RESUMO

Objective To explore the medication for lung cancer treatment at Hubei Cancer Hospital. Methods The electronic cases of 3 234 hospitalized lung cancer patients in Hubei Cancer Hospital were collected. The medication of the Peiyuan Guben method in treating lung cancer was analyzed with cluster and association analyses by the Apriori association rule algorithm in IBM SPSS modeler 18.0 and Qihuang data AI workstation software system. Results In the 11 293 pieces of traditional Chinese medicine used in 3 234 patients with lung cancer, the core prescription of 16 core drugs were Astragalus, Coix seed, Poria, Coke hawthorn, Ligustrum lucidum, Wolfberry, Bran fried atractylodes, Mushroom stem, Bran fried citrus husk, Bittersweet herb, Centipede, Rhodiola, Semiaquilegia adoxoides, Seaweed, Prunella and Pseudobulbus cremastrae seu pleiones. Conclusion "Peiyuan Guben" is an effective method of traditional Chinese medicine in treating lung cancer. Traditional Chinese medicine with the effects of Yiqi Jianpi and Bushen Yijing has an evident curative effect in treating lung cancer and can improve patients' quality of life.

In this study,a fluorescent(FL)aptasensor was developed for on-site detection of live Salmonella typhimurium(S.T.)and Vibrio parahaemolyticus(V.P.).Complementary DNA(cDNA)of aptamer(Apt)-functionalized multicolor polyhedral oligomeric silsesquioxane-perovskite quantum dots(cDNA-POSS-PQDs)were used as encoded probes and combined with dual-stirring-bar-assisted signal amplification for pathogen quantification.In this system,bar 1 was labeled with the S.T.and V.P.Apts,and then bar 2 was functionalized with cDNA-POSS-PQDs.When S.T.and V.P.were introduced,pathogen-Apt complexes would form and be released into the supernatant from bar 1.Under agitation,the two complexes reached bar 2 and subsequently reacted with cDNA-POSS-PQDs,which were immobilized on MXene.Then,the encoded probes would be detached from bar 2 to generate FL signals in the supernatant.Notably,the pathogens can resume their free state and initiate next cycle.They swim between the two bars,and the FL signals can be gradually enhanced to maximum after several cycles.The FL signals from released encoded probes can be used to detect the analytes.In particular,live pathogens can be distinguished from dead ones by using an assay.The detection limits and linear range for S.T.and V.P.were 30 and 10 CFU/mL and 102-106 CFU/mL,respectively.Therefore,this assay has broad application potential for simultaneous on-site detection of various live pathogenic bacteria in water.

Objective@#To investigate the effect of magnetic-induced cell targeted transplantation (MagIC-TT) on repair of bone defects in mice using therapeutic fluorescent gene labeled cells into bone tissue and its mechanism.@*Methods@#The proliferation, apoptosis and targeted migration ability were compared between magnetized and unmagnetized murine bone marrow stromal cells labeled with green fluorescent protein (GFP-BMSCs) (n=3). GFP-BMSCs were loaded into tissue engineering bone (TEB) by MagIC-TT in the experimental group (n=5) before the TEB was transplanted into the large femur defects in the model of red fluorescent protein (RFP) transgenic mice. In the control group (n=5) TEB was not loaded with GFP-BMSCs while in the blank group (n=3) the large femur defects were only fixated with intramedullary nails. The effects and mechanism of bone repair were explored 3 months after surgery using X-ray, micro-CT, semi-solid decalcification (SSD) and histology, respectively@*Results@#There were no significant differences between magnetized and non-magnetized GFP-BMSCs in proliferation (0.760±0.029 versus 0.733±0.033) or in survival rate (87.9%±1.0% versus 87.4%±2.0%) (P>0.05), but the mobility of magnetized GFP-BMSCs was significantly higher than that of non-magnetized GFP-BMSCs (P<0.05). The X-ray 3 months after surgery showed that the scaffolds in the experimental group were degraded and that the proximal and distal ends of the femoral defects were connected by new bone tissue. No new bone formation was found in the blank group while a small amount of bone formation was observed in the control group. The Micro-CT showed that stable new bone tissue formed in the femur defects after removal of intramedullary nails in the experimental group. The SSD showed that GFP-MSCs were densely distributed in the scaffolds with red fluorescent protein (RFP) recipient cells penetrating them, indicating involvement of both donor and recipient cells in the formation of new bone.@*Conclusions@#MagIC-TT can be used to promote introduction of therapeutic cells into bone tissue to achieve a fine effect on repairing bone defects. Dual fluorescence gene marking combined with SSD shows that both donor and recipient cells may take part in the bone repairing.

