RESUMO

Objective To investigate the relationship between the expression of glutathione peroxidase(GPX)genes and the clinical prognosis in glioma patients,and to construct and evaluate the model for predicting the prognosis of glioma. Methods The clinical information and GPX expression of 663 patients,including 153 patients of glioblastoma(GBM)and 510 patients of low-grade glioma(LGG),were obtained from The Cancer Genome Atlas(TCGA)database.The relationship between GPX expression and patient survival was analyzed.The key GPX affecting the prognosis of glioma was screened out by single- and multi-factor Cox's proportional-hazards regression models and validated by least absolute shrinkage and selection operator(Lasso)regression.Finally,we constructed the model for predicting the prognosis of glioma with the screening results and then used concordance index and calibration curve respectively to evaluate the discrimination and calibration of model. Results Compared with those in the control group,the expression levels of GPX1,GPX3,GPX4,GPX7,and GPX8 were up-regulated in glioma patients(all P<0.001).Moreover,the expression levels of other GPX except GPX3 were higher in GBM patients than in LGG patients(all P<0.001).The Kaplan-Meier curves showed that the progression-free survival of GBM with high expression of GPX1(P=0.013)and GPX4(P=0.040),as well as the overall survival,disease-specific survival,and progression-free survival of LGG with high expression of GPX1,GPX7,and GPX8,was shortened(all P<0.001).GPX7 and GPX8 were screened out as the key factors affecting the prognosis of LGG.The results were further used to construct a nomogram model,which suggested GPX7 was the most important variable.The concordance index of the model was 0.843(95%CI=0.809-0.853),and the calibration curve showed that the predicted and actual results had good consistency. Conclusion GPX7 is an independent risk factor affecting the prognosis of LGG,and the nomogram model constructed with it can be used to predict the survival rate of LGG.

RESUMO

Wound healing is an important physiological process as well as one of the problems to be solved in clinical medicine. Plant-derived exosomes are green, safe and efficient, and are expected to provide a new solution for wound healing. In this paper, ginseng exosomes were extracted and purified by hypervelocity centrifugation, and were identified to be consistent with the microscopic identification characteristics of exosomes. Fluorescence dye was used to label exosomes, which showed that exosomes could be ingested by skin cells. CCK-8 methods indicated that ginseng exosomes could promote the proliferation of skin cells at 24 h and 48 h (P<0. 05). Panax ginseng exosomes detected by real-time quantitative PCR increased the transcription levels of human collagen type 1 琢1 chain (COL1A1) and fibronectin-1 (human fibronecin-1) genes in skin cells (P<0. 05).Western blotting analysis showed that ginseng exosomes could increase the protein levels of TGF-β1 (transforming growth factor-β Ⅰ), Vimentin and Ki67 in skin cells (P<0. 05). The skin injury experiment in mice confirmed that ginseng exosomes could accelerate recovery after skin injury. Moreover, the inflammatory cytokines such as nuclear factor kappa B (NF-κB), INOS (nitric oxide synthase) and cycoxidase (COX-2) could be decreased (P < 0. 05). In this study, ginseng exosomes were successfully extracted and purified by hypervelocity centrifugation combined with density gradient centrifugation, which suggested that ginseng exosomes can regulate the skin cell cycle, activate the TGF-β pathway, promote the proliferation of skin cells, accelerate the recovery of damaged skin, and reduce the level of inflammatory factors.

