Objective • To determine clinical value of multi-voxel magnetic resonance spectroscopy (MVMRS) in the diagnosis of tongue carcinoma. Methods • A total of 56 cases that were pathologically diagnosed as having unilateral squamous cell carcinoma (SCC) or adenoid cystadenocarcinoma (ACC), from January to December 2017, were selected in this retrospective study. The characteristics of metabolic profiles in MVMRS were analyzed. The signals of choline (Cho), creatine (Cr), choline creatine ratio (Cho/Cr), and lactate/lipids (LL) and the apparent diffusion coefficient (ADC) were comparatively analyzed among the tumor parenchymal area, the paratumor area, and the contralateral control area in SCC/ACC group. The above indexes in the relative areas between SCC and ACC were also compared. Moreover, the relationship between the LL or Cho signal intensity with ADC in all the areas of two groups was investigated. Results • Higher Cho and LL signals were observed in the tumor parenchymal areas than those in the paratumor areas and the contralateral control areas in both SCC and ACC groups (P=0.000), while ADC values in the tumor parenchymal areas were obviously lower (P=0.000). Cho and LL signal intensities and ADC values in the paratumor areas were between the tumor parenchymal areas and contralateral control areas in both groups (P=0.000). However, no significant difference among the areas in Cr or Cho/Cr signal was found (P>0.01). There was no significant difference between SCC and ACC groups in the relative areas, either (P>0.01). The signal intensity of LL in the tumor parenchymal area in SCC group was negatively linearly correlated with the ADC value (P=0.000). Conclusion • MVMRS can detect the content of metabolites in tongue carcinoma, and LL and Cho signal intensities with ADC value can reflect the degree of tumor invasion in the paratumor areas.

<p><b>OBJECTIVE</b>This study aims to prepare docetaxel (DOC)-loaded multifunctional nanoparticles containing indocyanine green (ICG) and perfluorohexane (PFH) as targeted drug delivery system, which is supplemented with stromal cellderived factor-1 (SDF-1), and characterize their properties.</p><p><b>METHODS</b>Multifunctional nanoparticles were prepared by using the double emulsion method. SDF-1 was covalently conjugated to the surface of the nanoparticles through thioether bonding. Their particle size, distribution, and surface potential were determined with the Malvern measuring instrument. The conjugation of SDF-1 was evaluated by confocal laser scanning microscope. Encapsulation efficiency (ELC), drug loading capacity (DLC), and release regularity of the nanoparticles were determined by high-performance liquid chromatography (HPLC). In vitro photothermal property was recorded by a thermal imager. The in vitro imaging capacity was observed by a photoacoustic instrument and an ultrasonic diagnostic apparatus. Targeting capability was assessed by flow cytometry. The cell activity on SCC-15 cells was checked by CCK-8 method.</p><p><b>RESULTS</b>The targeted multifunctional nanoparticles showed regularly sphericity. The diameter was (502.88±17.92) nm. The zeta potential was (-11.5±3.15) mV. ELC was 54.12%±1.74%. DLC was 1.08 mg·mL-1. In vitro drug release was initially fast and subsequently slow. The photothermal characteristics were related to the concentration; the higher the concentration, the higher the temperature. Nanoparticles could detect significant photoacoustic and ultrasound signals. The in vitro targeting rate was 89.99%. No significant differences of cell viability in the SINPs groups were observed at each concentration (P>0.05). The inhibition effect of DOC-SINPs was stronger than that of SINPs whether or not in the presence of laser irradiation among the groups of 150 and 200 μg·mL-1 (P<
0.05).</p><p><b>CONCLUSIONS</b>Multifunctional nanoparticles for diagnosis and treatment were successfully prepared and displayed dualmode ultrasound/photoacoustic imaging and antitumor effects of chemotherapy and photothermal therapy.</p>

Objective:To evaluate the clinical applicability of CT angiography (CTA) for locating the perforator vessels of anterolateral thigh flap(ALTF) and the effects of individualized ALTF designed by CTA for the reconstruction of the soft tissue defects of tongue after the resection of tongue carcinoma.Methods:21 patients with tongue squamous cell carcinoma underwent CTA for locating the perforator vessels of ALTFs and for the design of individualized ALTFs before operation.The patients underwent soft tissue defect reconstruction with individualized ALTFs after tumor removal.Results:All the operations came off as preoperative designed,the intraoperative findings of the blood vessel alignment were consistent with the preoperative CTA results.The size of flaps was 6.0 cm × 5.0 cm-11.0 cm× 8.5 cm.20 ALTFs survived,2 ALTFs appeared vascular crisis,1 remained survival and the other was necrotic after surgical exploration.During 6 ~ 60 momhs of follow-up,the survival condition of flaps and the wound healing condition were both satisfactory.1 patient died because of distant metastasis 18 months after operation.20 patients reminded free of carcinoma and satisfied with the reconstructive effects of chewing,swallowing and linguistic function.Conclusion:CTA can accurately locate perforator vessels for the design of individualized AFLT.AFLT designed by CTA is an ideal choice for the reconstruction of postoperative soft tissue defects after resection tongue carcinoma.

