RESUMO

AIM: To investigate the effects of 0.01% atropine eye drops on macular blood flow density and retinal thickness in children with different degrees of myopia. METHODS: This was a prospective case-control study. Sixty-four patients (112 eyes) diagnosed with myopia for the first time with 0.01% atropine eye drops before and 6 months after medication were investigated with the uncorrected distance visual acuity (UCVA), axial length (AL), spherical equivalent (SE), macular ganglion cell-inner plexiform layer thicknes (mGCIPL) using slit lamp examination and optical coherence tomography (OCT), vascular density in the macular area and the area of the avascular in the fovea using optical coherence tomography angiography (OCTA) . Changes in various indicators before and after medication were compared. RESULTS: Compared with before medication, the AL of the three groups of myopia patients increased significantly (P0.05). The difference was statistically significant between the moderate myopia group and the high myopia group (P0.05). After 6 months of medication, the central circle macular vessel density (cCVD) increased in the low myopia group and moderate myopia group (P0.05). Before and after medication, there was no significant difference in outer circle macular vessel density (oCVD), inner circle macular vessel density (iCVD), and whole circle macular vessel density (wCVD) among the three myopia groups (P>0.05). The increase in mGCIPL was statistically significant in the low myopia group (P0.05). There was no significant difference in foveal avascular zone (FAZ) among the three myopia groups before and after medication (P>0.05). There was no correlation between CVD, AL, and SE in the three myopia groups (P>0.01). There was a low correlation between CVD and mGCIPL in the low myopia group (r=0.442, P0.01). CONCLUSION: 0.01% atropine can significantly reduce the rate of axial and refractive growth in children with low to moderate myopia, increase the density of central macular vessels, and increase the thickness of mGCIPL in children with low to moderate myopia.

The many-banded krait, Bungarus multicinctus, has been recorded as the animal resource of JinQianBaiHuaShe in the Chinese Pharmacopoeia. Characterization of its venoms classified chief phyla of modern animal neurotoxins. However, the evolutionary origin and diversification of its neurotoxins as well as biosynthesis of its active compounds remain largely unknown due to the lack of its high-quality genome. Here, we present the 1.58 Gbp genome of B. multicinctus assembled into 18 chromosomes with contig/scaffold N50 of 7.53 Mbp/149.8 Mbp. Major bungarotoxin-coding genes were clustered within genome by family and found to be associated with ancient local duplications. The truncation of glycosylphosphatidylinositol anchor in the 3'-terminal of a LY6E paralog released modern three-finger toxins (3FTxs) from membrane tethering before the Colubroidea divergence. Subsequent expansion and mutations diversified and recruited these 3FTxs. After the cobra/krait divergence, the modern unit-B of β-bungarotoxin emerged with an extra cysteine residue. A subsequent point substitution in unit-A enabled the β-bungarotoxin covalent linkage. The B. multicinctus gene expression, chromatin topological organization, and histone modification characteristics were featured by transcriptome, proteome, chromatin conformation capture sequencing, and ChIP-seq. The results highlighted that venom production was under a sophisticated regulation. Our findings provide new insights into snake neurotoxin research, meanwhile will facilitate antivenom development, toxin-driven drug discovery and the quality control of JinQianBaiHuaShe.

Objective To observe the effect of ultrafine herb moxibustion on nasal mucosal remodeling in rats with allergic rhinitis. Methods Forty SD rats were randomly divided into control group (Group D) and modeling group. When successfully induced by intraperitoneal injection, aerosol inhalation and nasal spray of ovalbumin, the modeling group were randomly subdivided into modeling group (Group A), fluticasone propionate group (Group B) and ultrafine herb moxibustion group (Group C). Comparisons were made between the groups in respect of HE staining, transmission electron microscopy and counts of EOS. Results (1) Compared with Group D, Group A had significant elevation in counts of EOS (P<0.01);mucosal epithelium swelling, apparent angiotelectasis and eosinophilic infiltrated by HE staining; incomplete mucosal surface with deformity cilia, swelling mitochondria and partial loss of the mitochondria ridge by TEM . ( 2 ) Counts of EOS in Groups B and C were significantly decreased than in Group A (P<0.05, and P<0.01). Those in Group C were significantly decreased than in Group B (P<0.05), and there was insignificant difference in the counts between Group C and Group D (P=0.539). The HE staining Group B showed mild telangiectasia, with EOS scattered. The HE staining in Group C showed epithelial minor swelling and no telangiectasia. TEM showed mucosal surfaces in Groups B and C were complete, the quantity of cilia and microvillus increased apparently and cell junctions were normal. Normal endoplasmic reticulum and mitochondria were evident in Group C. Conclusion The ultrafine herb moxibustion can relieve the clinical symptoms, slow down and even reverse the nasal mucosal remodeling.

