RESUMO

OBJECTIVE@#To evaluate in-vitro antioxidant, anti-inflammatory and antitumor abilities against human breast adenocarcinoma (MCF7) and human prostate cancer (PC3) as well as the suppressor effect of bacterial exopolysaccharide (BAEPS) on Ehrlich ascites carcinoma (EAC).@*METHODS@#In-vitro antioxidants characters of BAEPS were determined using various methods, while anti-inflammatory activity was estimated against cyclooxygenase (COX-1 and COX-2). In-vitro study, anticancer against MCF7 and PC3 were assessed by the mitochondrial dependent reduction of yellow MTT. In in-vivo study against EAC progression, mice were inoculated with EAC cells and then were orally administered BAEPS at 200 mg/kg after 24 h (equals to 0.10 of determined LD)/10 d.@*RESULTS@#BAEPS was acidic exopolysaccharide contained uronic acid (12.3%) and sulfate (22.8%) with constitution of glucose, galactose and glucuronic acid in a molar ratio 1.6:1.0:0.9, respectively, with a molecular mass of 3.76 × 10 g/mol. BAEPS appeared potent antioxidant characters as free radical scavenging, oxygen reactive species scavenging and metal chelation, while its reducing power was low. BAEPS showed selective anti-inflammatory activity against COX-2 than COX-1, COX-2 selective. BAEPS exhibited potent and selective effect to breast cell cancer MCF7, the death percentage was 65.20% with IC = 70 μg/mL and IC = 127.40 μg/mL. BAEPS decreased counted viable EAC cells and induced non-viable cells. BAEPS improved all assessed hematological parameters. These improvements were reflected in the increasing median survival time and significant increment (P < 0.05) in life span.@*CONCLUSIONS@#BAEPS has anti-tumor activity with a good margin of safety. The anti-tumor activity of BAEPS may be due to its content from sulfated groups and uronic acids and they have antioxidant and anti-inflammatory properties.

RESUMO

To evaluate the efficacy of the concomitant administration of antiglaucoma medications namely timolol 0.1% gel in cases with myopic regression after myopic laser in situ keratomileusis [LASIK]. Prospective observational clinical trial. Ninty five eyes of 75 patients were included in this study prospectively. The mean myopic regression was-1.29 +/- 0.83 diopters [range-0.5 to-4.62] after myopic LASIK. The eyes were divided into two groups: 50 eyes administrated timolol 0.1% gel once daily for 12 months [treated group], and 45 eyes were age matched [control group]. We assessed the amounts of myopic regression in terms of changes in the refraction [spherical equivalent and astigmatism], intraocular pressure [IOP], pachymetry and the refractive power of the cornea measurements for all participants. The refractive error and visual acuity were similar between the two groups at baseline. The treated group had an improvement in spherical equivalent significantly from-1.29 +/- 0.83 to-0.94 +/- 1.07 diopters [P=0.012]. Astigmatism was changed from-0.94 +/- 0.53 to-0.86 +/- 0.60 diopters but this change was not statistically significant [P=0.20]. The IOP was decreased significantly from 12.6 +/- 1.9 to 9.0 +/- 1.1 mm Hg [P<0.001]. Central corneal thickness was changed from 425.6 +/- 19.86 to 429 +/- 18.1 micro m but not statistically significant [P=0.56]. The central corneal power decreased significantly from 37.2 +/- 1.8 to 36.4 +/- 1.3 diopters [P<0.05]. Timolol 0.1% gel was effective for reduction and improvement of myopic regression especially the spherical errors after myopic LASIK

RESUMO

The present study was specifically designed to identify the effects of systemic arterial hypertension on the left ventricular and left atrial function analyzed by 2D and M-mode echocardiographic images. The study included two groups: Patient group included 30 hypertensive patients with age ranging from 35-70 years [51.5 +/- 10.4]; 11 males [36.7%] and 19 females [63.3%], were subjected to echocardiographic study including 2-dimensional and M-mode. Control group included 15 healthy volunteers with age ranging 35-70 years [51.2 +/- 9.75]; 7 males [46.7%] and 8 females [53.3%]. Statistically, there was no significant difference as regard the age. All patients in the study were subjected to full history taking with special emphasis on valvular heart diseases, congenital heart diseases, ischemic heart disease, medical therapy; especially antiarrhythmic drugs, thorough clinical examination with special emphasis on pulse rate, rhythm, systolic and diastolic blood pressure excluding patients on congestive heart failure, standard 12-lead ECG and echocardiographic examination including 2D and M-mode imaging. The study showed systemic arterial hypertension lead to impairment of left ventricular diastolic function due to increased after load but also the changes of left ventricular geometry and structure producing left ventricular hypertrophy. Also, the left atrium involved in this process by structural abnormalities of the left atrial chamber as regarding by the echo imaging. Individual with systolic arterial hypertension had; 1-ECG finding of LVH. 2-Echoparameters showed increased LAD, LAV, IVS, PWT, RWT, LVM and diastolic dysfunction. 3-Hypertension patients may have LVH or not which may be due to recent incidence or lower level of blood pressure. Hypertension patients without LVH did not show increase in LAD and LAV. 4-Left ventricular hypertrophy could be divided into concentric or eccentric LVH according to present or absence of increase RWT. However LA size and volume were similar in patients with concentric and eccentric LVH. There were positive correlation between LAD and degree of LVH represented by LVM and RWT also LAD positively correlate with LV internal dimension and DT and negatively correlate with E/A ratio