[ ABSTRACT] AIM:To study the expression of miR-203 in tongue carcinoma tissues and the effect of miR-203 over-expression on the viability and invasion ability of Tca 8113 cells.METHODS:Twenty-eight pairs of tongue carcinoma tissues and adjacent nontumor tissues were collected , and the clinicopathological characters were analyzed .miR-203 was detected in the tongue tissues of 28 patients with tongue carcinoma by real-time PCR.miR-203 mimics and scramble were transfected into Tca8113 cells by Lipofectamine 2000.The expression of miR-203 was detected in Tca8113, Tca8113-miR-203 mimics and Tca8113-scramble cells by RT-qPCR.The cell viability was measured by CCK-8 assay.The cell invasion ability was determined by Transwell chamber invasion experiment .RESULTS:miR-203 expression was significantly down-regulated in the tongue carcinoma tissues compared with those in the adjacent nontumor tissues .The expression of miR-203 was associated with TNM stage and lymph node metastasis .Up-regulation of miR-203 inhibited the viability and invasion a-bility of Tca8113 cells.CONCLUSION:miR-203 suppresses the growth and invasion of tongue carcinoma cells .miR-203 may be a potential therapeutic target for treating human tongue cancer .

An FDA approved drug for the treatment of type II diabetes, Troglitazone (TRO), a peroxisome proliferator–activated receptor gamma agonist, is withdrawn due to severe idiosyncratic hepatotoxicity. In the search for new applications of TRO, we investigated the cellular effects of TRO on YD15 tongue carcinoma cells. TRO suppressed the growth of YD15 cells in the MTT assay. The inhibition of cell growth was accompanied by the induction of cell cycle arrest at G₀/G₁ and apoptosis, which are confirmed by flow cytometry and western blotting. TRO also suppressed the expression of cell cycle proteins such as cyclin D1, cdk2, cdk4, cyclin B1, cdk1(or cdc2), cyclin E1 and cyclin A. The inhibition of cell cycle proteins was coincident with the up-regulation of p21(CIP1/WAF1) and p27(KIP1). In addition, TRO induces the activation of caspase-3 and caspase-7, as well as the cleavage of PARP. Further, TRO suppressed the expressions of Bcl-2 without affecting the expressions of Bad and Bax. Overall, our data supports that TRO induces cell cycle arrest and apoptosis on YD15 cells.

Objective:To study the effects of gossypol acetic acid(GAA)on the methylation level and the mRNA expression of hM-LH1 in human tongue cancer Tca8113 cells.Methods:Tca8113 cells were treated by GAA at various doses for 24 h,48 h and 72 h respectively.MTT assay was used to detect the cell proliferation.Nested methylation specific PCR(nMSP)was used to detect methyl-ation level of hMLH1 .Real-time fluorescence quantitative PCR(RFQ-PCR)was applied to investigate the mRNA expression of hM-LH1 gene.Results:GAA inhibited the proliferation of Tca8113 cells dose-and-time dependently,decreased the DNA methylation level of hMLH1(P<0.05)and increased hMLH1 mRNA expression in Tca8113 cells(P<0.05).Conclusion:GAA can suppress proliferation of Tca8113 cells by demethylation of hMLH1 gene and increase of hMLH1 mRNA expression.

Propósito: Determinar y comparar la expresión de la cicloxigenasa-2 (COX-2) en el carcinomaescamocelular de lengua (CECL) según el grado de diferenciación, con la finalidadde explorar si este puede ser un marcador molecular útil en el diagnóstico y pronóstico delcáncer de la cavidad oral. Métodos: Se utilizaron 45 especímenes con CECL, 15 de ellos biendiferenciados, 12 moderadamente diferenciados, 18 mal diferenciados, un control positivode carcinoma de colon y un control negativo de mucosa oral sana. La identificación de laCOX-2 se obtuvo por medio de inmunohistoquímica. Resultados: La muestra de mucosalingual sana expresó la COX-2 en bajo nivel; el 60 % de las quince muestras de carcinomaescamocelular diferenciado mostraron un bajo nivel de expresión, el 41,3 % de 12 muestrasde carcinoma moderadamente diferenciado mostraron una alta expresión, y el 74 % de 18muestras de carcinoma no diferenciado mostraron una alta expresión de la enzima. Conclusión:La expresión de la COX-2 aumenta si el carcinoma es indiferenciado, lo cual sugiereque esta enzima podría desempeñar un papel importante en el desarrollo histopatológicodel CECL, tanto en las etapas iniciales como en las tardías...