Objective:To explore the clinical effect of small incision endoscopic thyroidectomy in the treatment of differentiated thyroid cancer (DTC).Method:The clinical data of 63 patients with DTC in Anhui Wanbei Hospital were analyzed retrospectively. According to different surgical schemes, 33 patients who underwent complete endoscopic thyroidectomy were listed as the control group and 30 patients who underwent small oral endoscopic thyroidectomy were listed as the experimental group. The operation and postoperative recovery, immunosuppressive indexes (CD 3+, CD 4+, CD 8+, CD 4+/CD 8+) before operation and on the first and third day after operation, the levels of tumor indexes vascular endothelial growth factor (VEGF), thyroglobulin (Tg) before operation and one month after operation and the incidence of complications were compared between the two groups. Results:Compared with the control group, the operation time in the experimental group was shorter: (87.73 ± 15.35) min vs. (102.45 ± 20.78)min, t = 3.171, P = 0.002 ; the amount of intraoperative bleeding in the experimental group was shorter: (20.37 ± 5.26) ml vs. (44.25 ± 6.39) ml, t = 16.099, P<0.001 ; the incision length and the postoperative hospital stay in the experimental group were lower: (2.03 ± 0.14) cm vs. (2.58 ± 0.18) cm, t = 13.440, P<0.001; (4.58 ± 1.17) d vs. (6.02 ± 1.09) d, t = 5.057, P<0.001; the number of lymph node dissections in the experimental group was more: (6.20 ± 1.08) cases vs.(5.12 ± 1.13) cases, t = 3.869, P<0.001.Compared with before operation, the levels of CD 3+, CD 4+, CD 4+/CD 8+ decreased and CD 8+ increased in the two groups on the first day after operation, but the change range in the experimental group was small, and the experimental group recovered to the preoperative level on the third day after operation ( P>0.05). One month after operation, the levels of serum VEGF and Tg in the two groups were lower than those before operation ( P<0.05), but there was no significant difference between the two groups ( P>0.05). There was no significant difference in the incidence of complications between the two groups ( P>0.05). Conclusions:Small incision endoscopic thyroidectomy in the treatment of DTC can achieve the same tumor treatment effect as complete endoscopic surgery, significantly shorten the operation time, reduce incision trauma and intraoperative bleeding, and reduce immunosuppression, which is conducive to postoperative rehabilitation and good treatment safety.

To study the gene expression of cardiac mesenchymal cells in patients with type 2 diabetes mellitus (T2DM)based on a whole-genome high-throughput sequencing dataset,screen differentially expressed genes,analyze the genetics signature of cardiac mesenchymal cells in T2DM patients by bioinformatics analysis,and explore the environmental chemicals related to the key differentially expressed genes. The dataset GSE106177 was obtained from Gene Expression Omnibus (GEO) database.The dataset was pre-processed and analyzed by Network Analyst,Cytoscape 3.7.1,String11.0,CTD,and HMDD for screening for differentially expressed genes,enrichment analysis,establishment of protein-protein interaction (PPI) networks,and screening for relevant environmental chemicals. The gene expression pattern of cardiac mesenchymal cells in T2DM patients was significantly different from that in the control group.There were 135 differentially expressed genes,of which 58 (42.96%) were up-regulated and 77 (57.04%) were down-regulated.The differentially expressed genes mainly participated in biological processes such as multicellular organism development,anatomical structure development,and system development and were mainly involved in hepatocellular carcinoma,Cushing's syndrome,and cholesterol metabolism.PPI network showed that UBC was the core protein node.The microRNA-Gene interaction network showed that seven microRNAs,represented by hsa-mir-8485,interacted with the differentially expressed genes.Key T2DM related genes such as UBC,DNER,and CNTN1 interacted with bisphenol A. The gene expression profile of cardiac mesenchymal cells markedly changes in T2DM patients,during which UBC may play an important biological role.Bisphenol A exposure may also affect the development and normal function of cardiac cells in T2DM patients.

