RESUMO

Objective To determine the half-effective target effect-site concentration (EC50) of sufentanil inhibiting responses to insertion of Streamlined Liner of the Pharynx Airway (SLIPA) when combined with propofol given by target-controlled infusion (TCI).Methods Twenty-eight patients,aged 35-55 yr,with body mass index<30 kg/m2,of American Society of Anesthesiologists physical status Ⅰ or Ⅱ,scheduled for elective hysteroscopic surgery,were selected.Sufentanil was administered by TCI at a concentration for 4 min,and then propofol was given by TCI at a target effect-site concentration of 4 μg/ml.When the patients lost consciousness (bispectral index value<60) and the corresponding target effect-site concentrations of two drugs were achieved,SLIPA was inserted.Using the up-and-down sequential trial,the target effect-site concentration of sufentanil was determined according to patient's responses to SLIPA insertion.The initial target effect-site concentration of sufentanil was 0.3 ng/ml,and the ratio between the two successive concentrations was 1.1.The EC50 and 95% confidence interval of sufentanil inhibiting responses to SLIPA insertion were calculated by using Dixon-Mood formula.Results The EC50 of sufentanil inhibiting responses to SLIPA insertion was 0.17 ng/ml,and the 95% confidence interval was 0.15-0.19 ng/ml.Conclusion When combined with propofol (target effect-site concentration 4 μg/ml) given by TCI,the EC50 of sufentanil inhibiting responses to SLIPA insertion is 0.17 ng/ml.

AIM: To investigate the expression level of transcriptional factor Elf-1 in mononuclear cells, CD4~+ and CD8~+ T cells from umbilical cord blood (UCB). METHODS: Real-time PCR with SYBR green I technique was used for detecting the Elf-1 expression in mononuclear cells, sorted CD4~+ and CD8~+ T cells from 12 cases of umbilical cord blood. The relative mRNA expression level of Elf-1 was analyzed by a formula of 2~(-△Ct)×100%. The expression level of β_2-microglobulin gene (β_2M) was used as an endogenous reference. The peripheral blood from 10 cases of healthy adults was served as control. RESULTS: Elf-1 mRNA expressed in all blood samples collected from both UCB and healthy adults. The expression level showed apparent diversity in different individuals. The relative mRNA expression of Elf-1 in both mononuclear cells (18.55%±2.48%) and CD8~+ T cells (3.52%±0.45%) from UCB were significantly higher than those from healthy adults (9.16%±1.92%, 2.02%±0.27%, respectively, P<0.01, P<0.05). CONCLUSION: The results of Elf-1 expression level from umbilical cord blood indicate that the over-expression of Elf-1 gene in mononuclear cells and CD8~+ T cells might be one of the features of T cell immune state in umbilical cord blood.

RESUMO

Objective To investigate the effects of siRNAs targeted protein kinase C ?(PKC?) on the proliferation and apoptosis of A549 cell line.Methods Six PKC? siRNAs were designed and chemical synthesized. Candidate PKC? siRNA was transfected into A549 cells,PKC? mRNA level was determined by semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR). Then,the effects of more effective PKC? siRNAs on A549 were further investigated by using clone formation assay,Hoechst 33258 staining,propidium iodide (PI) staining,Annexin V-FITC and PI dual staining and western blot. Results Six candidate PKC? siRNAs were designed and named as No.1,No.2,No.3,No.4,No.5 and No.6 PKC? siRNA. Compared with controls,PKC? mRNA levels were all significantly downregulated and the cell proliferation was inhibited in A549 cells treated with candidate PKC? siRNAs except No.5 (P

RESUMO

Objective To detect genetic mutation of apolipoprotein B-100(apoB-100)in patients with primary hypercholesterolemia.Methods One special segment of apoB-100 gene from nucleotide 10549 to 10895 was amplified by polymerase chain reaction(PCR).The PCR products were denatured and hybridized with specific aligonucleotide labeled with digoxigenin,and were analyzed by single strand conformation polymorphism(SSCP)to detect the apoB-100 gene mutation 3531CGC→CGT or other mutations.Results Overall 41 members of 11 primary hypercholesterolemia families were detected,but the above genetic mutation was not detected.Conclusion This genetic mutation is unlikely to exist or of significantly low incidence in Chinese population,and might not be the main cause of primary hypercholesterolemia in the 11 primary hypercholesterolemia families.

AIM: To assess the antiradiation effect of oligomeric procyanidins. METHODS: The hemolysis, malondialdehyde(MDA) levels of mouse liver homogenates and the broken degree of DNA single-strain after being irradiated by ultraviolet were examined. RESUMO

AIM: To investigate the effect of tea polyphenols (TP) on alcohol-induced liver injury in mice.METHODS: 40% alcohol(0 5 mL) and TP (5 mg) were administered intragastrically, the liver MDA level and serum alanine transaminase (ALT) activity were determined. The effect of TP on MDA level in alcohol-treated liver in vitro was also examined.RESULTS: Pretreatment with TP significantly inhibited alcohol-induced liver MDA increase in mice in vivo and in vitro , the increase in serum ALT induced by alcohol was also reduced by pretreatment with TP (0 5 mg). TP at a dose of 5 mg, administered 1 h after alcohol treatment, also suppressed increase in liver MDA level stimulated by alcohol. CONCLUSION: These results demonstrated that TP has a protective effect on alcohol-induced liver injury in mice.