OBJECTIVE@#The aim of this study was to identify the differences in microbial diversity and community in patients with salivary adenoid cystic carcinoma (SACC).@*METHODS@#Saliva was collected from 13 patients with SACC confirmed by histopathological diagnosis and 10 healthy control subjects. Total metagenomic DNA was extracted. The DNA amplicons of the V3-V4 hypervariable regions of the 16S rRNA gene were generated and subjected to high-throughput sequencing. Microbial diversity and community structure were analyzed with Mothur software.@*RESULTS@#A total of 16 genera of dominant bacteria in the SACC group were found, including Streptococcus (36.68%), Neisseria (8.55%), Prevotella_7 (7.53%), and Veillonella (6.37%), whereas 15 dominant bacteria in the control group were found, including Streptococcus (18.41%), Neisseria (18.20%), Prevotella_7 (8.89%), Porphyromonas (6.20%), Fusobacterium (5.86%) and Veillonella (5.82%). The statistically different phyla between the two groups were Firmicutes, Proteobacteria and Fusobacterium (P<0.05). The statistically different genera between the two groups were Streptococcus, Neisseria and Porphyromonas (P<0.05), and Capnocytophaga was only detected in patients with SACC.@*CONCLUSIONS@#Significant differences were observed in the oral microorganisms between the two groups.

AIM To observe the effects of Tangzhi Kangping Granules (Astragali Radix,Ligustri lucidi Fructus,Schisandrae chinensis Fructus,etc.) on regulating blood sugar and lipid of rats with glucose and lipid metabolism disorders.METHODS Among the seventy rats for the trial,ten rats were randomly assigned to normal control group,the other 60 rats were injected with STZ citric acid-sodium citrate solution (60 mg/kg) combined with high-fat emulsion (10 mL/kg) for 4 weeks to be the models of glucose and lipid metabolism disorders.The model rats were divided into model control group,mefformin + fenofibrate group,Xiaoke Pills + Xuezhikang group,and high,medium and low dose Tangzhi Kangping Granules groups.After 4-week intragastric administration,the rats had their levels of fasting serum glucose (GLU),triglyceride (TG),total cholesterol (TC),high density lipoprotein cholesterol (HDL-C),low density lipids (LDL-C),insulin (INS),adiponectin (ADP),leptin (Leptin),interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α) measured.RESULTS Compared with the model control group,the rats in high,medium and low dose Tangzhi Kangping Granules groups displayed significantly decreased serum contents of GLU,TG and TC,lowered levels of LDL-C,ADP,Leptin,IL-6,TNF-α,and yet significantly increased levels of HDL-C and INS.CONCLUSION Tangzhi Kangping Granules,a blood sugar andlipid regulator may adjust the levels of insulin,adiponectin and leptin,and therefore improves the body's inflammatory environment to alleviate insulin resistance.

