RESUMO

OBJECTIVE: To investigate the improvement effects of Qishenlian eczema cream on the atopic dermatitis (AD) model guinea pigs, and to investigate its possible mechanism. METHODS: Female guinea pigs were randomly divided into normal control group, model control group, matrix control group (Qishenlian eczema cream matrix, 1 g/kg), Qishenlian eczema cream low-dose, medium-dose and high-dose groups (0.5, 1, 2 g/kg) and Hydrocortisone butyrate cream group (0.5 g/kg), with 10 guinea pigs in each group. Using ovalbumin as antigen, the skin allergy of guinea pigs was stimulated to induce AD model. Forty-eight hours after the last excitation, guinea pigs were smeared with normal saline in normal control group and model control group, while those were smeared with Qishenlian eczema cream matrix in matrix control group, and administration groups were treated with relevant medicine, twice a day, for consecutive 14 d. The scores of erythema, edema and scratch before and after administration were recorded in each group. The morphological characteristics of skin were observed by HE staining. TUNEL method was used to detect the apoptosis of keratinocytes in skin tissue. The level of IFN-γ in skin lesions was detected by ELISA. RESULTS: Compared with normal control group, inflammatory cell infiltration, hyperkeratosis of epidermis and thickening of spinous layer were observed in skin tissue of guinea pigs in model control group and matrix control group; the scores of erythema, edema and scratch and their total score, the level of IFN-γ in skin tissue were increased significantly; the apoptosis number of keratinocytes was decreased significantly (P<0.01). Compared with model control group and matrix control group, above symptoms of medication groups were relieved to different extents; the scores of erythema, edema and scratch and their total score after medication in medication groups, the level of IFN-γ in skin tissue in Qishenlian eczema cream medium-dose and high-dose groups and Hydrocortisone butyrate cream group were decreased significantly, and above scores in medication groups were significantly lower than before medication (P<0.05 or P<0.01); the apoptosis number of keratinocytes was increased significantly in medication groups (P<0.05 or P<0.01). CONCLUSIONS: Qishenlian eczema cream has certain improvement effect on AD model guinea pigs, the mechanism of which may be associated with promoting the keratinocyte apoptosis, reducing the level of INF-γ in skin tissue.

Objective@#To explore the effects of a complex made from lily,glutinous rice,black bean,red jujube,whey protein powder and spirulina on tumor growth and immune function in C57BL/6 tumor-bearing mice,so as to provide evidence for nutrition intervention on tumor sufferers. @*Methods@#Twenty-four C57BL/6 male tumor-bearing mice were divided into three dose groups and a control group. The mice in the low,medium and high dose group was fed with 8 g/kgbw,20 g/kgbw and 40 g/kgbw complex,respectively;the mice in the control group was fed with 0.5% CMC-Na. All mice were intragastrically administered for seven days,and the weight gain,tumor volume,tumor weight,tumor inhibition rate,thymus index,spleen index,T lymphocyte proliferation,NK cell killing rate and interleukin-2 expression were measured. @*Results@#The tumor weights of the mice in the three dose groups wereall less than that in the control group(P<0.05). The numbers of CD3+ T cells in the three dose groups were more than that in the control group(P<0.05). The tumor inhibition rates of the low,medium and high dose groupwere 32.46%,45.14% and 48.03%,respectively. The optical density,NK cell killing rates,IL-2 expression of spleen cells and the number of T lymphocyte subsets were dose-dependent(P<0.05). Compared with the control group,the mice in the medium and high dose group had higher thymus indexes,NK cell killing rates,IL-2 expression of spleen cells,CD4+T cell levels and CD4+ to CD8+ ratios,but lower tumor volumes and CD8+ T cell levels(P<0.05);the mice in the high dose group had higher optical density(P<0.05).@*Conclusion@#This nutritional complex may play an anti-tumor role by regulating the immune function of tumor-bearing mice.

RESUMO

Objective To investigate the effect of geraniin on expression of Wnt3a protein and mRNA in bone marrow stromal cell (BMSC) from osteoporotic rats. Methods The model of osteoporosis (OP) was duplicated by ovariectomy in rats. BMSCs were isolated and cultured. BMSCs from shamed rats were routinly cultured and taken as normal control, and BMSCs from OP rats were divided into model group, 1μmol/L simvastatin positive group, and geraniin group (0.01, 0.1, 1, 10, 100 μmol/L), respectively. The methods of realtime-PCR and western blot were used to assay the protein and mRNA expression of wnt3a, respectively.Results As compared with normal control group, the protein and mRNA expression of wnt3a in model group were significantly suppressed;Compared with model group, 1 μmol/L simvastatin, and 0.1, 1, 10 and 100 μmol/L geraniin significantly increased the expression of wnt3a protein and mRNA. Conclusion It is suggested that geraniin activates wnt/β-catenin pathway though increasing the expression of signaling protein wnt 3a in BMSCs from OP rats. It would be beneficial to osteogenic differentiation of BMSCs and osteogenesis.

