RESUMO

Background: Maintenance of hygiene is very important to lead a healthy, disease-free life. Lack of hygiene increases the chance of communicable and non-communicable diseases. Wastes generated by the hospitals increase the chance of nosocomial infections. Objective: The present study deals with the patient’s perception about hygiene and sanitation rendered by the hospitals. Materials and Methods: The present study was a questionnaire-based cross-sectional survey. The data were collected from the indoor patients admitted in the Department of Surgery, Obstetrics and Gynecology, and General Medicine during December 2016–January 2017 at a Medical College in Kolkata. Results: In the present study, a total of 150 patients were taken as study population, of which majority of them were female. The major age group of the study population was 32–39 years, 33 (22%). In the present study, 93.33% of patients responded that the food was hygienic, 90% of patients opined that all housekeeping staffs used their protective equipment while they were collecting the garbage in this hospital, and 78% of patients agreed that the dustbins were cleaned regularly. Conclusion: In order to improve patients’ compliance and reduce the sufferings, evaluation of to hospital hygiene is of utmost importance. Awareness programs should be implemented in hospitals and health-care centers about the importance and maintenance of hygiene. This would reduce the chances of hospital-borne infections not only for the patients but also for the hospital workers.

Background and Objectives: Emergence of multi‑drug resistant Neisseria gonorrhoeae resulting from new genetic mutation is a serious threat in controlling gonorrhea. This study was undertaken to identify and characterise mutations in the mtrR genes in N.gonorrhoeae isolates resistant to six different antibiotics in the quinolone group. Materials and Methods: The Minimum inhibitory concentrations (MIC) of five quinolones for 64 N.gonorrhoeae isolates isolated during Jan 2007–Jun 2009 were determined by E‑test method. Mutations in MtrR loci were examined by deoxyribonucleic acid (DNA) sequencing. Results: The proportion of N.gonorrhoeae strains resistant to anti‑microbials was 98.4% for norfloxacin and ofloxacin, 96.8% for enoxacin and ciprofloxacin, 95.3% for lomefloxacin. Thirty‑one (48.4%) strains showed mutation (single/multiple) in mtrR gene. Ten different mutations were observed and Gly‑45 → Asp, Tyr‑105 → His being the most common observed mutation. Conclusion: This is the first report from India on quinolone resistance mutations in MtrRCDE efflux system in N.gonorrhoeae. In conclusion, the high level of resistance to quinolone and single or multiple mutations in mtrR gene could limit the drug choices for gonorrhoea.

Angiogenesis plays an important role in the pathogenesis of haematological neoplasms and may be correlated with the prognosis. We recently evaluated the microvessel densities in trephine biopsy sections of seventeen patients of myelodysplastic syndromes (MDS). Of the 17 cases, 2 were RAEB-t, 3 were RAEB, one was RARS and 11 were of the subtype RA (FAB subtyping). The microvessel counts were measured in the bone marrow biopsy sections by immunohistochemical staining, using CD34 reactive monoclonal antibodies. MVD was significantly higher in the cases of RAEB and RAEB-t as compared to the cases of RA. The average MVD per x400 in the cases of RA was 5.7 +/- 4.7 with a median value of 4.65 (range 19) whereas it was 45.4 +/- 10.0 and 44.0 (range 27.3) respectively in RAEB and RAEB-t (p <.001), the 95% confidence interval being (2.94, 8.5) and (36.6, 54.3), for the two groups respectively. This finding may imply that subtypes of MDS with a higher tendency for converting to acute leukaemia are associated with increased angiogenesis as compared to other subtypes where the risk of progression to acute leukaemia is much lower.

RESUMO

Immature zygotic embryo explants of Carica papaya were cultured on MS medium supplemented with 2,4-D (2.0 mg/l) and formed globular embryos on explants without callus formation in 4-6 weeks. Maturation and conversion of somatic embryos was also achieved on the same medium. Cotyledonary stage embryos germinated to 63.66 and 68.33% in cv. honey dew and washington respectively in MS basal medium supplemented ABA (0.5 microm/l). Robust development and proliferation of plantlet roots in vitro was obtained on MS basal medium. Hardened plantlets have 60% survival rate.