Objective To explore the difference of gene expression profiling between normal basilar arteries and basilar arteries of cerebral vasospasm (CVS) after subarachnoid hemorrhage (SAH) in rabbits. Methods cDNA chip of normal basilar arteries and basilar arteries of CVS after SAH in rabbits were downloaded from GEO database. The chip was analyzed and screened by Bioconductor software, and function enrichment and pathway analysis of the differentially expressed genes were analyzed by Cytoscape software. Then 6 adult male Japanese rabbits were used, and randomly divided into normal control group (n=3) and SAH model group (n=3). Rabbit SAH models were established by cisterna secondary-blood-injection method. RNA data of normal basilar artery specimens on the 0 day and basilar artery specimens after SAH on the 5-day were used to validate the parts of differentially expressed genes by qRT-PCR. Results A total of 4356 differentially expressed genes were found in normal basilar arteries and basilar arteries of CVS after SAH in rabbits. Among them, 920 genes were considered to be significant with P-value<0.05, such as GRIK1, MYH13, ZNF45, SAA3, RLN1, MSR1 and others. Function enrichment analysis indicated that the differentially expressed genes were involved in regulation of Ca2+transmembrane transporter activity, negative regulation of ion transmembrane transport, regulation of potassium ion transport, positive regulation of JAK-STAT signaling cascades and other biological processes. Pathway analysis showed that calcium signaling pathway, cGMP-PKG signaling pathway, HIF-1 signaling pathway, PI3K-Akt signaling pathway and other signaling pathways maybe related with the differentially expressed genes. qRT-PCR verification showed that the expression of MSR1 in SAH model group was consistent with that of the chip result. Conclusion The gene expressions of basilar arteries of CVS after SAH in rabbits are significantly different, and MSR1 gene can be used as a potential target for studying the pathological mechanism of CVS.

An ultrasensitive immunoassay was developed based on As3+ and Hg2+ labeled SiO2 @ Au nanoparticles signal tags and hydride generation-atomic fluorescence spectrometry (HG-AFS) for the detection of carcinoembryonic antigen(CEA) and carbohydrate antigen 19-9 (CA 19-9) respectively. Firstly, amino SiO2@ Au NPs were synthesized for selective absorption of As3+ and Hg2+ ions respectively. Subsequently,the secondary antibody (Ab2) of CEA and CA 19-9 was respectively labeled on As3+ or Hg2+-SiO2 @ Au NPs to prepare the corresponding signal tags for CEA and CA 19-9. Based on the sandwich immunoassay scheme, the tags, two antigen and corresponding first antibodies were bio-conjugated on the bottom of 96-well plate at room temperature to form the immunocomplex. After it was dissolved in alkali solution, As3+ and Hg2+ ions were released in solution and detected by HG-AFS, which concentration was proportional with logarithms of CEA and CA 19-9. The reaction conditions were optimized and the tags were characterized. This assay was based on determination of the concentration of As3+ and Hg2+ for quantization of the corresponding CEA and CA 19-9 antigen. The assay showed a wide linear range from 0. 001 to 100. 0 μg / L for CEA and 0. 01-80 U/ mL for CA 19-9, and a lower detection limit of 0. 5 ng / L and 0. 005 U/ mL respectively. This proposed method was used in real serums samples, the results were consistence with that by ELISA. The immunoassay showed three orders of magnitude of sensitivity lower than that of ELISA, which provides a promising simultaneous immunoassay for the early diagnosis of cancer .

