RESUMO

Abstract Background Previous studies has shown that nucleotide-binding and oligomerization domain-containing protein 2 (NOD2) is expressed in Fibroblast-like synoviocytes (FLSs) of rheumatoid arthritis (RA) patients which is stimulated by muramyl dipeptide (MDP) present in the joint environment and induces inflammation via the NF-κB pathway. Also, other studies have shown that curcumin inhibits proliferation, migration, invasion, and Inflammation and on the other hand increases the apoptosis of RA FLSs. In this study, we aim to evaluate the effect of curcumin, a natural antiinflammatory micronutrient, on the expression of NOD2 and inflammatory cytokines. Methods Synovial membranes were collected from ten patients diagnosed with RA and ten individuals with traumatic injuries scheduled for knee surgery. The FLSs were isolated and treated with 40 μM curcumin alone or in combination with 20.3 μM MDP for 24 h. mRNA was extracted, and real-time PCR was performed to quantitatively measure gene expression levels of NOD2, p65, IL-6, TNF-α, and IL-1β. Results The study findings indicate that administering MDP alone can significantly increase the mRNA expression levels of IL-6 and IL-1β in the trauma group and TNF-α in the RA group. Conversely, administering curcumin alone or in combination whit MDP can significantly reduce mRNA expression levels of P65 and IL-6 in FLSs of both groups. Moreover, in FLSs of RA patients, a single curcumin treatment leads to a significant reduction in NOD2 gene expression. Conclusion This study provides preliminary in vitro evidence of the potential benefits of curcumin as a nutritional supplement for RA patients. Despite the limitations of the study being an investigation of the FLSs of RA patients, the results demonstrate that curcumin has an anti-inflammatory effect on NOD2 and NF-κB genes. These findings suggest that curcumin could be a promising approach to relieve symptoms of RA.

RESUMO

Abstract Introduction The isolation of captured peripheral blood mononuclear cells (PBMNCs) from leukoreduction filters (LRFs) can be of great importance in terms of bringing the lost cells back into use. Objective The aim of this study was to evaluate various methods based on their potential to recover the peripheral blood cells from LRFs with a focus on mononuclear cells (MNCs). Method For cell isolation from LRFs, three distinct methods (back-flushing, direct and vacuum pump) were compared through the calculation of the yield of isolated MNCs. The viability of extracted cells was determined by the flow cytometry technique. Moreover, the recovered MNCs were characterized regarding the presence of blood stem cell purification. The cell culture, microscopic observation, and immunophenotyping were employed to characterize the blood stem cells (hematopoietic, mesenchymal and progenitor endothelial stem cells). Results The yield of isolation obtained in the back-flushing, direct and vacuum pump methods were 17.7 ± 1.28, 17.3 ± 0.96 and 21.2 ± 0.90 percent, respectively. Although the highest potential for total blood cell recovery belonged to the vacuum pump method, the lowest cell viability (85.73 ± 4.84%) was observed in this method. However, the isolation process of the back-flushing and direct methods had less effect on cell viability. The characterization of the isolated MNCs displayed that the dominant positive phenotype was for CD34/CD45, indicating hematopoietic stem cells. In addition, the endothelial stem/progenitor cells were significantly detected as CD31/CD133 positive cells. Conclusion According to our results and considering the safety and efficiency potential of each of the applied methods, the back-flushing in comparison with the other methods can be considered a suitable procedure for MNC isolation from LRFs.

RESUMO

Abstract Glucose exposure induces toxic effects on the function of the pancreatic islets. Moreover, myricitrin as a flavonoid glycoside may have favorable effects on insulin secretion of Langerhans islets. The present study aimed to investigate the effect of Myricitrin and its solid lipid nanoparticles (SLN) on the insulin secretion as well as the content of isolated pancreatic islets from male mice. In this experimental study, Langerhans islets were separated from adult male NMRI mice using the collagenase method. The insulin secretion and content of islets were assessed in glucose-containing medium (2.8, 5.6, and 16.7mM). Further, islets treated were prepared by the administration of Myricitrin and its SLN (1, 3 and 10µM). Myricitrin 3µM, and SLN containing Myricitrin 3 and 10µM increased insulin secretion in medium containing glucose concentration 2.8mM. Accordingly, this variable increased in Myricitrin 3 and 10µM, SLN containing Myricitrin 1, 3, and 10µM utilization as well as glucose concentration 5.6mM. Afterward, the insulin secretion increased in medium containing 16.7mM glucose after the addition of Myricitrin and SLN containing Myricitrin 1, 3, and 10µM. Also, the insulin content increased in Myricitrin and SLN containing Myricitrin 1, 3, and 10µM administered groups in all medium containing glucose concentrations. Myricitrin and its SLN increased islets insulin secretion and content in low, moderate, and high glucose concentration mediums

