Résumé

La importancia que tienen para la avicultura cubana el virus de la enfermedad infecciosa de la bolsa (Gumboro) y el virus de la viruela aviar, así como la producción de vacunas que permitan controlar las enfermedades producidas por estos agentes biológicos, justifican la necesidad del establecimiento de una buena gestión de la bioseguridad, ya que el desconocimiento de los peligros y riesgos del personal que labora en estas vacunas puede provocar accidentes de consecuencias indeseables para el producto, escapes de estos microorganismos durante sus procesos productivos y la consecuente contaminación del medio ambiente. El objetivo de la presente investigación fue realizar un análisis de la percepción de riesgo existente en el personal responsable del proceso de producción de dos vacunas aviares. Para ello se utilizó el software RISKPERCEP en una instalación de producción de vacunas aviares; su aplicación mostró variables que demostraron subestimación del riesgo por el personal expuesto y variables con tendencia a la sobrestimación, asociadas fundamentalmente al incorrecto diseño de la instalación. Se concluye que existe la necesidad de una buena capacitación y que se impartan cursos de actualización de bioseguridad donde se tengan en cuenta todos los aspectos del diseño del laboratorio que puedan solucionarse(AU)


The importance of infectious bursal disease virus and fowl pox virus for Cuban poultry farming, as well as the production of vaccines to control the diseases caused by these biological agents, justifies the need for establishment of a good Biosafety management; since the ignorance of the dangers and risks on the part of the personnel that works in them can cause accidents with undesirable consequences for the product, escapes of these microorganisms during their production processes and the consequent contamination of the environment. The objective of the research was to carry out an analysis of the perception of risk in the personnel responsible for the production process of two avian vaccines. The RISKPERCEP software was used in an avian vaccine production facility; its application showed variables that demonstrated underestimation of the risk by the exposed personnel and variables with a tendency to overestimate; fundamentally associated with the incorrect design of the facility. Finally, it is proposed that biosafety update courses be given and that all aspects of the laboratory design that can be solved are taken into account(AU)

Résumé

Black-bone silky fowl, sweet, pungent, and hot-natured, is one of the valuable domesticated birds with special economic value in China's genebank of poultry breed, which has a long history of medicinal and edible uses. It has the effects of tonifying liver and kidney, replenishing Qi and blood, nourishing yin, clearing heat, regulating menstruation, invigorating spleen, and securing essence. Therefore, it has remarkable efficacy of enhancing physical strength, tonifying blood, and treating diabetes and gynecological diseases. Various local black-bone silky fowl breeds have been generated due to the differences in environmental conditions, breed selection, and rearing conditions in different areas of China, which are mainly concentrated in Taihe, Wan'an, and Ji'an in Jiangxi province and Putian, Jinjiang, and Yongchun in Fujian province. The indigenous chicken breeds in China have different body sizes, appearance, coat colors, etc. The complex lineages lead to extremely unstable genetic traits. The diverse breeds similar in appearance result in the confusion in the market of silky fowl breeds. With the rapid development of molecular biological technology, the genetics of black-bone silky fowls has been intensively studied. This article reviews the research progress of the germplasm resources, genetic diversity, and breed identification of black-bone silky fowl in China at the morphology, chromosome, protein, and DNA levels. Further, it introduces the principles, application status, and limitations of DNA markers such as mitochondrial DNA, microsatellite markers, and SNPs. This review provides a theoretical basis for the mining of elite trait genes and the protection and utilization of local black-bone silky fowl germplasm resources in China.

Résumé

Salmonelose é uma doença causada por bactérias do gênero Salmonella, com importância para saúde pública e animal. Dentre os sorotipos hospedeiro-específicos, destaca-se o Gallinarum, que possui os biovares Gallinarum e Pullorum adaptados às aves e amplamente difundidos pelo mundo. Os dados sobre a ocorrência de Salmonella spp. em criações avícolas alternativas no Brasil são escassos. O objetivo deste estudo foi pesquisar a ocorrência de Salmonella spp. em galinhas coloniais encaminhadas para necropsia ao LRD/FV/UFPel. Foram realizadas análises histopatológicas, microbiológicas e moleculares das colônias bacterianas isoladas de 12 amostras de órgãos de galinhas domésticas dos municípios de Pelotas e Piratini, no Rio Grande do Sul. Na análise microbiológica, foram isoladas bactérias do gênero Salmonella sorotipo Gallinarum das 12 amostras, sendo 10/12 bioquimicamente compatíveis com biovar Gallinarum e 2/12 com biovar Pullorum. Na análise molecular PCR 11/12, 91,7% foram identificadas genotipicamente como Salmonella spp. O presente estudo demonstrou uma elevada frequência de isolamento de Salmonella Gallinarum biovar Gallinarum em aves sintomáticas criadas em regime extensivo. Além disso, os dados epidemiológicos das aves analisadas demonstram que a infecção por Salmonella Gallinarum nesses casos está associada ao contato com aves silvestres e falhas de manejo sanitário.(AU)

