Résumé

ABSTRACT One of the greatest challenges facing humanity is the development of sustainable strategies to ensure food availability in response to population growth and climate change. One approach that can contribute to increase food security is to close yield gaps and enhancing genetic gain; to such end, what is known as "molecular breeding" plays a fundamental role. Since a crop breeding program is mainly based on the quality of the germplasm, its detailed genetic characterization is mandatory to ensure the efficient use of genetic resources and accelerating development of superior varieties. Deep genotyping is an essential tool for a comprehensive characterization of the germplasm of interest and, fortunately, the technology is now accessible at a reasonable cost. What must be ensured is the correct interpretation of the genotypic information and on that basis develop efficient practical molecular crop breeding strategies that respond to the real needs of the breeding program.


RESUMEN Uno de los mayores desafíos que enfrenta la humanidad es el desarrollo de estrategias sostenibles para asegurar la disponibilidad de alimentos en respuesta al crecimiento de la población y el cambio climático. Un enfoque que puede contribuir a aumentar la seguridad alimentaria es cerrar las brechas de rendimiento y mejorar la ganancia genética; para tal fin, lo que se conoce como "mejoramiento molecular" juega un papel fundamental. Dado que un programa de mejoramiento de cultivos se basa principalmente en la calidad del germoplasma, su caracterización genética detallada es fundamental para garantizar el uso eficiente de los recursos genéticos y acelerar el desarrollo de variedades superiores. La genotipificación profunda es una herramienta esencial para una caracterización integral del germoplasma de interés y, afortunadamente, en la actualidad se puede acceder a la tecnología a un costo razonable. Lo que debe asegurarse es la interpretación correcta de la información genotípica y sobre esa base desarrollar estrategias eficientes y prácticas de mejoramiento molecular de cultivos que respondan a las necesidades reales del programa de mejoramiento.

Résumé

As an important biological resource of a country or region, human genetic resources (HGR) are increasingly attracting attention and concern from many countries internationally, and even have been elevated to the dimension of national sovereignty by many countries. As a large country with rich and diverse HGR, relevant regulatory work in China started relatively late compared to foreign countries, but has developed rapidly. However, looking around the world, there is still room for further improvement and development of domestic HGR regulation. By reviewing the regulatory legislation of HGR in some countries abroad, this paper sorted out their advanced experience and highlighted measures in detail, and summarized their deep-seated legislative guidance. A detailed analysis and evaluation were conducted from seven aspects:the advanced level of HGR legislation and the enhancement of legal effectiveness, the systematization and systematization of policies and regulations, the unification of platforms and relevant standards, the highlight of the concept of ethics and the rule of law, the strengthening of anti-discrimination protection, the special protection of major human genetic family resources and national special protection areas of HGR, and the establishment of principled consensus or agreement on HGR in international and local regions, to provide reference for the construction of regulatory policies and legal systems for HGR in China.

Résumé

La producción peruana de cacao representa aproximadamente el 2% de la producción mundial; con una contribución muy reconocida por la calidad y diversidad de sus sabores y aromas. De milenario origen amazónico y probada utilización de hace por lo menos 5200 años, este cultivo recién ha sido visible para el desarrollo agrario en las dos últimas décadas, cuando se le ha utilizado con relativo éxito en la sustitución de cultivos ilegales de coca ¿Y qué tanto está contribuyendo la academia a la cadena de valor del cacao? Con carácter exploratorio, se registró y analizó una muestra de 44 tesis que tenían como objeto de estudio el cacao, obtenido por sus autores en los centros de producción y parcelas de productores. La mayor parte de estas tesis (41) estuvieron destinadas a obtener títulos profesionales, dos para optar el grado de magister y uno para obtener el grado de doctorado. Estas tesis se realizaron en universidades de 13 departamentos del Perú, contando Lima con el mayor número de tesis (12). Once tesis abarcaron actividades de investigación del germoplasma para la transformación y/o la innovación del cacao, confiriéndole así valor agregado con potencial uso comercial. Estas investigaciones estuvieron relacionadas con cuatro sectores productivos con un claro potencial comercial en el mercado global de recursos genéticos. En ningún caso, las investigaciones se realizaron bajo la seguridad jurídica del sistema de acceso a recursos genéticos y participación justa y equitativa de beneficios del Protocolo de Nagoya, del Convenio sobre la Diversidad Biológica.


