Electroacupuncture alleviates postoperative pain through inhibiting neuroinflammation via stimulator of interferon genes/type-1 interferon pathway / 中西医结合学报

OBJECTIVE@#This work explores the impact of electroacupuncture (EA) on acute postoperative pain (APP) and the role of stimulator of interferon genes/type-1 interferon (STING/IFN-1) signaling pathway modulation in the analgesic effect of EA in APP rats.@*METHODS@#The APP rat model was initiated through abdominal surgery and the animals received two 30 min sessions of EA at bilateral ST36 (Zusanli) and SP6 (Sanyinjiao) acupoints. Mechanical, thermal and cold sensitivity tests were performed to measure the pain threshold, and electroencephalograms were recorded in the primary somatosensory cortex to identify the effects of EA treatment on APP. Western blotting and immunofluorescence were used to examine the expression and distribution of proteins in the STING/IFN-1 pathway as well as neuroinflammation. A STING inhibitor (C-176) was administered intrathecally to verify its role in EA.@*RESULTS@#APP rats displayed mechanical and thermal hypersensitivities compared to the control group (P < 0.05). APP significantly reduced the amplitude of θ, α and γ oscillations compared to their baseline values (P < 0.05). Interestingly, expression levels of proteins in the STING/IFN-1 pathway were downregulated after inducing APP (P < 0.05). Further, APP increased pro-inflammatory factors, including interleukin-6, tumor necrosis factor-α and inducible nitric oxide synthase, and downregulated anti-inflammatory factors, including interleukin-10 and arginase-1 (P < 0.05). EA effectively attenuated APP-induced painful hypersensitivities (P < 0.05) and restored the θ, α and γ power in APP rats (P < 0.05). Meanwhile, EA distinctly activated the STING/IFN-1 pathway and mitigated the neuroinflammatory response (P < 0.05). Furthermore, STING/IFN-1 was predominantly expressed in isolectin-B4- or calcitonin-gene-related-peptide-labeled dorsal root ganglion neurons and superficial laminae of the spinal dorsal horn. Inhibition of the STING/IFN-1 pathway by intrathecal injection of C-176 weakened the analgesic and anti-inflammatory effects of EA on APP (P < 0.05).@*CONCLUSION@#EA can generate robust analgesic and anti-inflammatory effects on APP, and these effects may be linked to activating the STING/IFN-1 pathway, suggesting that STING/IFN-1 may be a target for relieving APP. A new phloroglucinol from Dryopteris fragrans and its antibacterial activity in vitro / 中国中药杂志

A new phloroglucinol was isolated from 50% ethanol extract of Dryopteris fragrans by silica gel column chromatography, Sephadex LH-20 gel column chromatography, thin-layer chromatography(TLC), and preparative liquid column chromatography. On the basis of MS, ~1H-NMR, ~(13)C-NMR, and reference materials, compound 1 was identified as 2,5-cyclohexadien-1-one, 2-{[2,6-dihydroxy-4-methoxy-3-methyl-5-(1-isobutyl)phenyl]methyl}-3,5-dihydroxy-4,4-dimethyl-6-(1-oxobutyl)(1), and named disaspidin BB. Compound 1 was evaluated for its antibacterial activity. The experimental results showed that compared with the commonly used topical antibiotics erythromycin or mupirocin, disaspidin BB exhibited significant antibacterial activities against Staphylococcus epidermidis(SEP), S. haemolyticus(SHA), and methicillin-resistant S. aureus(MRSA)(P<0.05). Additionally, disaspidin BB was sensitive to ceftazidime-resistant SEP1-SEP4, SHA5-SHA7, MRSA8, and MRSA9. The MIC values of disaspidin BB against SEP and SHA were 1.67-2.71 μg·mL~(-1) and 10.00-33.33 μg·mL~(-1) respectively. Disaspidin BB has good antibacterial activities and deserves development as a new anti-infective drug for external use.