BACKGROUND:Human-mammal chimeric liver chimera has been a vital significance for the proliferation and differentiation of bone marrow mesenchymal stem cells. OBJECTIVE:To establish an animal model of human-rhesus chimeric liver using adult bone marrow mesenchymal stem cells. METHODS:Adult bone marrow mesenchymal stem cells were isolated, purified and cultured for the sixth generation. The number of bone marrow mesenchymal stem cells was no less than 5×108. Bone marrow mesenchymal stem cells labeled with green fluorescent protein were transplanted into the liver of the embryo rhesus with pregnancy of 10 weeks under guided by type-B ultrasound. At the 1st and 3rd months of birth, the liver tissue of the infant rhesus was taken for biopsy. After routine pathological section, histological specimens were observed under fluorescence microscope to confirm if there were adult bone marrow mesenchymal stem cells positive for green fluorescent protein and their distribution, and detected by immunohistochemical staining to identify if human albumin expressed in the liver of infant rhesus. RESULTS AND CONCLUSION:Fluorescence microscope observation indicated that at the 1st and 3rd months after birth, there were surviving bone marrow mesenchymal stem cells derived from human with green fluorescence in the liver of infant rhesus, and these cells migrated to form more concentrated distribution. The immunohistochemical results demonstrated that functional liver cells expressing human albumin were observed in the liver of infant rhesus at the 1st and 3rd months after birth, and their distribution was in accordance with bone marrow mesenchymal stem cells with green fluorescence. Human-rhesus chimeric liver can be established using adult bone marrow mesenchymal stem cells, which can generate functional liver cells in the liver of infant rhesus.

Surgery time,technique the joint func-tion restores the situation , the bone fracture cicatrization time and the complication occurrence were ana-lyzed.Results TAFS grades:A group was 77.8%;B group was 96.3%;C group was 96.7%.D group was 96.2%( P<0.05 ) .Conclusion Because of different to loss degree palm , we should select and use the different inner or outside fixture respectively .

BACKGROUND: Rat models induced by unilateral nephrectomy combined with adriamycin are widely used in screening anti-drugs of glomerular sclerosis. However, few reports concerning this method on inducing rabbit model of glomerular sclerosisOBJECTIVE: To establish a rabbit model of glomerular sclerosis, and to observe the renal function and histopathological changes during model preparation. DESIGN, TIME AND SETTING: Randomized controlled animal experiment was performed in Laboratory Animal Center of Kunming General Hospital of Chengdu Military Region between December 2008 and May 2009. MATERIALS: Twenty-five Japanese big-ear rabbits, weighing 1.75-2.25 kg, half males and half females, were selected to establish glomerular sclerosis models, and randomly divided into normal (n=10) and model (n=15) groups. METHODS: The left kidney of rabbits in the model group was removed under anesthetized with 30g/L saline solution of sodium pentobarbital (1 mL/kg). Rabbits in the normal group underwent a similar surgical procedure without kidney removing. At 1 week after operation, 5 mg/kg adriamycin was injected into rabbits in the model group, 3 mg/kg adriamycin was reinjected 2 weeks later. Same volume of physiological saline was injected in the normal group. MAIN OUTCOME MEASURES: The renal blood biochemical indexes were detected prior to and at weeks 4, 6, and 8 after model preparation. One nephridial tissue was harvested from each group to undergo pathological observation at week 8 after the second medication. RESULTS: ①Kidney in the normal group presented slightly white, slightly tough texture with smooth surface. ②Pathological features of focal segmental glomerulosclerosis was showed in the model group under a light microscope, presented as extracellular matrix hyperplasia, mesangial region expansion, renal glomerular capillary wall ball mix adhesion, tubule degeneration, or even interstitial fibrosis, as well as interstitial infiltration of inflammatory cells. ③Compared to the normal group, the total protein and albumin was and decreased, triglyceride, total cholesterol, low density lipoprotein cholesterol, urea nitrogen, and creatinine were significantly increased in the model group. The urine protein content of rabbits in the model group was obviously increased at 4 weeks, gradually reached a platform after 8 weeks, which still greater than that of the normal group. ④SPECT showed that the glomerular filtration rate of the model group was notably decreased (33 mL/min) than the normal group (92.6 mL/min).CONCLUSION: The unilateral nephrectomy combined with adriamycin in rabbits results in the formation of glomerular sclerosis, renal function and 24 h urinary protein is more visible in the phase change.