Objective To investigate the change of the serum matrix metalloproteinase-13 (MMP-13) and tissue inhibitor of matrix metalloproteinase-1 (TIMP-1) and the correlations of MMP-13 and TIMP-1 with bone metabolic markers or bone mineral density(BMD) in aged postmenopausal Chinese women.Methods One hundred and twenty women aged 48-65 years with postmenopausal were selected as subjects.They were divided into normal group(n =28),low bone mass group (n =36) and osteoporosis group (n =56) accordance with the WHO standard.The levels of serum MMP-13,TIMP-1,estradiol (E2),Osteoprotegerin (OPG),Osteoprotegerin ligand (OPGL),Procollagen terminal propeptide of N (PINP),and Type Ⅰ collagen cross-linked C terminal peptide(CTX) were measured using enzyme linked immunosorbent assay(ELISA).And the ratios of MMP-13 to TIMP-1 were calculated.Bone mineral density(BMD) of the lumbar spine,femoral neck,Fahrenheit and greater trochanter were measured using dual energy X-ray absorptiometry.Results There was significant difference of serum MMP-13 concentrations in normal controls,low bone mass group and osteoporosis group((27.08 ± 1.41) μg/L,(45.64 ± 1.62) μg/L and (44.25 ± 1.21) μg/L; F =110.314,P =000),and serum MMP-13 concentrations in low bone mass group and osteoporosis group higher than that in normal controls (P < 0.05),serum MMP-13 concentrations in low bone mass group higher than that in osteoporosis group,but there was on significant difference(P > 0.05).But Serum TIMP-1 concentrations in three groups was no significant difference (F =10.721,P =0.801).The ratio between MMP-13 and IIMP-1 was 0.185 ± 0.062,0.311 ± 0.053,0.332 ± 0.063 respectively,and there was significant difference of the three groups (F =137.771,P =0.000),and the ratio of the low bone mass group and osteoporosis group higher than the normal group(P <0.05),but there was no significant difference between the two groups.In osteoporosis group,Negative correlations between MMP-13,and BMD,E2 and OPGL,were found (r =-0.296,-0.198,-0.301,-0.298,-0.233 respectively,P < 0.05).Meanwhile positive relations were found between MMP-13 and OPG,PINP,CTX (r =0.228,0.315,0.312 respectively,P < 0.05).In low bone mass group,the level of MMP-13 was significantly relative with BMD (the lumbar spine,Fahrenheit),E2 and CTX (r =-0.188,-0.196,-0.235,0.289 respectively,P < 0.05).Conclusion There are significant correlations between serum MMP-13,ratios of MMP-13 and TIMP-1 and bone metabolism marker and BMD.MMP-13 level may increase with increases in osteoporosis and osteopenia.The increases of serum MMP-13 and the decrease of ratio of MMP-13 to TIMP-1 might be biomarkers in high bone turnover state,such as postmenopausal osteoporosis and early stage of osteopenia.

Objective To apply team based learning (TBL) in the teaching of dental endodontics for undergraduates in order to expand and deepen the TBL teaching in stomatology education.Methods TBL was used among 2008 grade oral undergraduates in Chongqing Medical University.Average test scores of 2008 grade undergraduates before and intra class were calculated and questionnaire was designed.At the same time,final examination scores between 2007 grade and 2008 grade under-graduates were compared.Results There was no significant correlation in average test scores before and intra class (r =0.027,P > 0.05).Final examination scores were higher in 2008 grade than in 2007 grade.Based on the questionnaire survey,learning interests,sense of teamwork and classroom knowledge grasp of 2008 grade undergraduates were obviously elevated.Conclusions TBL teaching significantly improve students' learning effect and it can be promoted in stomotology education.

<p><b>OBJECTIVE</b>To evaluate accuracy of Propex, Raypex 5, Root ZX electronic apex locator in positions of the perforation with different irrigations in the root canal.</p><p><b>METHODS</b>Perforation lengths were measured with Propex, Raypex 5, Root ZX electronic apex locators in 19 extracted human teeth embedded in model after simulate perforation preparation by ultrasonic ET40 and compared with the actual canal length measurements taken before embedding the teeth in model. Measurements were taken with the different canal contents. RESULTS; Propex, Raypex 5, and Root ZX could locate the positions of the perforation with different irrigations in the root canal. The differences between actual root canal perforation length and measured root canal perforation length of same electronic apex locators in different irrigations were no statistically significant (P > 0.05). Meanwhile, the differences between actual root canal perforation length and measured root canal perforation length of three kinds of electronic apex locators in same irrigations were not statistically significant (P > 0.05).</p><p><b>CONCLUSION</b>Propex, Raypex 5, and Root ZX electronic apex locators can detect perforation accurately.</p>

