RESUMO

Attenuated Salmonella VNP20009 specifically colonizes and proliferates within tumor tissues and inhibits tumor growth. It has been used as drug delivery vehicle or in combination with other therapies (such as chemotherapy), which shows a good application potential in tumor therapy. In this paper, study was conducted to determine the physiological changes of growth curve and formation of bacterial biofilm of VNP20009 under various environmental stresses, such as temperature, pH, and H2O2. The results showed that VNP20009 could grow normally under the conditions of 42 ℃, pH 6.5, and 1 mmol·L-1 H2O2. Furthermore, the weak acid environment was beneficial to the biofilm formation of VNP20009. This study provides a basis for in-depth study of the survival mechanism and application of attenuated Salmonella.

RESUMO

Local focal adhesion kinase (FAK) is a non-receptor intracellular tyrosine kinase that plays an important role in tumor initiation, development, metastasis and invasion, and is considered to be an important target for the development of antineoplastic drugs. It has both kinase-dependent and non-kinase-dependent scaffolding functions. However, traditional small molecular inhibitors can only inhibit its kinase-dependent activity, so it is difficult to target the kinase-independent scaffolding function. Therefore, there is an urgent need for novel strategies to enhance FAK targeting to lay the foundation for determining the druggability and discovery of FAK inhibitors. Proteolysis targeting chimera (PROTAC) is a new drug development strategy that can recruit E3 ligase to specifically ubiquitinylate target proteins for degradation through the proteasome system. The unique mechanism of action of the PROTAC system could be used to target and degrade the FAK protein, thus eliminating the scaffolding function of FAK. In this review, FAK protein, the signaling pathway, and small molecule inhibitors are briefly described, and the latest research progress in targeting the degradation of FAK using PROTAC technology is summarized.

<p><b>OBJECTIVE</b>To grasp the clinical efficacy of acupuncture in the treatment of primary insomnia (PI), and analyze its future research directions.</p><p><b>METHODS</b>A total of 41 prospective randomized controlled trials (RCTs) were conducted in which the clinical efficacy of acupuncture for treatment of PI was compared with sedative and hypnotic drugs in recent six years by searching databases of CNKI, database, PubMed, and BioMed Central (BMC), the aspects of diagnostic criteria, efficacy standards, observation time, control drugs, characteristics of acupoint selection and regularity were used to review and analyse.</p><p><b>RESULTS</b>Acupuncture treatment for PI was mainly based on acupoints in the head, combined with selecting the points according to the different syndrome, showing short-term efficacy and safety advantages, but it was not well-established in many aspects such as diagnostic criteria, efficacy evaluation, observation time, and control drugs.</p><p><b>CONCLUSION</b>Current evidence shows that acupuncture treatment is effective, but it is necessary to add more stringent RCTs, and introduce objective monitoring indicators to strengthen the evidence and enhance the overall level of research.</p>

Injectable bone tissue engineering scaffold material has a certain form and mechanical strength.It can be loaded with seed cells for injection to the defected site of bone tissue in the form of fluid.After that,structural recovery and functional reconstruction are achieved,and the new bone is formed ultimately.This material has the characteristics of small trauma and good plasticity.It can repair the irregular bone defects and well associate with growth factor,which is the ideal method for repairing bone tissue defects up to now.Among the various injectable bone tissue engineering materials,bioceramic materials,polymer materials,etc.have been proved to have a high biological compatibility and good mechanical properties.That makes these materials become the focus of current research in bone tissue engineering materials.In this paper,the development and application of bioceramic materials,polymer materials,bioceramics and polymer composites were reviewed.