The Peruvian production of cocoa represents, approximately, 2% of the world production, with a very recognized contribution because of the quality and diversity of its flavors and aromas. With a millenary Amazonian origin and proved utilization by at least 5200 years ago, this crop just became visible for the agrarian development in the last two decades, when it was used with relative success for the substitution of illegal coca crops. And how much is the academy contributing to the value chain of cocoa? A sample of 44 theses which cocoa obtained from production centers and farmers plots was their object of study, has been registered and analyzed in an exploratory way. 41 theses were to get professional titles, two for magister degree and one for doctorate degree. These theses were done in universities of 13 Department of Peru, where Lima reached the highest number (12). Eleven theses addressed germplasm research activities aimed to cocoa transformation and/or innovation, thus conferring added value with potential commercial use. These researches were related to four productive sectors with a clear potential in the global market of genetic resources. In any case, the studies were done under the legal certainty of the system of access to genetic resources and fair and equitable benefit-sharing of the Nagoya Protocol to the Convention on Biological Diversity.

ABSTRACT Zephyranthes citrina is an ornamental American bulbous plant used as an ornamental garden crop for the aesthetic qualities of its yellow perigonium. The objective of this work was to characterize the species by classical chromosome staining and fluorochrome banding. A sporophytic chromosome number of 2n=8x=48 chromosomes was observed, being the karyotypic formula 20 m + 26 sm + 2 st. Satellites were detected in the short arm of metacentric chromosomes 8, 9, 11 and 12, which colocalized with constitutive heterochromatin CMA+/DAPI-/0 bands. The karyotype comprised chromosome pairs with terminal constitutive heterochromatin bands that included satellites and heteromorphic clusters indicating that it is an allooctoploid. These results will be used as a tool for monitoring genetic improvement, in interspecific crosses and its progenies and in biotechnological procedures by in vitro culture.


RESUMEN Zephyranhtes citrina es una planta bulbosa americana, ornamental, utilizada en jardines por las cualidades estéticas de su perigonio amarillo. El objetivo de este trabajo fue caracterizar citogenéticamente la especie con tinción clásica convencional y bandeo cromosómico. Se observó un número cromosómico esporofítico de 2n=8x=48, siendo la fórmula cariotípica 20 m + 26 sm + 2st. Se detectaron satélites en el brazo corto de los cromosomas metacéntricos 8, 9, 11 y 12, que co-localizaron con bandas de heterocromatina constitutiva CMA+/DAPI-. El cariotipo comprendió pares de cromosomas con bandas de heterocromatina constitutivas terminales que incluyeron satélites y grupos heteromórficos que indican que es un alooctoploide. Estos resultados serán usados como herramientas en el monitoreo del mejoramiento genético, en análisis de cruzamientos interespecíficos y progenies y en procedimientos biotecnológicos de cultivo in vitro.

Résumé

Abstract One of the texts in the "Biodiversity in the State of São Paulo" series, within the FAPESP-Biota Program, was dedicated to the Infrastructure for Biodiversity Conservation, with a focus on Biological Collections and Conservation Units. From the early 1960s, when FAPESP was established, to the present day, financial resources have been invested in the preservation of the biodiversity of the national genetic heritage, besides other fields. History of years of advances in scientific knowledge was built, which can be portrayed through the projects that resulted in high-quality data of national and international impact. Microbiological collections are centers that generate technology and specialized human resources, and act (among other things) as living repositories preserving reference material and as witnesses to the history of microbial biodiversity because they preserve what may no longer exist. They have enormous potential to promote the global bioeconomy and address problems that have resulted from the misuse of natural resources. This reading brings everyone the history, advances, and future perspectives of culture collections, within the efforts of 60-year scientific activities in Brazil.


Resumo Um dos textos da série "Biodiversidade do Estado de São Paulo", dentro do Programa FAPESP-Biota, foi dedicado à Infraestrutura para Conservação da Biodiversidade, com foco nas coleções biológicas e nas unidades de conservação. Do início dos anos 60, quando a FAPESP foi criada, até os dias atuais muito foi investido em pesquisa nas mais diversas áreas, incluindo a preservação da biodiversidade do patrimônio genético nacional. Uma história de longos anos de avanços no conhecimento científico foi construída, a qual pode ser retratada através dos projetos que resultaram em dados de alta qualidade com impacto nacional e internacional. As coleções microbiológicas são centros geradores de tecnologia e recursos humanos especializados, que atuam (dentre outros) como repositórios vivos, preservando material de referência, e como testemunho da história da biodiversidade microbiana, preservando o que pode não mais existir. Possuem enorme potencial para alavancar a bioeconomia global e tratar de problemas que resultaram do mau uso dos recursos naturais. Essa leitura traz a todos o histórico, os avanços e as perspectivas futuras das coleções de microrganismos, dentro dos esforços de 60 anos de atividades científicas no Brasil.

