RESUMO

Objective:To study the effects of Terra Flavausta on diarrhea mice with spleen yang deficiency based on metabonomics. Methods:Totally 30 mice were divided into normal group, model group and Terra Flavausta group according to random number table method. Mice in the model group and Terra Flavausta group were treated by the method of "diet disorder + clearing fire with herbs bitter in flavour and cold in property" to establish the diarrhea model of spleen yang deficiency. After successful modeling, Terra Flavausta group received Zaoxintu Decoction 12.0 g/kg for gavage, while normal group and model group were given equal volume of distilled water for gavage, for consecutive 7 d. The serum metabolites of each mouse were analyzed and identified based on UPLC-Q-Exective-MS. The differential metabolites were characterized by principal component analysis and orthogonal partial least squares discriminant analysis, and the potential biomakers were screened, and the KEGG pathway enrichment analysis was performed. Results:Totally 110 different metabolites were screened under the positive and negative ion mode. Terra Flavausta can effectively reverse the disorder of serum metabolism in diarrhea mice with spleen yang deficiency, and has a significant callback effect on 12 potential biomarkers related to diarrhea with spleen yang deficiency. KEGG pathway enrichment mainly involved HIF-1 signaling pathway, ascorbate and aldarate metabolism, platelet activation, etc. Conclusion:Terra Flavausta may play the effect of warming spleen and relieving diarrhea through down-regulation of L-ascorbic acid affecting HIF-1 signal pathway, ascorbic acid and aldose metabolism pathway, vitamin digestion and absorption pathway, up-regulation of prostaglandins G2 and H2 affecting platelet activation pathway, and down-regulation of jasmonic acid α linolenic acid metabolic pathway.

RESUMO

ObjectiveTo investigate the therapeutic effect of the water extract of Zanthoxylum bungeanum aqueous extract(ZBAE)on rheumatoid arthritis. MethodThe sixty SD rats were divided into normal group, model group [complete Freund's adjuvant (CFA), 10 mg·kg-1], methotrexate(MTX) group (0.25 mg·kg-1), low -, medium -, and high-dose ZBAE groups (90, 180, 360 mg·kg-1). The rats in MTX group were given intraperitoneal injection for two weeks, three times a week, and the rats in ZBAE group were administrated for 14 days. The swelling of the ankle joint and body weight were observed, and arthritis scores were also performed. Computed tomography (CT), hematoxylin-eosin (HE) staining and safranine-O and fast green staining were used to observe the effect of ZBAE on synovial hyperplasia and bone protection. Cell counting kit-8(CCK-8)method was used to detect the proliferation of the RA-FLSs cells treated with ZBAE. According to the results of CCK-8 experiment, the optimal concentration and time of administration were determined, blank group, low -, medium -, and high-dose ZBAE groups (0.08,0.10,0.12 g·L-1) were set up. The cell cycle distribution and apoptosis rate were analyzed by flow cytometry,the migration ability of RA-FLSs cells was examined by scratch test. Western blot was used to detect the effect of ZBAE on phosphatidylinositol 3-kinase (PI3K), protein kinase B (Akt), cyclin-dependent kinase 2 (CDK2), Cyclin A and phosphorylated PI3K, Akt (p-PI3K,p-Akt) protein expression in RA-FLSs cells. ResultCompared with the normal group,joint swelling index and arthritis score were increased in the model group (P<0.05),the bone of the ankle was seriously damaged, and there was obvious synovial hyperplasia. Compared with the model group, the ZBAE group could significantly reduce the joint swelling index (P<0.05), inhibit synovial hyperplasia and cartilage destruction. In vitro study showed that compared with the blank group, ZBAE could inhibit the migration of RA-FLSs (P<0.05), promoted cell apoptosis (P<0.05), and acted on RA-FLSs cells in S phase to inhibit cell proliferation. Moreover, the result of Western blot showed that compared with the blank group, the expression of p-PI3K, p-Akt, CDK2 and Cyclin A proteins were significantly decreased in the high dose group of ZBAE (P<0.05). ConclusionThese results suggest that ZBAE has a therapeutic effect on rheumatoid arthritis by inhibiting synovial hyperplasia, promoting synovial apoptosis and inhibiting its migration.