OBJECTIVE: To investigate the. METHODS: for locating and selecting the acupoints of "Taixi" (KI3), "Shuiquan" (KI5), "Fuliu" (KI7), "Jiaoxin" (KI8), "Zhubin" (KI9), and "Yingu" (KI10) and the morphological structure of these acupoints in rabbits. MethodsAccording to the WHO and national standards for human acupoints and rabbit X-ray images, acupoint locations were marked using the anatomical landmarks on body surface in 10 New Zealand rabbits. The acupoints were dissected to compare the homologous and analogous tissue between rabbits and human body and thus correct the locations of these acupoints. Potentials were measured for the 10 New Zealand rabbits at the corrected locations of the acupoints and around the acupoints, and the final locations of these acupoints were determined by comparing the anatomical results and the data of potentials. Anatomical observation was performed after marking, and the relationship between acupuncture needle and adjacent structure was observed. RESULTS: "Taixi" was located in the ankle area, at the midpoint between the prominence of the medial malleolus and the calca-neal tendon; "Shuiquan" was located in the calcaneal area below "Taixi" in the depression anterior to the calcaneal tuberosity; "Fuliu" was located at the medial side of the calf, at 2 cun above the prominence of the medial malleolus anterior to the calcaneal tendon; "Jiaoxin" was located at the medial side of the calf, at 2 cun above the prominence of the medial malleolus and in the depression posterior to the medial border of the tibia; "Zhubin" was located at the medial side of the calf, at 5 cun above the medial malleolus on the line between "Taixi" and "Yingu"; "Yingu" was located at the medial side of the knee, at the posterior-inferior border of the semitendinosus tendon on the popliteal crease. The results of skin potentials at the acupoints suggested that "Taixi", "Shuiquan", "Fuliu", and "Zhubin" were high-reliability acupoints, "Jiaoxin" was a medium-reliability acupoint, and "Yingu" was a low-reliability acupoint. CONCLUSION: Comparative anatomy combined with imaging, surface anatomy, and electrophysiological techniques of acupoints can help with the accurate localization and selection of acupoints in experimental animals, improve the reliability of acupoint location, and enrich the comparative anatomical data of acupoints.

OBJECTIVE@#To explore potential therapeutic targets other than androgen-deprivation treatment for prostate cancer by screening the proteins induced by androgen at palmitoylation modification level in LNCaP cells.@*METHODS@#The LNCaP cells were treated with androgen (Methyltrienolone, R1881, 5 nmol/L) or dimethyl sulfoxide (DMSO) for 24 h, and then labeled with alkynyl palmitic acid Alk-C16 (100 μmol/L). After that, the cells were collected, lysed, the total protein was extracted, agarose beads labeled with azide (1 mmol/L) were added, and the click-chemistry reaction was carried out at room temperature for 1 h. The covalent bond formed by click-chemistry reaction of azide and alkynyl group was used to enrich the palmitoylated proteins on agarose beads. Label-free quantitation (LFQ) was used to compare the protein palmitoylation level of R1881 treated and untreated cells to screen the proteins induced by androgen at palmitoylation modification level.@*RESULTS@#In this experiment, 907 potential palmitoylated proteins (mascot score>2, P<0.05) were identified, among which 430 proteins had LFQ values not zero at least twice. Among the 430 proteins, the palmitoylation levels of 92 candidates were increased by androgen treatment, and their LFQ values were significantly upregulated (>1.5-fold, P<0.05) in ≥2 samples of androgen-treated vs. untreated LNCaP cells. We also used the software of cytoscape to classify the 92 proteins, and found that the known functional proteins of them could be divided into three categories: metabolism related, protein folding related and translation initiation related. Among them, metabolism related proteins included lipid metabolism (6), glucose metabolism (7) and respiratory electron transport chain (8), and a small amount of amino acid metabolism (2) and other metabolism related proteins (2). Notably, the ratio of LFQ of cytochrome b-c1 complex subunit 2 (UQCRC2) was significantly (>3-fold, P<0.05) higher in androgen-treated cells compared with untreated cells, indicating that the palmitoylation level of UQCRC2 was enhanced by androgen most significantly than that of others. The second was long-chain acyl CoA dehydrogenase (ACADVL) related to lipid metabolism and glucose 6-phosphate dehydrogenase (PGD) related to glucose metabolism, but the LFQ ratio of them was less than 3-fold.@*CONCLUSION@#The research on palmitoylation mechanism of metabolism, especially the proteins related to respiratory electron transport chain, will provide a new guidance for the diagnosis and treatment of prostate cancer and the development of targeted drugs.