Objective:The management system of Investigator Initiated Clinical Trials (IIT) using human genetic resources in large hospitals is still not optimal. The purpose of this study is to sort out the existing problems of human genetic resources management in IIT of large hospitals, analyze the management practices, and provide reference for the standardized management of human genetic resources in IIT.Methods:The existing problems of human genetic resources management in IIT of large hospitals were sorted out by literature analysis, and the management practices since the establishment of human genetic resources management office were analyzed, and suggestions were put forward.Results:The problems of human genetic resources management in IIT of large hospitals mainly focused on the lack of awareness and attention of investigators and managers, the lack of process supervision, and the imbalance between input and output of genetic resources. A large hospital has implemented personnel and system construction, strengthened node management and control, and developed a training system. The approval rate of administrative approval and archival projects of human genetic resources of the Ministry of Science and Technology in 2021 increased by 52.69% compared with the previous year.Conclusions:Large hospitals should proceed from the actual situation, establish the human genetic resource management system jointly through multi-department linkage, improve management process, strengthen quality control, increase training efforts, improve the professional capacity and quality of researchers and managers, promote the high-quality, high-level and well development of IIT.

Résumé

Objective:To encourage the innovation of medical science and technology, regulate the Investigator Initiated Clinical Trials (IIT), this paper analyzed the problems faced by the management of human genetic resources in medical institution, and the corresponding management measures.Methods:Existing problems of human genetic resource management in IIT were discussed and summarized, taking into consideration of regulatory requirements and daily practices in the hospital.Results:Problems identified include imperfect supervision mechanism, incomplete policy interpretation, insufficient understanding of human genetic resources management policies by researchers, and how to support innovation with human gentic resources and so on.Conclusions:Medical institutions should establish and improve the supervision system and strengthen the whole process management of IIT. Accurately interpret regulatory requirements to improve application efficiency. Improve researchers′ recognition of human genetic resources to avoid omission and misrepresentation. Build a biobank to encourage legal and compliant use of human genetic resources to carry out high-quality IIT.

Summer apples are one of the most important plant community in Artvin province located Northeastern part of Turkey. In present study 22 local apple genotypes were characterized by phenological, morphological, biochemical and sensory properties. Harvest date was the main phenological data. Morphological measurements included fruit weight, fruit shape, fruit ground color, fruit over color, fruit over color coverage and fruit firmness, respectively. Sensory measurements were as juiciness and aroma and biochemical characteristics included organic acids, SSC (Soluble Solid Content), vitamin C, total phenolic content and antioxidant capacity. Genotypes exhibited variable harvest dates ranging from 11 July to 13 August and cv. Summered harvested 30 July 2017. The majority of genotypes were harvested before cv. Summered. Fruit weight were also quite variable among genotypes which found to be between 89 g and 132 g, and most of the genotypes had bigger fruits than cv. Summered. Pink, red, yellow and green fruit skin color was evident and main fruit shape were determined as round, conic and oblate among genotypes. ART08-9, ART08-4, ART08-21 and ART08-22 had distinct bigger fruits and ART08-1, ART08-2, ART08-5, ART08-12 and ART08-17 had higher total phenolic content and antioxidant capacity. The results of the study showed significant differences for most of the phenological, morphological, sensory and biochemical characteristics. Thus, the phonological, morphological, sensory and biochemical characteristics of summer apple genotypes were distinguishable and these results suggest that phonological, morphological, sensory and biochemical differences of the summer apple genotypes can be attributed to differences in genetic background of genotypes which placed different groups by PCoA analysis.


As maçãs cultivadas no verão são uma das culturas vegetais mais importantes da província de Artvin, localizada no nordeste da Turquia. No presente estudo, 22 genótipos locais de maçã foram caracterizados quanto às suas propriedades morfológicas, bioquímicas e sensoriais. As características analisadas foram peso do fruto, data da colheita, forma do fruto, coloração da casca, firmeza do fruto, suculência, aroma, teor de ácidos orgânicos e de sólidos solúveis, vitamina C, teor fenólico total e capacidade antioxidante. Os genótipos exibiram datas de colheita que variaram de 11 de julho a 13 de agosto, e a cultivar de verão foi colhida em 30 de julho 2017. A maioria dos genótipos foi colhida antes da cultivar de verão. O peso dos frutos também foi bastante variável entre os genótipos, apresentando entre 89 e 132 g, e a maioria dos genótipos apresentou frutos maiores que a cultivar de verão. As colorações rosa, vermelho, amarelo e verde da casca dos frutos foram as mais evidentes, e a forma principal dos frutos foi determinada como redonda, cônica e oblata entre os genótipos. ART08-9, ART08-4, ART08-21 e ART08-22 apresentaram frutos maiores distintos, e ART08-1, ART08-2, ART08-5, ART08-12 e ART08-17 apresentaram maior conteúdo fenólico total e capacidade antioxidante. Os resultados do estudo mostraram diferenças significativas para a maioria das características morfológicas, sensoriais e bioquímicas. Objective:This paper aims to explore the management of human genetic resources in medical institutions, according to reflections of the management mode of a particular hospital, providing possible reference for other medical institutions.Methods:The management system of human genetic resources was constructed refer to the McKinsey 7S model. Approved projects information includes the types of projects, characteristics of human genetic resources involved and the characteristics of principal investigator are analyzed.Results:A total number of 82 projects were approved Since the implementation of newly updated Regulation of the People′s Republic of China on the Administration of Human Genetic Resources (hereinafter referred to as the regulations), and majority of which are drug clinical trials. The human genetic resources materials and data involved are mainly blood, urine, serum, plasma, clinical data, imaging data, etc. Most of the principal investigators with senior professional title are from key disciplines.Conclusions:McKinsey 7S model provides a new reference path for medical institutions to carry out human genetic resources management.