RESUMO

ABSTRACT Introduction: Peripheral blood leukocytes are a suitable cell model for science research. However, blood samples from healthy volunteers are limited in volume and difficult to obtain due to the complexity of volunteer recruitment. Objective: Therefore, it is urgent to find an alternative source of peripheral blood leukocytes. Method: One of the possibilities is the use of leukocyte reduction filters (LRFs) in blood banks that is used for preparation of leukoreduced blood products. More than 90% of the leukocytes are trapped in the leukofilters allowing the desired blood product to pass through. Results: It has been reported that the biological function of leukocytes collected from the filters are no different from those isolated from buffy coats, leukapheresis products and whole blood (WB) cells. Moreover, LRFs are waste products that are discarded after leukoreduction. Conclusion: Thus, leukofilters represent an economic source of human cell populations that can be used for a variety of investigative purposes, with no cost. In the present study, we reviewed the different usage of LRFs in the research, clinical and commercial applications.

RESUMO

Abstract Introduction Gross's Emotion Regulation Questionnaire is one of the most widely-used and valid questionnaires for assessing emotion regulation strategies. The validity and reliability of the Persian version have not been determined and data on its psychometric properties are not available to Iranian mental health researchers. The purpose of this study was to determine the psychometric properties of the Emotion Regulation Questionnaire in Iranian students. Methodology In this cross-sectional study, 348 students (170 males and 178 females) were selected from Shahid Beheshti University of Medical Science and Tehran University of Medical Science. The following statistical procedures were conducted: correlation coefficients, factor analysis, Cronbach's alpha, and independent t tests. Results The results showed that men use suppression more than women (T = -2.62, p = 0.009). Cronbach's alpha coefficients were 0.76 for the cognitive reappraisal sub-scale and 0.72 for the suppression sub-scale (excluding question 9). Six questions related to the cognitive reappraisal factor explained 30.97% of emotion regulation variance, and 3 questions related to the suppression factor explained 22.59% of emotion regulation variance. Overall, these factors explained 53.5% of emotion regulation variance. There were significant correlations between suppression and difficulties in emotion regulation, trait anxiety, and affective control. Furthermore, there was a significant correlation between cognitive reappraisal and the Five-Facet Mindfulness Questionnaire. Conclusion The results indicate that the Persian version of the ERQ is a reliable and valid instrument that can be helpful for development of further important studies of emotional regulation.

RESUMO

Mycobacterium tuberculosis has infected more than two billion individuals worldwide, of whom 5%–10% have clinically active disease and 90%–95% remain in the latent stage with a reservoir of viable bacteria in the macrophages for extended periods of time. The tubercle bacilli at this stage are usually called dormant, non-viable, and/or non-culturable microorganisms. The patients with latent bacilli will not have clinical pictures and are not infectious. The infections in about 2%–23% of the patients with latent status become reactivated for various reasons such as cancer, human immunodeficiency virus infection, diabetes, and/or aging. Many studies have examined the mechanisms involved in the latent state of Mycobacterium and showed that latency modified the expression of many genes. Therefore, several mechanisms will change in this bacterium. Hence, this study aimed to briefly examine the genes involved in the latent state as well as the changes that are caused by Mycobacterium tuberculosis. The study also evaluated the relationship between the functions of these genes.

