RESUMO

Serum toxic metals have been implicated in development of many diseases. This study investigated the association between blood levels of lead and cadmium with abnormal bone mineral density (BMD) and incidence of osteoporosis. Sixty Saudi male adults age matching were assigned into two groups: A healthy control group (n = 30) and osteoporosis patients diagnosed according to T-score (n = 30). Serum calcium, vitamin D, osteocalcin, lead, cadmium were measured. Osteoporotic group showed a highly significant elevation of blood lead and cadmium levels compared to the control group (p <0.001). BMD was negatively correlated with serum osteocalcin level compared with control. There was a significant negative correlation between the cadmium and lead levels (r=-0.465 and p-value = 0.01) and calcium (p < 0.004). Our findings suggested that high cadmium and lead were negative correlated to BMD and increased the risk factor for osteoporosis.

Aim: To enhance the productivity of lovastatin from Fusarium nectrioides isolate with liquid cheese whey as a major carbon source and to optimize the media components using Response Surface Methodology (RSM) and Fuzzy Logic System (FLS). Methodology: Euphorbia hirta was collected, surface sterilized and incubated on potato dextrose agar medium amended with ampicillin and streptomycin sulphate. F. nectrioides was isolated from E. hirta and identified using morphological and molecular methods. Primarily, media components were screened by Plackett Burman design (PBD). Further, the effect of significant nutrients was predicted using RSM and FLS and compared with experimental yield. Results: Molecular identification by gene sequencing confirmed the isolate to be F. nectrioides, given an accession number (MH173849) the sequence was submitted in the gene bank. PBD revealed that peptonized milk (which is an enzymic digest of high grade skimmed milk powder), corn steep liquor, liquid cheese whey and histidine were significant variables. The optimum levels of these significant variables in different combinations were studied by RSM in which the predicted yield of lovastatin was 1.2 gl-1. Further, it was analyzed by FLS with 14 set of fuzzy rules and the maximum production obtained was 1.8 g100 ml-1 which was closer to the experimental yield of 1.75 g100 ml-1. Therefore, compared to RSM, FLS was more suitable technique to determine the optimum levels of significant nutrients for enhanced lovastatin production. Interpretation: This study suggests that F. nectrioides (MH173849) can be used as a potent producer of lovastatin and the production highly influenced by glucose, corn steep liquor, liquid cheese whey and histidine.

Background: However, lindernia species were considered as weeds, they occupy amost important place in the traditional system of medicine used worldwide especially in China.Linderniaceae is a family having high traditional medical summary with less pharmacognostical,phytochemical and biological profile. Objective: This study was designed to identify andcharacterize the phytochemical profile of different crude extracts of newly identified wholeplant Lindernia madayiparense, using gas chromatography–mass spectrometry (GC- MS).Method: Powdered plant material was extracted and evaluated for various in-vitropharmacological activities. Furthermore the potent extracts were analyzed by GC- MS. Results:The obtained Total ion chromatogram of potent extracts revealed the different types of havingsmall and moderate phytochemicals under the classification of alkaloids, glycosides, terpenoidsand phenols in major and minor amount. Conclusions: The study gives a detailed insight aboutthe phytochemical profiles of three crude extracts of Lindernia madayiparense. So thechemical entities found are may be responsible for the pharmacological activities that probablywill act as lead molecules for furthermore drug development process.

Chromium (Cr) contamination in soil is a growing concern in sustainable agriculture production and food safety. We performed pot experiment with chromium (30 mg/ soil) to assess the accumulation potential of Zea mays and study the influence of four fertilizers, viz. Farm Yard Manure (FYM), NPK, Panchakavya (PK) and Vermicompost (VC) with respect to Cr accumulation. The oxidative stress and pigment (chlorophyll) levels were also examined. The results showed increased accumulation of chromium in both shoots and roots of Zea mays under FYM and NPK supply, and reduced with PK and VC. While the protein and pigment contents decreased in Cr treated plants, the fertilizers substantiated the loss to overcome the stress. Similarly, accumulation of Cr increased the levels of antioxidant enzymes such as catalase (CAT), superoxide dismutase (SOD), and peroxidase (POD) indicating the enhanced damage control activity. However, these levels were relatively low in plants supplemented with fertilizers. Our results confirm that the maize can play an effective role in bioremediation of soils polluted with chromium, particularly in supplementation with fertilizers such as farm yard manure and NPK.

