Résumé

PURPOSE: This study evaluated powdered burn wound dressing materials from wild silkworm fibroin in an animal model. METHODS: Fifteen rats were used in this experiment. Full-thickness 2x2 cm burn wounds were created on the back of rats under anesthesia. In the two experimental groups, the wounds were treated with two different dressing materials made from silkworm fibroin. In the Control Group, natural healing without any dressing material was set as control. The wound surface area was measured at five days, seven days and 14 days. Wound healing was evaluated by histologic analysis. RESULTS: By gross observation, there were no infections or severe inflammations through 14 days post-injury. The differences among groups were statistically significant at seven days and 14 days, postoperatively (P<0.037 and 0.001, respectively). By post hoc test, the defect size was significantly smaller in experimental Group 1 compared with the Control Group and experimental Group 2 at seven days postoperatively (P=0.022 and 0.029, respectively). The difference between Group 1 and Group 2 was statistically significant at 14 days postoperatively (P<0.001). Group 1 and control also differed significantly (P=0.002). Group 1 showed a smaller residual scar than the Control Group and Group 2 at 14 days post-injury. Histologic analysis showed more re-epithelization in Groups 1 and 2 than in the Control Groups. CONCLUSION: Burn wound healing was accelerated with silk fibroin spun by wild silkworm Antheraea pernyi. There was no atypical inflammation with silk dressing materials. In conclusion, silk dressing materials can be used for treatment of burn wound.

Résumé

Objective: To investigate the effect of the extract from male zooid of Ahtheraea pernyi on the expression of hypoxia-inducible factor-1α (HIF-1α) in tumor-bearing rats after radiotherapy. Methods: W256 tumor model was established by continuous ig administration of the extract from the male zooid of A. pernyi (16.53 mg/kg) for 10 d after conventional radiotherapy. To observe the expression of HIF-1α and vascular endothelial growth factor (VEGF) in tumor tissue by immunohistochemical staining and the HIF-1α mRNA expression by in situ hybridization (ISH). Results: Compared with the model group, immunohistochemistry results showed the expression of HIF-1α and VEGF increased with tendency (P>0.05) in tumor tissue of the rats in radiotherapy group, and the HIF-1α and VEGF expression in the combination group and the extract from the male zooid of A. pernyi group decreased significantly (P0.05) in radiotherapy group compared with the model group. However, the HIF-1α mRNA expression in tumor tissue was lower in both the combined group and the extract from male zooid of A. pernyi group (P<0.05). Conclusion: The extract from male zooid of A. pernyi could down-regulate the HIF-1α expression in tumor tissue after radiotherapy or without radiotherapy.

ApNPV PstI-B was 7406 bp long, contained seven open reading frames (orfs)/genes, including p87, he65, pnk/pnl, odv-ec43 and Orgyia pseudotsugata multicapsid nucleopolyhedrovirus (OpMNPV) orf107,orf108 homologue on either strands of genomic DNA. ApNPV PstI-C was 6663 bp long, contained eleven orfs/genes, including pk-1, orf1629, polh, lef-2, ptp-2, ctl-1, ptp-1 and OpMNPV orf5, orf7, orf8, orf1 1 homologue on either strands of genomic DNA. Among the eighteen baculovirus genes identification, he65 and orf1629 were two diverse genes, while polh and lef-2 were two conserved genes. ApNPV was the third baculovirus found to contain pnk/pnl gene, the fourth baculovirus found to contain both ptp-1 and ptp-2 gene.
