbreak had been recently reported. Sera were tested for specific viral antibodies using plaque-reduction neutralisation test. From a total of 748 equids, of which 264 were immunised with vaccine composed of EEEV and WEEV and 484 had no history of immunisation, 10 (1.3%) were seropositive for MAYV and two (0.3%) for VEEV using criteria of a ≥ 4-fold antibody titre difference. Among the 484 equids without history of immunisation, 48 (9.9%) were seropositive for EEEV and four (0.8%) for WEEV using the same criteria. Among the sheep, five were sero- positive for equine encephalitis alphaviruses, with one (0.4%) for EEEV, one (0.4%) for WEEV and three (1.3%) for VEEV. Regarding free-ranging caimans, one (1.1%) and three (3.4%), respectively, had low titres for neutralising antibodies to VEEV and undetermined alphaviruses. The neurological disorder outbreak could not be linked to the alphaviruses tested. Our findings represent strong evidence that MAYV and all equine encephalitis alphaviruses circulated in the Pantanal.

The purpose of this research study was to develop 5-fluorouracil compression coated tablets by using biodegradable polysaccharide polymer locust bean gum (LBG) and hydroxyl propyl methyl cellulose (HPMC) as coating materials. The fast disintegrating core tablets containing 50 mg of 5-fluorouracil were compression coated with LBG and HPMC in different ratios (8:1, 7:2 and 6:3) with a coat weight of 300, 400 and 500 mg. In vitro dissolution data indicated that the formulation (CLH63) with a coat weight of 500 mg containing LBG and HPMC in the ratio 6:3 gave the best release profile (0% in first 5 hour and 96.18% in 24 hours). DSC and FTIR results indicated no possibility of interaction between drug and polymers or other excipients. In vivo human X-ray studies revealed that formulation CLH63 was able to resist breakdown in the stomach and small intestine. The disintegration of the tablet occurred in the colon between 8 to 16 hours of post dose. By the present study, it can be concluded that the LBG and HPMC based compression coated tablets of 5-fluorouracil will be useful strategy for colonic delivery of 5-fluorouracil without being released in upper gastrointestinal region for the safe and effective management of colon cancer.


O propósito desta pesquisa foi desenvolver comprimidos revestidos de fluoruracila utilizando polissacarídio biodegradável polymer locust bean gum (LBG) e hidroxipropilmetil celulose (HPMC) como materiais de revestimento. Os comprimidos de desintegração rápida contendo 50 mg de fluoruracila foram revestidos por compressão com LBG e HPMC em diferentes proporções (8:1, 7:2 e 6:3), com peso de cobertura de 300, 400 e 500 mg. Os dados da dissolução in vitro indicaram que a formulação (CLH63) com peso de cobertura de 500 mg contendo LBG e HPMC na proporção de 6:3 forneceu o melhor perfil de liberação (0% nas primeiras 5 horas e 96,18% em 24 horas). Os resultados de DSC e de FTIR não indicaram interação entre o fármaco e os polímeros ou outros excipientes. Os estudos de raios X in vivo revelaram que a formulação CLH63 foi capaz de resistir à quebra no estômago e no intestino delgado. A desintegração do comprimido ocorreu no cólon, entre 8 e 16 horas após a administração da dose. Pelo presente estudo, concluiu-se que os comprimidos de fluoruracila revestidos com LBG e HPMC por compressão se constituirão em estratégia útil na liberação de fluoruracila no cólon, para o tratamento seguro e efetivo do câncer de cólon, sem que o fármaco seja liberado na região gastrointestinal superior.

Total 95 isolates of Aureobasidium pullulans were isolated from different flowers and leaves samples, out of which 11 thermotolerant strains produced pullulan. One thermotolerant non-melanin pullulan producing strain, designated as RG-5, produced highest pullulan (37.1±1.0 g/l) at 42ºC, pH 5.5 in 48h of incubation with 3% sucrose and 0.5% ammonium sulphate in a non-stirred fed batch fermentor of 6 liters capacity. The two liters of initial volume of fermentation medium was further fed with the 2 liters in two successive batches at 5 h interval into the fermentor. The sterile air was supplied only for 10h at the rate of 0.5 vvm.

This study aimed at optimizing the medium of a new Ganoderma lucidum strain CAU5501 to enhance the yield of exopolysaccharides (EPS) and mycelial growth. Firstly, the suitable level of glucose, magnesium, phosphate and C/N ratio was determined by single factor experiment. Subsequently, the optimum concentrations of these medium components were investigated using the orthogonal matrix method. The results indicated that the higher levels of EPS were correlated with the level of cell growth when glucose concentration was studied (data no show). The optimum medium for EPS yield was found to be 70 g/l glucose, 5 C/N ratio, 2.5 g/l KH2PO4, 0.75 g/l MgSO4·7H2O, and for mycelial growth was 50 g/l glucose, 5 C/N ratio, 1.5 g/l KH2PO4, 0.5 g/l MgSO4·7H2O. When cultivated in the obtained optimal media in 3 L shake flask, compared to the basal medium, the EPS yield increased markedly from 1.003 to 1.723 g/l, and the mycelium formation was also markedly improved from 2.028 to 7.235 g/l. Results obtained in this study are beneficial to further study for enhancing the production of Ganoderma lucidum polysaccharides in large scale commercialized production.