Objective:By strengthening the management of external provision of data generated by clinical trials in medical institutions, to improve the effectiveness of supervision of human genetic resource information, and to promote the legal sharing and effective use of data.Methods:Analyzed problems identified in filing human genetic resource information in clinical trials since July 1, 2019, put forward possible solutions and suggestions.Results:Main problems were identified in external provision of human genetic resource information, including the specification and time limit of external provision of information, the information recipient, the storage location and the final disposal method of information.Conclusions:Hospital and regulatory authority need to carry out more tailored training, optimize management systems and procedures in order to strengthen the management of the external provision of human genetic resources information.

Résumé

Objective:To analyze the intellectual property risk of international cooperative scientific research involving human genetic resources, explore possible risk control measures regarding to intellectual property.Methods:By means of literature review, this paper analyzes the special attributes and strategic position of human genetic resources, reviews the policies and systems involving human genetic resources in international cooperative scientific research, identifies the intellectual property risk points, and puts forward suggestions on risk management and control from the perspective of intellectual property protection.Results:The management of human genetic resources in China is evolving quickly. However, there is still a lack of practical guidelines on intellectual property protection and development, more substantial engagement and contribution of Chinese investigators in the international collaborative research should be promoted, and the perception and awareness of the significance of human genetic resources should be enhanced.Conclusions:In the international cooperative scientific research involving human genetic resources, we should clarify the operating rules at the level of intellectual property protection, improve the substantive participation of Chinese investigators, enhance the strategic awareness and risk awareness of human genetic resources, and provide support at the level of executive management institutions.

ABSTRACT: The objective of this study was to estimate sugar content variation (°Brix) in 95 accessions from the elephant grass germplasm active bank of Embrapa Dairy Cattle research center, located in Coronel Pacheco, MG, Brazil. The accessions with the highest sugar content were identified, and the effect of plant age on sugar concentration was evaluated. The experiment was conducted in randomized blocks design with two replications. Sugar content analysis occurred twice during the growth-cycle (at 70 and 100 days) along two points of the stem, one 30 cm from the base and the other at the midpoint. The analysis of variance considered the split plot model, with accessions as main plots and plant age as subplots. The means were compared using the Scott Knott test. While there was sugar content variation between accessions, increased plant age had no significant effect on the sugar content. However, some accessions did have a significant increase or decrease in sugar content as they aged. The highest and lowest mean sugar content was 6.96% (in the accession BAG80) and 4.03% (in the accession BAG13) °Brix, respectively.


RESUMO: O objetivo deste estudo foi estimar a variação do teor de açúcar (°Brix) existente entre 95 acessos do banco ativo de germoplasma de capim-elefante da Embrapa Gado de Leite, localizado no campo experimental de Coronel Pacheco. Foram identificados os acessos com maior teor de açúcar, bem como avaliado o efeito da idade da planta sobre a variação da concentração de açúcar. O experimento foi conduzido em blocos ao acaso com duas repetições, 95 genótipos, duas idades de planta, 70 e 100 dias de crescimento, e análise do teor de açúcar em duas secções amostradas à 30 cm da base e na metade do colmo. A análise de variância considerou modelo de parcela subdividida no tempo, sendo os acessos, as parcelas e a idade da planta as subparcelas. As médias foram comparadas pelo teste de Scott Knott. Observou-se variação no teor de açúcar entre os acessos, porém não foi observado efeito significativo no teor de açúcar devido ao aumento da idade da planta. Entretanto, alguns acessos apresentaram interação significativa com aumento ou redução do teor de açúcar em decorrência do aumento da idade da planta. O acesso de maior média de teor de açúcares entre as duas idades foi BAG80, com 6,96%, e o de menor foi BAG13, com 4,03%.

