Résumé

OBJECTIVES@#To study the protective effect of breviscapine against brain injury induced by intrauterine inflammation in preterm rats and its mechanism.@*METHODS@#A preterm rat model of brain injury caused by intrauterine inflammation was prepared by intraperitoneal injections of lipopolysaccharide in pregnant rats. The pregnant rats and preterm rats were respectively randomly divided into 5 groups: control, model, low-dose breviscapine (45 mg/kg), high-dose breviscapine (90 mg/kg), and high-dose breviscapine (90 mg/kg)+ML385 [a nuclear factor erythroid 2-related factor 2 (Nrf2) inhibitor, 30 mg/kg] (n=10 each). The number and body weight of the live offspring rats were measured for each group. Hematoxylin-eosin staining was used to observe the pathological morphology of the uterus and placenta of pregnant rats and the pathological morphology of the brain tissue of offspring rats. Immunofluorescent staining was used to measure the co-expression of ionized calcium binding adaptor molecule-1 (IBA-1) and nucleotide-binding oligomerization domain-like receptor protein 3 (NLRP3) in the cerebral cortex of offspring rats. ELISA was used to measure the levels of interleukin-6 (IL-6), interleukin-8 (IL-8), and interleukin-1β (IL-1β) in the brain tissue of offspring rats. Western blotting was used to measure the expression of Nrf2 pathway-related proteins in the brain tissue of offspring rats.@*RESULTS@#Pathological injury was found in the uterus, and placenta tissue of the pregnant rats and the brain tissue of the offspring rats, and severe microglia pyroptosis occurred in the cerebral cortex of the offspring rats in the model group. Compared with the control group, the model group had significant reductions in the number and body weight of the live offspring rats and the protein expression levels of Nrf2 and heme oxygenase-1 (HO-1) in the brain tissue of the offspring rats (P<0.05), but significant increases in the relative fluorescence intensity of the co-expression of IBA-1 and NLRP3, the levels of the inflammatory factors IL-6, IL-8, and IL-1β, and the protein expression levels of NLRP3 and caspase-1 in the brain tissue of the offspring rats (P<0.05). Compared with the model group, the breviscapine administration groups showed alleviated pathological injury of the uterus and placenta tissue of the pregnant rats and the brain tissue of the offspring rats, significant increases in the number and body weight of the live offspring rats and the protein expression levels of Nrf2 and HO-1 in the brain tissue of the offspring rats (P<0.05), and significant reductions in the relative fluorescence intensity of the co-expression of IBA-1 and NLRP3, the levels of the inflammatory factors IL-6, IL-8, and IL-1β, and the protein expression levels of NLRP3 and caspase-1 in the brain tissue of the offspring rats (P<0.05). The high-dose breviscapine group had a significantly better effect than the low-dose breviscapine (P<0.05). ML385 significantly inhibited the intervention effect of high-dose breviscapine (P<0.05).@*CONCLUSIONS@#Breviscapine can inhibit inflammatory response in brain tissue of preterm rats caused by intrauterine inflammation by activating the Nrf2 pathway, and it can also inhibit microglial pyroptosis and alleviate brain injury.

Résumé

Objective: To understand the changes in pain and its effects in patients with the diagnosis of herpes zoster. Methods: A total of 3 487 patients diagnosed with herpes zoster (HZ) for the first time at the outpatient department of Miyun District Hospital from January 1, 2017, to December 31, 2019, were included in the study. The information of patients was registered and issued with a record card. Patients were required to record the time of pain and rash by themselves. Telephone follow-up was conducted at 21, 90, 180 and 365 days after the onset of rashes, including hospitalization, location of rash and pain, and the time of start and end. The impact of pain on life was evaluated by the Zoster Brief Pain Inventory (ZBPI). Results: The age of 2 999 HZ patients included in the analysis were (53±16) years old, including 1 377 (45.91%) males and 1 903 (63.45%) patients aged 50 years and older. After 21 days of rash, mild, moderate and severe pain accounted for 20.87% (626 cases), 37.98% (1 139 cases) and 33.81% (1 014 cases), respectively. Only 5.07% (152 cases) had no pain or discomfort, and 2.27% (68 cases) had no pain but discomfort. Most of the pain sites were consistent with the rash sites. The chest and back and waist and abdomen were the most common, accounting for 35.58% (1 067 cases) and 29.18% (875 cases), respectively, followed by the limbs and face and neck, accounting for 16.74% (502 cases) and 16.40% (492 cases), respectively. The M (Q1, Q3) of pain days in the HZ patients was 14 (8, 20) days, and the incidence of post-herpetic neuralgia (PHN) was 6.63% (171/2 580) (excluding 419 patients who refused to visit or lost to visit on 90 days after the onset of rash). The pain score of HZ patients within 21 days after the rash was (5.19±2.73) points, and the pain score of PHN patients was (7.61±2.13) points, which was significantly higher than that of non-PHN patients [(5.04±2.69) points] (P<0.001). Daily activities, emotions, walking ability, work, social interaction, sleep and recreation were affected for 21 days after the rash in HZ patients, ranging from 60.79% to 83.83%, with sleep being the most affected (83.83%). The impact scores of pain and life dimensions in PHN patients ranged from 4.59 to 7.61 points on the ZBPI scale, which were higher than those in non-PHN patients (2.49-5.04) (t values ranged from 8.86 to 11.67, all P values <0.001). Conclusion: The proportion of pain in HZ patients after the diagnosis is high, and the pain is more obvious in patients with PHN and HZ patients aged 50 and older, which has a greater impact on their daily lives.

