Résumé

Objective To study the efficacy of pemetrexed or fluorouracil in combination with irinotecan in the treatment of advanced colorectal cancer. Methods 68 patients with advanced colorectal cancer were selected from January 2014 to January 2016 in Qinghai Provincial People's hospital. Patients were divided into the control group and the observation group by random grouping, and 34 patients for each group. Patients in the control group were received second-line therapy with fluorouracil and irinotecan. The patients in the observation group were received second-line therapy with the combination of pemetrexed and irinotecan. After treatment, the treatment effects, adverse reactions and living conditions of two groups were compared. Results The total effective rate of the observation group was 38.24%, was higher than that of the control group 8.82% (P<0.05); the observation group's disease control rate was 76.47%, was higher than that of the control group 52.94% (P<0.05). The incidence of adverse reactions in the observation group was 200.00%, which was lower than 305.88% in the control group (P<0.05). Progression free survival time in the observation groupwas (6.81±2.31) months, was higher than the control group (3.75±1.06) months (P<0.05); the total survival time in the observation group was (14.69±4.28) month, was higher than the control group (8.76±2.27) month (P<0.05). Conclusion In the second-line treatment of advanced colorectal cancer, the application of raltitrexed combined with irinotecan treatment, could improve the total efficiency of treatment and disease control rate, reduce adverse reactions, and prolong the survival time of patients.

BACKGROUND:At present there is no specific therapy for the treatment of degenerateive disc diseases. The development of tissue engineering technology provides a new therapy idea for it. OBJECTIVE:To review the research progress of acelular matrix scaffold to construct tissue-engineered intervertebral disc. METHODS:A computer-based online search of PubMed, CNKI, Wanfang and Chinese Biomedical Database was performed for relevant articles published between 2005 to 2014 using the keywords of “intervertebral disc degeneration, extracelular matrix, tissue engineering, scaffold, nucleus pulposus, annulus fibrosus” in English and Chinese, respectively. RESULTS AND CONCLUSION:Construction of tissue-engineered intervertebral disc includes three essential elements: cel scaffolds, seed cels and cytokines. Scaffolds are the key. Acelular matrix scaffold is an important scaffold which has been a hot area of research in scaffolds for tissue-engineered intervertebral disc for several years, which can be divided into three types: acelular annulus fibrosus scaffold, acelular nucleus pulposus scaffold and integrated intervertebral disc scaffold. Different scaffolds have a variety of preparation methods. Acelular matrix scaffold has its own significant advantages, such as low antigenicity, good histocompatibility, and a most similarity with the three-dimensional structure of normal nucleus pulposus that can provide a proper microenvironment for cel growth. However, there are stil some disadvantages of acelular matrix scaffold in tissue-engineered intervertebral disc. So, it needs to be further improved.

Objective To construct tissue engineering nucleus pulposus by culture of rabbit bone marrow mesenchymal stem cells (rBMSCs)-nucleus pulposus acellular matrix scaffold (NPAMS)complexes (rBMSCs-NPAMS).Methods Several NPAMS were prepared,and rBMSCs was seeded into NPAMS.The scaffolds and complex were detected by general observation,HE stai-ning,immunohistochemical,qRT-PCR,scanning electron microscopy.Results The scanning electron microscopy showed the seed cell in nucleus pulposus ECM-derived scaffold could adhesion and growth.The cell attachment and proliferation were observed by HE staining.Immunohistochemical examination with typeⅡ collagen showed positive results.qRT-PCR revealed the time-depend-ent of the mRNA expression of collagen Ⅱ,and which was smaller than positive control(P0.05).Con-clusion Natural nucleus pulposus acellular matrix scaffold composite allogeneic bone marrow mesenchymal stem cells can be suc-cessfully built into tissue engineering nucleus pulposus.