Abstract Introduction: The fruit of the yellow mombin (Spondias mombin L.) is notable due to its sensory and functional qualities. However, there is little knowledge regarding the genetic diversity of this species, and this would aid the implantation of the cultivation of the fruit as a crop, since current production is based on extractivism. Objective: Evaluate the diversity and genetic structure of natural populations of S. mombin in the state of Mato Grosso, Brazil, through microsatellite molecular markers in order to assist in the implementation of conservation strategies and the collection of genetic resources. Methods: A total of 139 S. mombin individuals were sampled in ten natural populations. PCR amplifications were performed with seven fluorescence-marked microsatellite primers. Genetic diversity was evaluated by the number of alleles, expected (He) and observed heterozygosity (Ho), polymorphic information content (PIC), fixation index (ƒ), rare and exclusive alleles. The genetic structure was evaluated using AMOVA, UPGMA dendrogram and Bayesian statistical analysis. Results: 46 alleles were amplified, which had an average of 6.6 alleles per locus. He was higher than Ho and f was positive, indicating the presence of inbreeding. The PIC ranged from 0.048 to 0.700, and only two loci were poorly informative. We found 27 rare alleles and 16 unique alleles. The largest component of variation was intrapopulational (90.64 %). The estimated gene flow was 1.99, which indicates that there is no genetic isolation between populations, and justifies the FST value (0.0963). The ten populations were grouped into two groups, and two populations constituted an isolated group. The Mantel test demonstrated that the genetic structure is not related to the geographic distance between populations. Conclusion: There is genetic diversity in the populations of S. mombin, which must be conserved in situ or ex situ, due to the diversity they present and because they are promising sources for collection of germplasm.


Resumen Introducción: El fruto amarillo del jobo o yuplón (Spondias mombin L.) destaca por sus cualidades sensoriales y funcionales. Sin embargo, existe poco conocimiento sobre la diversidad genética de esta especie, lo que ayudaría a la implantación del cultivo del fruto como cultivo, ya que la producción actual se basa en el extractivismo. Objetivo: Evaluar la diversidad y estructura genética de poblaciones naturales de S. mombin en el estado de Mato Grosso, Brasil, a través de marcadores moleculares microsatélites para ayudar en la implementación de estrategias de conservación y recolección de recursos genéticos. Métodos: Se muestrearon un total de 139 individuos de S. mombin en diez poblaciones naturales. Las amplificaciones por PCR se realizaron con siete cebadores de microsatélites marcados con fluorescencia. La diversidad genética se evaluó por el número de alelos, heterocigosidad esperada (He) y observada (Ho), contenido de información polimórfica (PIC), índice de fijación (ƒ), alelos raros y exclusivos. La estructura genética se evaluó mediante AMOVA, dendrograma UPGMA y análisis estadístico bayesiano. Resultados: Se amplificaron 46 alelos, los cuales tenían un promedio de 6.6 alelos por locus. Fue más alto que Ho y f positivo, lo que indica la presencia de endogamia. El PIC osciló entre 0.048 y 0.700, y solo dos loci fueron poco informativos. Encontramos 27 alelos raros y 16 alelos únicos. El mayor componente de variación fue intrapoblacional (90.64 %). El flujo de genes estimado fue de 1.99, lo que indica que no hay aislamiento genético entre poblaciones y justifica el valor de FST (0.0963). Las diez poblaciones se agruparon en dos grupos y dos poblaciones constituyeron un grupo aislado. La prueba de Mantel demostró que la estructura genética no está relacionada con la distancia geográfica entre poblaciones. Conclusión: Existe diversidad genética en las poblaciones de S. mombin, la cual debe ser conservada in situ o ex situ, por la diversidad que presentan y porque son fuentes promisorias para la recolección de germoplasma.

Resumen Dentro de las estrategias de evaluación de especies forestales nativas por parte de la Corporación Colombiana de Investigación Agropecuaria - AGROSAVIA, se encuentra la caracterización fenotípica. Con el propósito de validar el uso de descriptores morfológicos y determinar la variabilidad fenotípica de la colección de trabajo del Centro de Investigación La Libertad ubicado en el piedemonte llanero (Villavicencio, Meta); se evaluaron 121 individuos de machaco (Simarouba amara Aubl.) de 6.2 años de edad, por medio de 34 descriptores de planta, hoja y fruto. Los datos cualitativos y cuantitativos se analizaron mediante análisis de correspondencias múltiples y análisis de componentes principales respectivamente, con el fin de reducir la dimensionalidad de los datos, seguido de análisis de conglomerados, utilizando el algoritmo de WARD para el agrupamiento de los individuos. Los resultados obtenidos permitieron identificar que el volumen de tronco presentó la mayor variabilidad (31.13%) y las variables cuantitativas asociadas al tamaño y diámetro de la copa, diámetro de fuste, volumen del tronco, longitud y ancho de la hoja, altura total y de copa presentaron las mayores correlaciones con los tres primeros componentes principales (57.82%). Se obtuvieron nueve conglomerados que reúnen el 95.73% de la variabilidad original y en los cuales se encontraron individuos potencialmente útiles con fines maderables, arboricultura y sistemas agroforestales. La forma del fuste, la altura de ramificación y de bifurcación, el tipo de corteza y el hábito de fructificación presentaron mínima variación en los individuos, contrario a la rectitud del fuste y la forma de la copa.