RESUMO

Purpose@#This study was performed to investigate the efficacy and safety of short-course radiation therapy (SCRT) and sequential chemotherapy followed by delayed surgery in locally advancer rectal cancer with subgroup analysis between the older and young patients. @*Materials and Methods@#In this single-arm phase II clinical trial, eligible patients with locally advanced rectal cancer (T3–4 and/or N1–2) were enrolled. All the patients received a median three sequential cycles of neoadjuvant CAPEOX (capecitabine + oxaliplatin) chemotherapy. A total dose of 25 Gy in five fractions during 1 week was prescribed to the gross tumor and regional lymph nodes. Surgery was performed about 8 weeks following radiotherapy. Pathologic complete response rate (pCR) and grade 3–4 toxicity were compared between older patients (≥65 years) and younger patients (<65 years). @*Results@#Ninety-six patients with locally advanced rectal cancer were enrolled. There were 32 older patients and 64 younger patients. Overall pCR was 20.8% for all the patients. Older patients achieved similar pCR rate (18.7% vs. 21.8; p = 0.795) compared to younger patients. There was no statistically significance in terms of the tumor and the node downstaging or treatment-related toxicity between older patients and younger ones; however, the rate of sphincter-saving surgery was significantly more frequent in younger patients (73% vs. 53%; p=0.047) compared to older ones. All treatment-related toxicities were manageable and tolerable among older patients. @*Conclusion@#Neoadjuvant SCRT and sequential chemotherapy followed by delayed surgery was safe and effective in older patients compared to young patients with locally advanced rectal cancer.

RESUMO

Plant growth-promoting rhizobacteria (PGPR) with multiple beneficial traits serve as potentially, ecofriendly, and cost-effective alternatives to chemical fertilizers and pesticides. They have both direct and indirectaffirmative impacts on overall plant growth and health. PGPRs are well known to directly improve the plantgrowth by phytohormone production and availability of minerals in soil. A total of nine soil samples were takenfrom near the rhizospheric zone of different crops and 56 rhizobacterial strains were isolated. Only 16 out of56 rhizobacterial strains were found positive for more than one beneficial trait that included solubilization ofphosphate, indole acetic acid (IAA), siderophore, ammonia and H2S production. Among all PGPR strains,RKM15 was observed having the highest phosphate solubilizing index (3.4), solubilized phosphorus (339mg L−1) and also siderophore unit (70.54 %). The maximum IAA production was observed by RKM25 strain(35.56 µg ml−1). The most promising RKM15 isolate was identified as Pantoea dispersa (MN629239) through16S rRNA gene sequencing technique. This characterized PGPR strain may be used for the development ofbiofertilizers to enhance crop productivity and improvement in soil fertility

RESUMO

Introduction:Obesity is closely associated with various types of illness, primarily caused by more calorie intake than body burn. In adipocytes, Calcium (Ca2+) is an important second messenger involved in theregulation of many physiological functions which are essential for survival. In the present research, we have investigated the role of Ca2+ions in obesity by manipulating cytosolic Ca2+ion concentration by selective blocking/advancing the Ca2+ions through the voltage-gated calcium channels. Voltage-gated calcium channel (vCa) plays a key role in regulating intracellular and extracellular Ca2+concentration.Cytoplasmic level of Ca2+was manipulated by supplying calcium carbonateand by using vCa blockers i.e. nifedipine-(N-type-vCa-CCB) and ethosuximide (T-type, vCa-CCB).Methods:Obesity was induced by progesterone in female mice and test drugs were co-administered with progesterone whereas sibutramine was used as standard. The treatment was carried out for 28 days, during and afterthetreatment periodvarious parameters were studied viz food consumption, change in body weight and temperature, the effect on WAT (white adipose tissue, adiposity index, histology of fat pad) and fecal lipid content. Results:Calcium carbonate treated group has shown promising effects in the decrease in body weight by increasing fecal lipid content and lipolysis which was reflected by an increase in body temperature. Ethosuximide also offered significant protection by decreasing the food intake but has not shown any notable effect on fecal fat content, whereas nifedipine has not offered any protection against the obesity induced by neurosteroid. Conclusion:Calcium carbonate has significant anti-obesity activity by including thermogenesis, and increasing fecal lipid content