Endangered animal medicine is an important part of traditional Chinese medicine, which is distinctive in the treatment of diseases. At present, the rare and endangered medicinal materials such as tiger bone, rhinoceros horn, pangolin, antelope horn, bear bile are listed as national key protected animals, so their clinical application is limited, the current solution is mainly based on the ideas and methods of similar pharmacological effects, close genetic relationships, artificial breeding, and artificial synthesis to find and develop alternatives for endangered animal medicinal materials. Although artificially cultured bear bile and musk, and artificially synthesized tiger bone, bezoar and musk can solve the shortage of endangered animal medicines to a certain extent, there are still some problems such as difficult breakthroughs in breeding technology and incomplete recognition in the substitute industry. According to this, based on summarizing the existing substitutes for endangered animal medicines, our group proposed the concept of homology, homogeneity and equivalent of substitutes, and constructed a new idea to develop and evaluate substitutes by combining frontier biotechnology with multi-omics detection, so as to provide some support for protecting rare and endangered animals and solving the shortage of endangered animal medicines.

OBJECTIVE:To study the difference of dissolution performance of chemical component and anticonvulsant effect of Bombyx mori decoction and powder taken with water,and to provide reference for the selection of application forms of B. mori. METHODS:The yield of dry extract was determined for decoction and powder biomimetic gastric juice of B. mori. HPLC method was used to detect the content of ammonium oxalate in decoction and powder biomimetic gastric juice of B. mori. The content of protein in decoction and powder biomimetic gastric juice of B. mori was determined with bicinchoninic acid(BCA)method. Mice was divided into normal group(1%Sodium carboxymethylcellulose solution),model group(1%Sodium carboxymethylcellulose solution),positive group (phenytoin,2 mg/kg) and B. mori decoction and powder suspensions high-dose,medium-dose and low-dose groups(0.75,1.5,3 g/kg by crude drug)according to random number table,with 20 mice in each group. After 60 min of intragastric administration,electric stimulation was conducted,and the rate of convulsion in mice was recorded. RESULTS:The yields of dry extract were 22.08% and 26.40% in decoction and powder gastric juice of B. mori (P<0.05);the contents of ammonium oxalate were 11.22% and 16.83% (P<0.05),and the contents of protein was 3.39% and 4.92% (P<0.01). Compared with normal group,convulsion rate of mice was increased significantly in model group (P<0.01);compared with model group,convulsion rates of mice were decreased significantly in administration groups (P<0.05 or P<0.01),and the convulsion rate of mice in B. mori powder suspensions group was lower than decoction group. CONCLUSIONS:The dissolution performance of the chemical component from gastric juice of B. mori powder is better than that of decoction, and the anticonvulsant effect of B. mori powder is better than decoction.

OBJECTIVE:To optimize extraction technology of protein from Cryptotympana pustulata and investigate its in vitro antioxidant activity,so as to provide reference for further research of protein from C. pustulata. METHODS:Using extraction amount of protein as response value,based on single factor test,Box-Behnken response surface methodology was used to optimize the ratio of liquid to material,ultraonic time and extraction times.Validation test was conducted. Using Vitamin C(VC)as positive control, in vitro antioxidant activity of protein from C. pustulata was evaluated by using scavenging rate of 1, 1-diphenyl-2-picrylhydrazyl(DPPH)and 2,2′-azino-bis(3-ethyl benzothiazoline-6-sulfonic acid)(ABTS)free radical as index. RESULTS:The optimal extraction condition of protein from C.pustulata was as follows as the ratio of liquid to material 28:1(mL/g), ultrasonic time of 65 min,extracting for twice. In validation test,average extraction amount of protein from C. pustulata was 65.45 mg/g(RSD=1.68%,n=3),relative error of which to predicted value was 5.48%. The protein from C. pustulata showed strong scavenging effect on ABTS and DPPH free radicals. When the concentration of protein from C. slough was 0.2 mg/mL,and the scavenging rate of it to ABTS free radicals was 97%,the effect of which was similar to VC.The protein from C. pustulata showed weak scavenging ability to DPPH free radical,IC50was 0.96 mg/mL,the effect of which was not as good as VC. CONCLUSIONS:The extraction technology of protein from C. pustulata optimized by Box-Behnken response surface methodology shows high accuracy and good reliability.The protein from C.pustulata shows certain antioxidant activity in vitro.