Analysis of vaccination certificate inspection and supplementary immunization among children entering kindergarten and school in Chaoyang District, Beijing from 2014 to 2020 / 中国学校卫生

Objective@#To understand the situation relating to vaccination certificate inspection and supplementary immunization among children entering kindergarten and school in Chaoyang District.@*Methods@#The data on vaccination certificate inspection and supplementary immunization among children entering kindergarten and school in Chaoyang District from 2014 to 2020 were reviewed and statistically analyzed.@*Results@#From 2014 to 2020, a total of 5 886 kindergartens and primary and secondary schools in Chaoyang District inspected the vaccination certificates of children entering kindergarten and school, and the coverage rate of inspection was 100%. A total of 605 675 children were inspected, and the coverage rate of inspection was 100%. The average certificate holding rate of children was 99.35%. The certificate holding rate of children in each year was between 99.29% and 99.97%, the difference was statistically significant ( χ 2=3 389.80, P <0.01), and an overall increasing trend was observed. There were statistically significant differences in children s certificate holding rates among different institutions ( χ 2=1 276.81, P <0.01) and different household registrations ( χ 2=1 476.78, P <0.01). The certificate replenishment rate for children entering kindergarten and school was 100%. As part of the national immunization plan, eight types of vaccines have been supplemented with 71 875 doses in total, with an average supplement rate of 99.71%. The vaccine supplement rate has been increasing annually, and the difference was statistically significant ( χ 2=414.10, P <0.01). After 2017, the vaccine supplement rate reached 100%. In respect to the eight national immunization program vaccines, the number of replenishing doses has been decreasing annually. In the case of replenishing doses, the top three vaccines were MMR vaccine, MPSV vaccine and DT vaccine.@*Conclusion@#The work carried out as part of vaccination certificate inspection and supplementary immunization among children entering kindergarten and school in Chaoyang District has continued to improve. The children s certificate holding rate, the certificate replenishment rate, and the vaccine supplement rate are maintained at a high level.

Application of quality control circle with project achievement style in the construction of family-centered care for premature infants / 中国实用护理杂志

Objective To discuss the application of quality control circle with project achievement style in the construction of family-centered care for premature infants. Provide theoretical support for family-centered care model. Methods Set up quality control circle group, select the topic as"construct a family-centered care for premature infants". Carry out activities according to the activity steps of the quality control circle with project achievement style. Advance activities by looking at the support of health care workers, family needs, carrying on the training and the examination to the family member, using the Failure Mode and Effect Analysis (FMEA) to prevent hospital infection and developing discharge plan by trinity model. Then evaluation of tangible and intangible results before and after the activity. Results The growth rate of body mass, breast-feeding rate, length of stay, family satisfaction and family care ability of premature infants after the quality control circle activities were respectively as follow (18.92±0.66) g/kg/d, 47.25％, (23.28 ± 1.39) d, 96.58％, (98.10 ± 12.57) points;before the quality control circle activities all the scores were respectively as follow (17.26 ± 0.38) g/kg/d, 13.45％, (26.40 ± 1.098)d, 80.67％, (61.45 ± 15.07) points. The difference between the two groups was statistically significant(t=7.31, 5.89, 7.52,χ2=34.41, 17.55, P<0.01). The incidence of hospital infection before and after quality control circle activities were respectively (6.11 ± 3.59)％, (5.61 ± 2.19)％,the difference was not statistically significant (χ2=0.262, P>0.05).In addition, we also gained economic and social benefits and many hospitals came to visit and study family-centered care model. Conclusion The family-centered care model is beneficial to the improvement of the nursing quality of premature infants. The activities of the Quality Control Circle with project achievement style can help to improve the participation and acceptance of all medical staff in this model and provide a foundation for the implementation of family-centered care model.

Application status of HPV vaccine in China / 中国实用妇科与产科杂志

The coverage rate of HPV vaccination in China is still very low,and the main reason may be related to people's confusion about the choice of vaccine types,the best age of vaccination,vaccination procedures and prices and other factors.This paper briefly describes the etiology of cervical cancer,the approval of HPV vaccine,the application status of HPV vaccine in foreign countries,and the current situation and challenges in the use of cervical cancer vaccine in China,and advocates that obstetrics and gynaecology experts/disease control experts actively promote the prevention and control of cervical cancer,so that young girls and their parents understand the knowledge related to cervical cancer and HPV vaccine,and young girls can be vaccinated with HPV vaccine as early as possible,so as to improve the coverage rate of HPV vaccination and eliminate cervical cancer.