OBJECTIVE@#To identify the chaperone of polypyrimidine tractor-binding protein-associated splicing factor (PSF) in myeloid leukemia cells, and to explore the mechanism and redistributive pattern to cell surface of PSF in chemo-sensitive HL60 cells and resistant HL60/DOX cells.@*METHODS@#The eukaryotic expression vector of PSF was transfected with liposomes transiently, then flow cytometry was used to detect the expression level of PSF on the cell surface 24 h, 48 h and 72 h after vector transfections. We constructed a chimeric expression vector, streptavidin binding peptide (SBP)-PSF, meanwhile this vector was transfected and made SBP-PSF fusion protein overexpress. In addition, we used streptavidin magnetic beads to precipitate the cellular chaperonin of PSF and then identified its chaperonin by mass spectrometry (MS). Lentiviral vectors containing cytokeratin18 (K18) interference sequences were transfected into 293T cells to prepare lentivirus. HL60 and HL60/DOX cells were infected with lentivirus to obtain stable interfering K18 cell lines. Next, flow cytometry was used to test the membrane relocation level of PSF. Together, these methods confirmed the similar or different mechanisms of the PSF redistributing to membrane synergistically mediated by K18 in HL60 and HL60/DOX cells.@*RESULTS@#The expression of membrane relocated PSF was detected every day for three days (at the end of 24 h, 48 h and 72 h) after transient overexpression. The expressing rate of PSF on the cell surface was 22.4%±3.5%, 37.9%±6.0%, 58.3%±8.8%, respectively in sensitive HL60 cells, while that was 4.7%±0.5%, 3.9%±0.6%, 2.9%±0.6% , respectively in resistant HL60/DOX cells. The difference of expressing rate on each day was significant, P<0.01. We identified K18 detected by co-immunoprecipitation and mass spectrum assay which was the cellular chaperone of PSF. We found that K18 knockdown decreased the PSF expression level which redistributed on cell surface from 48.9%±5.4% to 6.2%±1.0% in sensitive HL60 cells, and from 9.11%±1.2% to 2.21%±0.51% in resistant HL60/DOX cells, respectively.@*CONCLUSION@#K18 is the intracellular chaperonin of PSF. The interaction of PSF and K18 mediates its redistribution to cell membrane in sensitive cells. While in resistant cells, PSF failed to relocate at the cell surface and accumulated in cells, which mediated resistance to chemotherapeutics.

Objective :To analyze value of plasma levels of homocysteine (Hcy)and D-dimer(D-D)for early diagnosis and stag-ing in aged patients with myocardial infarction (MI) and their correlation .Methods :A total of 92 aged MI patients treated in our hospital from Jan 2014 to Jun 2016 were enrolled in MI group.Another 92 healthy aged people were simultaneously regarded as healthy control group .Plasma Hcy and D-D levels were measured and compared between two groups .Diagnos-tic value of Hcy ,D-D and their combination for MI were analyzed .According to clinical staging ,MI group was further di-vided into acute stage group (n=46) and non-acute stage group (n=46).Levels of Hcy ,D-D ,cardiac troponin I (cTnI ) and cTnT were measured and compared between two groups ,and correlation among Hcy ,D-D levels and occurrence and staging of MI ,cTnT and cTnI levels were analyzed .Results :Compared with healthy control group ,there were significant rise in plasma levels of Hcy [(8-76 ± 2-59)μmol/L vs.(22-72 ± 10-21)μmol/L]and D-D [(229-59 ± 31-60 )μg/L vs. (1057-88 ± 250-74)μg/L]in MI group , P= 0-001 both .Accuracies of Hcy ,D-D and Hcy+ D-D diagnosing MI were 90-76%,84-78% and 94-02% respectively.Compared with non-acute stage group ,there were significant rise in plasma levels of Hcy ,D-D ,cTnT and cTnI in acute stage group ,P=0-001 all.Correlation analysis indicated that plasma levels of Hcy and D-D were significant positively correlated with MI occurrence ,cTnT and cTnI levels (r=0-875-0-951) ,and sig-nificant inversely correlated with MI staging (r= -0-870 ,-0-914) , P=0-001 all.Conclusion :Hcy and D-D level detec-tions possess high accuracy for early diagnosis and clinical staging of MI .It can be extended and applied in clinic .