Abstract Myiopsitta monachus is an invasive psittacine with wide distribution due to the pet trade. Its large communal nests and synanthropic nature contribute to its successful colonization of cities, from where it seems to be expanding in range and numbers. This is relevant with regard to pathogens that invasive species may harbor, especially when host populations thrive. We aimed to identify an abundant mite found in invasive monk parakeet chicks that had been collected in Santiago during 2017 and 2018. Through morphological and molecular identification of the 18S ribosomal RNA gene, we confirmed the presence of Ornithonyssus bursa. This was the first report of this mite in Chile. This mite is common in native and invasive monk parakeet populations and may affect other birds, including domestic fowl. Further, this mite bites people and can be a potential vector of pathogens such as bacteria or viruses. We conclude that this parasite was likely introduced with the parakeet and discuss possible ecological, health and economic consequences of this new potential pest.


Resumo Myiopsitta monachus é um psitacídeo invasor amplamente distribuído devido ao tráfico de animais selvagens. Os grandes ninhos comunitários construídos e sua condição de espécies sinantrópicas contribuem para a colonização bem-sucedida das cidades, onde parece estar expandindo sua distribuição e número de indivíduos. Isso é relevante, quando se trata de patógenos que os invasores podem abrigar, especialmente quando as populações hospedeiras prosperam. O objetivo deste trabalho foi identificar um ácaro abundante, encontrado em filhotes de periquitos-monge introduzidos em Santiago, Chile, coletados durante 2017 e 2018. Por meio da identificação morfológica e molecular do gene do RNA ribossômico 18S, foi confirmada a presença de Ornithonyssus bursa, sendo o primeiro registro para o Chile. Esse ácaro é comum em populações nativas e introduzidas de periquitos-monge e pode afetar outras aves, incluindo aves domésticas. Além disso, esse ácaro pode picar pessoas e pode ser um potencial vetor de patógenos, como bactérias ou vírus. Conclui-se que esse parasita provavelmente foi introduzido com o periquito e foram discutidas as possíveis consequências ecológicas de saúde e econômicas dessa nova praga em potencial.

ABSTRACT Although the Colombian poultry industry has almost doubled its production in the last decade, our ability to diagnose and characterize avian pathogens is deficient, and there is little information of the circulating viral pathogens. One of these pathogens is Marek disease virus (MDV), which is an immunosuppressive agent that can cause high mortality rates and substantial production losses. Currently, there are few documented clinical cases due to the implementation of mass vaccination programs with GaHV-2 strains (serotype I) and HVT (serotype III). However, losses in production rates are likely occurring-due to immunosuppression and subclinical infections. The objective of this review is to describe MDV and the current status of the infection in Colombia.


RESUMEN Aunque la industria avícola colombiana ha crecido casi el doble en producción durante la última década, el diagnóstico de agentes infecciosos y caracterización de estos aún es escasa, y es poca la información acerca de las cepas virales circulantes en el país. Dentro de estos agentes se encuentra el Virus de la Enfermedad de Marek (VEM), el cual es un patógeno inmunosupresor que puede causar mortalidad elevada y graves pérdidas en la producción. Aunque es poco probable que ocurran casos clínicos de la enfermedad causada por el VEM, debido a los programas de vacunación generalizada con GaHV-2 (serotipo I) y HVT (serotipo III), la inmunosupresión que causan las infecciones subclínicas puede estar causando pérdidas considerables en la producción avícola nacional. El objetivo de esta revisión es describir brevemente la enfermedad de Marek y el estado actual del estudio de la infección en Colombia.

