RESUMO

Aim To observe the effect of epidurally application of osthole on the model of nucleus pulposusinduced inflammatory radicular pain and the expression of p38 MAPK signaling related pathway in the spinal dorsal horn of rats.Methods The model of radicular pain was generated by putting nucleus pulposus to the L5 dorsal root ganglion (DRG).50% MWT was measured using Von Frey filaments to calculate mechanical pain threshold before and after operation.50 μL of 20 g · L-1 osthole was administered epidurally in group Ost and 50 μL of 100 mL · L-1 DMSO in group DMSO at postoperative day (POD).The expression of phosphorylated p38 (p-p38),IL-18 and IL-18R in the lumbar spinal dorsal horn was detected by Western blot.IL-18 mRNA was assessed by real-time PCR.Results The mechanical pain threshold significantly decreased after operation (P < 0.05),while the expression of protein p-p38 MAPK,IL-18,IL-18R and IL-18 mRNA was significantly different.Compared with DMSO group,50% MWT was significantly increased and accompanied with the decrease of protein p-p38,IL-18,IL-lgR and IL-18 mRNA in Ost group after drug administration (P < 0.05).The correlation analysis between protein concentration of p38 MAPK and IL-18 mRNA showed that the Spearman correlation coefficient was 0.9 (P < 0.05).Conclusion p-p38 and IL-18 of spinal dorsal horn participate in the rat model with inflammatory radicular pain induced by nucleus pulposus,and IL-18R plays a role in maintenance of the pain.Osthole administered epidurally in the early stage of pain could alleviate the pain for a long time,which may be related with inhibiting p38 MAPK signaling related pathways.

RESUMO

Cerebrovascular diseases (CVD) with high morbidity, disability, mortality, and recurrence rate have become the focus in the medical research. Modern researches have indicated that Guizhi Fuling Prescription (GFP) possesses anti-inflammatory, immune-modulation and anti-oxidative effects and so on. Investigations showed that GFP had been used in the researches of ischemic and hemorrhagic cerebrovascular diseases, mainly used in the clinical research of ischemic cerebrovascular, in which the research of treating acute cerebral infarction gets the best clinical effect of all. In order to provide theoretical reference for intensive research of GFP in treatment of CVD, this article reviewed the research progress in clinical application and mechanisms of GFP in treatment of CVD.

Objective To study the inhibitory effect of Gallnut on the biofilm of Candida albicans. Methods MTT assay was used to determine the inhibitory effect of Gallnut on biofilm formation and mature biofilm. RT-PCR was used to determine the effect of Gallnut on the expressions of ALS1 and CPH1 genes. Results Gallnut has a stronger inhibitory effect on Candida albicans biofilm formation, the minimum inhibitory concentration (MIC) and the minimum fungicidal concentration (MFC) for inhibition of biofilm formation both are 128mg/mL. Gallnut also has a strong inhibitory effect on the mature biofilm of Candida albicans, the minimum inhibitory concentration (MIC) is 256mg/mL. Gallnut could significantly reduce the gene expressions of ALS1 and CPH1. Compared with the control group, 64 mg/mL Gallnut act on Candida albicans after 16h, the gene expression of ALS1 and CPH1 decreased by 79.2% and 82.0% respectively. Conclusion Gallnut has a significant inhibitory effect on the formation and maturation of Candida albicans biofilm. The inhibitory effect of Gallnut on the adherence and invasion of Candida albicans is mainly through inhibiting the expressions of ALS1 and CPH1 genes.

<p><b>OBJECTIVE</b>This study was to construct the lentivirus vector carrying hepatocyte growth factor (HGF) gene and to explore the condition for transfecting the adipocyte-derived mesenchymal stem cells (ADSC) by HGF lentivirus.</p><p><b>METHODS</b>The target gene was obtained from plasmid carrying HGF gene by PCR and was cloned into GV287 vector. The recombinant GV287-HGF vector plasmid and lentivirus-packing plasmid were co-transfected into 293 T cells to generate HGF lentivirus, and the virus titer was assayed, then the ADSC were transfected by using recombinant HGF lentivirus, and the optimal multplicity of infection (MOI) was detected.</p><p><b>RESULTS</b>The PCR product of HGF gene was consistent with expectant sizes, suggesting that the electrophoretic result of recombinant GV287-HGF plasmid PCR product was correct. The sequencing analysis of cleaved product showed consistance of obtained results with the sequences of target gene, suggesting correct construction of recombinant lentivirus carrying HGF gene. The ELISA showed that the virus tilter was 5×10(8) TU/ml. The optimal MOI for transfecting ADSC with recombinant lentivirus carrying HGF gene was 50.</p><p><b>CONCLUSION</b>The lentivirus vector expressing human HGF gene has been constructed, and transfected the ADSC succesfully. This study lays a foundation for further stadying the ADSC over-expressioning HGF, treating the radiation damage of bone marrow and impartant internal organs.</p>