Flavonoids are a group of secondary metabolites found in plants. They have many pharmacological functions and play an important role in Chinese sumac( Rhus chinensis),which is a well-known traditional Chinese medicinal plant. Chalcone isomerase( CHI,EC 5. 5. 1. 6) is one of the key enzymes in the flavonoids biosynthesis pathway. In this paper,the full-length c DNA sequence encoding the chalcone isomerase from R. chinensis( designated as Rc CHI) was cloned by RT-PCR and rapid-amplification of c DNA Ends( RACE). The Rc CHI c DNA sequence was 1 058 bp and the open reading frame( ORF) was 738 bp. The ORF predicted to encode a 245-amino acid polypeptide. Rc CHI gene contained an intron and two exons. The sequence alignments revealed Rc CHI shared47. 1%-71. 6% identity with the homologues in other plants. Real-time PCR analysis showed that the total flavonoid levels were positively correlated with tissue-specific expressions of Rc CHI mRNA in different tissues. The recombinant protein was successfully expressed in an Escherichia coli strain with the p GEX-6 P-1 vector. In this paper,the CHI gene was cloned and characterized in the family of Anacardiaceae and will help us to obtain better knowledge of the flavonoids biosynthesis of the flavonoid compounds in R. chinensis.

<p><b>BACKGROUND</b>White matter lesions (WMLs) are common findings in brain magnetic resonance imaging (MRI) and are strongly associated with stroke incidence, recurrence, and prognosis. However, the relationship between WMLs and transient ischemic attacks (TIAs) is not well established. This study aimed to determine the clinical significance of WMLs in patients with TIA.</p><p><b>METHODS</b>A total of 181 consecutive inpatients with first-ever TIA were enrolled. Brain MRIs within 2 days of symptom onset were used to measure WML volumes. Recurrent vascular events within 1 year of TIA onset were assessed. The relationship between WMLs and recurrent risk of vascular events was determined by a multivariate logistic regression.</p><p><b>RESULTS</b>WMLs were identified in 104 patients (57.5%). Age and ratio of hypertension were significantly different between patients with and without WMLs. The incidence of vascular events in patients with WMLs significantly increased in comparison to those without WMLs (21.15% vs. 5.19%, 95% confidence interval [CI]: 1.18-15.20, P = 0.027) after controlling for confounders. Furthermore, distributions of WML loads were found to be different between patients who developed vascular events and those who did not. WML volumes were demonstrated to be correlated with recurrent risks, and the fourth quartile of WML volumes led to an 8.5-fold elevation of recurrent risk of vascular events compared with the first quartile (95% CI: 1.52-47.65, P = 0.015) after adjusting for hyperlipidemia.</p><p><b>CONCLUSION</b>WMLs occur frequently in patients with TIA and are associated with the high risk of recurrent vascular events, suggesting a predictive neuroimaging marker for TIA outcomes.</p>

Objective To provide the basis for early diagnosis and clinical treatment for diabetic kidney disease(DKD)and cardiovascular disease by analyzing the causes of DKD and the relationship between them. Methods Microarray gene chip data from the glomerulus in DKD were downloaded from GEO Database.The gene expression profile,gene function,protein-protein interaction network,gene and signal pathway co-expression network of control and DKD groups were analyzed and the key node genes between the two groups were selected by R,QOE,GCBI,STRING and Genclip.Results The gene expression profile of glomerular tissue in DKD patients was significantly altered.Compared with control group,there were 844 differentially expressed genes in DKD patients.One protein-protein interaction network with the core of TNNT 2 occurred among top 20 differentially expressed genes.Differentially expressed genes were mainly related to inflammatory and stress responses,as well as to MAPK signal pathway.Conclusion The cause of DKD is mainly related to inflammatory and stress responses, which are also related to cardiovascular disease.

·AIM: To evaluate the effects of foldable capsular vitreous body (FCVB) implantation on the treatment of severe ocular trauma and late silicone oil-dependent eyes due to severe ocular trauma and recurrent retinal detachment. · METHODS: We retrospectively reviewed the four patients (four eyes) with FCVB implantation at our hospital since November 2017. Out of these 4 patients,2 were males and 2 were females, with an average age of 31.5 years and an average intraocular pressure (IOP) of 5.6mmHg pre-operatively. Among those 4 eyes,3 eyes underwent silicone oil tamponade due to severe ocular trauma and the other one was recurrent retinal detachment in silicone oil - filled eye. Standard pars plana vitrectomy (PPV) was performed, and the FCVB was triple folded and implanted in the vitreous cavity of four eyes. The retinal was assessed, as well as visual acuity,IOP, FCVB condition before and after treatment, and applied therapy. ·RESULTS: All of those 4 eyes underwent successfully implantation of FCVB, which remained its proper position. During 1-3mo follow-up, the mean visual acuity was no different compared with pre - operative values.