Résumé

Abstract Ornithonyssus bursa, known as the "tropical fowl mite", is a hematophagous mite of domestic and wild birds, occasionally biting humans. Infestation on humans occurs mainly when the abandoned nests are close to homes, or by manipulation of infested birds by humans. In Brazil, this species occurs in the south and southeast of the country. In the present study we are reporting bites on humans, new localities records, host associations, and molecular information of O. bursa.


Resumo Ornithonyssus bursa, conhecido como "ácaro tropical de galinha", é um ácaro hematófago de aves domésticas e silvestres, ocasionalmente picando humanos. A infestação em humanos ocorre principalmente quando os ninhos abandonados de aves estão próximos de casas, ou pela manipulação de humanos de aves infestadas. No Brasil, esta espécie ocorre na região sul e sudeste do país. No presente estudo, estamos relatando picadas em humanos, registros de novas localidades de ocorrência, novo hospedeiro e informações moleculares de O. bursa.

Fowl adenovirus serotype 4 (FAdV-4) strain SD1511 was isolated from chickens with severe inclusion body hepatitis and hydropericardium syndrome in Shandong Province, China. The isolate was cultured in primary chicken embryo kidney cells. A study of pathogenicity indicated that SD1511 readily infected 7-35-d-old chickens by intramuscular injection and intranasal and oral routes, causing 50%-100% mortality. The 35-d-old chickens suffered more severe infection than 7- and 21-d-old chickens with mortality highest in the intramuscular injection group. The serum from surviving chickens showed potent viral neutralizing capability. The complete genome of SD1511 was sequenced and analyzed. The strain was found to belong to the FAdV-4 cluster with more than 99% identity with the virulent FAdV-4 strains isolated in China in recent years except for some distinct variations, including deletions of open reading frame 27 (ORF27), ORF48, and part of ORF19. Our findings suggest that SD1511 might be used as a prototype strain for the study of pathogenesis and vaccine development.

Résumé

Avian malaria is one of the most important general blood parasites of poultry in Southeast Asia. Plasmodium (P.) juxtanucleare causes avian malaria in wild and domestic fowl. This study aimed to identify and characterize the Plasmodium species infecting in Thai native fowl. Blood samples were collected for microscopic examination, followed by detection of the Plasmodium cox I gene by using PCR. Five of the 10 sampled fowl had the desired 588 base pair amplicons. Sequence analysis of the five amplicons indicated that the nucleotide and amino acid sequences were homologous to each other and were closely related (100% identity) to a P. juxtanucleare strain isolated in Japan (AB250415). Furthermore, the phylogenetic tree of the cox I gene showed that the P. juxtanucleare in this study were grouped together and clustered with the Japan strain. The presence of P. juxtanucleare described in this study is the first report of P. juxtanucleare in the Thai native fowl of Thailand.

Abstract Salmonella Gallinarum is a host-restrict pathogen that causes fowl typhoid, a severe systemic disease that is one of the major concerns to the poultry industry worldwide. When infecting the bird, SG makes use of evasion mechanisms to survive and to replicate within macrophages. In this context, phoPQ genes encode a two-component regulatory system (PhoPQ) that regulates virulence genes responsible for adaptation of Salmonella spp. to antimicrobial factors such as low pH, antimicrobial peptides and deprivation of bivalent cations. The role of the mentioned genes to SG remains to be investigated. In the present study a phoPQ-depleted SG strain (SG ΔphoPQ) was constructed and its virulence assessed in twenty-day-old laying hens susceptible to fowl typhoid. SG ΔphoPQ did cause neither clinical signs nor mortality in birds orally challenged, being non-pathogenic. Furthermore, this strain was not recovered from livers or spleens. On the other hand, chickens challenged subcutaneously with the mutant strain had discreet to moderate pathological changes and also low bacterial counts in liver and spleen tissues. These findings show that SG ΔphoPQ is attenuated to susceptible chickens and suggest that these genes are important during chicken infection by SG.