Glycyrrhizin is the major bioactive compound in Glycyrrhiza uralensis. Farnesyl-diphosphate synthase (FPS) is one of the major enzymes involved in the glycyrrhizin biosynthetic. We cloned the full length of FPS coding sequence based on the transcriptome data of G. uralensis. The FPS gene of G. uralensis (GlyurFPS) is 1029 bp, coding 342 amino acid. The homology between the sequence of its protein and that of the FPS in Cicer arietinum, Medicago sativa, Medicago truncatula, Glycine max, Lotus japonicus was of 94%, 94%, 93%, 93% and 92%, respectively. Plant FPS gene is very conservative, therefore the phylogenetic tree is same with the plant classification consistent.

A total of 184 teeth obtained from 102 cases(27 males and 75 females aged 17-64 years)were selected from Department of Tooth and Dental Pulp.DalJan Stomatology Hospital from June to Septembet 2007.All the teeth with deep caries which resulted from caries showed no pulp.Ninety-two teeth obtained from 54 cases were regarded as Vitrebond group which were used by light-cured glass to cover pulp;while,the other 92 teeth obtained from 48 cases were regarded as dycal group which were cured by dycal calcium hydroxide.Finally,3MZ250 resin was used in this study.A half-year following up showed that the filling shed and loose were observed in the Vitrebond group(n=1)and the dycal group(n=3)(P>0.05).There were significant differenoe in color between Vitrebond group(n=1)and dycal group(n=8)(P<0.05),suggesting 3M Vitrebond light-cured glass Was superior to dycal calcium hydroxide in decreasing dental pulp stimulation and improving dental appearance.

AIM: Tongxinluo had showed effects of inducing proliferation and differentiation on neural stem cells in previous observations on animal models. The experiment would investigate the time-effect and dose-effect relationship of Tongxinluo on neurocyte lineage development from rat embryonic neural stem cells (NSCs). METHODS: Experiments were performed at the Laboratory of Functional Science of Southern Medical University from June to October 2007. ①Pregnant rats of 12-14 days were provided by Experimental Animal Center of Southern Medical University. The experiment was performed in strict compliance with all standards for ethics. Tongxinluo contained ginseng, leech, scorpion, ground beetle, centipede, pellicula cicadae, red peony root, borneol and so on. Tongxinluo was produced by Shijiazhuang Yiling Pharmaceutical Co., Ltd., No. Z19980015. Preparation of serum containing Tongxinluo: 1 g/kg per day Tongxinluo suspension was given by lavage in rats of high-dose Tongxinluo group, and 0.5 g/kg per day in rats of low-dose Tongxinluo group. 7 days later, sera were extracted from all the rats and sterilized with filtrating, followed by separate-loading and cryopreservation at –70 ℃. ②NSCs cultured were separated from 12th-14th day SD rat embryo. The third passage of cells was treated with serum containing Tongxinluo in the high-dose and low-dose Tongxinluo group. Rats were treated with common serum as a control. ③Proportion of various kinds of nerve cells was observed by immunofluorescence staining at days 1, 3 and 7. RESULTS: ①At day 1, there was few cells labeled with 5-bromodeoxyuridine (BrdU) (+) and glial fibrillary acidic protein (GFAP) (+) in high-dose Tongxinluo group, with rest cells all labeled with BrdU (+) and Nestin (+). While in low-dose Tongxinluo group and control group, all cells were BrdU(+)Nestin(+). ②At day 3, the ratios of cells labeled with Nestin (+), ?tubulin (+) or GFAP (+) differentiated with each other (P 0.05). ③The ratios of NSCs labeled with nestin (+) decreased soon after administered Tongxinluo, and increased after that. The ratios of ?tubulin (+) cells increased in the high-dose Tongxinluo group while increased firstly then decreased in the low-dose Tongxinluo group and control group. The ratios of GFAP (+) cells increased in the low-dose Tongxinluo group and control group while increased firstly then decreased in the high-dose Tongxinluo group. The ratios of GalC (+) cells had no remarkable change in the high-dose and low-dose Tongxinluo group, but increased significantly at day 7 in the control group. CONCLUSION: Tongxinluo may induce proliferation and neuronal differentiation of rat embryonic NSCs with time-effect and dose-effect relationship. The more Tongxinluo is administered, the more significant and permanent effects will be achieved.