Objective To investigate the role of IL-6/STAT3/miR-21 in the treatment of cerebral ischemia reper?fusion injury in mice by pine bark extract. Methods 120 KM mice were randomly divided into normal control group, sham operation group, 14 d and 28 d model groups, 14 d and 28 d treatment groups, each group with 20 mice. The model of cerebral ischemia?reperfusion injury model was made by bilateral common carotid artery occlusion. The mice in the treat?ment group were treated with pine bark extract procyanidins by intragastric administration for 7 days before the execution. The control group and sham operation group were treated with normal saline. The differences in learning and memory abili?ties and histopathological changes in the hippocampus of mice in each group were examined. The levels of interleukin 6, STAT3, p?STAT3 and miR?21 were measured. Results The learning and memory abilities of the mice in the treatment group were higher than that in the model group at the same time points. The treatment group had less irregularity in the ar?rangement of Nissl body, irregular shape of the cells, with relatively lighter cytoplasmic staining, but was better than the model group. The expression level of IL?6 in the 28 d model group was lower than that in the 14 d model group, The IL?6 in the hippocampus of the treatment group was significantly lower than that in the model group at the same time point. Therewere no significant differences in the contents of total STAT3 protein in the hippocampus between the groups (P>0. 05). The expression of p?STAT3 in the treatment group was lower than model group at the same time point. The expression of miR?21 in the hippocampus of 14 d, 28 d treatment groups was lower than that in the model group at the same time point, and decreased with time. Conclusions The procyanidins extracted from pine bark may effectively inhibit IL?6, then the phosphorylation level of STAT3 is significantly decreased and finally decreases the expression of miR?21, so as to alleviate the cerebral ischemia reperfusion injury in mice.

<p><b>OBJECTIVE</b>To analyze the current status of acupuncture consultation in Peking Union Medical College Hospital, which could provide references and suggestions for the development of acupuncture at comprehensive hospitals and the treatment of diseases.</p><p><b>METHODS</b>The source of departments, the number of patients, the number of consultation, treatment frequency, consultation reasons and western medicine diagnosis of 279 inpatients who received acupuncture consultation were analyzed.</p><p><b>RESULTS</b>A total of 26 departments, including department of internal medicine, department of surgery, gynecology department, etc. and emergency department, requested for acupuncture consultation, in which the request from neurology department was the highest. The distribution of diseases was quite wide, mainly gastrointestinal dysfunction, cerebrovascular diseases, urinary retention, hiccup and pain-related symptoms.</p><p><b>CONCLUSIONS</b>The indication of acupuncture is extensive, which could supplement the disadvantages of modern medicine and play a positive role in the adjuvant treatment of comprehensive hospital. It is suggested the communication and cooperation with other departments should be strengthened to promote development of acupuncture and improve the understanding and application of acupuncture for western medicine doctors.</p>

<p><b>OBJECTIVE</b>To observe therapeutic efficacy of osteoarthritis treated by electroacupuncture, and explore its function of promoting cartilage restoration.</p><p><b>METHODS</b>According to random digital table, sixty cases of knee osteoarthritis (60 knees) were randomly divided into an electroacupuncture group and a physiotherapy group, 15 cases (30 knees) in each one. The electroacupuncture was applied at Neixiyan (EX-LE 4), Dubi (ST 35), Heding (EX-LE 2) and Xuehai (SP 10) in the electroacupuncture group, once every other day. The physiotherapy group was treated by medium-frequency therapeutic apparatus every day. For both groups, 4 weeks of treatment were required. The Lysholm knee scoring scale (LKSS) was used to evaluate and compare the knee joints function before and after treatment. At the same time, the GE Signa EXCITE Twin Speed HD 1.5T was used to take MRI examination of knee joints, and measure the T2 values in 10 sub-regions of the cartilage of tibiofemoral joints.</p><p><b>RESULTS</b>Compared before treatment, the LKSS score of both groups were improved with significant differences except item demands for support (P < 0.01, P < 0.05). Between the two groups after treatment, there were significant differences on total score, item instability and swelling (all P < 0.05), the electroacupuncture group was better than the physiotherapy group, but no significant difference on the other items (all P > 0.05). In the electroacupuncture group after treatment, T2 value in anterior lateral tibial sub-region (LTa) was significantly lowered (P < 0.05), but no significant difference in the other nine sub-regions (all P > 0.05). In the physiotherapy group, T2 value in any sub-region was not significantly different before and after treatment (all P > 0.05).</p><p><b>CONCLUSION</b>Electroacupuncture could effectively improve the symptom, sign and knee joint's function of patients with knee osteoarthritis. Compared with physiotherapy, it has more superior effect and considered as a better non-operative treatment for osteoarthritis. Electroacupuncture also has positive influence on T2 value in cartilage, indicating that electroacupuncture may have the function of promoting cartilage restoration.</p>