Abstract Phenotyping is one of the strategies used by Corporación Colombiana de Investigación Agropecuaria (AGROSAVIA) for evaluating native forest species. One hundred and twenty-one 6.2 year-old mountain damson/stavewood/bitterwood/paradise trees (Simarouba amara Aubl. commonly known as machaco in Colombia) were evaluated using 34 plant, leaf and fruit descriptors to validate the use of morphological descriptors and determine the phenotype variability of AGROSAVIA's La Libertad Research Centre's working collection. The centre is located in the foothills of Colombia's eastern plains (Villavicencio, Meta). Multiple correspondence analysis (MCA) was used for analysing qualitative data and principal component analysis (PCA) for +data to reduce dataset dimensionality; this was followed by cluster analysis, using Ward's method (minimum variance method or Ward's minimum - agglomerative algorithm) for hierarchical cluster analysis for grouping the trees. The results led to identifying that the wood volume descriptor had the greatest variability (31.13%) and the quantitative variables associated with crown size and diameter, stem diameter, trunk volume, leaf length and width, total and crown height had the greatest correlation with the first three components (57.82%). Nine clusters were obtained (accounting for 95.73% of original variability) and in which trees were found for timber, arboriculture and agroforestry system purposes. The trees' stem shape, branch height and bifurcation, the type of bark and fruiting habits had minimum variation, contrary to the that found regarding stem straightness and crown shape.


Resumo Dentro das estratégias de avaliação de espécies florestais nativas pela Colombian Agricultural Research Corporation - AGROSAVIA, está a caracterização fenotípica. A fim de validar o uso de descritores morfológicos e determinar a variabilidade fenotípica da coleção de trabalhos do Centro de Pesquisa La Libertad localizado no sopé da planície, (Villavicencio, Meta); 121 indivíduos de machaco (Simarouba amara Aubl.) de 6,2 anos de idade foram avaliados por meio de 34 descritores de planta, folha e fruto. Os dados qualitativos e quantitativos foram analisados por meio da análise de correspondência múltipla e da análise de componentes principais, respectivamente, a fim de reduzir a dimensionalidade dos dados, seguida da análise de cluster, por meio do algoritmo WARD, para agrupamento dos indivíduos. Os resultados obtidos permitiram identificar que o volume do tronco apresentou a maior variabilidade (31,13%) e as variáveis quantitativas associadas ao tamanho e diâmetro da copa, diâmetro do caule, volume do tronco, comprimento e largura da folha, total a altura e a copa apresentaram as maiores correlações com os três primeiros componentes principais (57,82%). Foram obtidos nove aglomerados que atendem a 95,73% da variabilidade original e nos quais foram encontrados indivíduos potencialmente úteis para fins madeireiros, arboricultura e sistemas agroflorestais. O formato do caule, a altura da ramificação e bifurcação, o tipo de casca e o hábito de frutificação apresentaram variação mínima nos indivíduos, ao contrário da retidão do caule e do formato da copa.

ABSTRACT Plant genetic resources for food and agriculture are ex situ conserved in germplasm banks as samples (accessions) of natural or naturalized populations, either as the originally sampled propagules (mainly seeds) or their multiplications. The premises underlying ex situ conservation are that (a) it is the safest and cheapest alternative for germplasm preservation for future generations and (b) accessions are representative of the genetic diversity encountered in nature. In the past decades, ideas, alternatives and considerations have been put forward on the topic, and protocols have been devised for plant germplasm sampling, conservation and multiplication. However, limitations in the management efficiency of germplasm banks have been pointed out by international organizations. In our opinion, germplasm banks in general need to revise their functioning and management at the light of principles and methods of Genetics. To that end, it is necessary to consider the reproductive biology of higher plants -whose genetic consequences at both the individual plant and the population levels are not always either fully understood or taken into account in devising the protocols-, the genetic structures of wild and cultivated populations, and the course of the genetic material in the populations. In this paper, we discuss the three topics and provide an example of a national forage breeding program, from germplasm bank accessions as the germplasm of origin to the obtainment of commercial cultivars. Finally, we present a proposal as a base for discussion among curators, researchers and breeders.