Résumé

The present study aimed to document the seasonal variations in the testicular morphology of the adult guinea fowl (Numida meleagris) under tropical savannah climate of India. The study was conducted in 24 adult healthy guinea cocks. The testes were ovoid in shape throughout the year. The consistency of the testes was moderately firm with great resilience and creamy white in colour during summer and monsoon-I whereas soft in consistency and dull white to yellowish tinge in colour during monsoon-II and winter seasons. The testicular weight, length, width, thickness, volume and circumference were higher during summer and monsoon I than the winter and monsoon II seasons. The mean body weight of the birds did not show significant difference among the seasons. The testicular weight, length, width, thickness, volume and circumference were higher during the summer and monsoon I than the winter and least during monsoon II in the current study. The gonadosomatic index (GSI) and paired testicular weight were high and showed no significant difference during the summer and monsoon-I whereas during winter and monsoon-II, the values were low and showed significant difference between them. The diameter of the seminiferous tubules and its lumen, the diameter of the sustentacular cells (Sertoli cells) were much higher during summer and Monsoon I than winter and monsoon II seasons. The percentage area of interstitial cells and the thickness of the testicular capsule appeared less during the summer and monsoon I seasons. The decreased size of seminiferous tubules, increased amount of cellular debris in the tubular lumen, degenerating germ cells and collapse of seminiferous tubules observed during monsoon II and winter in the present study.


El presente estudio tuvo como objetivo documentar las variaciones estacionales en la morfología testicular de la gallina de Guinea (Numida meleagris) en el clima de la sabana tropical de la India. El estudio se realizó en 24 gallos adultos sanos de Guinea. Los testículos fueron ovoidedurante todo el año. La consistencia de los testículos fue moderadamente firme con gran elasticidad y color blanco cremoso durante el verano y durante el Monzón I, mientras que de consistencia blanda y color blanco pálido a amarillento durante el Monzón II y las temporadas de invierno. El peso testicular, la longitud, el ancho, el grosor, el volumen y la circunferencia fueron más altos durante el verano y el Monzón I en comparación con las temporadas de invierno y Monzón II. El peso corporal medio de las aves no mostró una diferencia significativa entre las estaciones. El peso testicular, la longitud, el ancho, el grosor, el volumen y la circunferencia fueron más altos durante el verano y el Monzón I en comparación con el invierno y menos durante el Monzón II. El índice gonadosomático (GSI) y el peso testicular apareado fue alto y no mostraron diferencias significativas durante el verano y el Monzón I, mientras que durante el invierno y el Monzón II, los valores fueron bajos y mostraron diferencias significativas entre ellos. El diámetro de los túbulos seminíferos y su luz, el diámetro de las células sustentaculares (células de Sertoli) fue mucho mayor durante el verano y el Monzón I que en las estaciones de invierno y el Monzón II. El área de porcentaje de células intersticiales y el grosor de la cápsula testicular aparecieron menos durante las temporadas de verano y Monzón I. Se identificaron en el presente estudio disminución del tamaño de los túbulos seminíferos, el aumento de la cantidad de restos celulares en la luz tubular, la degeneración de las células germinales y el colapso de los túbulos seminíferos observados durante el Monzón II y el invierno.

Fowl adenovirus (FAdV) is distributed worldwide and causes economic losses in the poultry industry. The objectives of this study were to determine the hexon and fiber gene changes in an attenuated FAdV isolate from Malaysia in specific pathogen-free chicken embryonated eggs (SPF CEE) and its infectivity in commercial broiler chickens. SPF CEE were inoculated with 0.1 mL FAdV inoculum via the chorioallantoic membrane (CAM) for 20 consecutive passages. The isolate at passage 20 (E20), with a virus titer of 10(8.7)TCID₅₀/mL (TCID₅₀, 50% tissue culture infective dose), was inoculated (0.5 mL) into one-day-old commercial broiler chicks either via oral or intraperitoneal routes. The study demonstrated that 100% embryonic mortality was recorded from E2 to E20 with a delayed pattern at E17 onwards. The lesions were confined to the liver and CAM. Substitutions of amino acids in the L1 loop of hexon at positions 49 and 66, and in the knob of fiber at positions 318 and 322 were recorded in the E20 isolate. The isolate belongs to serotype 8b and is non-pathogenic to broiler chickens, but it is able to induce a FAdV antibody titer. It appears that molecular changes in the L1 loop of hexon and the knob of fiber are markers for FAdV infectivity.

