Clinical application of combined detection of three indicators in pulmonary tuberculosis patients accompanied with infection / 国际检验医学杂志

Objective To explore the clinical significance of the three indicators combined detection that plasma endotoxin,pro-calcitonin (PCT)and (1,3)-β-D-glucan(BG)in pulmonary tuberculosis patients complicated with bacterial,fungal infection.Methods Retrospective investigation was conducted in 240 pulmonary tuberculosis patients,who were divided into gram negative bacteria (G- )group(39cases),gram positive bacteria (G+ )group(45 cases)including fungal,germ-free group(156 cases).Other 45 healthy people were selected into control group.The levels of plasma endotoxin,PCT and BG in the four groups were compared.Results The levels of plasma endotoxin(0.682±0.418)EU/mL,PCT(2.93±0.87)μg/L in the G- group were significant higher than those of the G+ group (0.063±0.034)EU/mL,(0.85±0.52)μg/L,the difference was significant (P <0.05).The levels of the plasma endotoxin,PCT in the G- group were higher than those of the germ-free group.The levels of plasma endotoxin,PCT and BG in the control group had no significant difference with those of the germ-free group.Conclusion The combined detection of plasma endo-toxin,PCT and BG have some clinical value on the early diagnosis of the patients with pulmonary tuberculosis complicated with G-fungal infection for the advantages of fast and sensitiveness.

Influence of betahistine on the expression of histamine H3 receptor in the medial vestibular nucleus following unilateral labyrinthectomy in guinea pigs / 中华耳鼻咽喉头颈外科杂志

<p><b>OBJECTIVE</b>To observe the influence of betahistine on the expression of histamine H3 receptor in the medial vestibular nucleus (MVN) following unilateral labyrinthectomy (UL).</p><p><b>METHODS</b>Fifty-six healthy guinea pigs were randomly divided into three groups:the sham-operated group (group I), the UL group[group II, and UL+betahistine (BET) group (group III)], BET was intraperitoneally injection at 2.17 mg×kg(-1)×d(-1) for 7 days. The expression of histamine H3 receptor was analyzed by immunohistochemistry at 1 day, 3 days and 7 days after UL.</p><p><b>RESULTS</b>H3 receptors were presented in the MVN and the expression of histamine H3 receptor were increased significantly in the ipsilateral MVN at 1 and 3 days after UL(P < 0.05), the change turned into the normal value at 7 days(P > 0.05). In the UL+BET group, the intensity of histamine H3 receptor was lower than that in the UL at 1 day and 3 days(4.25 ± 0.71, 3.50 ± 0.92 vs 5.75 ± 0.71, 5.50 ± 0.93, P < 0.05). However, the changes turned into the normal values at 3 and 7 days (P > 0.05).</p><p><b>CONCLUSIONS</b>The early stage of the vestibular compensation process may be associated with the change of H3 receptor expression in MVN. In the UL+BET group the histamine H3 receptor recovered quickly.</p>

Blocking NR2B Receptor Reversed the Process of Cytotoxicity to Spiral Ganglion Neurons Induced by Sodium Salicylate in Guinea Pig / 听力学及言语疾病杂志

Objective To investigate whether blocking NR2B receptor can reverse the process of cytotoxicity to spiral ganglion neurons induced by sodium salicylate in guinea pig by applying ifenprodil (a NR2B antagonist) at the round window niche .Methods Sixty healthy guinea pigs provided by the experimental animal center of Guangxi medical university were randomly and evenly divided into a control group (Group I ,no treatment) ,an APL group (Group II ,60μl APL directly applied to the round window ) ,a sodium salicylate group (Group III ,60 μl APL di-rectly applied to the round window and then be given intraperitoneal sodium salicylate injection ) ,and an ifenprodil group (Group IV ,60μl of 10μmol/l ifenprodil in APL directly applied to the round window and then be given intra-peritoneal sodium salicylate injection ) .Sodium salicylate was given at 400 mg · kg -1 · d-1 for 7 days .Auditory brainstem responses (ABRs) were recorded before animal sacrifice by decapitation .The left cochlea was removed and prepared for detection of caspase -3 expression in spiral ganglion neurons via immunohistochemistry .From each group ,6 cochleae were used to test apoptosis index in spiral ganglion neurons using the TUNEL technique .Results Before salicylate administration ,the ABR threshold was less than 40 dB SPL in all animals .After salicylate ad-ministration ,the ABR threshold was 33 .33 ± 5 .17 dB SPL in Group II ,64 .17 ± 7 .36 dB SPL in Group III and 49 .17 ± 5 .85 dB SPL in Group IV ,in contrast to 31 .67 ± 5 .16 dB SPL in Group I (controls) .The caspase -3 ex-pression was not changed obviously in Group I and Group II ,but was significantly changed in Group III and Group IV (P<0 .01) .The caspase-3 expression appeared to be decreased in Group IV compared to those in Groups III (P<0 .05) ,but still increased compared to those of in Group I and II (P<0 .05) .The apoptosis index among spiral ganglion neurons in Groups III and IV increased significantly compared to those of in Group I and II (P<0 .001) .It was however ,lower in Group IV than in Group III (P<0 .01) .Conclusion Blocking NR2B receptor with specificity can reverse the process of cytotoxicity to spiral ganglion neurons induced by sodium salicylate in guinea pig .