RESUMO

Portal hypertension is a leading cause of chronic illness in Egypt. It is responsible for a significant proportion of lost days from work and refractory ascites is the most frequent admitting diagnosis in the medicine and hepatology departments. To evaluate the saphenoperitoneal shunt [SPS] in the management of hepatic patients with intractable ascites. Prospective study. Mansoura University Hospital, Department [8], General Surgery. Hepatic patients with intractable ascites. For 15 patients with intractable ascites the great saphenous vein is reversed to form saphenoperitoneal shunt under spinal anaesthesia. Patients morbidity and mortality as reflected by patients outcome, clinical follow up, laboratory follow up [kidney functions - liver functions] Child-Pugh grade and the therapeutic follow up. There was significant relief of symptoms in [86.6%] of patients, shunt stenosis in a single case and partial thrombosis in 2 cases, significant reduction of patients weight, abdominal girth and increased blood pressure, improved liver and kidney functions and upgraded patients scoring with less requirments for diuretics. The saphenoperitoneal anastomosis [SPS] has a wide range safety, cost effective, favorable outcome, less morbidity and mortality. So, it is a definitive treatment but not a palliative one

RESUMO

Carbon tetrachloride [CCl[4]] is a well-known hepatotoxicant that is used to develop a model of liver cirrhosis in rats. The present study was conducted to investigate the effects of chronic treatment with natural or recombinant interferon [IFN], Ismafron or Roferon, respectively, on CCl[4]-induced liver cirrhosis in rats. Seventy male Sprague-Dawley rats were used in the current study; animals were divided into three major groups, Group I, non-treated animals; Group II, normal animals treated with Ismafron, Roferon or liquid paraffin [the vehicle of CCl[4]]; Group III, cirrhotic animals; the cirrhotic rats were divided into 3 subgroups, cirrhotic animals treated with Ismafron, cirrhotic animals treated with Roferon, and untreated cirrhotic animals. Liver cirrhosis was induced in rats after 10 weeks of treatment with CCl[4] at a dose of 0.3 ml/100 g body weight twice a week. Two weeks of treatment with Ismafron [10,000 IU/rat/day] or Roferon [50,000 IU/rat/day] could decrease the serum activities of the enzymes alanine aminotransferase [ALT], alkaline phosphatase [ALP], and gamma-glutamyl transferase [GGT], and suppress the serum level of procollagen III. Furthermore, treatment with Ismafron or Roferon decreased fibrosis and inflammation of cirrhotic livers. It is concluded that treatment with Ismafron or Roferon could normalize the above serum markers of liver cirrhosis and enhance the histological picture of the liver in CCl[4]-treated rats

RESUMO

Many studies have been conducted to further the understanding of factors influencing alcohol consumption. The purpose of the current study was to investigate: [1] The competitive behavior of food-deprived rats housed as fixed pairs and competing for food. [2] The correlation between the animal's specific competition score, within its specific dyad, and its individual chronic alcohol consumption, when given a free access to 10% [w/v] alcohol or water. [3] Differential chronic alcohol consumption of winners and losers of rat dyads. [4] The effect of alcohol withdrawal on the competitive behavior of animals. Forty male rats were used in this study; thirty animals composed 15 fixed dyads, and 10 animals served as a control. Each pair of food-deprived animals was tested in the home cage for food competition. Eleven out of 15 dyads established a stable dominance hierarchy, winners and losers. These animals were then housed individually and had free access to 10% [w/v] alcohol and water for 4 hours a day over a period of 14 days. Daily alcohol consumption was recorded for each subject. It was found that the total amount of alcohol consumed by winners was significantly higher than that of losers [Winners, 39.2 +/- 2.3 ml vs Losers, 31.8 +/- 2.27ml, p