RESUMEN Los recursos genéticos vegetales para la alimentación y la agricultura se conservan ex situ en bancos de germoplasma como muestras (introducciones) de poblaciones naturales o naturalizadas ya sea como propágulos originales (mayoritariamente semillas) o sus multiplicaciones. Las premisas subyacentes son que (a) es la alternativa más segura y barata de preservación de germoplasma para futuras generaciones y (b) las introducciones son representativas de la diversidad genética que se encuentra en la naturaleza En las últimas décadas, se han presentado ideas, alternativas y consideraciones sobre el tema y se han elaborado protocolos para el muestreo, conservación y multiplicación de germoplasma. Sin embargo, organizaciones internacionales han señalado limitaciones en la eficiencia del manejo de los bancos de germoplasma. En nuestra opinión, se necesita revisar el funcionamiento y manejo de dichos bancos en general a la luz de los principios y métodos de Genética. Para tal fin, es necesario considerar la biología reproductiva de las plantas superiores -cuyas consecuencias genéticas a nivel de planta individual y de población no se comprenden en su totalidad o no se consideran al idear los protocolos -, las estructuras genéticas de poblaciones naturales y cultivadas, y el curso del material genético en las poblaciones. En este trabajo discutimos los tres temas y proveemos un ejemplo de un programa nacional de mejoramiento de forrajeras, desde las introducciones como germoplasma de origen hasta la obtención de cultivares comerciales. Finalmente, presentamos una propuesta como base de discusión entre curadores, investigadores y mejoradores.

Résumé

The objective of this study was to quantify the superovulatory response and embryo production of Brazilian Bergamasca sheep and to evaluate the link to the follicular condition before superovulatory treatment, as a reference for selection of donors with potential for superovulation. Follicular population of twenty-three sheep was evaluated by ultrasound during metestrus phase of the estrous cycle and divided into groups of low, medium and high follicular population. Subsequently, they were synchronized, superovulated with 133mg of pFSH, mated and subjected to embryo collection. The superovulatory response (9.0±3.3 vs 10.7±6.2 vs 13.8±7.1) and embryo production (4.0±3.8 vs 2.6±2.0 vs 1,8±4.0) were similar between groups (P>0.05). There was a positive correlation between the number of follicles during the metestrus phase and the number of corpus luteum with premature regression (PLR) (0.52) and a negative correlation between the recovery rate and PLR (-0.44) (P<0.05). The sheep that presented PLR had more follicles during metestrus (16.9±7.8 vs 12.7±3.2) and lower embryo recovery rate (38.8±29.3 vs 72.2±29.9) than those with functional CLs (P<0.05). Follicular quantification during metestrus phases was unable to identify donors with high embryo production. Animals with PLR had a larger follicular population during metestrus and lower embryo recovery rate.(AU)


O objetivo deste trabalho foi quantificar a resposta superovulatória e a produção embrionária de ovelhas Bergamácia Brasileira e relacioná-las com a condição folicular antes do tratamento superovulatório, como referência para seleção de doadoras com potencial para superovulação. Vinte e três ovelhas foram avaliadas quanto à população folicular por ultrassonografia na fase de metaestro do ciclo estral e divididas em grupos com baixa, média e alta população folicular. Posteriormente foram sincronizadas, superovuladas com 133mg de pFSH, acasaladas e submetidas à coleta de embriões. A resposta superovulatória (9,0±3,3 vs. 10,7±6,2 vs. 13,8±7,1) e a produção embrionária (4,0±3,8 vs. 2,6±2,0 vs. 1,8±4,0) foram semelhantes entre os grupos (P>0,05). Houve correlação positiva entre o número de folículos no metaestro e o número de corpos lúteos com regressão prematura (RPCL) (0,52) e correlação negativa entre a taxa de recuperação e RPCL (-0,44) (P <0,05). As ovelhas que apresentaram RPCL tiveram mais folículos no metaestro (16,9±7,8 vs. 12,7±3,2) e menor taxa de recuperação embrionária (38,8±29,3 vs. 72,2±29,9) do que as que apresentaram CLs funcionais (P<0,05). A quantificação folicular nas fases de metaestro não foi capaz de identificar doadoras com alto potencial de produção embrionária. Animais com RPCL tiveram maior população folicular no metaestro e menor recuperação de embriões.(AU)

Résumé

Cajui (Anacardium spp.) is a native fruit tree (small cashew) of the Brazilian Cerrado and possesses the potential for commercialization. However, cajui exploitation occurs exclusively through extractivism in the absence of conservation strategies. The lack of conservation strategies may lead to a decrease in genetic diversity of Anacardium. In this work, the genetic diversity and population structure of three natural populations in Sete Cidades National Park (PNSC; PI, Brazil) were assessed using ISSR analysis of 56 cajui accessions and two A. occidentale accessions (outgroup) from Pacajus (CE, Brazil). A total of 112 markers were obtained, 93 (83.04%) of which were polymorphic. The diversity indices of these populations indicated moderate levels of genetic diversity. According to AMOVA, 96.17% of the genetic variability lay within populations, with low genetic differentiation among populations (ΦST = 0.03828). Furthermore, STRUCTURE analysis indicated the existence of four connected genetic groups. The findings show that the individuals from the three collection sites did not represent different subpopulations, likely due to the high gene flow (Nm = 6.7) favored by the floral biology of Anacardium, pollinators and small-animal seed dispersers. This research identifies genetically divergent individuals (C-03, C-05, C-22, C-26, C-34 and C-39), which should be considered priority individuals for conservation and can inform conservation programs for Anacardium spp.