OBJECTIVE:To provide reference for promoting the inherirance and development of TCM powder decoction piec-es. METHODS:By retrieving literature,the inheritance,overview of modernization development,and key points in industrial de-velopment of TCM powder decoction pieces were analyzed,and development suggestion was put forward. RESULTS & CONCLU-SIONS:Aiming at inheritance of TCM powder decoction pieces,suggestions were put forward as explaining scientific connotation of TCM powder decoction pieces,mining the essence of preparation technology,and cultivating comprehensive decoction pieces talents with basic knowledge of powder science. According to the particle size of powder,TCM powder decoction pieces can be di-vided into TCM common powder,TCM ultrafine particle powder,and TCM nano powder. Aiming at the key points of TCM pow-der decoction pieces,such as application scope,particle size and its correlation with the performance of TCM,grinding technology and equipment and quality standards,the developing strategies were put forward as adjusting the suitable application scope,select-ing appropriate particle size,strengthening its performance research,standardizing and innovating the preparation,improving the packaging and sterilization technologies,establishing and implementing the quality standard with characteristics.

OBJECTIVE:To establish HPLC fingerprint of stir-baked Manis pentadactyla.METHODS:HPLC method was conducted.The determination was performed on Capcell Pak Mg Ⅱ S5 C18 column with mobile phase consisted of acetonitrile-0.1% phosphoric acid solution (gradient elution) at the flow rate of 0.8 mL/min.The detection wavelength was set at 275 nm,and column temperature was 30 ℃.The sample size was 10 μL.Using oxyphenylaminopropionic acid as reference,HPLC chromatograms of 11 batches of medicinal materials were determined.TCM Chromatogram Fingerprint Similarity Evaluation System (2004 A edition) was used for common peak identification and similarity evaluation.RESULTS:There were 23 common peaks in 11 batches of stir-baked M.pentadactyla,with similarity>0.90.After validation,HPLC chromatograms of 11 batches of medicinal materials were in good agreement with control fingerprints.CONCLUSIONS:Established HPLC fingerprint can provide reference for the identification and quality evaluation of stir-baked M.pentadactyla.

Rhizoma Pinelliae Fermentata(RPF)wasoneofthecommonlyusedChinesemateriamedica(CMM). According to the ancient and modern literatures on RPF, the historical evolution, fermentation methods, chemical compositions, efficacy and microbes of RPF were systematically summarized in this paper. Through the analysis on existing problems of fermentation strains, effective components, quality standard and fermentation process, the corresponding solutions were proposed. This work may provide an idea and reference for the further study of RPF.

This study was aimed to establish a rapid discrimination method of Pinellia ternataand its adulterants based on the odour fingerprints analysis.Typhonium flagelliforme and Arisaema Rhizome,which were the common adulterants of Pinellia ternata,were collected.The adulterants were mixed with Pinellia ternatain different proportions.E-nose technology was used to obtain the odour fingerprints of Pinellia ternataand its adulterants of different types and proportions.Chemometrics methods,such as the analysis of variance (ANOVA),principal component analysis (PCA) and discriminant factor analysis (DFA) were used in the analysis and discrimination on sensors response data collected by sensors.The results showed that there were obvious differences on the odour characteristics between Pinellia ternateand its adulterants.PCA can obviously discriminate Pinellia ternateand its adulterants.And the odour difference became obvious along with the increasing of the adulteration proportion.There was a linear relationship between e-nose signal and the proportion of Typhonium flagelliforme.The cumulative proportion in ANOVA of the DFA model was 100%.The correct recognition rate was not less than 97%.It was concluded that e-nose can be used for rapid discrimination of Pinellia ternataand its adulterants.This study provided new technology and method for the discrimination of adulterants of Chinese materia medica.

RESUMO

Presently, there are many issues in traditional Chinese medicine (TCM) decoction, such as the uncertain sources of TCM, the lack of reminder for medication taboo, the nonstardard herb operation, and difficult supervision, etc. A digital service system of TCM decoction was established to solve the problems mentioned above. The digital service system mainly includes automatic coding for checking in & out, drug medication taboo database, digital operation in decoction, distribution through 2D code, the corresponding application for mobile phone, and the information supervision platform for TCM decoction. The digital service system of TCM decoction can track the quality & duty of the pieces, remind decoction medicine contraindications, improve the standard operation process of decoction, develop decoction distribution & tracking through cell phone, save the waiting time, and hence provides a new supervising method for TCM decoction. The digital service system of TCM decoction solves the key issues for the formula, operation, delivery and supervision of TCM. In the same vein, this system will expand the market share of TCM decoction and promote the development of TCM.

Because of the relationship between the quality and the processing technology of Chinese herbal pieces was very close, we proposed concerning the problems of Chinese herbal pieces characters and processing experi-ence. Then, we made use of intelligent sensory technique to objectively express the characters of Chinese Herbal pieces, and explored to set the methods and models about evaluating the quality of Chinese herbal pieces of appear-ance characters and internal quality. To realize the expression of experience terms such as huoli and huohou in the process of Chinese herbal pieces objectively, we adopted on-line infrared temperature measurement and biologi-cal evaluation technologies, and established new technologies and methods of Chinese herbal pieces process quality control in the process.We established a workable monitoring technology, which could not only achieve the controlla-bility and stability of processing quality, also ensured the safety and effectiveness of Chinese herbal pieces.