Objective To use detergents and nucleic acid enzyme to prepare scaffold of extracellular ma-trix , then assess the morphological and cytotoxic changes in vitro , and explore the feasibility of this type of scaffold as an ideal tissue-engineering scaffold. Methods Fifty pieces of fresh nucleus pulposus were randomly divided into a fresh control group and a decellularized group. The specimens in decellularized group were treated with 0.3%Tri-ton X-100, 0.5%sodium deoxycholate, and nuclease for 24 h. Morphological changes were studied by macroscopy, pathological staining and scanning electron microscopy. Cytotoxicity was determined by CCK-8 and LIVE/DEAD Viability/Cytotoxicity Assay Kit in vitro. Results The shape of scaffold was maintained,and the extracellular ma-trix was presented while the cells disappeared after decellularization. As compared with the fresh tissue , the scaffold and its extracts had no cytotoxicity to rabbit bone marrow stem cells. Conclusions Almost all the cells have been removed while the extracellular matrix is reserved , and the scaffold has no cytotoxicity to the seed cells. The decel-lularized scaffold can be used as an ideal substance to fabricate tissue-engineering nucleus pulposus.

<p><b>OBJECTIVE</b>To develop an approach to the determination of saponins in Radix Cynanchi Atrati, and to optimize the parameters for purified the preparation of total saponins by macroporous resin column chromatography.</p><p><b>METHOD</b>Using cynanversicoside A as a reference, the determination of saponins was performed; according to the elution rate and the purity of the products, the preparation performance of total saponins by macroporous resin was investigated, and its parameters were optimized.</p><p><b>RESULT</b>The saponins in Radix Cynanchi Atrati were successfully determined at 518 nm by vanillin-perchloric acid as spray reagent. The macroporous resin HP-20 showed static absorption ratio of 59. 3 mg x g(-1); the 70% ethanol extraction of Radix Cynanchi Atrati was eluted from column of macroporous resin HP-20 by water and 30% ethanol, and the saponins were concentrated in 90% ethanol solution. The content of saponin part eluted from HP-20 column was 77.62%.</p><p><b>CONCLUSION</b>The proposed approach allows convenient and efficient preparation and purification of saponin in Radix Cynanchi Atrati.</p>

Objective To observe the effect of early intervention on the oligodendrocyte-myelin glycoprotein(OMgp) mRNA expression of brain injury neonatal rats caused by intrauterine infection.Methods 1.Twenty-eight Wistar pregnant rats were randomly divided into 2 groups:lipopolysaccharide(LPS)-treated group and saline control group.Pregnant rats were consecutively injected with LPS (450 ?g?kg-1) or saline on the 18th gestation age.After birth,the placentas were taken out and made HE staining to observe intrauterine infection.2.Thirty neonatal rats in the saline control group and 55 rats in the LPS-treated group were randomly selected which were divided into intervention group (n=25) and no-intervention group (n=25).The second post-natal day (P2) rats in intervention group were treated by early touch and enriched environment.The neonatal rats in the no-intervention group and saline control group were fed in a routine way.Five cases of P1 rats were selected respectively from the LPS-treated group and saline control group,and brain tissue pathological section was made to observe the condition of brain injury.3.Five cases of P1,P3,P7,P28 and P42 rats were selected from the saline control group,intervention group and no-intervention group to detect the OMgp mRNA expression levels by using the real time polymerase chain reaction me-thod.Results 1.There were a great number of neurophilic granulocytes in the placentas in the LPS-treated group.2.Brain tissue pathological of P1 in the saline control group had complete substantia alba structure,ordered disposition and lightly stained clear chromatospherite.While in the LPS-treated group, there existed brain tissue looseness,colloid cell aggregation and oligodendrocyte cytoreduction in the position of substantia alba,callositas and capsula interna.Intraventricular hemorrhage,substantia alba blood vessel dilatation and blood capillary angiorrhexis and hemorrhage could also be found.3.There was a higher increase in OMgp mRNA expressions of brain tissue in the LPS-treated group at P1,P3,P7,P28,P42 than those in the saline control group (Pa

AIM: To explore the effects of processing on components group of Semen Raphani. METHODS: By comparing HPLC maps of Semen Raphani samples prepared with different methods and computing similarity,(analysis) the change law of the components group. RESULTS: The mechanism of inhibiting the enzyme by processing Semen Raphani was found and C3 was able to produce new compounds A209,B221. CONCLUSION: Processing Semen Raphani can change the components group through influencing the decoction process,eventually produce different clinical effects.
