Résumé

Zika en Embarazadas y Niños (ZEN) is a prospective cohort study designed to identify risk factors and modifiers for Zika virus (ZIKV) infection in pregnant women, partners, and infants, as well as to assess the risk for adverse maternal, fetal, infant, and childhood outcomes of ZIKV and other congenital infections. ZIKV infection during pregnancy may be associated with longterm sequelae. In the ZEN cohort, 1,519 pregnant women and 287 partners were enrolled from 3 departments within Colombia between February 2017 and January 2018, as well as 1,108 infants born to the pregnant women who were followed to 6 months. The data include baseline questionnaires at enrollment; repeated symptoms and study follow-up questionnaires; the results of lab tests to detect ZIKV and other congenital infections; medical record abstractions; infant physical, eye, and hearing exams; and developmental screening tests. Follow-up of 850 mother-child dyads occurred at 9 months, 12 months, and 18 months with developmental screenings, physical exams, and parent questionnaires. The data will be pooled with those from other prospective cohort studies for an individual participant data meta-analysis of ZIKV infection during pregnancy to characterize pregnancy outcomes and sequelae in children.

Résumé

Antiretroviral therapy has been associated with side effects, either from the drug itself or in conjunction with the effects of human immunodeficiency virus infection. Here, we evaluated the side effects of the protease inhibitor (PI) indinavir in hamsters consuming a normal or high-fat diet. Indinavir treatment increased the hamster death rate and resulted in an increase in triglyceride, cholesterol and glucose serum levels and a reduction in anti-oxLDL auto-antibodies. The treatment led to histopathological alterations of the kidney and the heart. These results suggest that hamsters are an interesting model for the study of the side effects of antiretroviral drugs, such as PIs.

O objetivo deste estudo foi testar o PM2.5 de São Paulo sobre o ECG, parâmetros hematológicos e histopatológicos de ratos saudáveis. Os animais receberam instilação de:salina, filtro branco, 100 e 500μg de PM2.5. O SDNN diminuiu 60 minutos após a instilação nos grupos de PM2.5 (p=0.025). Os reticulócitos aumentaram em ambas as doses de PM2.5 (p<0.05). Segmentados, neutrófilos e fibrinogênio diminuíram, enquanto que os linfócitos aumentaram com 100μg de PM2.5 (p<0.05) / The objective of this study was to test Sao Paulo PM2.5 on ECG, hematological and histopathological parameters of healthy rats. Animals received instillation of saline, blank filter, 100 and 500μg of PM2.5. SDNN decreased 60 minutes after instillation in PM2.5 groups (p=0.025). Reticulocytes significantly increased at both PM2.5 doses (p<0.05). Segmented, neutrophils and fibrinogen decreased, while lymphocytes increaseded with 100 μg of PM2.5 (p<0.05). A significant dose-dependent decrease of intra-acinar pulmonary arterioles Lumen/Wall ratio (L/W) was observed in PM groups (p<0.001). Peribronchiolar arterioles L/W showed a significant decrease in the 500μg group (p<0.001)...

Objetivo: Com a finalidade de estabelecer uma linha de pesquisa em cirurgia redutora de volume pulmonar, foi proposta a reprodução de um modelo experimental de enfisema em ratos através da instilação intratraqueal de papaína. Métodos: Foi feita a instilação orotraqueal de papaína (20mg/kg) dissolvida em 3,5ml/kg de solução fisiológica a 0,9 por cento. Após 40 dias da instilação, os animais foram submetidos a mecânica ventilatória, com medidas de elastância e resistência do sistema respiratório, e sacrificados com retirada dos pulmões. O tecido pulmonar dos animais foi analisado qualitativamente com coloração de hematoxilina-eosina e submetido à análise morfométrica com medida do diâmetro alveolar médio. O tecido pulmonar foi também submetido à coloração de resorcina-fucsina, para identificação de fibras elásticas, que foram quantificadas em septos alveolares através de análise digital de imagem. Resultados: A análise histológica dos pulmões dos animais submetidos à instilação de papaína mostrou um enfisema pan-acinar, com rotura de septos alveolares e hiperdistensão alveolar. A análise morfométrica revelou médias superiores de diâmetro alveolar médio nos pulmões dos animais submetidos à papaína (149,08mim e 100,56mim), em comparação com o grupo de solução fisiológica (64,08mim e 75,90mim). A quantificação de fibras elásticas de septos alveolares de animais tratados com papaína foi 70 por cento menor do que a de animais submetidos à solução fisiológica. A mecânica ventilatória não mostrou diferença na resistência do sistema respiratório de animais submetidos à papaína ou à solução fisiológica. Já no caso da elastância do sistema respiratório, esta foi menor nos animais do grupo com papaína, em comparação com o grupo com solução fisiológica, demonstrando comportamento funcional do grupo com papaína compatível com enfisema pulmonar, apresentando diminuição da capacidade de recolhimento elástico do tecido pulmonar. Conclusão: Foi possível reproduzir um modelo experimental de enfisema pulmonar pan-acinar em ratos, através da instilação de papaína pela árvore respiratória, com comprovação funcional e morfológica