Fowl Cholera (FC) is a disease caused by Pasteurella multocida. The severity of this disease is partly caused by virulence factors. Genes encoding fimbriae, capsule, sialidases and proteins for iron metabolism may be related to P. multocida's ability to infect the host. Besides to examining DNA for the presence of virulence genes, DNA is essential for the diagnostic and FTA cards are an alternative for genetic material transport. The study aims to evaluate the viability of P. multocida DNA transport using the cards and to detect 14 virulence genes in 27 strains isolated from FC cases in the United States by multiplex-PCR. No growth was observed in any of the FTA cards, which was essential to assess the security. Furthermore, DNA detection was possible in 100% of the samples, independent of the storage period (7 to 35 days) and temperature (4°C and 37°C). ptfA, exbd-tonB, hgbA, nanB, oma87, hyaD-hyaC, sodC, hgbB, sodA, nanH and pfhA genes were detected in more than 80% of the samples. FTA cards have proven to be a viable and safe tool for DNA transport of P. multocida. A majority of genes showed a high frequency, which was similar to strains isolated from FC cases.(AU)


Cólera aviária (CA) é uma doença causada pela bactéria Pasteurella multocida e a severidade dos casos é em parte justificada por fatores de virulência. Genes codificando fímbrias, cápsulas, sialidases, dismutases e proteínas do metabolismo férrico podem ser relacionados à capacidade do agente em infectar o hospedeiro. Além da obtenção do DNA para pesquisa de genes de virulência, o material genético é fundamental para o diagnóstico, e os cartões FTA seriam uma alternativa no transporte de microrganismos. Os objetivos da presente pesquisa foram avaliar a viabilidade do transporte de DNA de P. multocida através dos cartões e detectar 14 genes de virulência em 27 cepas isoladas de CA nos Estados Unidos, por meio de multiplex-PCR. Nenhuma das amostras para análise microbiológica da segurança dos cartões apresentou crescimento. Foi possível a detecção do DNA em 100% das amostras, independentemente do tempo de estocagem (sete a 35 dias) e das temperaturas (4°C e 37°C) avaliadas. Genes ptfA, exbd-tonB, hgbA, nanB, oma87, hyaD-hyaC, sodC, hgbB, sodA, nanH e pfhA foram detectados em mais de 80% das amostras. Os cartões FTA demonstraram ser uma ferramenta viável e segura para o transporte do DNA de P. multocida. A maioria dos genes apresentou uma alta frequência, compatível com isolados de CA.(AU)

SUMMARY: The morphology of the interstitial tissue of sexually active and resting testis of the guinea fowl were studied. Six adult health birds of active and resting phases of reproductive cycle were used for this study. The interstitial tissue consisted of loose connective tissue, interstitial cells (Leydig cells), few connective cells, blood vessels and adrenergic nerve fibres in the present study in both active and resting testes. The interstitial tissue was compact in sexually active testis whereas, the volume of the tissue was found to be increased in resting testis. The loose connective tissue of the interstitial tissue composed of mainly of collagen fibres and few reticular fibres whereas elastic fibres were absent in both groups studied. The interstitial cells appeared in clusters of a few cells and were relatively less in the active testis than the resting testis. The interstitial cells were pale staining or polygonal cells with euchromatic nuclei with few large lipid droplets in the active testis whereas the cells were flat and highly heterochromatic with numerous small lipid droplets in resting testis. Few macrophages were found only in resting testis. Interstitial cells showed negative reaction to alkaline, acid phosphatases and PAS in both groups studied but positive for lipids. The interstitial tissue was well vascularised with centrally located blood vessels in the active testis whereas the blood vessels were small and inconspicuous in the resting testis. The lymphatic vessels were not identified in both groups studied.


RESUMEN: Se estudió la morfología del tejido conectivo intersticial en testículos sexualmente activos y en reposo de la gallina de Guinea (Numida meleagris). Se utilizaron seis gallinas de Guinea machos adultos sanos, en fase activa y de reposo del ciclo reproductivo. El tejido conectivo intersticial estaba formado por tejido conectivo laxo, células endocrinas intersticiales, pocas células conectivas, vasos sanguíneos y fibras nerviosas adrenérgicas, tanto en testículos activos como en reposo. El espacio intertubular en los testículos sexualmente activos era menor en comparación a los del testículos en reposo. El tejido conectivo laxo estaba compuesto principalmente de fibras colágenas y en menor cantidad de fibras reticulares. Las fibras elásticas estaban ausentes en ambos grupos. Las células endocrinas intersticiales se organizaban en racimos de pocas células y se observaban con menor frecuencia en los testículos sexualmente activos. Las células endocrinas intersticiales de los testículos activos presentaban forma poligonal, citoplasma levemente eosinofílico con algunas gotas lipídicas de gran tamaño en su interior y nucleos redondos con cromatina laxa. Las células intersticiales de los testículos en reposo eran planas y altamente heterocromáticas, con numerosas gotas lipídicas pequeñas en su citoplasma. Las células intersticiales mostraron una reacción negativa a las fosfatasas ácidas, alcalinas y PAS en ambos grupos, Sin embargo presentaron reacción positivas para los lípidos. El tejido conectivo intersticial estaba bien vascularizado con vasos sanguíneos situados centralmente en el testículo activo y vasos sanguíneos pequeños y discretos en el testículo en reposo. Los vasos linfáticos no fueron identificados en los dos grupos estudiados.Los macrófagos fueron observados solo en los testículos en reposo, aunque en escasa cantidad.