Résumé

Objective To investigate the AIDS-related high risk behaviors of men who havesex with men (MSM) who ever exchanged money for the same sex. Methods Target sampling forcross-sectional study and valid anonymous questionnaires were adopted to compare the differences ofAIDS-related high risk behaviors between MSM with the experience of exchanging money for thesame sex and those without that experience. 1959 rstadied samples were recruited in nine cities. SPSS13.0 was used and t, x 2 and Mann-Whitney tests were taken for statistical analysis. ResultsCompared to corresponding ones without "buying" the same sex experience, MSM who ever engagedin "buying" sex had the characteristics of being prone to in marriage and living in larger cities witholder age and higher income (P<0.01). They also had a significant larger number in the followingevents: total sexual panners, anal sex and oral sex episodes with same sex, number of sexualpartners, anal sex and oral sex in the previous six months, with the figure of median 50.0, 20.0,20.0, 5.0,4.0,5.0 respectively. 31.5% had ever participated in 'group sex', 48.0% had sex with malepartners away from his own region in the previous year, 70.5% had sexual intercourse withstrangers at MSM avenues in the last six months. The OR (95% CI) values were 2.288( 1.702-3.077), 3.231 (2.462-4.241 ), 2.840 (2.140-3.770). All the above mentioned figures werehigher than those without the experience, with significant differences. They had a significant lower rate of 45.7% of condom use while having sex with female partners (P<0.05). Conclusion MSM with the experience of exchanging money for the same sex would have more AIDS-related high risk behaviors, AIDS preventative measures should be taken to target different subgroups.

Résumé

Objective To explore the risks on sexual behaviors related to AIDS among men who having sex with men (MSM) in floating population and to develop relevant interventions. Methods Study objectives were recruited through target sampling and anonymous questionnaires were adopted. Male sexual behaviors were compared between floating population and permanent registered residents. Results Among those MSM, the average numbers of same-gender partners, oral sex, anal sex and sex with females during the past 6 months among floating population were 9.61, 8.61, 7.52 and 1.17 respectively. Numbers of partners on oral sex and anal sex during the past month were 2.89 and 3.69 respectively. Both figures were higher than those of the permanent registered residents (P<0.05 or P< 0.01). The rates of condom use among floating population when having anal sex with men or with non-marital female partners were 37.15% and 23.28% respectively. The figures were all higher than those of permanent registered residents' (P<0.05 or P<0.01). Among floating population, the percentages of men who had sex with male sex workers or who had sex with men acquainted through inter-net or with stable female sex partners were 17.89%, 12.56%, and 31.86% respectively. The rates of men who had group sex among gay men, who bled when having sex with men or who had sex with men from other areas during the past year were 22.13%, 31.38% and 32.36% respectively. The figures were all higher than those of permanent registered residents' (P<0.05 or P<0.01). Conclusion Sex-related high risk behaviors were more ubiquitous among floating population than those of permanent registered residents because they were more likely to transmit HIV to people from/to other areas. More attention should be paid to the floating population with MSM in particular, when health interventions are carried out.

The effects of antisense LNA on mouse organs were investigated by HE staining.

RESULTS5 days after LNA injection, serum HBsAg levels in the dual-target group were reduced by 72.8%, and serum HBV DNA levels were decreased by 52.9%. These values were significantly higher than those in the control groups (all P < 0.05). No significant differences were noted in serum ALB, ALT, BUN and CR between the experiment groups and the control groups (all P > 0.05). The expression levels of HBsAg and HBcAg in the liver of dual-target group were significantly lower than those in the control groups. No significant histopathological abnormality was found in liver and kidney tissues in all groups.

CONCLUSIONAntisense LNA targeting to both S and C genes can significantly inhibit HBV replication in transgenic mice.

Résumé

BACKGROUNDThe balance between vasodilation and vasoconstriction plays a major role in maintaining vascular homeostasis. However, the underlying mechanisms are unclear. More and more evidence suggested that there was an interaction in the regulation of vasorelaxation between nitric oxide (NO) and hydrogen sulfide (H(2)S). We explored the interaction between and effects of NO and H(2)S on the relaxation of pulmonary arteries in rats.

METHODSSeven male Sprague-Dawley rats were anaesthetized with chloral hydrate and the pulmonary arteries of each rat separated for the study of vascular activities. The vasorelaxing activities of pulmonary artery rings in response to different doses of a NO donor, sodium nitroprusside (SNP), or a H(2)S donor, sodium hydrogen sulfide (NaHS), were measured in vitro. When pulmonary artery rings were treated with a cystathionine-gamma-lyase inhibitor, DL-propargylglycine, in the presence of SNP or a nitric oxide synthase inhibitor, N(omega)-nitro-L-arginine methyl ester, in the presence of NaHS, the changes in relaxing activities were analyzed.

RESULTSThe relaxation of pulmonary artery rings was in a dose dependent manner in response to either SNP or NaHS. The relaxation rates of pulmonary artery rings increased from (30.90+/-4.62)% to (60.50+/-8.08)% when the concentration of SNP increased from 1 micromol/L to 3 micromol/L and from (26.13+/-4.12)% to (53.09+/-14.01)% when the concentration of NaHS increased from 25 micromol/L to 100 micromol/L. However, when appropriate inhibitor was added, the relaxation responses to SNP and NaHS decreased.

CONCLUSIONSThe results suggested that similarly to NO, H(2)S acted as a vasorelaxant either independently of, or synergistically with NO in the regulation of vasorelaxation. The interaction between NO and H(2)S played an important role in regulating relaxing activities of pulmonary arteries.
