RESUMO

Hepatic fibrosis characterized by various chronic liver injuries can lead to abnormal activation of hepatic stellate cells, unbalanced production and degradation of extracellular matrix proteins, and excessive deposition that destroys the normal structure of the liver. The aggravated liver fibrosis can cause irreversible cirrhosis and hepatocellular carcinoma, becoming a great challenge to the global health. Ferroptosis is a new form of iron-dependent cell death discovered in recent years, which mainly involves abnormal iron metabolism, lipid peroxide accumulation, and weakening of the antioxidant defense system. A number of studies have reported that inducing ferroptosis in hepatic stellate cells or alleviating ferroptosis in the liver can ameliorate liver fibrosis and reduce liver injury. Chinese medicine widely applied in the treatment of chronic liver diseases has demonstrated good safety, wide therapeutic effects, and easy access compared with Western medicine. Therefore, The intervention of hepatic stellate cells or hepatic ferroptosis by Chinese medicine may be a new direction for the prevention and treatment of liver fibrosis in the future. This paper summarized the various regulatory mechanisms of ferroptosis and expounded how ferroptosis affected the progression of liver fibrosis, providing theoretical support for the prevention and treatment of liver fibrosis with Chinese medicine in the future.

RESUMO

Based on the relevant guidelines published by the International Atomic Energy Agency and the Ministry of Ecology and Environment of the People's Republic China, this article summarizes the process of conducting a comprehensive radiological emergency exercise by reviewing the achievements and experience of radiological emergency exercises in various provinces of China since 2014. In addition, based on the actual combat requirements of the exercises, this article details the technical requirements and standard methods of exercise planning, preparation, implementation, and evaluation, so as to provide a reference for conducting radiological emergency exercises.

OBJECTIVES@#This study was performed to review the efficacy of curcumin in the treatment of oral submucous fibrosis systematically.@*METHODS@#We searched seven databases, including Web of Science, PubMed, EBSCO, The Cochrane Library, CNKI, WanFang Data, and VIP, to obtain randomized controlled trials related to the treatment of oral submucous fibrosis by curcumin. Each database was searched from inception to 30 June 2019. RevMan 5.3 software was used for the meta-analysis.@*RESULTS@#Six randomized controlled trials involving 350 patients were included in this study. Meta-analysis showed that curcumin can increase the maximal mouth opening and improve burning sensation compared with placebo treatment. Curcumin was not as effective as the controls in achieving maximal mouth opening after 1 month of treatment. However, no statistically significant difference was observed between the treatments from 2 months to 6 months. Curcumin significantly improved burning sensation compared with the controls after 3 months of treatment. No statistically significant diffe-rence in burning sensation was observed between the curcumin and control groups after 1, 2, and 6 months of treatment.@*CONCLUSIONS@#The current evidence shows that curcumin is an effective treatment for improving maximal mouth opening and burning sensation in patients with oral submucous fibrosis. Given the limited number and low quality of the included studies, however, more high-quality studies are needed to verify these findings.

<p><b>OBJECTIVE</b>To explore the relationship between the expression of Survivin and Ki67 with prognosis of pancreatic endocrine tumors (PETs).</p><p><b>METHODS</b>Immunohistochemistry for Survivin and Ki67 was performed in 25 cases of normal pancreatic tissues and 81 cases of PETs by tissue microarrays and to observe the expression and evaluate the relationship with prognosis.</p><p><b>RESULTS</b>(1)The expression of Survivin and Ki67 in PETs was significantly higher than that in normal pancreatic tissues (P <0.01); (2)The expression of Survivin and Ki67 in PETs was correlated with tissue grading and the TNM-staging (P < 0.05), but not related with tumor size, location and functional status. In addition, the expression of nuclear Survivin was association with lymph node metastasis (P < 0.05). (3)The high expression of Ki67 was related with the expression of nuclear Survivin, but not related with the expression of cytoplasmic Survivin.</p><p><b>CONCLUSION</b>Survivin and Ki67 were both expressed in PETs, which were closely related to the clinical pathological characteristics. They could be used as new indicators in the evaluation of prognosis of PETs. The expression of Survivin in nucleus had more diagnostic significance than that in cytoplasm, and that could be highly correlated with lymph node metastasis, which would be used as a new marker of poor prognosis.</p>

