RESUMO

Objective:To investigate the effects of interleukin-34(IL-34)on the expression of CC chemokine ligand 28 (CCL28) by fibroblast-like synoviocytes(FLS) in rheumatoid arthritis(RA) patients.Methods:FLS were isolated from 6 RA patients and stimulated with IL-34,IL-34 receptor antagonist /IL-34,inhibitors of signaling pathway/IL-34 in vitro respectively.CCL28 mRNA expression was measured by reverse transcription polymerase chain reaction(RT-PCR).The level of CCL28 in the supernatant of RA FLS culture was detected by enzyme linked immunosorbent assay(ELISA).Statistical analysis between groups was performed by t test.Results:Compared with unstimulated FLS,CCL28 expression was increased obviously in IL-34-stimulated group(P<0.05).The level of CCL28 was significantly decreased when anti-IL-34 antibody was added into IL-34-administrated RA FLS(P<0.05).While after adding of nuclear factor κB(NF-κB) and mitogen-activated protein kinase38(p38 MAPK) signaling pathway inhibitors into the cell culture system,CCL28 expression was remarkably reduced (P<0.05).Conclusion: The secretion of CCL28 by RA FLS can be promoted by the binding of IL-34 with its specific receptor via the activation of NF-κB and p38 MAPK signaling pathways,which suggests that CCL28 might be involved in the pathogenesis of RA.

Objective:To investigate the effect of electroacupuncture (EA) at Ganshu (BL 18) and Shenshu (BL 23) on vascular endothelial growth factor (VEGF) and platelet endothelial cell adhesion molecule-1 (PECAM-1)/CD31 around the cerebral infarction focus in middle cerebral artery occlusion (MCAO) rats and the possible mechanism, thus to provide a new strategy for the treatment of cerebral ischemic stroke by acupuncture. Methods:A total of 180 healthy male Sprague-Dawley (SD) rats were randomly divided into a sham operation group, a model group, an acupoint group and a non-acupoint group, 45 rats in each group. MCAO model was established using the modified line-embolus method in all rats except for those in the sham operation group; rats in the acupoint group were treated with EA at Ganshu (BL 18) and Shenshu (BL 23); rats in the non-acupoint group were treated with EA at the control points; rats in other 2 groups were only subjected to bundling without treatment. Ten rats in each group were randomly selected on the 3rd day, the 14th day and the 21st day after acupuncture stimulation to test the neurological function impairment. The expression levels of CD31 and VEGF were also detected. Results:Compared with the model group and non-acupoint group, the neurological function score of the acupoint group was decreased at each time point, and the differences were statistically significant (P<0.05,P<0.01). The expressions of VEGF and CD31 in each group were the lowest on the 3rd day, reached the peak on the 14th day and still remained at high level on the 21st day. And the differences among groups were statistically significant both on the 14th day and the 21st day (P<0.05,P<0.01). Compared with the model group and the non-acupoint group, the expressions of VEGF and CD31 in the acupoint group were increased, and the differences were statistically significant (allP<0.05). Conclusion: EA at Ganshu (BL 18) and Shenshu (BL 23) can significantly improve the neurological function score of MCAO model rats, and shows protective effect on cerebral ischemia. The protective mechanism may be related to the up-regulation of CD31 and VEGF expression around the cerebral infarction focus in the MCAO model rats and induction of angiogenesis.

