RESUMO

ObjectiveTo study the effect of Shenling Baizhusan on electrogastrogram in children with spleen deficiency diarrhea. To clarify the occurrence of gastric electrical rhythm disorder in children with this disease, and to study whether Shenling Baizhusan can improve the abnormal gastric motility in children with diarrhea (spleen deficiency) MethodA total of 125 children with spleen deficiency diarrhea in the outpatient department of Children's Hospital of Shanghai from October 2019 to March 2021 were selected as the research objects, and they were randomly divided into a control group (60 cases) and an observation group (65 cases). The children in the control group were treated with Montmorillonite powder combined with probiotics treatment, and the children in the observation group were additionally treated with Shenling Baizhusan. The course of treatment for both groups was 1 week. The clinical efficacy of the two groups of children after treatment and the scores of main traditional Chinese medcine(TCM) symptoms before and after treatment were compared, and the changes in the main parameters of electrogastrogram in children before and after treatment were compared. ResultAfter treatment, the total effective rate of observation group (90.77%, 59/65) was higher than that of control group (76.67%,46/60) (χ2=4.617, P<0.05). After treatment, scores of fecal morphology, frequency of defecation, fatigue, inappetence, and other symptoms in both groups were lower than that before treatment (P<0.05), and the observation group was lower than the control group (P<0.05). As compared with before treatment, the main frequency, the percentage of normal slow wave, and the percentage of normal gastric electrical rhythm in the two groups increased after treatment (P<0.05), and the control group was lower than the observation group (P<0.05). The proportion of children with slow gastric rhythm decreased (P<0.05) as compared with before treatment, and the control group was higher than the observation group (P<0.05). ConclusionShenling Baizhusan can significantly relieve the diarrhea symptoms in children with spleen deficiency diarrhea and improve gastric motility with good clinical effects.

Objective To explore the biological characteristics and biofilm formation of a strain of Klebsiella pneumoniae isolated from the stool of a patient with clinical inflammatory bowel disease .Methods The TH6 strain was identified as Klebsiella pneumoniae by 16S RNA, MALDI-TOF and automatic biochemical instrument .The morphology, growth rate, biofilm formation, capsid virulence factors and other biological characteristics were analyzed .Results and Conclusion The TH6 strain was short rod-shaped with a capsule under the transmission electron microscope , and was of K57 type according to the capsule serotype test .K.pneumonia TH6 grew rapidly under aerobic and anaerobic conditions , almost at twice the rate of standard strain ATCC2146, and reached the logarithmic growth phase at 2 h.The strain had multi-drug resistance.Biofilm formation was observed under a confocal laser scanning microscope (CLSM) at 4, 8, 12, 24, 48, 72, 96 and 120 h and the biofilm was formed at 72 h.The ability of fast growth , biofilm formation and other characteristics of K.pneumonia TH6 showed that this strain has a strong ability to adapt to the new environment is difficult to cure after colonization in vivo.

<p><b>BACKGROUND</b>Dehydroepiandrosterone (DHEA) is widely known for its beneficial effect on postmenopausal osteoporosis, although the underlying mechanisms remain mainly unclear. In this study, we tried to determine the activation of mitogen-activated protein kinase signal pathways during DHEA treatment and the indirect role of osteoblasts (OBs) on osteoclasts under the DHEA treatment of postmenopausal osteoporosis.</p><p><b>METHODS</b>Primary human OBs and osteoclast-like cells were cultured and, we pretreated OBs with or without U0126 (a highly selective inhibitor of both MEK1 and MEK2). The OBs were treated with DHEA. We then tested the effects of DHEA on human osteoblastic viability, osteoprotegerin production and the expression of phosphor-ERK1/2 (extracellular signal-regulated kinase). In the presence or absence of OBs, the function of osteoclastic resorption upon DHEA treatment was calculated.</p><p><b>RESULTS</b>DHEA promoted the human osteoblastic proliferation and inhibited the osteoblastic apoptosis within the concentration range of 10(-8) - 10(-6) mol/L (P < 0.05, P < 0.01, respectively). Within the effective concentration range, the expression of phosphor-ERK1/2 and osteoprotegerin was increased by DHEA and blocked by U0126. In the presence of OBs, DHEA could significantly decrease the number and the area of bone resorption lacuna (P < 0.05 and P < 0.01, respectively). Without OBs, however, the effects of DHEA on the bone resorption lacuna were almost completely abolished.</p><p><b>CONCLUSIONS</b>DHEA could indirectly inhibit the human osteoclastic resorption through promoting the osteoblastic viability and osteoprotegerin production, which is mediated by mitogen-activated protein kinases signal pathway involving the phosphor-ERK1/2.</p>

<p><b>OBJECTIVE</b>To construct eukaryotic expression vectors for HA-tagged receptor for advanced glycation end products (RAGE) mutants.</p><p><b>METHODS</b>Site-directed mutagenesis was applied to wild-type RAGE gene cloned in the pcDNA3 vector with HA tag to obtain the mutants pcDNA3-HA-RAGE(S391A), pcDNA3-HA-RAGE(S399A), pcDNA3-HA-RAGE(S400A), and pcDNA3-HA-RAGE(T401A). After identification by sequencing, the mutants were transfected into HEK293 cells, and the expression of these mutants were detected by Western blotting using anti-HA antibody.</p><p><b>RESULTS</b>The HA-tagged RAGE mutants constructed were verified successfully by sequencing, and highly expressed in HEK293 cells.</p><p><b>CONCLUSION</b>The success in constructing HA-tagged RAGE mutants, which are highly expressed in eukaryotic cells, may facilitate the functional study of RAGE in cell signal transduction.</p>

<p><b>OBJECTIVE</b>To construct a mammalian expression vector for HA-tagged receptor of advanced glycation end products (RAGE).</p><p><b>METHODS</b>Human RAGE cDNA codon region was amplified by PCR from human cDNA library and cloned into the pcDNA3 vector following routine procedures. After identification by enzyme digestion, PCR and sequencing, the correct recombinant vector RAGE/pcDNA3 was inserted with HA sequence behind the signal peptide sequence of RAGE. After identification by sequencing, HA-RAGE/pcDNA3 was transfected into HEK293 cells, and its expression was detected by Western blotting.</p><p><b>RESULTS</b>Identification by enzyme digestion, PCR and sequencing, confirmed the validity of the recombinant vector RAGE/pcDNA3, and HA-RAGE/pcDNA3 was highly expressed in HEK 293 cells.</p><p><b>CONCLUSION</b>HA-tagged RAGE is successfully constructed and expressed in mammalian cells, which may facilitate functional study of RAGE in cell signal transduction.</p>

RESUMO

<p><b>OBJECTIVE</b>To investigate the role of p53 gene in serum-induced cell migration.</p><p><b>METHODS</b>The effects of p53 knockout on serum-induced formation of lamellipodia and cell migration were observed using Transwell cell migration system.</p><p><b>RESULTS</b>p53(+/+) cells developed lamellipodia upon serum stimulation and showed enhanced activity of cell migration, but these effects were not observed in p53 knockout cells after serum stimulation.</p><p><b>CONCLUSION</b>p53 plays a role in serum-induced cell migration.</p>