Objective To analyze the differentially expressed genes and key proteins in T cells between acute and chronic idiopathic thrombocytopenic purpura (ITP) in children and provide the basis for the prevention and therapies of this disease. Methods Microarray gene chip data from T cells of children with acute or chronic ITP were downloaded from the GEO Database. The gene expression profiles,gene function,and protein interaction network were analyzed by R,QOE,Networkanalyst,GCBI,and GenClip. Results The gene expression profiles between these two groups were significantly different. Among the 54 675 genes analyzed,there were 457 (0.84%) differentially expressed genes between these two groups. In the protein interaction networks among top 20 differentially expressed genes,the core was JUN(down-regulated) and ITCH(up-regulated),which were both related to the tumor necrosis factor signaling pathway;differentially expressed genes were mainly related to the activation and tolerance of T cell. Conclusions The gene expression profiles differ between acute and chronic ITP patients,suggesting that the gene transcription profile plays a regulatory role in the different stages of ITP. JUN and ITCH may play a role in predict the progression of ITP and may exert their biological functions by regulating the tumor necrosis factor signaling pathway.

Objective To evaluate the magnetic resonance imaging (MRI) findings in differential diagnosis between the adult reversible splenial lesion syndrome (RESLES) and ischemic infarction of the splenium of the corpus callosum (SCC). Methods The MRI findings and clinical data of 7 RESLES patients and 13 patients with ischemic infarction of SCC who were clinically diagnosed and treated in our center from May 2015 to June 2017 were analyzed retrospectively. The main MRI findings included location,morphology,signal intensity,maximum cross-sectional area,diffusion weighted imaging (DWI),and apparent diffusion coefficient (ADC) value. Results On the MRI findings of 7 RESLES patients (5 males and 2 females),the centers of all lesions of the SCC were located in the midline of SCC,the lesion shapes were round,ellipse,or spindle,and the distribution of the lesions was bilateral and symmetric as the center of the midline of SCC. The lesions were hyperintense on DWI,and the mean maximum cross-sectional area of lesions was (56.9±32.6) mm and the mean ADC value was (0.3963±0.0715) ×10 mm/s. On the review MRI,all the lesions disappeared (mean interval:10 days). On the MRI findings of 13 patients with ischemic infarction of SCC (10 males and 3 females),the lesions were irregular or patchy in shape and were almost laterally and asymmetrically distributed. The lesions were hyperintense on DWI,and the mean maximum cross-sectional area was (55.1±43.9) mm and the mean ADC value was (0.4978±0.0123) ×10 mm/s. The mean maximum cross-sectional area (t=0.096,P=0.925) and the ADC value (t=-1.988,P=0.062) were not significantly different between RESLES group and ischemic infarction of SCC group. Conclusions The location,morphology,and distribution of the SCC lesions and the co-existence of other lesions in the brain are helpful for the differential diagnosis between RESLES and ischemic infarction of SCC. However,the mean maximum cross-sectional area and the ADC value show no obvious difference between these two diseases.