<p><b>OBJECTIVE</b>To investigate the chemical constituents of Desmodium blandum and their cytotoxic activity against the growth of several tumor cells.</p><p><b>METHOD</b>Various chromatographic techniques including silica gel, Sephadex LH-20 column chromatography were employed for the isolation and purification of the constituents. The structures of compounds were elucidated by spectral analyses (IR, UV, NMR, MS). Their cytotoxic activity was then studied.</p><p><b>RESULT</b>Eight compounds were isolated from the stems of D. blandum and identified as N, N-dimethyltryptamine (1), 5-methoxy-N, N-dimethyltryptamine (2), citrusinol (3), yukovanol (4), (Z)-1-(4-hydroxy-2, 3-dimethoxyphenyl)-3-(4-hydroxyphenyl) propene (5), (Z)-1-(3-hydroxy-2, 4-dimethoxy-phenyl)-3-(4-hydroxy-3-methoxy-phenyl) propene (6), methylprotocatechuate (7), katuranin (8).</p><p><b>CONCLUSION</b>Among these compounds, compound 6 was isolated from D. blandum for the first time. In the MTT test, compounds 2 and 6 exhibit cytotoxic activities against the KB cell, and compounds 3 and 6 exhibit the same activities against the HepG2 cell.</p>

RESUMO

Objective To identify the key factors triggering medical disputes induced by differences in cognition between doctors and patients in order to control the occurrence and development of medical disputes. Methods From 2002 to 2006,the outpatients,inpatients and medical staff of 3 hospitals in Shanghai(class 3 first level general hospital,n=2;class 3 first level special hospital,n=1) were investigated,and random sampling method was employed for the research analysis. Results There were significant differences between doctors and patients in cognition of influential factors triggering medical disputes such as health law,medical services,rights and obligations. Conclusion The differences in cognition between doctors and patients are important causes for medical disputes,and effective prevention and intervention measures must be taken.

OBJECTIVE: Researches on total hip replacements are always the hot spots in recent years. The replacements have obtained satisfactory outcomes in clinic. However, the long-term effect is limited because of the material and technical problems. This paper is to summarize the developmental trend and the prospect of clinical application for total hip replacements in 10 years.DATA SOURCES: A computer-based online search of Medline database was undertaken to identify relevant articles published in English from January 1996 to January 2006 by using the keywords of "total hip replacements, surgical robot, tissue engineering, virtual endoscopy". Meanwhile,Chinese relevant articles published from January 1990 to December 2005 were searched in Wanfang database, the keywords were "total hip replacements, material, virtual endoscopy, surgical robot, tissue engineering" in Chinese.STUDY SELECTION: The articles were primarily checked, and the references in each literature were reviewed. Articles about the study on total hip replacements were involved; Meanwhile, the repetitive studies or Meta analysis were excluded.DATA EXTRACTION: Totally 39 relevant articles were collected, and 22 of them accorded with the. Inclusive criteria, 17 old or repetitive ones were excluded. Of the 22 articles, 5 were about the influence of artificial joint material on prosthetic loosening and its carcinogenesis; 2 about the microinvasion of joint replacement induced by computer-aided surgery navigation; 1 about the introduction of surgical robot; 3 about the development of custom-made and specially made artificial joint; 9 about the possibility for the surface replacement of hip; and 2 about the combination of tissue engineering technique and artificial joint.DATA SYNTHESIS: Modern researches on total hip replacements are exploring wearing tolerant prosthesis to reduce the prosthetic loosening caused by osteolysis and bone resorption. The computer-aided design/made technique is used for special design and manufacture according to the need of disease, so as to solve the fracture around prosthesis and meet the need of limb salvage surgery. The computer-aided surgery navigation realizes the microinvasion of joint replacement and the fast recovery after surgery. The successful rate of total hip replacements is improved by developing localized surgical robot in China. By using the virtual endoscopy, we can accurately know the damaged conditions of intraarticular cartilage and sub-cartilage bone, which are also able to be quantifized, so that more suitable and reliable prosthesis can be selected to increase the accuracy. The tissue engineering technique in repairing defect can increase the synostosis of prosthesis, and perfect the technique of joint replacement.CONCLUSION: In the coming 10 years, total hip replacements will develop towards stability, harmlessness, microinvasion, mechanization, individualization and diversification, so that the total hip replacements will be much stander and safer with higher successful rate.

