RESUMO

Objective: To investigate the association between congenital hypothyroidism (CH) and the adverse outcomes during hospitalization in very low birth weight infants (VLBWI). Methods: This prospective, multicenter observational cohort study was conducted based on the data from the Sino-northern Neonatal Network (SNN). Data of 5 818 VLBWI with birth weight <1 500 g and gestational age between 24-<37 weeks that were admitted to the 37 neonatal intensive care units from January 1st, 2019 to December 31st, 2022 were collected and analyzed. Thyroid function was first screened at 7 to 10 days after birth, followed by weekly tests within the first 4 weeks, and retested at 36 weeks of corrected gestational age or before discharge. The VLBWI were assigned to the CH group or non-CH group. Chi-square test, Fisher exact probability method, Wilcoxon rank sum test, univariate and multivariate Logistic regression were used to analyze the relationship between CH and poor prognosis during hospitalization in VLBWI. Results: A total of 5 818 eligible VLBWI were enrolled, with 2 982 (51.3%) males and the gestational age of 30 (29, 31) weeks. The incidence of CH was 5.5% (319 VLBWI). Among the CH group, only 121 VLBWI (37.9%) were diagnosed at the first screening. Univariate Logistic regression analysis showed that CH was associated with increased incidence of extrauterine growth retardation (EUGR) (OR=1.31(1.04-1.64), P<0.05) and retinopathy of prematurity (ROP) of stage Ⅲ and above (OR=1.74(1.11-2.75), P<0.05). However, multivariate Logistic regression analysis showed no significant correlation between CH and EUGR, moderate to severe bronchopulmonary dysplasia, grade Ⅲ to Ⅳ intraventricular hemorrhage, neonatal necrotizing enterocolitis in stage Ⅱ or above, and ROP in stage Ⅲ or above (OR=1.04 (0.81-1.33), 0.79 (0.54-1.15), 1.15 (0.58-2.26), 1.43 (0.81-2.53), 1.12 (0.70-1.80), all P>0.05). Conclusion: There is no significant correlation between CH and in-hospital adverse outcomes, possibly due to timely diagnosis and active replacement therapy.

OBJECTIVE Bergapten (BG), is a furanocoumarin derived from herbal and citrus extracts can act as antioxidant and selective anticancer agents.The current study aimed to investigate whether bergapten would attenuate immunosenescence and to exploreits immunomodulatory effects on immune responses in D-galactose-induced aging BALB/c mice.METHODS Firstly,mice were given D-galactose(180 mg·kg-1)subcutaneous injections for 30 d.To evaluate the establishment of the aging-related effect in mice, serum samples of BALB/c mice were collected from tail vein. Aging BALB/c mice were freely divided into three groups: negative control group received 1% Tween 80 solution only, named D-gal group. Positive groups were received BG administration at the dose of 20 and 100 mg·kg-1, named D-gal+BG(20)group and D-gal+BG(100)group,respectively.Effects of bergapten on T lympho-cyte proliferation and flow cytometry were assessed by using the splenic cell suspension. Enzyme linked immunospot kits were used to quantitatively determine interferon-γ(IFN-γ)and interleukin-4(IL-4) levels of the isolated serum. Immunophenotype was determined by using mixture of antibodies includ-ing anti-CD3,anti-CD4,and anti-CD8.RESULTS Bergapten(20 mg·kg-1)therapy can modulate immu-nity against viral epidemics and attenuate aging-induced immune deficiency(P<0.01),which was correlat-ed with the decline in the activation of the Th and Tc responses in D-galactose induced aging BALB/c mice.According to the in vivo results,bergapten exposure up-regulated the secretion of IFN-γ and IL-4 in T-helper 1(Th1)and T helper 2(Th2)cells(P<0.05,P<0.01).Additionally,BG(20 mg·kg-1)restored antigen-specific CD4+and CD8+T cells in aging models (P<0.05, P<0.01), which may help to curing chronic infections. CONCLUSION The beneficial effect of bergapten in D-galactose induced aging BALB/c mice may be due to the Th and Tc responses activation.

Human tongue cancer (TC) is an aggressive malignancy with a very poor prognosis.There is an urgent need to elucidate the underlying molecular mechanisms involved in TC progression.mRNA expression profiles play a vital role in the exploration of cancer-related genes.Therefore,the purpose of our study was to identify the progression associated candidate genes of TC by bioinformatics analysis.Five microarray datasets of TC samples were downloaded from the Gene Expression Omnibus (GEO) database and the data of 133 TC patients were screened from The Cancer Genome Atlas (TCGA) head and neck squamous cell carcinoma (HNSC) database.The integrated analysis of five microarray datasets and the RNA sequencing data of TC samples in TCGA-HNSC was performed to obtain 1023 overlapping differentially expressed genes (DEGs) in TC and adjacent normal tissue (ANT) samples.Next,Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis was conducted to enrich the significant pathways of the 1023 DEGs and PI3K-Akt signaling pathway (P=0.011) was selected to be the candidate pathway.A total of 23 DEGs with |log2 fold change (FC)| ≥1.0 in phosphatidylinositol 3-kinase-serine/threonine kinase (PI3K-Akt) signaling pathway were subjected to survival analysis of 125 eligible TC samples in TCGA database,indicating increased integrin-α3 gene (ITGA3) expression was significantly associated with poorer prognosis.Taken together,our study suggested ITGA3 may facilitate the development of TC via activating PI3K-Akt signaling pathway.