Résumé

A comparative survey between non-systemic (paratyphoid Salmonellae) and systemic (S. Pullorum and S. Gallinarum) Salmonella strains was performed to produce a virulence gene profile for differentiation among the groups. The following virulence genes were evaluated: invA, spvC, sefC, pefA, fimY, sopB, sopE1, stn and avrA. There are substantial differences among paratyphoid Salmonellae, S. Pullorum, and S. Gallinarum regarding the genes sefC, spvC, sopE1 and avrA. A higher frequency of sefC, spvC, sopE1 and avrA genes were detected in S. Gallinarum and S. Pullorum when compared with strains from the paratyphoid group of Salmonella. These results may be useful for differentiating among different groups and serotypes.(AU)


Uma investigação comparativa entre amostras de Salmonella não-sistêmicas (grupo paratifoide) e sistêmicas (S. Pullorum and S. Gallinarum) foi desenvolvida para produzir um perfil de genes de virulência para diferenciação entre os grupos. Os seguintes genes de virulência foram avaliados invA, spvC, sefC, pefA, fimY, sopB, sopE1, stn e avrA. Detectou-se uma diferença substancial entre Salmonella do grupo paratifoide, S. Pullorum e S. Gallinarum considerando os genes sefC, spvC, sopE1 e avrA. Os genes sefC, spvC, sopE1 e avrA foram detectados, em maior número, em S. Gallinarum e S. Pullorum quando comparados com as amostras de Salmonella do grupo paratifoide. Estes resultados podem ser úteis para a diferenciação entre os diferentes grupos e sorotipos de Salmonella.(AU)

ABSTRACT: Salmonella Gallinarum (S. Gallinarum) and Salmonella Pullorum (S. Pullorum) are poultry host-specific, agents of fowl typhoid and pullorum disease, respectively. These biovars cause septicemic infections, resulting in high mortality. Outbreaks are frequently reported worldwide, causing losses due to the elimination of infected flocks and treatments. The use of antimicrobial agents is frequent in poultry farms to prevent or treat gastrointestinal infections. In the present research it was evaluated the antimicrobial susceptibility of 50 S. Gallinarum and S. Pullorum isolates, from outbreaks that occurred between 1987 to 1991 and 2006 to 2013. The comparison of the susceptibility profiles showed that all isolates were susceptible to β-lactams. All isolates from 1987-1991 were susceptible to all antibiotics tested except NAL and CIP (78%). The susceptibility profile of S. Gallinarum (2006 - 2013 period) was the following NAL (58%), CIP (63%), ENR (67%), TET (92%), FFC (96%) and SXT (96%). S. Pullorum isolates (2006 - 2013 period) showed the following susceptibility rates to NAL (65%), CIP (71%), ENR (94%) and TET (94%). All isolates were susceptible to β-lactams tested, however, resistance to quinolones and fluoroquinolones increased over time. Furthermore, low levels of resistance to other antibiotics were found in recent isolates, such as tetracyclines.