Objective: To observe the effect of acupuncture plus thunder-fire moxibustion on the expressions of matrix metalloproteinase-3 (MMP-3), tissue inhibitor of metalloproteinase-1 (TIMP-1) and transforming growth factor-β1 (TGF-β1) in cartilage of knee osteoarthritis (KOA) rats, and to explore the mechanism of acupuncture plus thunder-fire moxibustion in the treatment of KOA. Methods:Thirty Sprague-Dawley (SD) rats were randomly divided into a blank control group, a model group and an acupuncture-moxibustion group by random digits table, 10 rats in each group. Rats in the model group and the acupuncture-moxibustion group were injected with papain in the right posterior knee joint to prepare the models. The levels of MMP-3 and TIMP-1 in rat synovium of each group were measured by enzyme-linked immunosorbent assay (ELISA) after 2 weeks of treatment. The level of TGF-β1 was determined by Motic B5 Micro-camera system. Results:The levels of MMP-3 and TIMP-1 in the cartilage of the model group were significantly higher than those in the blank control group (allP<0.01); the levels of MMP-3 and TIMP-1 in the acupuncture-moxibustion group were lower than those in the model group, and the between-group differences were statistically significant (all P<0.05). The levels of MMP-3 and TIMP-1 in the acupuncture-moxibustion group were higher than those in the blank control group, and the differences were statistically significant (all P<0.05). The level of TGF-β1 in cartilage tissues of the model group was significantly lower than that in the blank control group (P<0.01); the level of TGF-β1 in the acupuncture-moxibustion group was higher than that in the model group (P<0.05), but it was lower than that in the blank control group, and the between-group difference was statistically significant (P<0.05). Conclusion: Acupuncture plus thunder-fire moxibustion can effectively recover the abnormal expressions of MMP-3 and TIMP-1 in KOA model rats and somewhat up-regulate TGF-β1, which may be one of its mechanisms of acupuncture plus thunder-fire for KOA.

RESUMO

Objective To observe the efficacy of acupoint thread embedding in treating bronchial asthma in remission stage and how it controls the acute attack, the length of interval, and severity of symptoms during the attack. Method Thirty subjects were selected as the treatment group by using the random number table following the visiting sequence, to receive acupoint thread embedding plus Western medication; thirty cases in the control group were given Western medication alone, both for 3 months. The pulmonary function and eosinophil count were compared before and after the intervention, as well as the symptoms scores during the acute attack. Result There were significant differences in most parameters between the two groups (P<0.05). Conclusion Acupoint thread embedding can produce a positive effect in preventing and treating bronchial asthma in remission stage.

<p><b>OBJECTIVE</b>This study attempts to test the total amount of 41 inorganic elements in traditional Chinese medicine materials (TCMM), to analyze to surpass the set standard of 5 toxic heavy metal elements and to explore possible method of modification.</p><p><b>METHOD</b>By using microwave as digestion tool, ICP-MS and ICP-AES as detective instrument, 41 elements in 466 samples are tested. While the amount of elements Al, Ca, Fe, K, Mg, Mn, P, S, Sr are determined by ICP-AES, the amount of the rest 32 elements are determined by ICP-MS. SPSS 16.0 is used as the statistical tool for data analysis.</p><p><b>RESULT</b>Testing of reference material Astragalus showed that the measurements of each element are within the reference range, and except for element I, RSD of determination for each element is less than 10%, thus the accuracy and precision of the study method is confirmed. Using above mentioned instruments and methods, general information of elements in 466 TCMM are obtained.</p><p><b>CONCLUSION</b>The concentrations of each elements in TCMM varied a lot and are different among each TCMM. The over standard rate of 5 heavy metal elements are lower than previous results, however, the over standard rate is still descending in the order of Cd > Cu > Pb > As > Hg. The content of heavy metal elements in TCMM varies among different areas, especially, over standard of heavy metals are not detected in locality as Gansu, Qinghai, Shanxi, Xinjiang and Hainan provinces.</p>

<p><b>OBJECTIVE</b>To assess the effectiveness of vertical control with high-pull headgear in the treatment of high angle cases.</p><p><b>METHODS</b>Forty high angle cases (MP-SN >40 degrees) were included in this study and all the patients were treated with extraction. Twenty patients in the study group and control group, respectively. Totally 14 cephalometric measurements were selected for the assessment Paired t-test and independent t-test were used in the analysis.</p><p><b>RESULTS</b>No significant differences were found between the two groups (P > 0.05). In the study group, after treatment Y angle, MP-SN angle, MP-FH angle and U6-PP distance increased 1.9 degrees, 1.5 degrees, 2.3 degrees and 1.1 mm, respectively (P < 0.01).</p><p><b>CONCLUSIONS</b>High-pull headgear could not provide effective vertical control in high angle cases.</p>