OBJECTIVE@#To explore the methods for identification of sibling brothers with Y-STR locus mutation by detection of genetic markers on autosome and Y-biallelic.@*METHODS@#Goldeneye 20A and 18NC kit were used to genotyped the 35 STRs on autosome from two men. PowerPlex Y kit and Yfiler kit were used to genotyped the 16 STRs on Y chromosome full sibling index was calculated by ITO method. Twenty Y-biallelic markers were genotyped by fragment length discrepant allele specific PCR or general PCR.@*RESULTS@#Relationship of sibling brothers was found to have mutation of 2 loci on 16 Y-STR and the identical genetype of 20 Y-biallelic markers as well as a cumulative full sibling index of 4.3149 x 10(6) from 35 STRs on autosome.@*CONCLUSION@#In identification of paternal linage of Y-STR mutation, more genetic information can be acquired by detection of Y-biallelic markers including SNP and InDel.

<p><b>OBJECTIVE</b>To explore the influence of acupuncture treatment on proinflammatory cytokines of osteoarthritic (OA) cartilage.</p><p><b>METHODS</b>Forty female SD rats were randomly and averagely divided into four groups: the normal, model, acupuncture and control group. The method of heel tendon resection for unilateral hind limb was used to establish OA animal models-that was to cut off the rat's heel tendon of left hind limb. The acupuncture group and control group were respectively treated by electro-acupuncture and Diclofenac on right hind limb of each animal for 2 weeks. Immunohistochemical staining was used to observe the expression characteristic of IL-1beta and TNF-alpha in cartilage of each group,and the differences were compared among the groups.</p><p><b>RESULTS</b>Both IL-1beta and TNF-alpha expression in model group were significantly up-regulated than those in the normal group (P<0.01). Their expression in model, acupuncture and control group were significantly different (P< 0.01). IL-1beta and TNF-alpha expression were significantly down-regulated in acupuncture and control group than that in model group (P<0.01), while the difference between the two former groups was not significant (P>0.05).</p><p><b>CONCLUSION</b>Acupuncture can down-regulate the expression of IL-1beta and TNF-alpha in osteoarthritic chondrocytes, and the regulating effect is the same as Diclofenac. This shows that acupuncture has a certain protective effect on osteoarthritic cartilage.</p>

<p><b>OBJECTIVE</b>To explore the curative mechanism of acupuncture treatment on osteoarthritis (OA).</p><p><b>METHODS</b>Forty cases of female SD rats were randomly divided into a normal group, a model group, an acupuncture group and a medication group, 10 cases in each group. OA animal model was established by using the method of heel tendon resection for unilateral hind limb. The acupuncture group was treated with electroacupuncture at "Xiqian"(ST 35) and "Housanli"(ST 36), and the medication group with inunction of Diclofenac cream, and the normal group and the model group without any treatment. The expression of matrix metalloproteinase-1, 3 (MMP-1, MMP-3) and tissue inhibitor of metalloproteinase-1 (TIMP-1) in the cartilage were observed by immunohistochemistry.</p><p><b>RESULTS</b>There were significant differences among four groups. The expressions of MMP-1, MMP-3 and TIMP-1 in the model, acupuncture and medication groups were all significantly stronger than those in the normal group (all P < 0.01). The expressions of MMP-1 and MMP-3 in the acupuncture and medication groups were down regulated and TIMP-1 expression up-regulated with significant differences as compared with the model group (all P < 0.01), and the expressions of MMP-1 and MMP-3 in acupuncture group were significantly lower, while TIMP-1 expression significantly higher than that in the medication group (all P < 0.01).</p><p><b>CONCLUSION</b>Acupuncture can down-regulate the expression of MMP-1 and MMP-3 and up-regulate the expression of TIMP1, which is superior to that of Diclofenac cream, showing that acupuncture has a certain protective effect on cartilage from OA.</p>