A new trend in cosmetic formulations is the use of biotechnological raw materials as the polysaccharides from Klebsiella pneumoniae, which are supposed to enhance cell renewal, improve skin hydration and micro-relief. Botanical extracts of Myrtus communis leaves contain different sugars, which may provide the same benefits. Thus, the objective of this study was to evaluate through objective and subjective analysis the immediate and long-term effects of cosmetic formulations containing polysaccharides biotechnologically-originated and / or the ones contained in Myrtus communis extracts. Three polysaccharide-based and placebo formulations were applied on the forearm skin of 40 volunteers. Skin hydration, transepidermal water loss (TEWL), viscoelasticity and skin micro-relief measurements were made before and 2 hours after a single application and after 15 and 30 day-periods of daily applications. Answers to a questionnaire about perceptions of formulation cosmetic features constituted the subjective analysis. All polysaccharide-based formulations enhanced skin hydration. Formulations with isolated or combined active substances improved skin barrier function as compared to placebo, in the short and long term studies. Formulations containing Myrtus communis extracts had the highest acceptance. Results suggest that daily use of formulations containing these substances is important for protection of the skin barrier function.


Uma nova tendência em formulações cosméticas é a utilização de matérias-primas biotecnológicas como os polissacarídeos de Klebsiella pneumoniae, que pode aumentar a renovação celular e melhor a hidratação e micro-relevo da pele. Por outro lado, o extrato vegetal de Myrtus communis contém diferentes polissacarídeos, que também podem proporcionar benefícios à pele. Assim, o objetivo do estudo foi a avaliação dos efeitos imediatos e em longo prazo, de formulações cosméticas contendo polissacarídeos obtidos por processo biotecnológico e/ou de extrato de M. communis por meio de análises objetivas e subjetivas. Três formulações contendo os polissacarídeos e um placebo foram aplicadas na pele dos antebraços de 40 voluntários. As medidas foram realizadas antes e após 2 horas da aplicação das formulações e após 15 e 30 dias de aplicações diárias em termos de hidratação da pele, perda transepidérmica de água (TEWL), viscoelasticidade e micro-relevo da pele. Para a análise subjetiva, os voluntários responderam um questionário a fim de obter-se informações sobre a percepção relativa à qualidade de cosméticos. Todas as formulações provocaram aumento da hidratação cutânea. As formulações que continham os polissacarídeos melhoraram a função barreira da pele, em curto e em longo prazo. A formulação contendo extrato de M. communis apresentou maior aceitação. Os resultados sugerem que o uso diário dos polissacarídeos avaliados é importante na proteção da função barreira da pele.

The xylanase production by Cochliobolus sativus strain Cs6 was improved under solid state fermentation (SSF). High xylanase activity (1079 U/g) was obtained when wheat straw was used after 8 days of incubation. Combinations of sodium nitrate with peptone or yeast extract resulted in an increased xylanase production (1543 and 1483 U/g, respectively). The Cs6 strain grown in SSF in a simple medium, proved to be a promising microorganism for xylanase production.


A produção de xilanase por Cochliobolus sativus cepa Cs6 SATIVUS por fermentação em estado sólido (SSF) foi melhorada. Com o emprego de palha de trigo, obteve-se elevada atividade de xilanase (1079 U/g) após 8 dias de incubação. Combinações de nitrato de sódio com peptona ou extrato de levedura aumentaram a produção de xilanase (1543 e 1483 U/g, respectivamente). Comprovou-se que a cepa Cs6, cultivada em SSF em meio simples, é um microrganismo promissor para produção de xilanase.

Degraded glucomannan was isolated from Abrus precatorius Linn. seed polysaccharide (Papilionaceae). Acid hydrolysis and methylation studies produced certain degraded methyl sugars as, 2, 3, 6-tri-O-methyl-D-glucose and 2, 3, 6-tri-O-methyl-D-mannose in 1:4 molar ratio. On the basis of hydrolysis and methylation experiments, a polysaccharide structure has been assigned to the degraded glucomannan and to the parent glucomannan of Abrus precatorius Linn. seed.