RESUMO: Salmonella Gallinarum (S. Gallinarum) e Salmonella Pullorum (S. Pullorum) são patógenos hospedeiro-específico de aves, agentes do tifo aviário e pulorose, respectivamente. Estes biovares causam infecções septicêmicas, resultando em alta mortalidade. Surtos são frequentemente relatados em diversos países, causando prejuízos devido à eliminação de lotes infectados e tratamentos. Agentes antimicrobianos são utilizados frequentemente em granjas avícolas para prevenir ou tratar infecções gastrointestinais. No presente trabalho, foi avaliada a susceptibilidade antimicrobiana de 50 isolados de S. Gallinarum e S. Pullorum, obtidos durante surtos que ocorreram entre 1987 a 1991 e entre 2006 a 2013. A comparação dos perfis de sensibilidade mostrou que todas as amostras são sensíveis aos β-lactâmicos. Todos os isolados de 1987-1991 foram sensíveis a todos os antibióticos testados, exceto NAL e CIP (78%). O perfil de susceptibilidade de S. Gallinarum (surtos de 2006 a 2013) foi NAL (58%), CIP (63%), ENR (67%), TET (92%), FFC (96%) e SXT (96%). Isolados de S. Pullorum (surtos de 2006 a 2013) apresentaram as seguintes taxas de sensibilidade: NAL (65%), CIP (71%), ENR (94%) e TET (94%). Todas as amostras foram sensíveis ao β-lactâmicos testados, no entanto, a resistência às quinolonas e fluoroquinolonas aumentou ao longo do tempo. Além disso, baixos níveis de resistência a outros antibióticos foram encontrados em isolados recentes, tais como as tetraciclinas.

PURPOSE: Salmonella enterica serovar Gallinarum (SG) ghost vaccine candidate was recently constructed. In this study, we evaluated various prime-boost vaccination strategies using the candidate strain to optimize immunity and protection efficacy against fowl typhoid. MATERIALS AND METHODS: The chickens were divided into five groups designated as group A (non-immunized control), group B (orally primed and boosted), group C (primed orally and boosted intramuscularly), group D (primed and boosted intramuscularly), and group E (primed intramuscularly and boosted orally). The chickens were primed with the SG ghost at 7 days of age and were subsequently boosted at the fifth week of age. Post-immunization, the plasma IgG and intestinal secretory IgA (sIgA) levels, and the SG antigen-specific lymphocyte stimulation were monitored at weekly interval and the birds were subsequently challenged with a virulent SG strain at the third week post-second immunization. RESULTS: Chickens in group D showed an optimized protection with significantly increased plasma IgG, sIgA, and lymphocyte stimulation response compared to all groups. The presence of CD4+ and CD8+ T cells and monocyte/macrophage (M/M) in the spleen, and splenic expression of cytokines such as interferon γ (IFN-γ) and interleukin 6 (IL-6) in the immunized chickens were investigated. The prime immunization induced significantly higher splenic M/M population and mRNA levels of IFN-γ whereas the booster showed increases of splenic CD4+ and CD8+ T-cell population and IL-6 cytokine in mRNA levels. CONCLUSION: Our results indicate that the prime immunization with the SG ghost vaccine induced Th1 type immune response and the booster elicited both Th1- and Th2-related immune responses.

Résumé

To develop a live vaccine strain against fowl typhoid and paratyphoid caused by Salmonella serovar Gallinarum biovar Gallinarum (Salmonella Gallinarum) and Salmonella serovar Enteritidis (Salmonella Enteritidis), respectively, several nalidixic acid resistant mutants were selected from lipopolysaccharide (LPS) rough strains of Salmonella Gallinarum that escaped from fatal infection of a LPS-binding lytic bacteriophage. A non-virulent and immunogenic vaccine strain of Salmonella Gallinarum, SR2-N6, was established through in vivo pathogenicity and protection efficacy tests. SR2-N6 was highly protective against Salmonella Gallinarum and Salmonella Enteritidis and safer than Salmonella Gallinarum vaccine strain SG 9R in the condition of protein-energy malnutrition. Thus, SR2-N6 may be a safe and efficacious vaccine strain to prevent both fowl typhoid and paratyphoid.