RESUMO

The aim of this study was to contrast the effects of drought stress on polyamine oxidases gene expression andactivity as well as photosynthetic efficiency in relatively tolerant (Karoon) and sensitive (260) maize genotype.d Reduction in leaf relative water content as a result of drought led to increase in root growth, butdiminished shoot growth indices. Under drought stress, activity of antioxidant enzyme, catalase, significantlyincreased in both genotypes, whereas significant higher activity of superoxide dismutase and peroxidase wasonly observed in Karoon genotype. Expression of polyamine oxidase (PAO) genes (zmPAO1, zmPAO2,zmPAO3, zmPAO4, zmPAO5, zmPAO6) and activity of enzymatic polyamine oxidation was increased in bothgenotypes under drought stress. The enhancement in PAO gene expression and enzyme activity was moreprominent in Karoon cultivar compared to 260. Chlorophyll a fluorescence and fast induction kinetics werenegatively influenced by drought stress. These parameters were more affected in 260 cultivar compared withKaroon. Our results suggest that under drought stress, higher activity of polyamine oxidase pathway in backconversion of Spermine and spermidine to putrescine (protectant of photosynthetic apparatus) as well as higherantioxidant enzymes activity in Karoon cultivar, may play a role in higher efficiency of photosynthetic processin this cultivar

RESUMO

Background: There is paucity of data regarding the relationship between the severity of left ventricular dysfunction and renal function impairment in diabetic patients. Aims and objectives was to study the relation between eGFR and ejection fraction in type 2 diabetes mellitus of more than 5 years duration. Methods: This cross sectional was carried out from May 2017 to May 2018 at Yenepoya Medical college a tertiary care center in the southern Indian state of Karnataka. The data on 220 diabetic patients were studied. A detailed clinical history physical examination was carried out on the study population as per the approved proforma. Apart from base line investigations, eGFR was calculated using serum creatinine levels. 2D echocardiography was done for the assessment of ventricular function. Results: Out of 220 patients, 138 were men (63%) and 82 were women (37%) and the mean age of the patients was 59.60±11.145 years. The mean duration of diabetes was 10.08±5.28 years. We found a progressive reduction in ejection fraction from 57.74±9.97% to 50.64±14.7 as the eGFR declined from <90 mL/min/1.73m2 to 30 mL/min/1.73m2. There was significant correlation between eGFR and RWMA (p value 0.001) and LVDD (p value 0.029) in this study cohort. Micro/macroalbuminuria was found to be in 55% of patients. Proteinuria had significant correlation with longer duration of diabetes, blood urea, serum creatinine and eGFR.Conclusions: This study found positive clinical correlation between LVEF and eGFR. However, it did not reach significant levels statistically. Further studies may be carried out to confirm the association.

RESUMO

ABSTRACT Colorectal cancer is one of the most important malignancies in the classification of gastrointestinal cancers. One of the predisposing factors at molecular level for this cancer is via WNT signaling which is associated with the vast numbers of different genes. Thus, in this study, we aimed to investigate whether Adenomatous Polyposis Coli gene (APC) mutation of rs41115in two locations such as 132.002 and 131.989 acts as a trigger or cause of colorectal cancer. Relatively, 30 blood samples of colorectal cancer patients and 30 normal blood samples as control group after colonoscopy and also confirmation of pathology report at Rohani Hospital in Babol (Iran) were investigated. The primers were designed in order to be included the rs41115 to identify the particular polymorphisms of gene. The polymerase chain reaction (PCR direct sequencing method) was used. Conclusively, deletion of adenine in two specific locations such as 131.989 and 132.002 has been identified, but there was no relationship between rs41115 polymorphisms located in adenomatous polyposis coli gene and colorectal cancer.