Plain X-ray for locations of intracochlear electrode arrays during cochlear implantation / 临床耳鼻咽喉头颈外科杂志

OBJECTIVE@#To evaluate the position and shape of inserted electrode arrays by plain X-ray during cochlear implantation.@*METHOD@#Cochlear view of implanted electrode arrays by plain X-ray were observed during operation in 54 patients received cochlear implant.@*RESULT@#The image in cochlear view could provide the information about position and shape of inserted electrode arrays. The spiral-shape electrode arrays without distortion and folding were found in 52 cases. The complete insertion of intracochlear electrode in these patients were confirmed by surgery. Of 54 cases, the other 2 cases showed improper position and C-shape of intracochlear electrodes, one was partial insertion and the other was bending inside the cochlea. The bending electrode array was reinserted immediately during operation.@*CONCLUSION@#Routine plain X-ray after inserted electrode during operation was a best way of knowing the shape and depth of the electrode array, especial for the difficult cochlear implantation, and also could avoided anesthetization again.

The diagnosis and treatment of the simple congenital malformation of ossicular chain / 临床耳鼻咽喉头颈外科杂志

OBJECTIVE@#To investigate the diagnosis and treatment of the simple congenital malformation of ossicular chain.@*METHOD@#Curative effect analysis for 4 cases of treatment of simple congenital malformation of ossicular chain were performed in our department from June 2005 to August 2009, retrospectively. 4 patients underwent middle ear exploration and ossicular chain reconstruction were followed up for 1-4 years, and air conduction threshold of speech frequency(0.5, 1.0, 2.0 kHz) were detected, 15 dB increase of which were as the standard of successful operation.@*RESULT@#Three cases was abnormal incus, 2 cases were absence of instruction on the stapes footplates, and stapes footplates fixed, one case was absence of the incus. Hearing level had significantly improve after hearing reconstruction in 3 patients. The averaged postoperative air conduction threshold of speech frequency had 47 dB increase, bone conduction threshold have no change, while one case of hearing level without change. Tinnitus, vertigo, facial paralysis and other complications occurred were found in all the patients.@*CONCLUSION@#The congenital conductive hearing loss without external ear malformations should be suspected of congenital malformation of ossicular chain, which could be diagnosed by tympanic exploration. Ossicular chain reconstruction can improve the hearing effectively.

The expression and significance of HIF-1alpha and COX-2 in laryngeal squamous cell carcinoma / 临床耳鼻咽喉头颈外科杂志

OBJECTIVE@#To investigate the protein and mRNA expression of HIF-1alpha and COX-2 in laryngeal squamous carcinoma, and their correlation with clinicopathologic parameters.@*METHOD@#The protein and mRNA expression of HIF-1alpha and COX-2 in fresh laryngeal squamous carcinoma tissues and adjacent normal tissues from 76 patients were examined by immunohistochemistry and reverse transcription polymerase chain reaction.@*RESULT@#1) The protein positive expression rate of HIF-1alpha and COX-2 in tumor samples were significantly higher than that in normal tissues (P 0.05); 2) The mRNA positive expression rate of HIF-1alpha and COX-2 were 67.11% and 75.00% respectively, their expression were significantly higher in tumors than that in normal tissues (P < 0.05). The mRNA expression of COX-2 were not detected in normal tissues. The mRNA positive expression rate of HIF-1alpha were associated with T stage, but were not associated with differentiated clinicopathologic stage. 3) The protein positive expression rate of HIF-1alpha were associated with that of COX-2.@*CONCLUSION@#HIF-1alpha and COX-2 could up regulate each other to promote the development and metastasis of laryngeal squamous carcinoma.

Construction of recombinant plasmid pEGFP-NGB and its expression in cultured neuroglial cells / 中国病理生理杂志

AIM: To establish recombination plasmid pEGFP-NGB and to investigate the expression of pEGFP-NGB in culture neuroglia cells. METHODS: The NGB cds was isolated by using RT-PCR method with total RNA extracted from fetal Kunming mouse brain, then the NGB cds was cloned into the eukaryotic expression vector pEGFP-C1 of EGFP reported green fluorescence protein. The expression vector of recombinant plasmid pEGFP-NGB was successfully constructed. GeneJamer transfection reagent was used to transfer recombinant plasmid pEGFP-NGB into culture neuroglial cells. The mRNA and protein expression of pEGFP-NGB in culture neuroglial cells were investigated. RESULTS: The positive clone sequencing was consistent with the sequence of Genbank. The NGB mRNA and protein expression of pEGFP-NGB in culture neuroglial cells were detected at high levels. The high expression of green fluorescence protein was observed by fluorescence microscope in culture neuroglial cells. CONCLUSION: The expression vector of recombinant plasmid pEGFP-NGB was successfully constructed and green fluorescence protein was expressed in cultured neuroglial cells.

Effects of taurine on left ventricular hypertrophy in renovascular hypertensive rats / 中国药理学通报

AIM To investigate the preventive effects of taurine on the left ventricle hypertrophy in renovascular hypertensive rats. METHODS Two-kidney 1-clip (2K1C) rats were used to establish the model of renovascular hypertension. The myocardial local aldosterone(Ald) and AngⅡconcentration (MDC,MAC) were measured by radioimmunoassay. Expression of oncogene c-fos mRNA was analyzed by means of in situ hybridization. The myocardial collagen concentration (MCC) was measured by biochemical method.
