<p><b>OBJECTIVE</b>To examine the expression of lymph vessel endothelial hyaluronan receptor-1 (LYVE-1) in human colorectal carcinoma and to evaluate the relationship of LYVE-1 with lymph mode metastasis and prognosis.</p><p><b>METHODS</b>Colonic cancer samples of 40 cases were collected. The expression of LYVE-1 was determined by RT-PCR and quantified by real-time quantitative PCR. LVD and MVD were detected by immunohistochemistry staining. The relationship of LYVE-1 and LVD with lymph mode metastasis and prognosis were analyzed. All the patients were followed up for at least 3 years.</p><p><b>RESULTS</b>The expression of LYVE-1 and the count of LVD were significantly higher in tumor tissue than those in common colon tissue (P<0.05). In the majority of tumors, the higher count of LVD indicated lymphangiogenesis. The recurrence rates in low LVD group and high LVD group were 46.7% and 60.0% respectively (P<0.05). The survival rates in the above two groups were 90.1% and 56.7% respectively (P<0.05). No significant correlation was found between LYVE-1 and recurrence rate (P>0.05) or overall survival (P>0.05).</p><p><b>CONCLUSION</b>LYVE-1 indicates an increase of lymphangiogenesis in colorectal carcinoma and LVD can be used to evaluate the prognosis for colorectal cancer patients.</p>

<p><b>OBJECTIVE</b>To study the possible role of human thrombospondin (hPWTSR) in gastric cancer and explore its potential to serve as the target for gastric cancer diagnosis and intervention.</p><p><b>METHODS</b>Using pLexA-hPWTSR as the bait, a premade pB42AD-based fetal brain cDNA library was constructed to identify the interacting proteins. The expression pattern of hPWTSR in gastric cancer tissues and a gastric cancer cell line was observed to investigate the correlation between hPWTSR expression and the biological behaviors of the tumor. The possibility of hPWTSR as a potential gastric cancer marker was evaluated.</p><p><b>RESULTS</b>Fifty-seven independent clones were isolated from 107 clones screened. Sequence analysis indicated that the 57 positive clones represented the products of 12 genes. A RT-PCR-based expression pattern revealed that the expression of hPWTSR in gastric cancer tissues and a gastric cancer cell line was lower than that in the corresponding normal tissues, but no mutations were identified by the subsequent sequence analysis.</p><p><b>CONCLUSIONS</b>hPWTSR interacts with adhesion-related proteins and tumor-related genes, and its expression is lowered in gastric cancer tissues and gastric cancer cell line. hPWTSR might play a role in gastric cancer development, especially in metastasis and might be used as a potential gastric cancer marker. The exact functions of hPWTSR and its potential clinical value still await further study.</p>

Objective: To study retrovirus (RV)-mediated transduction of gastric carcinoma cells with the herpes simplex virus thymidine kinase (HSV-tk) gene and the subsequent treatment with ganciclovir(GCV). Methods: The TK gene was transfected into human gastric carcinoma cell line MKN28 using HSV-TK that packed with PA317 cell, the sensitivity of MKN28TK cells to GCV was examined in vitro. Results: The retroviral-mediated HSV-TK gene can be transfected to MKN28 cells. The growth rate of MKN28 cells transfected with HSV-TK gene did not change. MKN28TK cells became significantly sensitive to GCV and had bystander effect. Conclusion: Transfection of gastric carcinoma with HSV-TK has higher transfection efficiency. MKN28TK cells are significantly sensitive to GCV.