<p><b>BACKGROUND</b>Ligament reconstruction tendon interposition (LRTI) is the most commonly performed surgical procedure for first carpometacarpal joint osteoarthritis. The purpose of this study was to examine the radiographic and clinical outcomes of LRTI arthroplasty and document the clinical results based on metacarpal subsidence.</p><p><b>METHODS</b>From January 2008 to January 2011, 19 patients (21 thumbs) underwent surgery for thumb carpometacarpal arthritis using ligament reconstruction tendon interposition arthroplasty with flexor carpi radialis (FCR) in Kleinert Kutz Hand Care Center of Louisville University, USA. The follow-up period was an average of 13.9 months. Pain, grip strength, tip pinch strength, range of motion, and radiographic measurements were recorded. Based on first metacarpal subsidence, the cases were classified in to mild, moderate, and severe. Clinical outcomes of the groups were evaluated and compared.</p><p><b>RESULTS</b>Grip strength improved from 18.6 kg to 20.5 kg, and tip pinch strength increased from 4.4 kg to 4.5 kg after the surgery. Radial abduction and palmar abduction improved after surgery. Radial abduction increased from 55.7° to 60.6° and palmar abduction improved from 56.7° to 63.5° after the procedure. Visual analogue scores (VAS) were significantly reduced, from 6.6 to 0.5. Compared with the preoperative radiographs the first metacarpal had subsided about 54.6% of the arthroplasty space. The height of arthroplasty space and index of the arthroplasty space significantly decreased from 12.4 mm to 5.6 mm and from 0.27 to 0.12 respectively. Between the various groups (mild, moderate and severe metacarpal subsidence), there was no difference in grip strength, tip pinch strength, thumb range of motion, and VAS.</p><p><b>CONCLUSIONS</b>Ligament reconstruction tendon interposition arthroplasty resulted in excellent relief of pain and increase in range of motion. However, LRTI cannot maintain the arthroplasty space. Compared with the preoperative radiographs, the metacarpal subsided more than 50%. The amount of first metacarpal subsidence has no bearing on the results.</p>

To study the immunomodulatory activity of saline extracts of leaves of Aloe vera Linn. (Family: Liliaceae) on the albino mice. The saline extract of leaves of Aloe vera was administered orally according to their body weight in mice. The assessment of immunomodulatory activity on specific and nonspecific immunity was studied by administration of test extract. The method of pyrogallol induced immunosupression was employed with slight modification to study the immunomodulatory potential of the extract. Humoral antibody response to SRBC measurement of antibody titer by haemagglutination reaction was done and cellular immune response (Foot pad reaction test) the edema was induced in the right paw of mice by injecting SRBC (0.025x109 cells) in the sub planar region. Pyrogallol-induced suppression of humoral as well as cell mediated immune response was significantly attenuated by daily oral treatment with saline extract of Aloe vera. Vitamin E treated group exhibited similar attenuation of the suppression in immune responses. Aloe vera extract at the dose of 100 mg/kg was found to suppress delayed type hypersensitivity reaction induced by SRBCs in mice. As evidenced by marked increase in haemagglutination titers in mice was also observed. The study demonstrates that A. vera triggers both specific and non-specific responses to a greater extent. The study comprised the acute toxicity and preliminary phytochemical screening of A. vera. From the results obtained and phytochemical studies the immunostimulant effect of Aloe vera could be attributed to the alkaloids content.

Background. Deliberate self-harm (DSH) ranges from behaviours aiming to communicate distress or relieve tension; but where suicide is not intended, to suicide. Not all individuals are prone to DSH, which suggests that there are factors that protect against it. Identifying these could play an important role in the management and prevention of DSH. Objectives. This study examined whether religious beliefs, coping skills and responsibility to family serve as factors protecting against DSH in Kota Kinabalu, Sabah, Malaysia. Method. A cross-sectional comparative study assessed DSH patients consecutively admitted or directly referred to Queen Elizabeth General Hospital and Hospital Mesra Bukit Padang during the period December 2006 - April 2007. DSH patients (N=42) were matched with controls (N=42) for gender, age, religion, race, occupation and marital status. The DSH and control groups were compared using psychosocial tests that assess coping skills, religious beliefs and responsibility to family. Results. There were significant differences in religious beliefs (p=0.01) and responsibility to family (p=0.03) between the DSH patients and the control group. There were also significant differences in coping skills, DSH patients tending to use emotion-orientated coping (p=0.01) as opposed to taskand avoidance-orientated coping. caution is required in generalising the results owing to limitations of the study. Further extensive research on religious and psychotherapeutic interventions and prospective studies on protective factors will be helpful

RESUMO

Late Dr RES Muthiah started the very first eye bank in India and the first corneal transplantation took place successfully by him in India in 1948. From then on a movement started for donation of eyes. The prime concept of Eye Bank Association of India (EBAI) is to motivate the people for eye donation. A vast propaganda throughout the country is going on for eye donation. The community should come forward shedding all inhibitions. EBAI has envisaged a master plan of action to regulate eye bank activities. Under this plan eye donation movement is catching up in the country.