Root nodules were collected from the existing plantation of Casuarina equisetifolia from three different agroclimatic zones of Tamil Nadu, India. Morphological variation, histology, histochemistry and nutrient contents of the nodules were studied. The nodules were found to be simple, dichotomously branched and coralloid. The stele in simple and branched nodules reached up to the tip of the nodules. Structure of Frankia nodules was described using histochemical methods to elucidate the nature of micro-symbiont within the nodules. The nodule consisted of a distinct periderm enclosing the cortex. The cortex showed patches of infected cells interspersed among the uninfected ones. Inside the cortex was a stele bound by an endodermis, which contained high amount of tannin. The micro-symbiont was observed in the nodule tisues in hyphal form and spore packed sporocorp was also seen in the cortex. The histochemical tests revealed the presence of proteins, insoluble polysaccharides and polyphenols.

Hidrogéis são potencialmente viáveis para atuar como sistema de liberação específica e controlada de fármacos. Hidrogéis baseados em polissacarídeos são, especialmente, atrativos para serem utilizados como sistema de liberação colo específica, pois a matriz é degradada por bactérias presente no colo. Nesta dissertação, foram desenvolvidos vários hidrogéis, sintetizados a partir de dois diferentes polissacarídeos, como arábica (sistema 1) e galactomanana (sistema 2) quimicamente modificados: Para isso, goma arábica, GA, um exsudato comoso dessecado dostroncos e dos ramos da Acacia senegal e galactomanana (GG) extraída do exsudatoda Psosopis juliflora, foram modificadas com metacrilato de glicidila (GMA). Desde que os reservatórios de flúor (F), cálcio (Ca) e fósforo inorgânico (Pi) na placa dental são suscetíveis a quedas de pH, este estudo in situ cruzado foi conduzido para testar a hipótese de que baixas concentrações destes íons na placa, formada na presença de sacarose, poderiam ser atribuídas simplesmente à fermentação deste açúcar. Onze voluntários utilizaram dispositivos palatinos contendo seis blocos de esmalte dental humano durante duas fases. Em cada fase os tratamentos foram solução de sacarose a 20 por cento ou água destilada deionizada, que foram gotejadas sobre os blocos 8 vezes ao dia. Após 28 dias, em cada fase, a placa dental formada sobre dois blocos foi coletada, o tratamento foi invertido e após um tempo adicional de 24 e 48 horas, o biofilme formado foi coletado dos outros blocos. A concentração de F, Ca e Pi solúvel em ácido e a concentração de polissacarídeo insolúvel (PI) foram determinadas na placa dental. Concentrações estatisticamente menores de F, Ca e Pi, e uma concentração maior de PI foram encontradas no biofilme de 28 dias formado na presença de sacarose do que na sua ausência; após a inversão do tratamento a mudança no F, Ca e Pi não foi estatisticamente significante, mas a concentração de PI mudou significativamente. A hipótese foi rejeitada porque a mudança na concentração de F, Ca e Pi não é devida à fermentação da sacarose.

In many tumors, the amount of chondroitin sulfate in the extracellular matrix has been shown to be elevated when compared to the corresponding normal tissue. Nevertheless, the degree of chondroitin sulfate increase varies widely. In order to investigate a possible correlation between the amount of chondroitin sulfate and tumor size, several individual specimens of human leiomyoma, a benign uterine tumor, were analyzed. The glycosaminoglycans from eight tumors were extracted and compared with those from the respective adjacent normal myometrium. The main glycosaminoglycan found in normal myometrium was dermatan sulfate, with small amounts of chondroitin sulfate and heparan sulfate. In leiomyoma, both dermatan sulfate and chondroitin sulfate were detected and the total amounts of the two galactosaminoglycans was increased in all tumors when compared to normal tissue. In contrast, the heparan sulfate concentration decreased in the tumor. To assess the disaccharide composition of galactosaminoglycans, these compounds were incubated with bacterial chondroitinases AC and ABC. The amounts of L-iduronic acid-containing disaccharides remained constant, whereas the concentration of D-glucuronic acid-containing disaccharides increased from 2 to 10 times in the tumor, indicating that D-glucuronic acid-containing disaccharides are responsible for the elevation in galactosaminoglycan concentration. This increase is positively correlated with tumor size

An enzyme-linked immunosorbent assay was standardized for the detection of cryptococcal antigen in serum and cerebrospinal fluid. The system was evaluated in clinical samples from patients infected by human immunodeficiency virus with and without previous cryptococcosis diagnosis. The evaluated system is highly sensitive and specific, and when it was compared with latex agglutination there were not significant differences. A standard curve with purified Cryptococcus neoformans antigen was settled down for the antigen quantification in positive samples

A crude polysaccharide obtained from mycelium of Fusarium solani by treatment with 2 per center KOH/2h/100§C and fractionated by gel filtration chromatography yielded three fractions denoted L1,L2 and L3. Chemical analysis of the crude polysaccharide showed the presence od 89,5 per center total carbohydrate, 4 per center protin 14 per center uronic acid, traces of phosphate and hexosamine. Mannose, galactose, glucose and unidentifid pentose, were present in a 27.5:34:34.5:4 molar ratio.