Assessment of the results of syndrome in clinical trials of dementia treated by Chinese herbal medicine / 中国中西医结合杂志

Chinese medical syndrome efficacy, as a second efficacy indicator, has been widely used in clinical trials of treating dementia by Chinese herbal medicine. The syndrome assessment tool is a key point in assessing the efficacy of Chinese medical syndrome. The syndrome assessment tool for dementia used nowadays needs to be optimized in content, reliability, and validity. In this paper, the authors reviewed some problems correlated with the design of Chinese medical assessment questionnaire on the basis of Chinese medical theories by combining the common requirements for questionnaire development.

An experimental study on static and dynamic friction of XH1-type self-ligating bracket / 中国组织工程研究

BACKGROUND:XH1 type self-ligating bracket is a novel appliance of straight wire appliance.Currently,the majority of clinically applied self-ligating brackets are imported,which has low friction,short course of treatment,simple and efficient clinical practice.However,its cost is also very high.Therefore,we designed and developed the bracket.OBJECTIVE:Through the scanning electron microscopy (SEM) observation of XH1-type self-ligating bracket grooves,to perform the energy spectrum analysis of bracket,compare with the other bracket,and test the static and dynamic friction of this bracket.METHODS:A total of 4 kinds of upper right canine brackets,XH1-type self-ligating brackets,2E1 brackets (Shanghai),TP Straight Wire Bracket (USA),and AO self-ligating bracket (USA) were used.Brackets and arch wire were combined,respectively and tested on the XF-1-type friction tester.The friction mv-ms change marked and saved by Fluke190-type oscilloscope was recorded.The compile software graphics Matlab2007 was used to smooth the graph to remove abnormal data.The maximum static friction and a corresponding voltage (mV) value dynamic friction were obtained,and converted into force values,followed by statistical analysis.RESULTS AND CONCLUSION:Different brackets had different static and dynamic frictions.The static and dynamic friction TP-type bracket was higher than XHl-type bracket's.There was no difference between 2E1 and TP bracket,AO and XH1 bracket.This XH1-type self-ligating brackets is a self-developed product,with small static and dynamic friction,simple sterilization,and low cost,which allow widely clinical application.

Deep fascia composite autologous red bone marrow transplantation for the treatment of fracture nonunion / 中国骨伤

<p><b>OBJECTIVE</b>According to bone regeneration under the membrane and the bone regeneration deep fascia composite autologous red bone marrow transplantation applied in the treatment of fracture nonunion, in order to find a simple and effective clinical treatment of nonunion.</p><p><b>METHODS</b>Since March 2006 to March 2009,17 patients of fracture nonunion were treated by the deep fascia composite autologous bone marrow transplantation,included 10 males and 7 females, aged from 7 to 52 years old (means 32 years). There were 10 cases of tibia, 5 cases of radius, 2 cases of clavicle. Injured to admission time was from 7 to 36 months (means 12 months). Ten cases underwent operation for 1 time,5 cases for twice and 2 cases for 3 times. The position of nonunion were all at bone shaft and the condition of the skin and soft tissue was good. X-ray film showed 11 cases of hyperplasia nonunion, 6 cases of shrinking. The original fixation were removed and the intramedullary nail or plate fixation were re-used, and fracture ends were sutured closed by autogenous deep fascia and implanted with autologous red bone marrow.</p><p><b>RESULTS</b>Seventeen patients were followed-up for from 5 months to 2 years with an average of 1 year. Fracture healing time was from 12 to 20 weeks (means 16 weeks). According to the criteria of fracture healing to assess efficacy, the results were excellent in 14 cases, good in 2 cases and poor in 1 case.</p><p><b>CONCLUSION</b>Deep fascia composite autologous autologous red bone marrow transplantation for the treatment of fracture nonunion is suitable at the bone shaft and good condition of skin and soft tissue. The method has been observed that the fracture healing time is short.</p>