@#Objective To explore the effect and mechanism of electroacupuncture at Baihui(GV20) and Zusanli(ST36) of patients with depression.Methods90 patients were randomly divided into group A electroacupunctured at Baihui(GV20) and Zusanli(ST36), group B electroacupunctured at Taichong(LR3), Sanyinjiao(SP6), Neiguan(PC6), and Shenmen(H7) and group C treated with Fluoxetine (20 mg/d, 6 weeks). The therapeutic effect was assessed by the Hamilton Depression Rating Scale (HAMD) before and after treatment. Serum cytokine levels (IL-2, IL-4, and IFN-γ) were measured by ELISA assay at admission and after treatment (6 weeks).ResultsCompared to pre-treatment, the mean levels of HAMD ( P<0.01)and IL-4 (P <0.01) decreased significantly in all groups after treatment. Rank distribution of HAMD scores had obvious difference between all groups. The mean levels of IL-2 and IFN-γ had no significant difference in group C after treatment (P>0.05). And the mean levels of IL-2 and IFN-γ increased significantly in groups A and B after treatment(P<0.01, P<0.05).ConclusionElectroacupuncture takes effect earlier than fluoxetine for depression. Electroacupuncture at Baihui(GV20) and Zusanli(ST36) was more effective than the other two methods. The three types of treatments can restore three types of cytokines and improve depressive syndrome.

For the problems existed in the teaching of acupuncture manipulation and moxibustion method in western medicine college, for instance, the lack of credit hours and divorce from theory and practice, in addition to visualization teaching, this paper introduced case study and problem-centered teaching approaches. Besides clinical teaching, this paper emphasized anatomical knowledge which strengthen memory. In this way, the quality of teaching and teaching effects will be improved in the education of acupuncture manipulation and moxibustion method in western medicine college.

<p><b>OBJECTIVE</b>To investigate the immunologic mechanism of acupuncture at Baihui (GV 20) and Zusanli (ST 36) for treatment of depression.</p><p><b>METHODS</b>Eighty-four cases of depression patients were randomly divided into an electroacupuncture observation group (group A), an electroacupuncture control group (group B) and a medication control group (group C), 28 cases in each group. Baihui (GV 20) and Zusanli (ST 36) were used in the group A; Taichong (LR 3), Sanyinjiao (SP 6), Neiguan (PC 6) and Shenmen (HT 7) were used in the group B; and the group C was treated with oral administration of Fluoxetine. The scores of Hamilton Depression Scale (HAMD) were tested and the level of serum interleukin 1 (IL-1 beta), interleukin 6 (IL-6), and tumor necrosis factor alpha (TNF-alpha) were measured by Enzyme Linked Immunosorbent Assay (ELISA) before and after treatment.</p><p><b>RESULTS</b>Their scores of HAMD were obviously decreased after treatment in three groups (all P<0.01). In the group A, 2 cases were cured, 19 cases were markedly effective, 5 cases were effective and 2 cases were ineffective. In the group B, 16 cases were markedly effective, 12 cases were effective. While in the group C, 1 case was cured, 17 cases were markedly effective, 7 cases were effective and 3 cases were ineffective. The grade distribution of clinical effect showed that the effect of group A was better (P<0.05). The levels of serum IL-1 beta and IL-6 in the three groups were obviously decreased after treatment (P<0.05, P<0.01), while there was no significant difference between the level of serum TNF-alpha before and after treatment.</p><p><b>CONCLUSION</b>The effect of electroacupuncture observation group is superior to those of electroacupuncture control group and medication control group. All of these three methods can clear the inflammatory cytokines such as IL-1 beta and IL-6 away and improve the symptoms of depression.</p>

<p><b>OBJECTIVE</b>To study the advantages of Chinese medicine (CM) in treating insulin resistance and disorders of glucose and lipid metabolism in patients with polycystic ovarian syndrome (PCOS), and to explore its underlying mechanisms.</p><p><b>METHODS</b>One hundred PCOS patients were assigned to three groups: 40 patients in the CM group treated by CM, 30 in the WM1 group treated by metformin, and 30 in the WM2 group treated by cyproterone. Before treatment and at 3 cycles and 6 cycles after treatment, changes of body mass index (BMI), fasting serum insulin (FINS) and fasting blood sugar (FBG) levels as well as lipid spectrum were measured and the homeostasis model of insulin resistance (HOMA-IR) was calculated. Meanwhile, the recovery of ovulation was observed.</p><p><b>RESULTS</b>There were 30, 22 and 23 patients in the CM, WM1 and WM2 group respectively completed their 6-month treatments. Levels of FINS, FBG, HOMA-IR, total cholesterol (TC) and low-density lipoprotein cholesterol (LDL-C) were lowered and high-density lipoprotein cholesterol (HDL-C) level increased in the CM group after 6-month treatment, showing significant difference as compared with the baseline (P < 0.05), and the difference in comparing with the WM2 group was statistically significant in terms of MBI, FINS, FBG, HOMA-IR, TC and LDL-C (P < 0.05). The ovulation rate was 53.3% (16/30) in the CM group, 27.3% (6/22) in the WM1 group and 21.7% (5/23) in the WM2 group, comparison between them showed a significant difference between the CM group and the WM2 group (P < 0.01).</p><p><b>CONCLUSION</b>CM is effective for the treatment of PCOS in improving insulin resistance, adjusting blood sugar and lipids levels and recovering ovulation.</p>