<p><b>OBJECTIVE</b>To study the characters of concentration of 42 elements in Radix Astragali and make an attempt at looking for relationship between the element concentration and regions where samplers are obtained.</p><p><b>METHOD</b>Determining the content of elements in 40 Radix Astragali samples from 7 different provinces by ICP-MS, AFS and ASS. Analyzing the correlation of elements in Radix Astragali using statistic software (Spss).</p><p><b>RESULT</b>Firstly, similar lines of element concentration have been acquired in our research. Secondly, it is observed that the content of elements in the samples shows regional diversity. Thirdly, there are 346 correlative element pairs in correlate analysis. And some of them indicate remarkable correlativity.</p><p><b>CONCLUSION</b>With the aid of obtained results, it is concluded that element content in Radix Astragali display special distributing line. Remarkable correlation is presented in some element pairs. The quality of Radix Astragali gained from Neimeng, Shanxi and Gansu are better than those from other regions.</p>

<p><b>OBJECTIVE</b>To provide basis of reference values for relevant parameters of Chinese Reference Man.</p><p><b>METHODS</b>Eighteen kinds of major organ or tissue samples, including muscle, rib, liver, and so on, were obtained from 4 areas (Hebei, Shanxi, Jiangsu, and Sichuan provinces) with different dietary patterns in China in autopsy of 16 healthy adult men, who had just encountered sudden deaths. At the same time, whole blood samples were collected from 10 volunteers living in each of these areas. The concentrations of 56 elements in these samples were detected by using Inductively Coupled Plasma Mass Spectrometry (ICP-MS), Inductively Coupled Plasma Atomic Emission Spectrometry (ICP-AES), and Graphite Furnace Atomic Absorption Spectrometry (GF-AAS) techniques. Based on obtained concentrations and reference values of these organ or tissue weights for Chinese Reference Man, the relative elemental burdens in these organs or tissues as well whole body were also estimated.</p><p><b>RESULTS</b>The concentrations of 56 elements in 18 main organs or tissues were determined all together and their elemental organ or tissue and whole body burdens were estimated. Furthermore, the distributions of important elements for radiation protection in these organs or tissues were emphatically discussed.</p><p><b>CONCLUSION</b>By summing with past related results, the total results obtained from the series of research may provide more reliable and better representative basis of these reference values for Chinese Reference Man than before.</p>

Objective Our previous study has shown that porcine antigen-primed and CD4 + T cells activated macrophages are capable of the ecognition and rejection of porcine xenografts but not mouse allografts, and therefore suggested the involvement of signaling between the graft and macrophages in this specific graft recognition and destruction. Methods NOD-SCID mice were transplanted with fetal pig pancreatic fragment (FPP) before adoptive transfer with exogenous macrophages isolated from rejecting FPP xenografts of BALB/c recipient mice. The exogenous macrophages were tracked by Ly5.1 surface antigen or via CSFE staining. Gene expression of CCR2 and CCR5 and their chemokines in transplanted FPP xenografts was evaluated by real-time PCR. Results After the adoptive transfer, recently transplanted but not established FPP xenografts were rejected by exogenous activated macrophages. In the meantime, greater level of chemokine gene expression was detected in recently-transplanted compared with the established xenografts. Furthermore, expression of both CCR2 and CCR5 genes was enhanced significantly in activated macrophages when compared with non-activated macrophages. Conclusion Upregulated chemokines were associated with macrophage recruitment and destruction of islet xenografts.

Objective To establish a method for the determination of 14 kinds of lanthanide (La) series elements in human feces at the same time by ICP-MS.Methods In January 2009,human feces were collected for three consecutive days from 30 subjects in Tianjin area,the fecal samples for the three-day period were weighed and homogenized;then dried;grinded and ashed,the samples were digested by HNO3 before determination.The solution was directly analyzed by ICP-MS to determine the concentrations of the 14 kinds of La series elements in human faces with rhenium (Re) internal standard calibration.Results The linear ranges were 0-10 ?g/L with a correlation coefficient for each element of more than 0.999.The detection limits of 139La,140Ce,141Pr,143Nd,147Sm,151Eu,158Gd,159Tb,163Dy,165Ho,166Er,169Tm,174Yb and 175Lu in human feces were 0.48,0.49,0.47,1.6,1.9,0.46,0.79,0.18,0.65,0.39,0.38,0.17,0.09 and 0.09 ng/L respectively.The recoveries of this method were 93.42%-108.21%,and RSDs were 2.91%-9.20%.The analytical values of the certified reference material of human hair GBW 09101a by this method showed closed agreement with the reference values.Conclusion This method has relatively higher sensitivity and less interference,and is applicable to the rapid determination of 14 kinds of La series elements in human faces at the same time.