Objective@#The strong antioxidant activity of Commiphora mukul prompted us to conduct the present study to explore whether treatment with C. mukul extract (CME) would have any protective influence on sperm parameters, testosterone levels, and plasma glucose levels in streptozotocin (STZ)-induced diabetic rats. @*Methods@#Male Wistar rats were randomly divided into four groups: control, control animals treated with CME, diabetic animals, and diabetic animals treated with CME. CME extract (300 mg/kg) was administered for 60 days by daily gavage. Diabetes was induced by an intraperitoneal injection of 50 mg/kg STZ. The epididymal sperm count, weight, motility, morphology, viability, and serum testosterone and glucose levels were determined. @*Results@#In the diabetic animals, CME decreased blood glucose levels (p<0.05), increased the total sperm count (p<0.05), and decreased the proportion of sperm with abnormal morphology (p<0.05). Diabetes reduced sperm motility (p<0.001), and CME supplementation partially reversed this effect of diabetes (p=0.003). Furthermore, in diabetic animals, CME decreased the proportion of immotile sperm (p<0.001). In rats, diabetes caused a significant decrease (p<0.05) in serum testosterone levels (F[3, 28]=3.283, p=0.035), but treatment of diabetic animals with CME increased serum testosterone levels. @*Conclusion@#The present study demonstrated that C. mukul possesses proandrogenic activity and exerts a beneficial effect on sperm parameters in diabetic rats.

RESUMO

Abstract INTRODUCTION: The human T-lymphotropic virus type 1 (HTLV-1) has a single-stranded RNA genome and expresses specific proteins that have oncogenic potential. Approximately 15 to 20 million people worldwide have been infected by this virus. Changes in protein or gene expression are the effects of single nucleotide polymorphisms (SNPs) within the Toll-like receptor 3 (TLR3) gene. The function and efficacy of signal transduction also lead to modified immune responses. The present study aimed to investigate the association of SNPs within TLR3 (rs3775291 and rs3775296) with susceptibility to HTLV-1 infection in Iranian asymptomatic blood donors. METHODS: This study was performed on 100 HTLV-1-infected asymptomatic blood donors and 118 healthy blood donors. Genomic DNA from all participants was purified and then amplified using specific PCR primers. SNPs within TLR3 were evaluated using the restriction fragmentation length polymorphism technique, and the results were analyzed using SPSS software (version 22). RESULTS: The frequencies of the TLR3 (rs3775296) CC, CA, AA genotypes were 70%, 24%, and 6% in the patient group, and 50.8%, 44.9%, and 4.2% in the control group, respectively. There was a significant difference in the frequency distribution of TLR3 (rs3775296) genotypes and alleles, but not in the frequency distribution of TLR3 (rs3775291) genotypes between the patient and control groups. CONCLUSIONS: The TLR3 SNP rs3775296 was significantly associated with HTLV-1 infection and may be a protective factor against this viral infection.

RESUMO

Introduction: the current COVID-19 pandemic has occasioned the increased adoption of telemedicine. This study reports the uptake and trend of a new teleconsultation service in a Tanzanian hospital. Methods: this is a retrospective observational study that profiled requests for teleconsultations and uptake of the service between April 1, 2020, and June 30, 2020. Results: two hundred and eighteen telephone inquiries were received over the 3 months. One hundred and sixteen (53.2%) individuals followed through with the teleconsultations. Paediatric (38.8%) and Internal medicine (32.8%) were the subspecialties with the highest number of teleconsultations. In a frame of 3 months, teleconsultation uptake was highest in May and lowest in June. Conclusion: there was a steady rise and a rapid fall in requests and uptake of teleconsultation services over the period under evaluation. Lack of insurance coverage for teleconsultations was a significant barrier. We propose a re-education and reiteration of the benefits of telemedicine to all stakeholders. This is important for the current era and beyond