<p><b>OBJECTIVE</b>To understand the related knowledge, discrimination attitudes toward HIV/AIDS among medical college students, and to provide scientific evidence for further HIV/AIDS anti-discrimination intervention.</p><p><b>METHODS</b>By means of stratified cluster sampling to classes, 2844 undergraduate students were randomly selected from medical colleges. A self-designed and self-administered questionnaire survey was conducted, and SPSS 13.0 software was used for data analysis.</p><p><b>RESULTS</b>2501 valid questionnaires had been collected. The overall HIV/AIDS knowledge coverage rate of the respondents was 73.1% (1828/2501); The HIV/AIDS discrimination rates in different questions were varying, the discrimination rate of infected with AIDS by bad sex and sharing needles was 83.1% (2078/2501) and 77.7% (1943/2501) respectively, the discrimination rates in term of contacting with HIV patients and their daily necessities, sharing desks, personal social were all exceeding 40%.</p><p><b>CONCLUSION</b>The medical students held serious discrimination attitudes to HIV infected persons and patients; it is necessary to strengthen anti-discrimination education about HIV/AIDS among medical students.</p>

@#Objective To investigate the effect of electro-acupuncture on cartilage renovation of knee osteoarthritis.Methods 40 female SD rats were randomly divided into the normal group,model group,electro-acupuncture treatment group and control group.The left knees of the treatment group and right ones of the control group were treated by electro-acupuncture for two weeks from the fifth week since operation.Collected left knees' cartilage of all rats at the seventh week since the beginning of experiment.HE and immunohistochemical staining were used to observe the expression characteristic of transforming growth factor-β1(TGF-β1)in cartilage.Results The expression of TGF-β1 in the treatment group was very significantly stronger than all other groups(P<0.01).Conclusion Electro-acupuncture can obviously up-regulate the level of TGF-β1 in cartilage of experimental rats with knee osteoarthritis,and promote the repair of cartilage.

<p><b>OBJECTIVE</b>To observe the influence of acupuncture on sebum thickness and the characteristic in the female obese patient of different types.</p><p><b>METHODS</b>Ninety-eight female obese patients were divided into 2 groups: an abdominal obesity group (n = 40) and an obesity group (n = 58). All of the patients in the two groups were treated by acupuncture at Zhongwan (CV 12), Chengman (ST 20), Tianshu (ST 25), Shuidao (ST 28), Qihai (CV 6), Zusanli (ST 36), Sanyinjiao (SP 6), etc. with electric stimulation, and in combination with ear point taping and pressing, for one month. Body weight index (BMI), waist circumference (WC), the sebum thickness of the upper limb (point A: at the inferior border of the right deltoid muscle and the midpoint of the lateral of arm), the trunk (point B: the inferior angle of right scapula) and the abdomen (point C: 3 cm right lateral to the navel; point D: the right anterior superior iliac) were observed and compared before and after treatment in the two groups.</p><p><b>RESULTS</b>There were significant differences before and after treatment in BMI, WC and the sebum thickness of points A, B, C and D in the two groups (P < 0.01), and the sebum thickness in the point C significantly changed as compared with those in other 3 points (P < 0.05) in the two groups. In the abdominal obesity group, change of BMI was very significantly positively correlated with changes of WC and sebum thickness in the point C, and significantly positively correlated to point A (P < 0.05), with the correlated coefficient [r(WC/BMI)] > [ r(C/BMI)] > [r(A/BMI); change of WC was very significantly positively correlated to point C (P < 0.01). In the obesity group, the change of BMI was very significantly positively correlated to WC (P < 0.01) and significantly positively correlated to the sebum thickness's change of point B (P < 0.05), while WC change was very significantly positively correlated to point B and C (P < 0.01) and significantly positively to point D (P < 0.05).</p><p><b>CONCLUSION</b>Acupuncture can effectively reduce the body weight, WC and subcutaneous fat in various parts of the body and selectively reduce the over fat of different places in both the obesity patient and the abdominal fat patient with exceeding standard of WC.</p>