<p><b>OBJECTIVE</b>To elucidate the relationship between azoospermia factor(AZF) microdeletion of Y chromosome and azoospermia, the exact breakpoint of a severe Y-chromosome deletion was determined according to the physical map of AZF.</p><p><b>METHODS</b>Multiplex polymerase chain reaction was used to amplify fifteen sequence tagged sites (STS), namely sY82, sY84, sY86 in AZFa, sY124, sY127, sY128, sY133, sY134, sY143 in AZFb, sY239, sY242 sY254, sY255 in AZFc, and sY145, sY152 in AZFd; sex-determining region Y(SRY) was taken as an internal control. And then sY82,sY86,sY85,sY84 were further analyzed using the sample of the patient who had Y-chromosome deletion by G band analysis to map the breakpoint at molecular level.</p><p><b>RESULTS</b>All 15 STS and sY85 were amplified in positive control while only sY82, sY86 were amplified in the clinical sample, thus the breakpoint was found to be between sY86 and sY85.</p><p><b>CONCLUSION</b>This study on the patient provided the direct biomolecular evidence of the exact breakpoint of the severe Y-chromosome deletion and established the deletion map of acrocentric chromosome. It also proved that the patient's azoospermia was due to the deletion of AZF.</p>

<p><b>OBJECTIVE</b>To obtain peptide mimicking epitopes of Schistosoma japonicum (S. japonicum) through screening of a phage peptide library and to test their potential for induction of protection.</p><p><b>METHODS</b>S. japonicum infected sera from Microtus fortis (IMFS) and normal sera from Microtus fortis (NMFS) were used respectively to screen a 12-mers random peptide library by testing the reactivity of anti-S. japonicum serum with the phagotopes. After three rounds of biopanning, the pooled phages were used to immunize mice, after which challenge infection was performed.</p><p><b>RESULTS</b>Of 12 randomly picked clones, 10 clones selected using IMFS and 7 clones selected using NMFS were shown to be antigenic. Significant reduction in adult worms (22.6%) and a high reduction (68.9%) in liver eggs were achieved following immunization with phages screened with IMFS. However, no protection was elicited by those selected with NMFS.</p><p><b>CONCLUSION</b>The results show that the phagotopes are both antigenic and immunogenic, suggesting a potential use of phage displayed peptide as novel vaccines against S. japonicum.</p>

RESUMO

The effect of trypsin on the separation an subculture of the keratinocytes was investigated in this work. It was found that when 0.25% trypsin was employed for 5 minutes to separate keratinocytes, the number of active keratinocytes and the cells capable of forming colony were higher than those of other experimental conditions. The maximum attached ratio of primary keratinocytes was obtained when skin tissues were treated at 0.05% concentration of trypsin. With the increase of the trypsin concentrations, the attached ratio, attachment rate constant, and colony forming efficiency were all increased. Thus, 0.25% concentration of trypsin was recommended for separating and subculturing the keratinocytes.

RESUMO

The effect of antioxidants on the in vitro life span of mouse keratinocytes was investigated in this work. It was found that the life span of the keratinocytes cultured in the medium supplemented with antioxidants was extended significantly. The most beneficial antioxidant used in this work was the mercaptoethanol, followed by the catalase and SOD. However, the growth rates of keratinocytes in vitro under all the experimental conditions still declined with the culture time. It was also found that the antioxidants added in the medium were also helpful to enhance the keratinocyte colony formation. In addition, the aging kinetics of the mouse epidermal keratinocytes in vitro were analyzed, and finally the aging rate constants corresponding to antioxidants used were calculated.