In this work, the morphological features of liver in Chinese Taihe black-bone silky Fowl (BSF) were analyzed by light microscope and transmission electron microscopy. The results showed that two kinds of hepatocytes were present in Taihe BSF liver, i.e., the dark and the light hepatocyte. The dark hepatocyte was electron-dense and lager, with many organelles, mitochondria especially. The light hepatocytes were smaller than the dark. They had electron-lucent cytoplasm with a small number of organelles. Furthermore, there were lipolysosomes in the light hepatocyte. The numerous long and serried finger-like microvilli spread into bile canaliculus lumen. The glycogen granules intensely stained, spread in some hepatocytes. Numerous glycogen granules scattered in cytoplasm especially near bile canaliculi. However, lipid droplets were not observed in any hepatocytes. The natural apoptotic hepatocytes were observed in Taihe BSF liver. The hepatocytes which contain abundant uesicae-like endoplasmic reticulum closed the apoptotic hepatocytes and spread the process to approach the cell residual bodies. Besides, there was a macrophage with several phagosomes. In conclusion, the dark and the light hepatocyte were present in Chinese Taihe BSF liver. They were different from electron-dense and organelles. The hepatocytes of Taihe BSF could undergo natural apoptosis, regeneration and renew ability.


Fueron analizadas las características morfológicas ultraestructurales de hígado en la gallina sedosa china de hueso negro por microscopía óptica y microscopía electrónica de transmisión. Los resultados mostraron que se encontraron hepatocitos claros y oscuros en el hígado de la gallina china Taihe. El hepatocito oscuro era denso y de mayor tamaño. Tenía numerosos organelos, especialmente mitocondrias. Los hepatocitos claros eran más pequeños que los oscuros. El citoplasma presentó un pequeño número de organelos. Además, había lipolisosomas en los hepatocitos claros. Numerosas microvellosidades se extendían hacia los canalículos biliares. En algunos hepatocitos se observó una tinción marcada en los gránulos de glucógeno. Sin embargo, no se observaron gotas de lípidos en los hepatocitos. Se observaron los hepatocitos apoptóticos naturales en el hígado de la gallina Silky Taihe. Aquellos hepatocitos que contenían abundante retículo endoplásmico, cerraban los hepatocitos apoptóticos y extendían el proceso de acercamiento a cuerpos residuales celulares. También hubo un macrófago con varios fagosomas. En conclusión, los hepatocitos claros y oscuros estaban presentes en el hígado de la gallina Taihe china. Estos diferían de electrones de alta densidad y organelos. Los hepatocitos de la gallina Taihe presentaron una apoptosis natural y capacidad de regeneración.

Résumé

Commercial broiler flocks from a farm located in the State of São Paulo, Brazil, presented diarrhea, depression, increased mortality and poor weight gain. Upon post-mortem examination, classical signs of Inclusion Body Hepatitis/Hydropericardium Syndrome (IBH/HPS) were observed, including enlarged pale yellow-colored livers and straw-colored liquid in the pericardial sac. In addition, gross lesions were also observed in the kidneys, pancreas, thymus, intestines and gallbladder. Samples of these organs were analyzed by PCR for the detection of the hexon gene of the Fowl Adenovirus (FAdVs) Group I. The results were positive for both flocks (A and B) assayed by PCR. The macroscopic lesions associated with the detection of FAdV Group I by PCR in several of these affected organs allowed for the identification of IBH/HPS. In fact, this is the first report in Brazil of IBH/HPS in broilers, which identifies FAdVs group I as a causal agent of the disease. These findings may contribute to the worldwide epidemiology of the adenovirus-mediated hepatitis/hydropericardium syndrome...


Lotes comerciais de frangos de uma granja localizada no Estado de São Paulo, Brasil, apresentavam diarreia, depressão, aumento de mortalidade e baixo ganho de peso. Após o exame post-mortem, sinais clássicos da síndrome de hepatite por corpúsculo de inclusão/hidropericárdio (IBH/HPS) foram observados incluindo hepatomegalia com aspecto amarelado pálido e líquido de coloração amarelo palha no saco pericárdio. Além disso, as alterações macroscópicas foram também observadas nos rins, pâncreas, timo, intestinos e vesícula biliar. Amostras destes órgãos foram analisadas pela técnica de PCR para detectar o adenovírus aviário do grupo I através do gene Hexon. Os resultados foram positivos para ambos os lotes (A e B) utilizando-se a técnica de PCR. As lesões macroscópicas associadas à detecção do adenovírus aviário do grupo I pela técnica de PCR em vários destes órgãos acometidos permitiu a identificação da síndrome de hepatite/hidropericárdio em frangos no Brasil. Ao nosso conhecimento, este é a primeira descrição da síndrome de hepatite/hidropericárdio causado por adenovírus aviário do grupo I, no Brasil. Estes achados podem contribuir com a epidemiologia mundial do adenovírus mediando a síndrome de hepatite/hidropericárdio...