RESUMO

<p><b>OBJECTIVE</b>To compare therapeutic effects between body acupuncture and scalp acupuncture combined with body acupuncture on atherosclerotic cerebral infarction at acute stage.</p><p><b>METHODS</b>Forty-two cases of atherosclerotic cerebral infarction at acute stage were divided into 2 groups. They were treated respectively by scalp acupuncture combined with body acupuncture and simple body acupuncture. The comprehensive scores of mind, speech, limb motor function, etc. of the patients before and after treatment were recorded and compared.</p><p><b>RESULTS</b>After treatment, the cumulative scores in the two groups reduced significantly (P < 0.01); the difference of scores before and after treatment in the scalp-body acupuncture group was significantly higher than that in the body acupuncture group (P < 0.05); there was no significant difference between the two groups in clinically effective rate (P > 0.05), but there was a very significantly difference between the two groups in the markedly effective rate (P < 0.01), the scalp-body acupuncture group being significantly higher than that in the body acupuncture group.</p><p><b>CONCLUSION</b>Acupuncture can improve mind, speech and limb motor function of the patient with atherosclerotic cerebral infarction at acute stage, and the therapeutic effect of scalp combined with body acupuncture is better than that of simple body acupuncture.</p>

RESUMO

<p><b>OBJECTIVE</b>To study the therapeutic effects of electroacupuncture for the treatment of osteoarthritis of knee joint.</p><p><b>METHODS</b>Forty patients with knee osteoarthritis were randomly divided into two groups: the electroacupuncture group (20 patients)and the control group (20 patients). The patients in electroacupuncture group (8 male and 12 female, with an average age of 62.50 +/- 9.23 years and mean course of disease 9.85 +/- 8.17 years)were treated with electroacupuncture for one month. At the same time, the patients in control group (7 male and 13 female, with an average age of 61.15 +/- 6.75 years and mean course of disease 8.60 +/- 6.89 years) were given Diclofenac sodium orally for one month. The motor function of knee joints were evaluated before and after treatment according to Lysholm knee scoring scale.</p><p><b>RESULTS</b>Most scores in the electroacupuncture group improved significantly compared with those of before treatment (P < 0.01 or P < 0.05) except for the item demands for support. While in the control group, compared with those of before treatment, there were significant difference on the item of pain, inter locking and instability (P < 0.01 or P < 0.05), but there were no significant differences on other five items (P > 0.05). Between the two groups after treatment, differences on item of inter locking, instability, swelling, stair activity and squat were significant (P < 0.05), but no differences on limp, demands for support and pain (P > 0.05).</p><p><b>CONCLUSION</b>Acupuncture can effectively improve the clinical symptoms and knee joint's motor function of patients with knee osteoarthritis, and more superior than Diclofenac sodium oral. It is a proved effective method for osteoarthritis.</p>

@#Objective To observe effects of acupuncture on different traditional Chinese medicine(TCM) syndrome types in patients with apoplectic hemiplegia.Methods 57 cases of apoplectic hemiplegia were divided into the Qi deficiency with blood stagnation group(35 cases) and blockage of the meridians and collaterals by phlegm and blood group(22 cases).All patients were treated by scalp acupuncture combined with body acupuncture.The clinical therapeutic effect was evaluated according to the scores of manner,speech,motor function of limbs,and so on.Results In the Qi deficiency and blood stagnation group,2 cases were basically cured,17 were remarkably effective,12 were effective,with a total effective rate of 88.6%;while,in the blockage of the meridians and collaterals by phlegm and blood group,3 cases were basically cured,14 were remarkably effective,3 were effective,with a total effective rate of 90.9%.There was a significant difference between the two groups in clinical remarkably effective rate(P<0.01),but no difference in total effective rate( P>0.05).Conclusion Acupuncture can obviously improve the consciousness,language and limb function of patients with apoplectic hemiplegia.The effective rate of type of blockage of the meridians and collaterals by phlegm and blood is superior to type of Qi deficiency with blood stagnation.