BACKGROUND & OBJECTIVES: Although Escherichia coli heat stable enterotoxin (STa) causes diarrhoea in laboratory animals, no studies were done to find out the species specific variation of distribution of the STa receptors in laboratory animals. The present investigation evaluates the density of STa receptors and the guanylyl cyclase (GC) activity in the small intestinal epithelial cells of hamsters and guinea pigs. METHODS: Brush border membrane (BBM) was prepared from the small intestines of hamsters and guinea pigs. Receptor binding assay, GC assay and autoradiography were performed to determine the density of STa receptors, the GC activity and molecular weights of the STa binding proteins respectively. RESULTS: The receptor densities, per mg BBM protein at equilibrium, were found to be 4.1 x 10(9) and 1.5 x 10(12) in hamsters and guinea pigs respectively. The GC activity was found to be lower in STa treated hamster BBM compared to that of guinea pig. Scatchard analysis of the stoichiometric data showed a linear plot, and STa bound with association constants of 0.31 x 10(12) M-1 and 1.04 x 10(12) M-1 in hamsters and guinea pigs respectively. Autoradiographic analysis of the SDS-PAGE, revealed that 125I-STa bound apparently to a 45 kDa membrane protein in hamster and a 115 kDa membrane protein in guinea pig. INTERPRETATION & CONCLUSIONS: It appears that a lower density of STa receptor exists in hamsters compared to that in guinea pigs. STa binds with a single population of STa receptors in each species with different ligand binding affinities. Also, the molecular weights of the STa binding proteins differ in these species. Moreover, the GC activity was found to be lower in hamsters than in guinea pigs.

RESUMO

Susceptibility tests were carried out with DDT (4%), dieldrin (0.4%) and malathion (5%) using the WHO test kits against Phlebotomus argentipes sandflies collected from Dankuni, situated in Hoogly district, 21 km from Calcutta, West Bengal. P. argentipes was found to be susceptible to all the three insecticides tested. LT 50 was calculated to be 10.6 minutes, 10.2 minutes and 2.8 minutes for DDT, dieldrin and malathion against the flies.

RESUMO

A simple growth medium for primary isolation and subsequent cultivation of Leishmania donovani promastigotes without using whole blood is described. This medium is modified from Aljeboori's biphasic medium (used originally only for cultivation), containing only beef extract, peptone, sodium chloride, bactoagar and foetal calf serum (FCS). We have modified the medium by adding glucose and ascertaining the pH in the solid phase and by drastically reducing (91%) FCS in the liquid phase. The medium helps in isolation of L. donovani promastigotes from kala-azar patients, in addition to luxurious growth of parasites. The medium is simple, reliable, reproducible and convenient, with minimal interference in using the parasitic cells for immunological, molecular and isoenzyme studies.

RESUMO

Cryopreservation of promastigotes of L. donovani in NNN and Tobie's media was attempted. The media containing the promastigotes were kept directly at -80 degrees C. The cryopreserved media were examined for living promastigotes after 9, 12, 15, 20, 23, 26, 29, 32, 42, 56, 65 and 126 days after the culture tubes were brought directly from -80 degrees C to room temperature and examined after 30 min. All tubes showed living promastigotes which were used for further growth with no apparent morphological changes in subsequent subcultures. Viability was optimum. This method of short term cryopreservation is simple, reliable and reproducible for cryopreservation of culture adapted parasites and also important because different strains can be stocked without using any other chemical, or special equipment and liquid nitrogen chamber, for further use in immunological or other purposes.

The effect of propranolol and acebutolol therapy on serum lipoproteins was studied in thirty patients (mean age 43.9 +/- 8.7 years) with essential hypertension and/or stable myocardial ischaemia. Patients received six weeks therapy with propranolol (80-160 mg per day) and acebutolol (400-800 mg per day) each in random order and were changed over from one to the other beta-blocker after 6 weeks therapy. On propranolol treatment there was no significant effect on total cholesterol and LDL-cholesterol but the concentration of serum triglycerides (94.2 +/- 37.7 to 129.1 +/- 41.2 mg/dl) and VLDL cholesterol (18.9 +/- 7.8 to 26.1 +/- 8.1 mg/dl) significantly increased, concentration of HDL cholesterol (49.5 +/- 9.4 to 42.4 +/- 8.7 mg/dl) significantly decreased (p less than 0.01). Atherogenic index also worsened from 4.17 +/- 0.48 to 5.15 +/- 0.54 (p less than 0.05) on propranolol therapy. Acebutolol therapy, on the other hand, produced no significant change in the concentration of total cholesterol, triglycerides, HDL cholesterol, LDL cholesterol and VLDL cholesterol.
