RESUMO

Polyphyllin Ⅰ(PPⅠ)and polyphyllin Ⅱ(PⅡ)are the main active substances in the Paris polyphylla. However, liver toxicity of these compounds has impeded their clinical application and the potential hepatotoxicity mechanisms remain to be elucidated. In this work,we found that PPⅠ and PⅡ exposure could induce significant hepatotoxicity in human liver cell line L-02 and zebrafish in a dose-dependent manner. The results of the proteomic analysis in L-02 cells and transcriptome in zebrafish indicated that the hepa-totoxicity of PPⅡ and PⅡwas associated with the cholesterol biosynthetic pathway disorders, which were alleviated by the cholesterol biosynthesis inhibitor lovastatin.Additionally,3-hydroxy-3-methy-lglutaryl CoA reductase(HMGCR)and squalene epoxidase(SQLE),the two rate-limiting enzymes in the choles-terol synthesis, selected as the potential targets, were confirmed by the molecular docking, the over-expression,and knockdown of HMGCR or SQLE with siRNA.Finally, the pull-down and surface plasmon resonance technology revealed that PPⅠ could directly bind with SQLE but not with HMGCR.Collectively, these data demonstrated that PPⅠ-induced hepatotoxicity resulted from the direct binding with SQLE protein and impaired the sterol-regulatory element binding protein 2/HMGCR/SQLE/lanosterol synthase pathways, thus disturbing the cholesterol biosynthesis pathway. The findings of this research can contribute to a better understanding of the key role of SQLE as a potential target in drug-induced hepatotoxicity and provide a therapeutic strategy for the prevention of drug toxic effects with similar structures in the future.

RESUMO

Nutrient agar medium was exposed in 0.085-0.092 T static magnetic fields for 12 h. Then we densities the optical densities at lamda = 600 (OD600) of Escherichia coli, Staphylococcus aureus and Bacillus subtilis in different culturing stage. The results were compared with those of control group in the normal geomagnetic field. The OD600 values of experimental groups of these three kinds of aerobes were significantly higher than those of control groups from 3h to 9h. However, after 11 h, there was no remarkable difference regarding the OD600 values between the two groups. The dissolved oxygen content of nutrient agar medium was determined by microtitration. The dissolved oxygen of nutrient agar medium under static magnetic for 12h increased 15% in average and there was significant difference when compared with the control. The results showed that the ferro-magnetic fields increased the dissolved oxygen content of nutrient agar medium significantly. These findings suggest that the effects of static magnetic fields on Escherichia coli, Staphylococcus aureus and Bacillus subtilis are related to the dissolved oxygen.

RESUMO

Object To set up a determination method for aucubin in three medicinal plants of Pedicularis L Methods The medicinal herbs were defatted by Et 2O, then extracted by MeOH and determined by HPLC method. Varian C 18 column, mobile phase: MeOH H 2O (15∶85); detection wavelength: 204 nm, flow rate: 0.5 mL/min. Results The content of aucubin and its recovery in three medicinal plants were as follows: aucubin in P. decora, in P. davidii and P. artselaeri were 0.90%, 1.94%, 0.95%; recovery rates were 102.3%, 98.2%, 97.6% in P. artelaeri. Conclusion Medicinal plants of Pedicularis Linn. has a good exploiting prospect, the research can provide the reference for evaluating of these medicinal herbs

RESUMO

Object To provide the theoretical basis for the artificial planting of Hypericum perforatum L., the effects of light, gibberellin and ethephon on the germination of H. perforatum seeds were studied. Methods The method of direct germination was used. Results The seeds germinated to 79% in the light, while didn't germinate in the dark. Gibberellin and ethephon not only promoted the germination in the light, but also induced the germination in the dark. But ethephon inhibited the growth of seedling radicals severely. Conclusion The seed of H. perforatum is light-dependent seed, its light-dependence is related to gibberellin and ethephon.

RESUMO

Objective:To study the effects of nourishing yin and supplementing kidney of Peikun Capsule. Methods: The Kindney yin vacuity model was made with thyroid hormone. Results: It was found that Peikun Capsules could reduce the oxygen consumption, raise the pain threshold, lower the body temperature and increase water intake of mice with kidney yin vacuity. Conclusion: Peikun Capsule has obvious effects of nourishing yin and supplementing kidney.

RESUMO

Objective: To study the anti oxidative effects of PLF. Methods: The ?OH scavenging effect was measured by salicylic acid method, the content of MDA in liver mitochondria and microsome was measured by TBA assay; hemolysis of RBC and the swelling of mitochondria were detected by spectrophotometric methods. Results: PLF can scavenge ?OH, inhibit production of MDA, swelling of mitochondria and hemolysis of RBC. Conclusion: PLF has anti oxidation effects invitro.

RESUMO

Objective: To study the anti-senile and anti-oxidative activities of Forsythia suspense leaves tea(FSLT) in mice. Methods: 36 ICR mice(7 mo age) were randomly divided into three groups: senior control group,ig distilled water ; FSLT1 group,ig FSLT 0.3 g ?kg-1?d-1 ; and FSLT2 group,ig FSLT 0.9 g?kg-1?d-1 .The other 12 ICR mice(young control group) were given ig distilled water. MDA and MAO-B in brain and mitochondria were measured. SOD,MDA and POD in heart, liver index, thymus index and spleen index were determined. Results: Forsythia suspense leaves tea may reduce the level of MDA and MAO-B, and increase the activities of SOD and POD. Conclusion: Forsythia suspense leaves tea may have anti-senile and anti-oxidative activities.
