Résumé

20C, a bibenzyl compound isolated from Gastrodia elata, possesses antioxidative properties in PC12 cells, but its in-depth molecular mechanisms against rotenone-induced neurotoxicity remains unknown. Recent studies indicate that without intact DJ- 1, nuclear factor erythroid 2- related factor (Nrf2) protein becomes unstable, and the activity of Nrf2-mediated downstream antioxidant enzymes are thereby suppressed. Therefore, increasing the nuclear translocation of Nrf2 by DJ-1 may present a helpful means for the prevention and treatment of chronic diseases related to oxidative stress. Our results showed that 20C clearly protected PC12 and SH-SY5Y cells against rotenone-induced oxidative injury in a concentration-dependent manner. Furthermore, 20C markedly up-regulated the levels of DJ-1, which in turn activated phosphoinositide-3-kinase (PI3K)/Akt signaling and inhibited glycogen synthase kinase 3β (GSK3β) activation, eventually promoting Nrf2 nuclear translocation and inducing the expression of Nrf2-mediated downstream antioxidative enzymes such as HO-1. The antioxidative effects of 20C could be partially blocked by ShRNA-mediated knockdown of DJ-1 and inhibition of the PI3K/Akt pathways with Akt1/2 kinase inhibitor in PC12 and SH-SY5Y cells, respectively. Conclusively, our findings confirm that DJ- 1 is necessary for 20C- mediated protection against rotenone- induced oxidative damage, at least in part, by activating PI3K/Akt signaling, and subsequently enhancing the nuclear accumulation of Nrf2. The findings from our investigation suggest that 20C should be developed as a novel candidate for preventing or alleviating the consequences of PD in the future.

Infecções de corrente sanguínea por leveduras do gênero Candida são uma das principais causas de morbidade e mortalidade em pacientes imunocomprometidos. Candida albicans permanece a espécie mais isolada nestas infecções e é de fácil e rápida identificação. Contudo, existem outras espécies, como C. parapsilosis, C. tropicalis, C. glabrata e C. krusei, que são encontradas com menor frequência e que necessitam de maior período de tempo e de metodologias comerciais automatizadas ou semi-automatizadas para sua identificação. Neste estudo foram analisadas 146 cepas de leveduras quanto à capacidade do Sistema API 20C AUX® (Biomerieux®, França) em identificar corretamente o gênero e a espécie de microrganismos em diferentes períodos de leitura, visando-se a liberação do resultado em menor tempo. C. parapsilosis, C. guilliermondii, C. pelliculosa, C. colliculosa, Rhodotorula mucilaginosa, Saccharomyces cerevisae, Trichosporon mucoides e T. asahii foram as leveduras cujos resultados finais puderam ser liberados nos períodos de tempo de 96, 120 e 144 h. Oitenta por cento das C. glabrata e 69 % das C. tropicalis também foram identificadas nos períodos além do tempo estabelecido. Com os resultados obtidos é possível antecipar a identificação do gênero e de algumas espécies de leveduras...

O desenvolvimento de técnicas de criopreservação utilizando temperaturas comerciais (-20 °C) na conservação de Basidiomicetos é uma vantagem técnica e econômica para a produção de biomassa e/ou biocompostos, porém geralmente associada a problemas de danos celulares. O objetivo deste trabalho foi avaliar o uso de substratos para o crescimento do fungo associado a agentes crioprotetores na criopreservação a -20 ºC e a -70 ºC do gênero Pleurotus. O fungo, crescido em ágar-batata-dextrose ou em grãos de aveia, foi criopreservado a -20 °C e a -70 °C na presença dos crioprotetores glicerol, dimetilsulfóxido, glicose, sacarose, extrato de malte e polietilenoglicol. Foram discutidas as vantagens do substrato em associação com o crioprotetor, nas diversas temperaturas de crioproteção. Concluiu-se que o substrato utilizado para o crescimento do micélio influencia na viabilidade de recuperação do fungo criopreservado, em ambas as temperaturas, sendo que grãos de aveia promovem melhor recuperação do fungo e melhor vigor micelial. Os resultados sugerem uma simplificação técnica e redução de custos de manutenção de linhagens de Basidiomicetos.


The development of cryopreservation techniques using commercial temperatures (-20 °C) to preserve Basidiomycetes is an economical advantage for biomass and/or biocompound production; however, it is usually associated to cellular damages. The objective of this work was to evaluate the use of substrates for the fungus growth associated to cryoprotectants for the cryopreservation of Genus Pleurotus at -20ºC and -70ºC. The fungus, grown in either potato dextrose agar or oat grains, was cryopreserved at -20 °C or -70 °C with glycerol, dimethyl sulfoxide, glucose, sucrose, malt extract and polyethyleneglycol. The advantages of the substrate associated with the cryoprotectant at different cryopreservation temperatures were discussed. It was concluded that the substrate used for the mycelial growth positively influences the viability of the recovery of the cryopreserved fungus on both temperatures – oat grains promote better recovery and mycelial vigor. Those results suggest technical simplification and reduction of maintenance costs of culture collections of Basidiomycetes.


El desarrollo de técnicas de criopreservación utilizando temperaturas comerciales (-20ºC) en la conservación de Basidiomicetos es una ventaja técnica y económica para la producción de biomasa y/o biocompuestos, pero generalmente está asociada a problemas de daños celulares. El objetivo de esta investigación fue evaluar el uso de substratos para el crecimiento del hongo, asociado a agentes crioprotectores en la criopreservación a -20ºC y a -70ºC del género PleurotUs. Elhongo, crecido en agar-patata-dextrosa o en granos de avena, fue criopreservado a -20ºC y a -70ºC en la presencia de los crioprotectores glicerol, dimetilsulfóxido, glucosa, sacarosa, extracto de malte y polietilenoglicol. Fueron discutidas las ventajas del substrato en asociación con el crioprotector, en las diversas temperaturas de crioprotección. Se concluyó que el substrato utilizado para el crecimiento del micelio influencia en la viabilidad de recuperación del hongo criopreservado, en ambas las temperaturas, siendo que granos de avena promueven mejor recuperación del hongo y mejor vigor del micelio. Los resultados sugieren una simplificación técnica y reducción de costos de manutención de liñajes de Basidiomicetos.

BACKGROUND: Rapid and reliable identification of Candida albicans becomes more important because the incidence of yeast infections has increased in recent years. Murex C. albicans 50(CA50) assay was developed for rapid and presumptive identification of C. albicans. We evaluated the CA50 assay to identify C. albicans. METHOD: Seventy four yeast isolates from clinical specimens were tested with CA50 and germ tube test. They were identified to species level by API 20C and chlamydospore agar test. RESULT: Of the 74 isolates, 52 were C. albicans and 22 were non-albicans Candida. The sensitivity and specificity for CA50 were 88.5% and 86.4%, respectively. The sensitivity and specificity of the germ tube test using human serum were 88.5% and 90.9% respectively, and those using fetal bovine serum(FBS) were 92.3% and 90.9% respectively. CONCLUSION: CA50 was a rapid and easy-to-perform test, and it was comparable to germ tube test in regard of sensitivity and specificity.