RESUMO

Abstract Introduction: Anxiety sensitivity plays a prominent role in the etiology of anxiety disorders. This construct has attracted widespread interest from experts and researchers. The Anxiety Sensitivity Index (ASI-3) is the most common scale for measuring anxiety sensitivity. Objective: To analyze the psychometric properties and factor structure of the ASI-3 in Iranian student samples. Methods: 220 students (135 women, 85 men) from Kermanshah University of Medical Sciences were selected by the convenience sampling method to evaluate the psychometric properties and analyze the factor structure of the ASI-3. The subjects were also asked to complete the Acceptance and Action Questionnaire-II (AAQ-II), Whiteley Index, Intolerance of Uncertainty, and Neuroticism scales. LISREL and SPSS were used to analyze the data. Cronbach's alpha and correlation coefficients were calculated and confirmatory factor analysis was conducted. Results: The results of the confirmatory factor analysis revealed a three-factor structure with physical, cognitive, and social components (comparative fit index = 0.94; normed fit index = 0.91; root mean square error of approximation = 0.09). The ASI-3 had positive and significant correlations with health anxiety (0.59), intolerance of uncertainty (0.29), and neuroticism (0.51). Furthermore, the ASI-3 had a negative and significant correlation with the AAQII (-0.58). Cronbach's alpha coefficients for the whole scale and for the physical, cognitive, and social concerns factors were 0.90, 0.74, 0.79, and 0.78, respectively. The invariance of the index was significant compared to the original English version. Conclusion: In general, the results support the adequacy of the psychometric properties of the Persian version of the ASI-3. Theoretical and applied implications will be discussed.

Eles foram solicitados a completar os seguintes instrumentos: Acceptance and Action Questionnaire-II (AAQ-II), Whiteley Index, Intolerance of Uncertainty e Neuroticism.

Objective:Nonalcoholic fatty liver disease (NAFLD) is rapidly going to be one of the most common metabolic diseases which can negatively affect the liver function and needs to be more explored within the context of new and efficient therapies. The aim of this study was to determine the benefit of Cynara cardunculus var scolymusand Cinnamomum zeylanicummixture infusion (1.5 and 0.25 g/100 mL and twice/day) on patients with NAFLD. Methods:The study was designed as a before-after clinical trial and performed on 20 patients having NAFLD. They were asked to use the mixture of extracts prepared in special bags twice a day for 30 days. Before and after using the mixture, Liver markers of NAFLD including alanin aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphates (ALP), cholesterol (chol), triglycerides (TG), high-density lipoprotein (HDL) and low-density lipoprotein (LDL) and also, fatty liver ultrasonograhic grading were measured. Results:Treatment with mixture of extracts in patients with NAFLD resulted in a significant decrease in ALT, AST, and ALP. Also there was a significant improvement in lipid profile of the patients given by decreased TG, Chol, and LDL and increased HDL in serum. Moreover, a significant decrease in grade of sonographic examination was showed. No significant linear correlation was found between age, BMI and gender one way side and biochemical parameters and liver enzymes. Conclusion:Taken together, it is concluded that the mixture of Cynara cardunculusvar scolymus, Cinnamomum zeylanicumcan have beneficial effects on hepatic function and lipid profiles of patients with NAFLD

RESUMO

To study the effects of some new facilitators on the vegetative and morphological traits of Thymus kotschyanus, nine treatments were tested in the experimental rangeland field at the University of Mohaghegh Ardabili, Ardabil, Iran. Treatments included control, potassium silicate nanoparticles (PSN) with two levels of 500 and 1000 mg/lit, superabsorbent hydrogel (SH) with two levels of 10 and 30 g/kg, animal manure (AM) with two levels of 100, 200 g/kg, and effective microorganisms (EM) with two levels of 1 and 2%. Data were subjected to one-way analysis of variance (ANOVA). Results of mean comparisons of treatments for Thymus kotschyanus characteristics showed that the highest amount of studied traits were observed in the treated SH 30 g/kg. These traits include plant height (19.44 cm), basal area (4.66 cm), canopy cover (99.11%), number of secondary branches (9.44) and depth of rooting (16.49 cm), aerial parts volume (26.77 cm3), root volume (17.66 cm3), aerial parts fresh weight (14.40 g), aerial parts dry weight (7.18 g), root fresh weight (3.98 g), and root dry weight (2.07 g). In general, the impact of treatments on Thymus growth traits were ranked as follows: SH 30 g/kg, PSN 1000 mg/lit, AM 200 g/kg, SH 10 g/kg, PSN 500 mg/lit, AM 100 g/kg, EM 2%, and EM 1%. In addition, the lowest plant traits were found in control. Overall, it is recommended extending the cultivation of this native medicinal plant by considering ecological conditions in each region. In addition, to promote the establishment and facilitate the growth of planted species, it is recommended using the facilitators utilized in the present work.