<p><b>OBJECTIVE</b>To investigate relation between displaced inferior ramus fractures and posterior pelvic ring injury.</p><p><b>METHODS</b>From August 2012 to August 2015, 51 patients of pubic ramus fractures with complete record were retrospective reviewed including 27 males and 24 females with an average age of(49.1±19.0) years old ranging from 9 to 90 years old. The time from injury to treatment ranged from 0.3 to 48 hours with an average of 10.1 hours. According to Tile classification of pelvic fractures, 28 cases were type A, 17 cases were type B, 6 cases were type C. Pelvic radiographs and computed tomography scans were detailed and evaluated for whether there were posterior pelvic ring injury, meanwhile pubic rami fractures were divided into 4 groups as follow: displaced inferior ramus fractures group, undisplaced inferior ramus fractures group, displaced superior ramus fractures group, undisplaced superior ramus fractures group;the incidence rate of association of posterior pelvic ring injury was determined and compared.</p><p><b>RESULTS</b>Twenty-six patients had displaced inferior ramus fractures, all of them (100%) were combined with posterior pelvic ring injury. Twenty patients had undisplaced inferior ramus fractures, 6 of them(30%)were combined with posterior pelvic ring injury. Twenty-eight patients had displaced superior ramus fractures, 22 of them(78.5%) were combined with posterior pelvic ring injury. Twelve patients had displaced superior ramus fractures, 5 of them(41.6%) were combined with posterior pelvic ring injury. Compared with undisplaced inferior ramus fractures group, there was statistic difference(=0.028 8<0.05) on the incidence rate of posterior pelvic ring injury, there were no statistic difference(=0.055 8>0.05;=0.168 3>0.05) while compared with other undisplaced superior ramus fractures group and displaced superior ramus fractures group, but the incidence rate of association with posterior pelvic ring injury much higher than both of two groups (100% vs 41.6%, 78.5%).</p><p><b>CONCLUSIONS</b>Displaced inferior pubic ramus fractures have the highest incidence rate of association with posterior pelvic ring injury, frequently prompted injury to the posterior pelvis. Displaced inferior ramus fractures were an indirect evidence of posterior pelvic injury.</p>

Objective To explore the effect and potential mechanism of suppressors of cytokine signaling 3 (SOCS3) knockout on cell viability and apoptosis of retinal ganglion cells (RGCs) after optic nerve injury.Methods The optic nerve transection was used to construct optic nerve injury model of rats,and RGCs were isolated after optic nerve injury.The experimental animals were divided into optic nerve transection injury (ONT) group and sham-operation (Sham) group.The expression of SOCS3 in RGCs was detected by Western blot and RT-PCR in each group.Subsequently,SOCS3 siRNA was transfected into RGCs of Sham group and ONT group,and the experimental were further subdivided into blank control group,negative control group and SOCS3 silence groups.Cell viability was measured by CCK8 and MTr methods.Apoptosis was detected by Hoechst 33342 staining and flow cytometry.Furthermore,the mTOR siRNA and SOCS3 siRNA were co-transfected into RGCs,and cell viability and apoptosis were detected.Results The expression of SOCS3 was dramatically increased at 3 days after injury in the ONT group when compared with Sham group (P =0.049),and it showed an increased tendency gradually along with the extension of injury time.Compared with the blank control in the ONT group,SOCS3 silence markedly promoted cell viability [(49.47 ± 7.35) % vs.(73.24 ± 8.70) %],reduced cell growth inhibition [(27.25 ±0.75)% vs.(10.96 ± 1.07)%] and apoptosis [(23.06 ± 1.43)% vs.(10.65 ± 1.77)%].The result of Hoechst 33342 staining indicated that SOCS3 silence ameliorated the cell apoptosis induced by ONT.In addition,SOCS3 silence significantly improved pS6 expression at 2 weeks after injury,and mTOR and SOCS3 co-silence reduced cell viability,increased cell growth inhibition and apoptosis compared with SOCS3 silence group after injury.Conclusion SOCS3 silence promotes injury-induced cell viability of RGCs and suppresses injury-induced apoptosis of RGCs via up-regulating mTOR activity in the later period of injury.