<p><b>OBJECTIVE</b>To observe the protective effect and mechanism of intermittent hypobaric hypoxia (IHH) on cardiomyocytes induced by hydrogen dioxide.</p><p><b>METHODS</b>Male guinea pigs were divided randomly into two groups (n = 10): intermittent hypoxia group (IHH), and control group (non-IHH). The IHH guinea pigs were exposed to a simulated 5,000 m high altitude and hypoxia in hypobaric chamber for 28 d, 6 h/d. The control guinea pigs were kept in the same environment as IHH except hypoxia exposure. Cardiomyocytes were enzymatically isolated from left ventricle of non-CIHH or CIHH guinea pigs. The contractile was assessed in guinea pigs by a video-based motion edge-detection system. The contents and activities of malondialdehyde(MDA), lactate hydrogenase(LDH) and antioxidant enzymes were evaluated by using biochemical methods.</p><p><b>RESULTS</b>1. Hydrogen peroxide could induce contractile and diastole dysfunction, the latent period was longer in IHH cardiac myocytes. 2. After hydrogen peroxide(300 micromol/L, 10 min) perfusion, LDH and MDA contents in supernatant increased significantly in non-IHH and CIHH cardiomyocytes (P < 0.01), Whereas the contents of MDA and LDH in IHH cardiomyocytes were lower than those in non-IHH cardiomyocytes (P < 0.01). 3. The activities of superoxide dismutase (SOD) and catalase (CAT) were significantly increased in the myocardium of IHH guinea pigs, after hydrogen peroxide (300 micromol/L, 10 min) perfusion, SOD and CAT activities decreased significantly in non-IHH and CIHH cardiomyocytes (P < 0.01), whereas the activities of SOD and CAT in CIHH cardiomyocytes were still higher than those in non-IHH cardiomyocytes.</p><p><b>CONCLUSION</b>IHH had a protective effect on cardiomyocytes injury induced by hydrogen peroxide, which might relate with its antioxidation effects.</p>

<p><b>OBJECTIVE</b>To investigate the effects of the cadmium chloride on the DNA damage and the expression of the gadd153 and gadd45beta promoter and mRNA in HepG2 cells.</p><p><b>METHODS</b>DNA damage induced by cadmium chloride was detected by comet assay. The plasmids (pGADD153-Luc and pG45-Luc) containing DNA damage and repair inducible gene 153 and 45 (gadd153 and gadd45beta) promoter and luciferase and gadd45beta reporter gene were constructed. The activity of gadd153 and gadd45beta promoter were represented by the luciferase activity, the inducible luciferase activities was detected by bioluminescence. The expression of gadd153 and gadd45beta mRNA was detected by RT-PCR.</p><p><b>RESULTS</b>The results of comet assay indicated that Olive Tail Moment induced by the cadmium chloride increased significantly at the dose of 100, 300 micromol/L, compared with the control (P < 0.05). The luciferase activity analysis showed that the expression levels of gadd153 promoter increased significantly in 1, 5, 10 micromol/L treatment group, compared with the control (P < 0.05). The expression levels of gadd45beta promoter in 5, 10 micromol/L treatment group were significantly higher than that in control group (P < 0.05). The expression levels of gadd153 mRNA induced by cadmium chloride at the doses of 1, 5, 10 micromol/L and the expression levels of gadd45beta mRNA induced at the doses of 5, 10 micromol/L were significantly higher than thoae in control group (P < 0.05).</p><p><b>CONCLUSION</b>The cadmium chloride can induce the DNA damage and increase the expression levels of the gadd153 and gadd45beta promoters in HepG2 cells.</p>

According to the method used in our laboratory,our group synthesized (DIPP-Trp)2-Lys-OCH 3. It inhibited the proliferation of K562 and HeLa cells in a dose-and time-dependent manner with an IC 50 of 15.12 and 42.23 microM, respectively. (DIPP-Trp) 2-Lys-OCH3 induced a dose-dependent increase of the G2/M cell population in K562 cells, and S cell population in HeLa cells;the sub-G0 population increased dramatically in both cell lines as seen by PI staining experiments using a FACS Calibur Flow cytometer (BeckmanCoulter,USA). Phosphatidylserine could signi?cantly translocate to the surface of the membrane in (DIPP-Trp)2-Lys-OCH3-treated K562 and HeLa cells.The increase of an early apoptotic population was observed in a dose-dependent manner by both annexin-FITC and PI staining.It was concluded that (DIPP-Trp) 2-Lys-OCH3 not only induced cells to enter into apoptosis,but also affected the progress of the cell cycle.It may have arrested the K562 and HeLa cells in the G 2/M,S phases,respectively.The apoptotic pathway was pulsed at this point,resulting in the treated cells entering into programmed cell death.(DIPP- Trp)-Lys-OCH is a potential anticancer drug that intervenes in the signalling pathway.