BACKGROUND:It is suggested that glucose metabolism of frontal lobes,cingulate gyri and caudate nuclei in cerebral functional area of patients with depression is low,and the blood flow in cerebellum decreases.OBJECTIVE:To observe the effect of scalp acupuncture on cerebral glucose metabolism in depression by positron emission tomography.DESIGN:Randomly controlled and verifying experiment.SETTING:Department of Acupuncture and Aoxibustion in Nanfang Hospital and Center of Positron Emission Tomography in a Guangdong People's hospital.PARTICIPANTS:Twelve patients of primary depression,from Nanfang Hospital,Zhujiang Hospital and Center of Positron Emission Tomography in Guangdong People's Hospital from October 2002 to October 2003,were selected randomly with the diagnostic criteria of [F32] in the Third Edition of the Chinese Classification and Diagnostic Criteria of Mental Disease.Among them,3 were male and 9 were female.Their ages were from 36 to 66 years,with the average of 48.METHODS:Giving scalp acupuncture in middle line of vertex(MS5),middle line of forehead(MS1) and bilateral lateral line 1 of forehead(MS2).All the patients received positron emission tomography examination before and after acupuncture treatment for 6 weeks.Then cerebral Regions of Interest,bilateral frontal lobes,bilateral caudate nuclei,bilateral cingulate gyri and bilateral cerebella,were chosen and their average glucose metabolisms were counted by half-quantifying analysis.MAIN OUTCOME MEASURES:Glucose metabolism of bilateral frontal lobes,bilateral caudate nuclei,bilateral cingulate gyri and bilateral cerebella.RESULTS:According to intension,all 12 patients with primary depression sults of right frontal lobes(2.91 ±0.34),left frontal lobes(2.96±0.26),right cingulate gyri(2.68±0.20),right caudate nuclei (2.61±0.39) and left cerebellum(2.55±0.18)were obviously higher than those of right frontal lobes(2.53±0.31),left frontal lobes(2.53±0.36),right cingulate gyri(2.34±0.31),right caudate nuclei(2.40±0.33) and left cerebellum(2.35±0.25)before treatment(P< 0.05-0.01).However, the results of left cingulate gyri (2.69 ±0.38),left caudate nuclei(2.66±0.01) and right cerebellum(2.50±0.27) were similar to those of left cingulate gyri(2.43±0.38),left caudate nuclei(2.45±0.21) and right cerebellum(2.30±0.34) before treatment(P > 0.05).CONCLUSION:Scalp acupuncture on middle line of vertex,middle line of forehead and bilateral lateral line 1 of forehead could improve the glucose metabolism in depressive patients of certain cerebral regions,and it might be a mechanism related to improvement of frontal lobes,caudate nuclei,cingulate gyri and cerebellum.

OBJECTIVE:To investigate the recent use of immunosuppressive agents for renal transplantation patients during our hospital.METHODS:Case history of124renal transplantation patients in our hospital in2004was reviewed ret-rospectively,and the categories and the associated costs of immunosuppressive agents were statistically analyzed,which were compared with that of2000.RESULTS:In2004,patients’mean hospitalization cost was74462.77yuan and mean immuno-suppressive agents cost was25809.93yuan,increased by75.12%and175.35%,respectively as compared to2000.Of the total drug costs,the immunosuppressive agents accounted for61.37%in2004and46.4%in2000.Dominated the first3places in the list of DDDs of immunosuppressive agents in2004were Methylprednisolone,ciclosporin A and dehydrocortisone,respec-tively.CONCLUSIONS:As compared with2000,the application categories and costs of neotype immunosuppressive agents for renal transplantation patients in our hospital increased in2004,thus to find a more effective immunosuppressive remedy that of less adverse drug reaction and lower cost is of great significance.

Objective:To explore the effect of jiudaiqingyuan oral solution in the treatment of chronic hepatitis-B(CHB) and in the elimination of HBV when used alone and combined with lamivudine.Methods:108 CHB patients were randomly divided into three groups:treatment group with Jiudaiqingyuan Oral Solution (Jiudai group),Lamivudine treatment group (lamivudine group),and treatment group with the combination of Jiudaiqingyuan Oral Solution and lamivudine (combination group),with each group including 36 cases.Laboratory tests were done before treatment,3,6,9 and 12 months after treatment,and at the 3rd,6th and 12th month of follow-up.The laboratory tests consisted of ALT,HBV-M and HBV-DNA.Results:At the 12th month of treatment and 12th month of follow-up,cases with normal ALT conversion cases with negative HBV-DNA conversion,cases with negative HBeAg conversion,HBeAg/anti-HBe serum conversion rate,and total effective rate of the combination group were respectively 94.4%,86.1%,55.6%,52.8%,75.0% and 83.3%,72.2%,47.2%,47.2%,72.2%,which were markedly better than those of Jiudai group or lamivudine group,with significant statistc difference in each index (P

RESUMO

Gwe, an active ingridient derived from Chinese herb, possess profound effects on immune functions in mice in vivo and human peripheral lymphocytes (PBL)in vitro. The mitogenesis stim ulated with ConA or LPS, LAK cytotoxicity, spontaneous proliferation of thymocytes and prodution of IL-2 may all be increased markedly after treatment with Gwe in mice, but the CIC (circulating immune complexes) in sera decreased conpared to saline controls. The results shows that GWe influence preferentially on T-cell in a dose-dependent manner.

In this paper, it has been found that such stressors as operation or swimming in cold watermay suppress the mitogenic response of murine spleen cells to ConA or LPS, whichis closely related to the action of suppressor cells.Using the plastic-adherence method, the plastic-adherence cells in spleen(PAC)was removed or obtained. The difference of mitogenic responsivebefore and after the removal of PAC is very apparent in stressed mice, being much lower thannormal before the renioval of PAC and near to normal after removing PAC. It is interestingthat the lymphocyte transformation stimulated by LPS is suppressed predominantly after swim-ming stress instead of those by ConA in operation stress.It has also been observed that theindomethacin may block the induction of suppressor PAC by stress, which means that the pro-staglandins may play an important role in the induction and action of suppressor PAC.
