Résumé

Objective To observe the effects of moderate intensity exercise training combined with Xianlinggubao capsule on bone mineral density (BMD), bone metabolism and femoral biomechanics of ovariectomized rats, so as to provide lab references for osteoporosis prevention. Methods Fifty female SD rats were randomly divided into 5 groups, 10 rats in each group. Group A was the normal control; Group B was given 1 mL normal saline by gavage after ovariectomy for one week; Group C was given moderate intensity exercise training (exercise speed was 20 m/min, lasting for 60 min per day, continuous 5 days per week); Group D was given 1 mL Xianlinggubao capsule [0.4 g/(kg · d)] after 1-week ovariectomy; Group E was was given both 1 mL Xianlinggubao capsule and moderate intensity exercise training after 1-week ovariectomy. After 8 weeks of continuous treatment, blood biochemical indexes, BMD, micro CT and biomechanics of the femur and L5 vertebral body were detected. Results Compared with group B, the blood biochemical indexes of Group C-E were improved in varying degrees, the BMD of L5 vertebral body and femur were increased, the bone volume fraction, trabecular number and trabecular thickness of femur (or L5 vertebral body) were increased, the trabecular space and structural model index were decreased, the maximum load, maximum deflection and maximum stress of L5 vertebral body were increased, and the maximum stress of femur was increased. The maximum load, elastic load, elastic deflection, elastic modulus, elastic stress, maximum stress and elastic deflection increased, and the effect of Group E was the most obvious. Conclusions Moderate intensity exercise training combined with Xianlinggubao capsule can improve BMD, bone metabolism and bone microstructure, and improve bone mechanical properties of ovariectomized rats.

Objective To assay Icariine, Epimedin C, asperosaponin Ⅵ, psoralen and angelicin in Xianlinggubao capsules via multi-wavelength HPLC method.Methods Separation was carried out on Welch Ultimate○R XB-C18 column.The mobile phase was acetonitrile-water system and a linear gradient elution was used.The column temperature was 30 ℃.The detection wavelength for Icariine, Epimedin C,asperosaponin Ⅵ was set at 212 nm, psoralen and angelicin at 246 nm.Results Five components reached baseline separation, the linearity was good when sample size was in the range of 0.008 2-0.328 μg for Icariine(r=0.999 5), 0.055 6-2.224 μg for Epimedin C (r=0.999 6), 0.144 1-5.764 μg for asperosaponin Ⅵ(r=0.999 6), 0.005 4-0.215 2 μg for psoralen(r=0.998 0), 0.006 6-0.265 6 μg for angelicin(r=0.998 5).The average recoveries were 97.59%, 98.58%, 98.11%, 97.86%, 98.22% respectively.The RSDs of recovery were all less than 2.0%.Conclusion This method is simple, accurate, with good separation, high sensitivity for the assay of multiple components in Xianlinggubao capsule.

OBJECTIVE:To observe clinical efficacy and safety of Xianlinggubao combined with alendronate sodium in the treatment of osteoporosis. METHODS:112 patients with osteoporosis were randomly divided into treatment group and control group with 56 cases in each group. Both group were given routine treatment as calcium carbonate,vitamin D and life style. Control group were given alendronate sodium 10 mg orally,qd;treatment group was additionally given Xianlinggubao capsule 1.5 g,bid, on the basis of control group. Both groups received 12 weeks of treatment. The levels of S-Ca,S-P,24 h urinary calcium(U-Ca), alkaline phosphatase(ALP),osteocalcin,IL-6,bone mineral density(BMD)of L2-4 lumbar spine and femoral neck were observed in 2 groups before and after treatment. Clinical efficacy and toxic reaction were compared. RESULTS:There was no statistical sig-nificance in the levels of S-P and S-Ca between 2 groups after treatment(P>0.05);the level of ALP、osteocalcin、BMD increased in treatment group after treatment,the level of U-Ca and IL-6 decreased compared to before treatment,and the treatment group was better than the control group,with statistical significance(P<0.05). Total effective rate of treatment group(92.86%)was sig-nificantly higher than that of control group (73.21%),with statistical significance (P<0.05). No obvious toxic side effects was found in 2 groups. CONCLUSIONS:Xianlinggubao combined with alendronate sodium shows significant therapeutic efficacy for patients with osteoporosis with good safety.

Résumé

Icariin and total flavonoids in "Xianlinggubao Capsule", a Chinese herbal preparationcontaining Epimedium L. as the main ingredient for the treatment of osteoporosis, were determined quan-titatively by HPLC and UV spectrophotometry respectively. It was found that the contents of icariin were4.16~27.67 mg/g and that of flavonoids were 62. 56~109.78 mg/g. The recoveries were 96.17% and103.58% respectively. The method could be used for the quality control of the drug.

Résumé

Objective: To determine the contents of three effective components in Xianlinggubao Capsules. Methods: A RP HPLC was developed for the determination of psoralen, isopsoralen and icariin in Xianlingbao Capsules. Results: The contents of these compoments were 0.93mg/g, 0.95mg/g and 9.2mg/g, respectively. Conclusion: This Methods is simple and reliable.