Aim: Determine and compare the expression of cyclooxygenase-2 (COX-2) in squamouscell carcinoma of the tongue (SCCT) in three degrees of differentiation, in order to verify ifthis may be a molecular marker useful in the diagnosis and prognosis of oral cavity cancer.Methods: The sample consisted of 45 specimens with SCCT (15 well-differentiated, 12 moderatelydifferentiated, 18 poorly differentiated), a positive control (colon carcinoma) and anegative control (healthy oral mucosa). The identification of COX-2 was obtained throughimmunohistochemistry. Results: Samples of healthy lingual mucosa showed a low expressionof COX-2, 60 % of the 15 samples of well-differentiated squamous cell carcinoma showeda low expression of COX-2, 41.3 % of 12 samples of moderately differentiated carcinomashowed high expression, and 74 % of the 18 non-differentiated carcinoma samples showeda high expression of the enzyme. Conclusion: The expression of COX-2 increases in lessdifferentiated squamous cell carcinoma, which suggests that identification of COX-2 in thehistologic development of squamous carcinoma of the tongue might be important in thedifferentiation of both, the early and late stag es...

Aberrations on the short arm of chromosome 8 (8p) are frequently observed in several human cancers. In this study, 20 squamous cell carcinoma (SCC) specimens from the tongue were examined in order to evaluate the role of 8p in SCC of the tongue. Microsatellite analysis using 14 markers demonstrated two commonly deleted regions (CDRs) on 8p. Reverse transcription-polymerase chain reaction (RT-PCR) revealed frequent down-regulation of the FEZ1 gene, mapped to 8p22, and frequent over-expression of the cathepsin B gene, mapped to 8p-21-22. These results suggested that genetic aberrations are involved in the development of SCC of the tongue. However, no significant relationship was observed to be established between the genetic alterations and clinicopathological features. Thus, further investigation is necessary in order to clarify the clinical role of 8p in carcinoma of the tongue.

Résumé

Cellular effects of ethanol in YD-15 tongue carcinoma cells were assessed by MTT assay, caspase activity assay, Western blotting and flow cytometry. Ethanol inhibited the growth and proliferation of YD-15 cells in a dose- and time-dependent manner in an MTT assay. The effects of ethanol on cell cycle control at low percent range of ethanol concentration (0 to 1.5%), the condition not inducing YD-15 cell death, was investigated after exposing cells to alcohol for a certain period of time. Western blotting on the expression of cell cycle inhibitors showed that p21 and p27 was up-regulated as ethanol concentration increases from 0 to 1.5% whilst the cell cycle regulators, cdk1, cdk2, and cdk4 as well as Cyclin A, Cyclin B1 and Cyclin E1, were gradually down-regulated. Flow cytometric analysis of cell cycle distribution revealed that YD-15 cells exposed to 1.5% ethanol for 24 h was mainly arrested at G2/M phase. However, ethanol induced apoptosis in YD-15 cells exposed to 2.5% or higher percent of ethanol. The cleaved PARP, a marker of caspase-3 mediated apoptosis, and the activation of caspase-3 and -7 were detected by caspase activity assay or Western blotting. Our results suggest that ethanol elicits inhibitory effect on the growth and proliferation of YD-15 tongue carcinoma cells by mediating cell cycle arrest at G2/M at low concentration range and ultimately induces apoptosis under the condition of high concentration.

Objective:To investigate the feasibility of modified combined radical neck dissection remaining integrality of mandible in the surgical management of advanced tongue squamous cell carcinoma.Methods:39 cases of advanced tongue squamous cell carcinoma received modified combined radical operation remaining integrality of mandible,7-year follow-up was conducted.Results:Integrality of mandible and the function of mastication were remained in all the cases.1 and 3 years after operation there were 1 case and 9 cases of recurrence respectively.Metastasis was found in 5 cases.The 3-year survival rate was(61.72?5.44)%.Conclusion:The modified combined radical neck dissection remaining integrality of mandible is feasible in the treatment of advanced tongue squamous cell carcinoma.

113 cases of postoperative patients with tongue carcinoma were analyzed to investigate the correlative factors of cervical lymphnode metastasis and the prognosis.Results showed that the prognosis of tongue carcinoma was related to cervical lymph node metastasis remarkably.