<p><b>OBJECTIVE</b>To study the chemical constituents of Eriophyton wallichii.</p><p><b>METHOD</b>Compounds were separated and purified by column chromatographic methods, and their structures were elucidated by spectroscopic methods.</p><p><b>RESULT</b>Eight phenylpropanoids were isolated and identified as martynoside (1), leucosceptoside A (2), citrusin B (3), (+)-dehydrodiconiferyl alcohol-4, 9-beta-D-glucopyranoside (4), liriodendrin (5), velutinoside 11[ (6), jionoside B, (7), stachysoside D (8), respectively.</p><p><b>CONCLUSION</b>The eight compounds were firstly isolated from E. wallichii.</p>

RESUMO

<p><b>OBJECTIVE</b>To investigate the feasibility of endoscope assisted transaxillary subglandular breast augmentation in order to minimize the bleeding and tissue injury.</p><p><b>METHODS</b>Since 2005, 27 patients underwent endoscope assisted transaxillary subglandular breast augmentation. Intraoperative bleeding and dissection injury were markedly decreased.</p><p><b>RESULTS</b>There was no hematoma, seroma and infection except for one case with partial poor incision healing. 16 cases were followed-up for six months to one year. Only one case developed fibrous capsular contracture (Baker II). Good results achieved in all the other cases.</p><p><b>CONCLUSIONS</b>Endoscope assisted breast augmentation can reduce the pain, bleeding and tissue injury. It can also help to put implants to right position.</p>

Objective: To investigate the inhibitory effect of Epibueropyridinium A on aldose reductase. Methods: Aldose reductase was extracted from cattle crystalline lens. Besides Epibueropyridinium A, the reactive system also contained DL-glyceraldehyde, aldose reductase, and NADPH. The activity changes of aldose reductase were detected at 340 nm. Epalrestat was taken as the positive control. The inhibitory type, Ki and IC50 were determined by double reciprocal plot, quadratic drawing, and drawing of inhibitor's concentration to inhibitory ratio, respectively. Results: Epibueropyridinium A significantly inhibited the activity of aldose reductase in a competitive manner, with IC50 being 4.2 μg/ml and Ki being 4.88 μg/ml. Conclusion: Epibueropyridinium A is competitive inhibitor of aldose reductase.

RESUMO

Objective:To investigate the effect of Epibueropyridinium A (EA), extracted from Scrophularia ningpoensis, in prevention of D-galactose-induced cataract in rats. Methods: SD rats were randomly divided into 5 groups (n= 10), namely, the normal control, cataract modelq 20 mg/kg EA, 10 mg/kg EA, and positive control groups. The cataract model was induced by intraabdominal administration of D-galactose into rats. Rats of the 2 EA groups received corresponding amount of EA and those in the positive control group received epalrestat (10 mg/kg) or VitE (30 mg/kg). The changes of the lens were examined with slit lamp microscope at defined time points. The activities of superoxide dismutase(SOD) and the content of malonaldehyde (MDA) and sorbitol were determined in the lens 3 and 6 weeks later. Results: EA obviously improved the lens opacification in the 2 EA groups compared to cataract model group, and the improvement in high EA group was more obvious than that in the low EA group(P<0.01). The activities of SOD raised from (32.1±11.2) Nu · mg-1 in cataract model group to (65.9±16.7) Nu · mg-1 in low EA group and (83.2±15.6) Nu · mg-1 in high EA group(P<0.01). The content of MDA reduced from (26.72±5.36) nmol · mg-1 to (18.34±4.29) nmol · mg-1 in low EA group and (15.6±5.47) nmol· mg-1 in high EA group(P<0.01). The content of sorbitol in lens reduced from (24.33±2.57) μmol · g-1 to (15.46±2.07) μmol · g-1 in the low EA group and (11.45±1.39) μmol · g-1 in the high EA group after 3 weeks(P<0.01); and after 6 weeks, it reduced from (20.04±1.59) μmol · g-1 to (15.82± 1.42) μmol · g-1 and (13.22 ± 1.37) μmol · g-1, respectively (P<0.05 or 0.01). Conclusion: Epibueropyridinium A has preventive effect on development of sugar cataract in experimental models.

Objective:To study the pharmacokinetics,thissue distribution and secretion of nerve growth factor(NGF)in mice.Methods:The conecntration of NGF in various body fluids and tissue were determined by isotope tracer combined SDSPAGE method.Results:The plasma concenmtration-time curve was in accordance with the two-compartment pharmacokinetic model.The elimation half-life(t1/2β)was 3.1.The half-life of distribution(t1/2ka)was 5min.Tpeak was 25 min.AUC was 72.4 mg·kg-1·h-1.The concentrations of NGF were high in thyroid,blood,submaxillary glands,superior cervical ganglion,adrenasl and kidneys.Conclusion:NGF has a wide distribution,high tissue concentrationa nd excrtet mainly through the urine.