Background Studies showed that microRNA (miR)-375 suppresses the growth,apoptosis,migration and adhesion of tumor cells,and it plays a regulation to the changes of vascular endothelial growth factor (VEGF) in tumor tissue to arrest neovascularization.However,whether miR-375 intervenes the formation of new blood vessel in eyes is unelucidated.Objective This study was to explore the effects of miR-375 on human retinal capillary endothelial cells (HRCECs) function induced by hypoxia.Methods HRCECs were cultured using IMDM and divided into normal control group,CoCl2 model group,CoCl2 +miR-375 mimic group,CoCl2 +miR-375 mimic control group,CoCl2+miR-375 inhibitor group and CoCl2 +miR-375 inhibitor control group,and hypoxia cell models were created by adding 200 μmol/L CoCl2.MiR-375 and frizzled 4 (FZD4) small interfering RNA (siRNA) were transfected into the cells by 50 nmol/L miRNA lipidosome for 48 hours.The proliferation of the cells was detected by MTT assay;migrated number of the cells was examined by Transwell chamber assay;ELISA was employed to detect the concentrations of VEGF and VE-cadherin in the medium;the expression of β-catenin,cyclinD1,matrix metalloproteinase-2 (MMP2) and VEGF proteins were analyzed by Western blot;tube length of vessel formation was evaluated by Matrigel assay.Cultured cells were divided into normal control group,CoCl2 model group,CoCl2 +mock group and CoCl2 + FZD4 siRNA group,the relative expression of FZD4,a miR-375 targeted gene,was detected by luciferase reporter.Results The relative expression of miR-375 mRNA was significantly increased in the CoCl2 +miR-375 mimic group compared with the CoCl2 + miR-375 mimic control group and reduced in the CoCo2 + miR-375 inhibitor group compared with the CoCo2 + miR-375 inhibitor control group (t =-19.237,8.764,both at P<0.01),with a higher transfected efficacy for miR-375.The cell proliferative fold,migrated cell number,VEGF and VE-Cadherin contents in the medium and the tube length were significantly different among the CoCl2 model group,CoCl2 +miR-375 mimic group,CoCl2 +miR-375 mimic control group,CoCl2 +miR-375 inhibitor group and CoCl2 +miR-375 inhibitor control group (F =24.324,26.776,14.113,19.225,15.040,all at P<0.001),and those in the CoCl2 +miR-375 mimic group were evidently reduced in the CoCl2 +miR-375 mimic group compared with the CoCl2 +miR-375 mimic control group,while those in the CoCl2 +miR-375 inhibitor group were considerably elevated in comparison with the CoCl2 +miR-375 inhibitor control group (all at P<0.01).The expressions of β3-catenin,cyclinD1,MMP2 and VEGF protein were significantly different among the normal control group,CoCl2 model group,CoCl2 +miR-375 mimic group,CoCl2+miR-375 mimic control group,CoCl2 +miR-375 inhibitor group and CoCl2 +miR-375 inhibitor control group (F=11.753,13.283,16.770,10.334,all at P<0.001).In addition,the cell proliferative fold,migrated cell number and the tube length were significantly increased in the CoCl2 model group and CoCl2+mock group,and those in the CoCl2+FZD4 siRNA group were decreased in comparison with the CoCl2 +mock group (all at P<0.05).Conclusions MiR-375 inhibits the growth,migration and tube formation ability of HRCECs in hypoxic status probably by regulating the activation of Wnt pathway via directly targeting FZD4.

RESUMO

Objective To develop a portable hanging perfusion support used in the medical tent of field medical unit.Methods The support was composed of a support body,a hanging hook at one end of the body,hanging components and an automatic locking mechanism.The hanging components included two screw hangers and two inverted cone-shaped sleeves.The support body consisted of three stainless steel tubes with different sizes,whose height could be regulated through connecting thread to adapt the support to hanging the dropping bottle.Results The support gained advantages over the traditional one in convenience as well as deployment and withdrawal time,and enhanced medical training efficacy and casualty treatment timeliness greatly.Conclusion The support behaves well in structure,operation and portability,and can be used for patient treatment in battlefield,disaster relief and etc.

Objection To improve the central oxygen supply system at field conditions to realize rapid transport of oxygen cylinders and enhanced application of medical oxygen.Methods The modified central oxygen supply system was composed of the 40-L oxygen cylinder,triple ball valve,flowmeter,dichotomantheshos hose,disposable nasal catheter for oxygen inhalation,pressure relief valve,immovable hanger and L-shaped trolley for oxygen cylinder.The trolley was composed of a bearing frame designed according to the size of oxygen cylinder,a fixing band and two wheels,which could be used for efficient fixation and rapid transport of oxygen cylinder.Results The improved system met the requirements for materials storage,loading,deployment and withdrawal during oxygen supply,and saved manpower and space.Conclusion The improved system enhances the efficiency of medical oxygen application and the supportability of field medical unit,and thus is worthy promoting in mobile medical unit.