A simple and reliable electroanalytical method for the fast determination of aluminum in natural and drinking waters by a. c. oscillopolarography using solochrome violet RS(SVRS) is described. The alkaline buffer solution used was 0.85 mol/L NH3·H2O- NH4 Cl(pH 8.8) containing 5 × 10-5 mol/L SVRS. A sensitive incision due to the redox reaction of Al-SVRS complex adsorption wave on the Hg-film electrode was observed on the dE/dtE oscillogram at - 1.05 V. The incision depth was linearly proportional to the A1 concentration in the range of 1 ×10-7 ~6 × 10-6 mol/L. No serious interference was found. The detection limit of this method was 5 × 10-8 mol/L,and the relative standard deviation was 5.5% for 2 × 10-7 mol/L Al ( n = 10). This method was successfully applied to the determination Al in 22 real samples. The results were found to be in good agreement with those obtained by inductively coupled plasma-atomic emission spectrometry.

RESUMO

Objective:To evaluate the accuracy and influence factors of a triple-united method used for the determination of root canal working length.Methods:Root canal working length was detemined by manual measurement,tooth average length and radiographic measurement(triple-united method).In vitro experiment was conducted in 40 teeth and clinical study in 96 teeth by experienced specialists and postgraduates.Results:The accuracy of triple-united method was 82.5% and 62.5% by spcialist and poststudents in vitro, 87.1% and 75.5% in clinic,respectively. The accuracy was interfered by experience of the examiner, tooth integrity and apical disease.Conclusion:Triple-united method can meet the clinic requirement in the determination of root canal length.

RESUMO

Objective To investigate the chemical constituents in Desmodium blandum and their antitumor bioactivity.Methods Various chromatographic techniques were employed for the isolation and purification of the constituents by butanol at 50 ℃ including silica gel, Sephadex LH-20.The compounds structures were elucidated by spectral analyses(IR,UV,NMR,and MS).Then their cytotoxic activity was studied.Results They were identified as N,N-dimethyltryptamine (Ⅰ),5-methoxy-N,N-dimethyltryptamine(Ⅱ),citrusinol(Ⅲ),yukovanol(Ⅳ),(Z)-1-(4-hydroxy-2,3-dimethoxyphenyl)-3-(4-hydroxyphenyl) propene(Ⅴ),(Z)-1-(3-hydroxy-2,4-dimethoxy phenyl)-3-(4-hydroxy-3-methoxy-phenyl) propene(Ⅵ),methyl protocatechuate(Ⅶ),katuranin(Ⅷ).Conclusion Eight compounds are isolated from the stems of D.blandum for the first time.In the MTT antitumor experiments,compounds Ⅰ and Ⅲ—Ⅵ have the cytotoxic activity to KB cell.

RESUMO

Objective: The method for separation by capillary zone electrophoresis (CZE) and electrochemical determination of organic acids in fruit juices was developed. Method: In this system, 0.2mmol/L cetylpyridinium bromide (CPB) was used as an electro osmotic flow (EOF) modifier to reverse the direction of EOF. cyclodexfrin ( -CD) was added into running buffer to improve the separation efficiency. Then, the optimal separation conditions were achieved and successfully employed to separate six organic acids, including oxalic, malic, tartaric, succinic, fumaric acid and citric acids. One nano palladium modified carbon paste electrode was used for determination. Results: The calibration curves for all organic acids studied were linear with 2-3 orders (1 10–6-1.0 10–3 mol/L) of magnitude and all the detection limits (S/N=3) were below 2 mol/L. The RSD was between 1.05%-2.15% , and recovery was 93.4% . Conclusion: CZE separation combined with electrochemical detection is suitable for simultaneous determination of organic acids in fruit juices quickly with high sensitivity and selectivity.