This study was aimed to establish an experimental mouse model of combined transgenic inhibition of both multifunctional Ca(2+)/calmodulin-dependent protein kinase II (CaMKII) and inward rectifier potassium current (Ik1), and to observe whether the specific inhibition of both CaMKII and Ik1 can bring about any effects on cardiac remodeling. Mice were divided into 4 groups: wild type (WT), CaMKII inhibited (AC3-I), Ik1 inhibited (Kir2.1-AAA) and combined inhibition of both CaMKII and Ik1 (AC3-I+Kir2.1-AAA). Mice in each group received electrocardiogram (ECG) and echocardiography examination. ECG in the condition of isoproterenol (ISO) injection was also checked. The whole cell patch clamp technique was used to measure Ik1 and the transient outward potassium current (Ito) from enzymatically isolated myocytes of left ventricle. In the condition of basal status, no significant changes of heart rate, PR interval and QRS interval were observed. No mouse showed ventricular arrhythmias in all of the 4 groups. After ISO injection, each group presented no significant ventricular arrhythmias either. The indexes measured by M-mode (motion-mode) and two-dimensional echocardiography had no significant differences among the four groups. Ik1 in AC3-I group was significantly higher than those in other three groups (P < 0.01) because of the results brought about by CaMKII inhibition. Among the latter three groups, both Kir2.1-AAA group and AC3-I+Kir2.1-AAA group had a significant reduced Ik1 compared with that of WT group, which was due to the Ik1 inhibition (P < 0.01). Ito in AC3-I group was higher than that of the other three groups (P < 0.01), but there were no significant differences in Ito among WT, Kir2.1-AAA and AC3-I+Kir2.1-AAA groups. Thus, combined transgenic myocardial CaMKII and Ik1 inhibition eliminated the up-regulation of Ik1 in CaMKII inhibited mice, and had no effects on cardiac remodeling including heart structure and function as well as arrhythmias at the basic and ISO conditions. The results of this study may provide a basis for the further investigation of combined inhibition of CaMKII and Ik1 in pathogenic cardiac remodeling.

<p><b>OBJECTIVE</b>To investigate the pathogenic infection and its drug resistance in expressed prostatic secretion (EPS) and its correlation with serum PSA, and provide some evidence for the systematic and normalized diagnosis and treatment of prostatitis.</p><p><b>METHODS</b>Three EPS swabs were collected from each of the 320 prostatis patients following measurement of the serum PSA level, 1 for bacterial culture and identification, 1 for detection of Mycoplasma and drug sensitivity, and the other for examination of Chlamydia trachomatis antigen by colloidal gold immunoblot.</p><p><b>RESULTS</b>Totally 244 strains were isolated from the 320 EPS samples, including 188 bacterial strains (dominated by Staphylococcus and sensitive to vancomycin or linezolid) and 44 Mycoplasma and Chlamydia strains (mainly Ureaplasma urealyticum and susceptible to josamycin or doxycycline). The serum PSA level was significantly higher in the pathogen-positive than in the pathogen-negative group ([6.98 +/- 0.56] microg/L vs [2.32 +/- 0.12] microg/L, P < 0.05).</p><p><b>CONCLUSION</b>Prostatitis may lead to the elevation of the serum PSA level and the pathogens involved vary in their resistance to different antibacterial spectrums. Therefore, appropriate and individualized antibiotic therapy should be selected according to etiological diagnosis and the results of drug sensitivity test.</p>

<p><b>OBJECTIVE</b>To develop the quality control standard of Zhishidaozhi Tabloid Pills.</p><p><b>METHOD</b>Applying TLC to identify Radix et Rhizoma Rhei, Fructus Aurantii Immaturus, Rhizoma Coptidis, Radix Scutellariae, and HPLC to determine the content of emodin of Radix et Rhizoma Rhei.</p><p><b>RESULT</b>Radix et Rhizoma Rhei, Fructus Aurantii Immaturus, Rhizoma Coptidis and Radix Scutellariae could be indentified by TLC. Emodin showed a good linear relationship at a rang of 0.0612-0.612 microgram, r = 0.9999. The average recovery was 97.9%, and RSD was 2.1%.</p><p><b>CONCLUSION</b>The methods are accurate and quick, and can be used for the quality control of Zhishidaozhi Tabloid Pills.</p>