Carbonic anhydrase I (CAI) is one out of ten CA isoenzymes that have been identified in humans. X-ray crystallographic and inhibitor complex studies of human carbonic anhydrase I (HCAI) and related studies in other CA isoenzymes identified several residues, in particular Thr199, GlulO6, Tyr7, Glull7, HislO7, with likely involvement in the catalytic activity of HCAI. To further study the role of these residues, we undertook, site-directed mutagenesis of HCAI. Using a polymerase chain reaction based strategy and altered oligonucleotide primers, we modified a cloned wild type hCAI gene so as to produce mutant genes encoding proteins with single amino acid substitutions. Thrl99Val, Thrl99Cys, Thr199Ser, GlulO6Ile, GlulO6Gln, Tyr7Trp, Glu.117Gln, and His I 07Val mutations were thus generated and the activity of each measured by ester hydrolysis. Overproduction of the Glu 117Gln and His I 07Val mutant proteins in Escherichia coli resulted in a large proportion of the enzyme forming aggregates probably due to folding defect. The mutations Thr199Val, GlulO6Ile and GlulO6Gln gave soluble protein with drastically reduced enzyme activity, while the Tyr7Trp mutation had only marginal effect on the activity, thus s.uggesting important roles for Thr199 and GlulO6 but not for Tyr7 in the catalytic function of HCAI.

Immunoaffinity column using Setaria digitata antigens coupled to cyanogen bromide activated Sepharose 4B beads were developed to purify antibodies from sera of filarial patients. Chaotropic (KSCN) ion elution was more efficient for purifying specific antibodies from the column in comparison to ]c elution. Dot blot analysis indicated that purified antibodies showed a high degree of reactivity with cattle filarial antigen and recombinant filarial protein but not with bacterial proteins of E. coli suggesting that the antibodies are specific.

Ubiquitin has been purified to homogeneity, through a dialysis membrane having a NMW cutoff of 12 kDa, by taking advantage of its non-dialysable nature under these conditions. The dialysate was continuously recycled through a CM-52 cation exchange column at pH 4.5. The adsorbed fraction was eluted selectively at pH 7.2. Ubiquitin (25 mg) was obtained from 500 ml of packed RBCs. On SDS PAGE, ubiquitin showed varying mobility depending on the time of boiling in SDS. With 2 min of boiling, the molecular weight seemed to be 10.5 kDa, whereas 10 min of boiling resulted in a molecular weight of 8.5 kDa. Ubiquitin showed a slow intrinsic proteolytic activity against SDS-denatured beta-galactosidase in the absence of ATP. For the first 4 hr, there was no detectable degradation, but degradation was nearly complete after 8 hr. These data are not in agreement with those of Freid et al. [Proc. Natl Acad. Sci, USA, 84 (1987), 3685] who have reported a proteolytic activity comparable to that of other proteolytic enzymes.

Adenosine deaminase (ADA) activity was studied in serum and peripheral blood lymphocytes of leprosy patients and healthy controls. Serum ADA levels were found to be elevated in tuberculoid as well as lepromatous cases compared to control subjects. Serum ADA activity was significantly higher in tuberculoid cases than in the lepromatous group. Lymphocyte adenosine deaminase activity showed a similar trend. These results suggest that, since the overall activity of the enzyme is not deficient in leprosy, the cellular immune abberation seen in the different types of leprosy may be due to abnormal proliferation of different subsets of lymphocytes in response to M. leprae.

Arginase activity was estimated in serum and lymphocytes of 22 healthy controls and 50 untreated leprosy patients across the spectrum. The patients included 21 lepromatous/borderline lepromatous (LL/BL); 20 borderline borderline/borderline tuberculoid (BB/BT) and 9 tuberculoid (TT) cases. Mean serum arginase levels were 1.51 +/- 0.43, 1.41 +/- 0.43, 1.24 +/- 0.43 and 1.10 +/- 0.026 mu moles/min/ml in LL/BL, BB/BT and TT patients and healthy controls respectively. The lymphocyte arginase activity showed a similar increasing trend from TT to LL/BL. The mean lymphocyte arginase levels were 0.87 +/- 0.31 mu moles/min/10(6) cells in healthy controls and 1.81 +/- 0.40, 2.54 +/- 0.60 and 5.48 +/- 0.56 mu moles/min/10(6) cells in TT, BB/BT and LL/BL patients respectively. The increasing trend specially in lymphocyte arginase levels across the spectrum of leprosy correlated with the degree of impairment in the protective cell mediated immune response and also the extent of disease. The role of these pathophysiological alterations in relation to defect in immune response calls for investigation.