Objective: To investigate the effects of 3'-hydroxy puerarin on improving insulin resistance in 3T3-L1 adipocytes and their mechanisms. Methods: The proliferation of 3T3-L1 preadipocytes was tested by MTT assay and the differentiation by oil red O staining. The insulin resistance model was induced by dexamethasone. Cellular glucose consumption was determined by GOD-POD assay and the concentration of FFA by colorimetric methods. The expression of PPARγ and PTP1B genes in insulin resistant adipocytes was analyzed by qPCR. The PPARγ-transactivation activity of 3'-hydroxy puerarin was examined by using a hybrid reporter gene assay and the activity of PTP1B by colorimetric methods. Results: Compared with the medium control group, 3'-hydroxy puerarin significantly activated PPARγ at 0.1 and 10 μmol/L (P 0.05); increased the proliferation and differentiation of 3T3-L1 preadipocytes at 1-10 μmol/L (P 0.05). Conclusion: 3'-Hydroxy puerarin can improve the insulin resistance via up-regulating the expression of PPARγ.

For preferable authentication and regulation of material quality of Celosia argentea, HPLC fingerprints of different habitats were studied. Analysis was carried out on a Hypersil ODS2 column (4.6 mm x 250 mm, 5 microm) with acetonitrile-0.1% glacial acetic acid as the mobile phase, and eluates were detected by an evaporative light scattering detector (ELSD). The similarity evaluation system for chromatographic fingerprint of traditional Chinese medicine ( Version 2004 A) was applied to analyses the similarity of the fingerprint of diverse habitats. The similarity results were verified by SPSS. The chromatographic profiles of the samples from different regions were very similar. HPLC fingerprints of Semen C. argentea 12 common peaks and each peak in the fingerprint was well separated under the chromatographic condition above. The different habitats of C. argentea can be grouped to two types: the middle region and the south region. The chemical constituents of C. argentea vary with different habitats so selection of material habitat is very important for quality control of C. argentea. The fingerprint with high individuality and specificity could be applied in the identification and quality control of the material of C. argentea.

<p><b>OBJECTIVE</b>To explore the feasibility of muscle-derived cell autotransplantation in the treatment of post-prostatectomy urinary incontinence.</p><p><b>METHODS</b>Skeletal muscle-derived cells (MDC) were isolated and purified by replate technique from 6 female SD rats, and then transduced with adenovirus carrying Lac-Z gene. About 5 x 10(6) of the transduced cells were injected autologously into the bladder neck of the animals. Tissues were harvested after 5 and 15 days for histological examination and X-gal staining.</p><p><b>RESULTS</b>At 5 and 15 days after the autologous MDC transplantation, histological examination revealed no apparent sign of inflammation and inflammatory cell invasion, and X-gal staining showed a large number of cells dyed blue, indicating the survival of the autologous cells.</p><p><b>CONCLUSION</b>Autotransplanted MDCs can survive permanently. Autologous muscle stem cell injection can be an effective treatment for post-prostatectomy urinary incontinence.</p>

<p><b>OBJECTIVE</b>To study the changes of the contralateral testicular histology and germ cell apoptosis after unilateral testicular torsion (UTT) and to determine whether the contralateral testis is injured or not.</p><p><b>METHODS</b>Sixty SD male rats were divided into control group (12 rats) and experimental group(48 rats). The former underwent sham operation of the left testis under general anaesthesia. The latter underwent left testis torsion(720 degrees) for 6 h, and then 4 of them were sacrificed and the other 44 were subdivided into the torsed testis untwisted group (22 rats) and the torsed testis removal group (22 rats), 7-8 rats were sacrificed and both testes (twisted and untwisted) were removed 1 day, 1 week and 4 weeks after surgery. All testes underwent histological and germ cell apoptosis examination.</p><p><b>RESULTS</b>There were significant histological changes in the contralateral testis, and the germ cell apoptosis was increased greatly in the contralateral testis.</p><p><b>CONCLUSIONS</b>UTT can cause contralateral testicular injury, whose mechanism may be related to reperfusion, and torsed testis removal can prevent or reduce damage to the contralateral testis.</p>