RESUMO

Odor and taste are characteristic properties of traditional Chinese medicine (TCM). They are closely related to the chemical compositions in TCM. They are also the outer reflection of TCM quality and an important part of the character identification. This article reviewed the current research on odor and taste identification of TCM, which indicated that odor and taste identification had not been paid enough attention to and its research lagged behind modern social development. Based on current analytical techniques and methods, this article proposed a new idea for odor and taste identification of TCM. The idea emphasized the objectification of odor and taste and TCM identification research should be combined with chemical components analysis.

RESUMO

Traditional Chinese medicine placebo has been put more and more attention to. However, there is no ac-cepted quality evaluation method for TCM placebo. How to evaluate TCM placebo objectively and quantitatively is a common problem in the industry of Chinese medicine. New technologies such as the Intelligent Sensory Technique have been used to establish the placebo evaluation methods which are suitable for TCM characteristics. This article provided the basis for establishing scientific, rational and objective evaluation guiding principles for TCM.

<p><b>OBJECTIVE</b>The color difference meter and electronic nose fingerprint analyzer were introduced to discriminate processed betel nut's different huohou and quantize empirical index during the processing of Chinese medicine.</p><p><b>METHOD</b>By collecting the color's and the smell's characteristic parameters of betel nut and its processed products to set up huohou's math discrimination modules and 90% reference value scale.</p><p><b>RESULT</b>We set up a huohou's math discrimination module and 90% reference value range base on L a*b*, which discrimination coincidence was 100% and 90% reference value range respectively was Shengpin: L* (50.07-55.03), a* (4.136-6.906), b* (6.65-8.82); Chaopin: L* (43.874-47.998), a* (3.816-6.732), b* (3.786-6.558); Jiaopin: L* (38.744-40.616), a* (1.11-3.01), b* (-1.434-0.538). At the same time, we set up a huohou's math discrimination module base on smell's characteristic parameters too.</p><p><b>CONCLUSION</b>The betel nut and its processed products' color and smell measurement based on the color difference meter and electronic nose fingerprint analyzer may be feasible, and provides a reference to better study on traditional Chinese medicine theory.</p>

RESUMO

Objective To study the optimum extraction process of Compound Ganmaoling Capsules by orthogonal design. Methods The rate of extraction and the content of chlorogenic acid were chosen as the assessment indexes.The amount of water, boiling duration and boiling times were studied to examine the extraction process by orthogonal design. Results The optimum water extraction process conditions were adding ten times amount of water for the first time and eight times amount of water for the second time, 1.5 h each time. Conclusion The preparation method is reasonable and stable.

METHODS: The problems existing in the General Regulations for the Processing of Crude Drugs in the supplement in China Pharmacopeia were analyzed, aimed at which, some constructive suggestions were presented. RESULTS & CONCLUSION: It's urgent to improve the feasibility of the General Regulations for the Processing of Crude Drugs in the supplement in China Pharmacopeia and standardize the production of Chinese cut crude drugs.

Objective To establish a HPLC method for the determination of gastrodine in Tianma Formula Granule.Methods A HPLC was performed on Shim-pack VP-ODS(4.6mm? 150mm,5? m).The mobile phase consisted of acetonitrile and 0.1 % H3PO4(3∶ 97).Detected wavelength was at 220 nm,the flow r ate was 0.5 mL? min-1,column temperature was at 25 ℃,and the injection volume was 5 ? L.Results The linearity was good in the range of 0.1020~0. 6120 ? g for gastrodine(r=0.9999).The average recovery rate was 100.5 %, and RSD was 1.01 %(n=5).Conclusion The established HPLC method is simple,accurate,and suitable for the determination of gastrodine in Tianma Formula Granule.

Objective: To establish a determination for Ricinus communis L.and Carthamus tinctorius L.could in Anchan Emulsion. Methods: The determinations were carried out by GC. Chromatographic conditions were: using Chromosorb DEGS as a stationary phase column temperature 180 ?C , flame ionization detecter.Rusults:The content limit of Linoleic acid wasn't lower than 20%, The content limit of Ricinoleic acid wasn't lower than 20%.Conclusion: The established methods is simple, feasibl and reproducible. This study provides a method for the quality control of Anchan Emulsion.