The clinical analysis of 51 patients with thoracic aortic dissection / 中华外科杂志

<p><b>OBJECTIVE</b>To summarize the clinical experience about surgical treatment of aortic dissection.</p><p><b>METHODS</b>The clinical data of 51 patients with aortic dissection admitted from December 2004 to December 2006 were analyzed retrospectively. There were 35 male and 16 female patients with a mean age of 55.7 years (ranged from 18 to 83-years-old). Twenty-seven patients of type I was performed under deep hypothermic circulatory arrest and selected cerebral perfusion with stent-graft which was implanted into the descending aorta through aorta arch. Five patients of type II was performed including Bentall operation in 3 patients, Wheat operation in 1 patient, ascending aorta replacement in 1 patient. Nineteen patients of type III was performed with stent-graft which was implanted into the descending aorta through aorta arch under deep hypothermic circulatory arrest.</p><p><b>RESULTS</b>The time of cardiopulmonary bypass (CPB) in type I patients was 250 to 290 min with an average of (274 +/- 53) min, and the arrest time was 40 to 59 min with an average of (53 +/- 14) min. CPB time of type II patients was 130 to 159 min with an average of (146 +/- 43) min, and the cross clamp time was 60 to 79 min with an average of (66 +/- 15) min. CPB time of type III patients was 240 to 280 min with an average of (260 +/- 28) min, and the arrest time was 20 to 27 min with an average of (24 +/- 3) min. The mean hemorrhage volume of the entire group was (500 +/- 250) ml. The mean ICU retention time was (5.0 +/- 1.5) d and the length of stay was (15.0 +/- 2.5) d. Three patients died during perioperative period. Two patients appeared cerebrovascular accident after operation. One patient appeared descending aorta dilation in the follow-up of 2 to 21 months.</p><p><b>CONCLUSION</b>Different clinical manifestations and treatment should be selected according to the different condition of aortic dissection aneurysm.</p>

Study of reduction rat T-lymphocytes activation and proliferation by inhibition inducible co-stimulator gene expression / 中华外科杂志

<p><b>OBJECTIVE</b>To observe the change of activation and proliferation ability of rat T-lymphocytes after suppress ICOS gene expression by RNA interference.</p><p><b>METHODS</b>Four interference sites targeting at rat ICOS gene were designed and four pairs of oligonucleotide fragments were cloned into the pSilencer 4.1-CMV neo plasmid vectors then transfected into rat lymphocytes with cationic liposome. The expression of mRNA and protein of ICOS was detected by RT-PCR and flow cytometry. The alteration of lymphocyte proliferation ability was evaluated by mix lymphocyte reaction, and the secretion levels of IFN-gamma and IL-4 were measured by ELISA procedure.</p><p><b>RESULTS</b>After transfection, the expression of mRNA and protein of ICOS in test groups were lower than that in control groups (P < 0.05). The ability of T-lymphocytes in proliferation was poor and the levels of IFN-gamma and IL-4 were reduced with ICOS gene shut down.</p><p><b>CONCLUSIONS</b>RNA interference plasmid vector can suppress ICOS expression in rat T-lymphocytes significantly, and it may be useful for further study on transplantation immunity tolerance.</p>

Studies on the properties and co-immobilization of manganese peroxidase / 生物工程学报

White-rot fungus manganese peroxidase (MnP) oxidizes a wide range of substrates, rendering it an interesting enzyme for potential applications. The stability of MnP can be improved by immobilization. With sodium alginate, gelatin, or chitosan as a carrier, and glutaraldehyde as the crosslinking agent, MnP was co-immobilized using the embed-crosslinked method and the adsorb-crosslinked method. The immobilization conditions and the partial properties of the three immobilized enzymes were investigated. When compared with the free enzyme, the optimum pH values and the temperatures of the three immobilized MnPs carried by alginate, gelatin, and chitosan were respectively shifted from 7.0 to 5.0, 5.0, 3.0 and from 35 degrees C to 75 degrees C , 55 degrees , 75 degrees C . The thermostabilities of the three immobilized MnPs were considerably better than that of the native enzyme. The chitosan-decreased by less than 5% even after repeated use for 6 - 9 times. The ability of decolorizing azo dyes in static and shaky situation by gelatin-immobilized MnP approached to the free enzyme, and there was no loss of enzyme activity during 2 repeated batch reactions.