Propósito/Contexto. En este artículo se exploran los principios bioéticos de autonomía y justicia, a la luz de tres casos de uso de la biodiversidad, tanto para fines investigativos como de aprovechamiento comercial, que involucran territorios colectivos de comunidades indígenas, afrodescendientes y predios de población campesina. Metodología/Enfoque. En cada caso se examinaron sus características, los actores, las relaciones entre los actores y los beneficios contemplados para las partes. El estudio comprendió la recopilación y análisis de documentación y la realización de 10 entrevistas semiestructuradas con investigadores, representantes de instituciones, empresas y organizaciones indígenas y afrocolombianas. La investigación se realizó entre 2015 y 2017 e incluyó comunidades del Amazonas, Risaralda, Chocó y Antioquia. Resultados/Hallazgos. Se encontró que las instituciones de investigación y los investigadores tramitan y obtienen las autorizaciones reglamentarias, mientras que las entidades del Estado registran una progresiva omisión de responsabilidades que afecta los derechos de las comunidades, por ejemplo, a través de la inobservancia del derecho a la consulta previa. Adicionalmente, en los casos de estudio, las relaciones que los actores establecen con las comunidades INAPRR-CL (sigla de indígenas, negros, afrodescendientes, palenqueros, raizales, Rrom, campesinas y locales) varían en la observancia de principios bioéticos. Discusión/Conclusiones/Contribuciones. Debido a que el régimen jurídico tiene un alcance limitado para el uso de la biodiversidad en investigación o innovación tecnológica, se evalúa y se discute si la bioética podría servir de guía para el respeto de la autonomía y la participación de beneficios de las comunidades en proyectos de investigación y bioprospección.


Purpose/Context. This article explores the bioethical principles of autonomy and justice in the light of three cases of use of biodiversity, both for research purposes and for commercial use, involving collective territories of indigenous communities, afro-descendants, and proper-ties of the peasant population. Methods/Approach. In each case, we examined the characteristics, agents, relationships be-tween agents, and benefits contemplated for the parties. The study included document compilation and analysis and ten semi-structured interviews with researchers, representatives of indigenous and Afro-Colombian institutions, companies, and organizations. The research was carried out between 2015 and 2017 and involved Amazon, Risaralda, Chocó, and Antioquia communities. Results/Findings. Research institutions and researchers process and obtain regulatory authorizations, while State entities report a progressive omission of responsibilities that adversely affects communities' rights, for example, through non-observance of the right to prior con-sultation. Besides, in the study cases, the agents' relationships with the INAPRRCL (acronym in Spanish for indigenous, black people, Afro-descendants, palenqueros, raizales, Rrom, peasants, and locals) vary regarding the application of bioethical principles. Discussion/Conclusions/Contributions. Because the legal system has a limited scope on the use of biodiversity in research or technological innovation, we assessed and discussed whether bioethics could serve as a guide for respecting communities' autonomy and benefit share in research and bioprospecting projects.

Metodologia/Abordagem. Em cada caso, foram examinadas as suas características, os intervenientes, as relações entre os intervenientes e os benefícios previstos para as partes. O estudo incluiu a coleta e análise de documentação e a realização de 10 entrevistas semiestruturadas com pesquisadores, representantes de instituições, empresas e organizações indígenas e afro-colombianas. A pesquisa foi realizada entre 2015 e 2017 e incluiu comunidades do Amazonas, Risaralda, Chocó e Antioquia. Resultados/Descobertas. Verificou-se que as instituições de investigação e os investigadores processam e obtêm as autorizações regulamentares, no entanto que as entidades do Estado registram uma progressiva omissão de responsabilidades que afeta os direitos das comunidades, por exemplo, através da inobservância do direito à consulta prévia. Adicionalmente, nos casos de estudo, as relações que os atores estabelecem com as INAPRRCL (sigla de indígenas, negros, afrodescendentes, palenqueros, raizais, Rrom, camponesas e locais) variam na observância de princípios bioéticos. Discussão/Conclusões/Contribuições. Dado que o regime jurídico tem um âmbito limitado para a utilização da biodiversidade na investigação ou inovação tecnológica, se avalia e se discute se a bioética poderia servir de guia para o respeito da autonomia e a participação de benefícios das comunidades em projetos de pesquisa e bioprospeção.