Screening and detecting of proteins interacting with mPem / 生物工程学报

mPem, a homeobox gene, is expressed in a time and stage specific manner during murine ontogeny. Pem transcripts are abundant in 7- and 8-day mouse embryos, but decrease precipitously thereafter. On Day 9 they become abundant in placenta and yolk sac, persisting there until parturition. Although Pem transcripts are not detectable in most of adult tissues, they are present in reproductive system such as testis, epididymis and ovary. This indicates a important role for Pem during embryogenesis and reproductive development. To study the function of mPem protein, we used a GAL4 based yeast two-hybrid assay to screen a 7-day mouse embryo library with full-length of mPem. 3 proteins were found interacting with mPem protein. One of theses is Mdfic. We confirmed the interaction between mPem and Mdfic in yeast and in vitro. Mdfic, MyoD family inhibitor domain containing, encodes the myoD family inhibitor domain (I-mfa domain). The interaction between mPem and Mdfic suggested they maybe form the transcriptional regulator complex to regulate embryo differentiation.

Study the mechanisms and inducing transplantation immune tolerance of FTY720 / 中华外科杂志

<p><b>OBJECTIVE</b>To explore the operational mechanisms and potential approach to inducing transplantation immune tolerance of FTY720.</p><p><b>METHODS</b>Mouse splenocytes were incubated with FTY720, then the DNA was extracted and analyzed using gel electrophoresis. Hearts of inbred BALB/c (H-2(d)) mice were transplanted heterotopically in C57BL/6 (H-2b) mice. Recipients were randomly divided into six groups. Group-1 (n = 6) was the nil-treated control. Groups-2, 3 and 4 were given FTY720 at the dose of 3 mg.kg(-1) by oral gavage once a day with different time courses. Group-2 (n = 14) were administrated from 3 days before transplantation (day-3) to the 11th day after the transplantation (day 11); Group-3 (n = 6) from day 0 to day 14; Group-4 (n = 6) from day-3 to day 0. Group-5 (n = 5) and 6 (n = 5) were treated with Cyclosporine A (10 mg.kg(-1)) and 40-O-(2-hydroxyethyl)-rapamycin (RAD) (3 mg.kg(-1)) respectively by daily gavage from day 3 to day 11. The long survivors (> 100 d) in Group-2 were detected with their IL-4 and IFN-gamma levels and their tolerant state was challenged with second graft: the donor type skin.</p><p><b>RESULTS</b>Apoptosis changes of the mouse splenocytes incubated with FTY720 was showed by typical DNA ladders. The median survival time (MST) of Group-1 was 8 d. MST of Group-2 was 55 d and grafts in six mice survived more than 100 d. MST of Group-3 was 16.5 d. Group-4 has a MST of 14 d with one case exceeded 100 d. MST of Group-5 and 6 were 10 d and 13 d respectively. Long survivors of Group-2 can accept donor-type skin graft and the level of IL-4 in their serum is up-regulated while IFN-gamma remained stable.</p><p><b>CONCLUSIONS</b>Pretreatment of FTY720 bring about effect on the early events of transplantation immune responses. This effect might be mediated by apoptosis induction in lymphocytes using this drug. We originally designed the regime of FTY720 monotherapy, which started pre-operationally and maintained for a short period of time, and induced stable tolerance the allo-graft in mouse.</p>

Methylation status of promoter of mismatch repair genes hMLH1 and hMSH2 in epithelial ovarian cancer / 中华检验医学杂志

Objective To explore the methylation status of hMLH1 and hMSH2 promoter region in the epithelial ovarian cancer and its role in oncogenesis.Methods Methylation status of hMLH1 and hMSH2 promoter region was assayed in 20 normal ovarian tissues,25 benign epithelial tumor,56 malignant epithelial tumor and cell lines SKOV3,3AO by methylation-specific PCR (MSP).SKOV3 and 3AO were analyzed before and after 5-aza-2′-deoxycytidine (5-Aza-CdR) treatment.In addition,an alterations of mRNA expression of hMLH1 and hMSH2 was observed by reverse transcription polymerase chain reaction (PT-PCR).Results No methylation of hMLH1 and hMSH2 promoter was found in normal ovarian tissues. CPG islands methylation of hMLH1 and hMSH2 was observed in 4% (1/25),8% (2/25) respectively in benign epithelial tumor,30.4% (17/56),51.8% (29/56) respectively in malignant epithelial tumor.
