RESUMO

Objective:To investigate the effect of body mass management and mindfulness therapy on obese patients with polycystic ovary syndrome (PCOS).Methods:A total of 68 patients with PCOS admitted to Haikou People's Hospital(Haikou Hospital Affiliated to Central South Xiangya Medical College) from January to December 2018 were divided into observation group and control group by random digits table method, 34 cases in each group. The observation group was treated with body quality management combined with mindfulness therapy, and the control group was treated with routine nursing intervention. The total cholesterol (TC), triglyceride (TG), fasting blood glucose (FPG), 2 h postprandial blood glucose (2 hPPG), insulin (PINS), fasting insulin (FINS), Self Perceived Burden Scale (SPBS), quality of life score in the two groups were evaluated.Results:There was no significant difference in TC and TG between the two groups before intervention ( P>0.05); after 12 weeks of intervention, TC, TG were (4.21 ± 0.20), (1.60 ± 0.20) mmol/L in the observation group and (4.50 ± 0.31), (1.95 ± 0.55) mmol/L in the control group, there were significant differences between the two groups ( t values were 4.584, 3.487, P<0.01). There was no significant difference in FPG, 2 hPPG, PINS and FINS between the two groups before intervention and FPG, 2 hPPG after 12 weeks of intervention ( P>0.05); PINS, FINS were (72.60±28.46),(16.02±3.00)mU/L in the observation group and (102.61±50.10),(18.10±4.22)mU/L in the control group, there were significant differences between the two groups ( t values were 3.037, 2.342, P<0.01 or 0.05). There was no significant difference in the SPBS score between the two groups before intervention ( P>0.05); after 12 weeks of intervention, the SPBS score was (26.15 ± 2.02) in the observation group and (31.03 ± 1.00) in the control group, there was significant difference between the two groups ( t value was 12.624, P<0.01). There was no significant difference in the quality of life score between the two groups before intervention ( P>0.05); after 12 weeks of intervention, the scores of quality of life energy, sleep emotion, social life and physical activity were (80.62 ± 3.50), (79.10 ± 3.62), (75.02 ± 3.00), (77.26 ± 2.87) in the observation group and (71.23±4.02),(72.59±3.65),(69.20±4.11),(69.20±4.47) in the control group, there were significant differences between the two groups( t values were 7.234-10.272, P<0.01). Conclusions:The application of body mass management and mindfulness therapy can effectively improve the glucose and lipid metabolism level of obese PCOS patients, reduce the burden of psychological self perception, and improve the quality of life.

RESUMO

As the most common active brain-computer interaction paradigm, motor imagery brain-computer interface (MI-BCI) suffers from the bottleneck problems of small instruction set and low accuracy, and its information transmission rate (ITR) and practical application are severely limited. In this study, we designed 6-class imagination actions, collected electroencephalogram (EEG) signals from 19 subjects, and studied the effect of collaborative brain-computer interface (cBCI) collaboration strategy on MI-BCI classification performance, the effects of changes in different group sizes and fusion strategies on group multi-classification performance are compared. The results showed that the most suitable group size was 4 people, and the best fusion strategy was decision fusion. In this condition, the classification accuracy of the group reached 77%, which was higher than that of the feature fusion strategy under the same group size (77.31%

RESUMO

Inflammatory bowel disease (IBD) is a chronic, recurrent, nonspecific inflammatory disease of gastrointestinal tract, and includes ulcerative colitis (UC) and Crohn's disease (CD). Many studies have shown that gut microbiota and its mediated immune responses are the key factors driving the development of IBD. Fecal microbiota transplantation (FMT) is widely recognized as a new and potentially effective method for the treatment of IBD, but the mechanism is unclear. This article reviewed the current application of FMT in IBD, the immune response and the microbiota-immune interaction mechanism after FMT for the treatment of IBD, so as to discuss the potential mechanism of FMT in IBD.

BACKGROUND:A lot of work has been carried out on the development of the primary cultured rat myocardial cel s at home and abroad. The primary culture technology of rat myocardial cel s becomes more mature, but myocardial cel s from neonatal mice are not easy to be obtained under the same experimental conditions. The mouse genome has more similarities with the human genome, which has a higher research value. OBJECTIVE:To improve the primary culture method of neonatal mouse myocardial cel s, and to obtain myocardial cel s with high purity, vitality and original structure and function. METHODS:The mouse cardiac tissues were treated using an enzyme digestion method to isolate isolated single myocardial cel s:first, the cardiac tissues were digested using trypsin, and then col agenous fibers were treated with col agenase to isolate single myocardial cel s. The concentration and action time of trypsin and type II col agenase were adjusted, and the pH values of reagents and temperature of each step were strictly control ed. RESULTS AND CONCLUSION:At 24 hours after inoculation, the myocardial cel s began to be adherent;at 48 hours, independent pulsation of myocardial cel s could be observed;at 72 hours, myocardial cel s were cross-linked;and at 96 hours, myocardial cel s formed cel clusters and presented with consistent beating. The survival rate and purity of myocardial cel s were both over 95%. This modified method could successful y culture myocardial cel s with high purity and viablility from neonatal mice, and the structure and function of myocardial cel s could be retained. Therefore, it is a feasible culture method.

<p><b>OBJECTIVE</b>To explore the best approach to treatment of cesarean scar pregnancy (CSP).</p><p><b>METHODS</b>A total of 138 patients with CSP treated between January and December, 2013 were retrospectively analyzed. The patients were treated with conservative drug therapy, direct curettage, uterine curettage after embolization, or open or transvaginal surgery. The amount of blood loss, proportion of patients with blooding loss greater than 50 mL, hospitalization days, and hospitalization expenses were compared among the groups.</p><p><b>RESULTS</b>The median volume of blood loss was 370 mL in the conservative treatment group, 59 mL in direct curettage group, 67 mL in interventional therapy group, and 1425 mL in the surgical group, and the proportion of patients with blood loss over 50 mL was 76.9%, 38.8%, 27.5%, and 100% in the 4 groups, respectively. The midian hospital stay of the 4 groups was 9.0, 4.0, 6.0 and 10.0 days, with median hospitalization expenses of 12281.0, 3843.5, 14805.0, and 17202.2 RMB Yuan, respectively. All these data were significantly different among the 4 groups (P<0.05).</p><p><b>CONCLUSION</b>Direct curettage surgery should be encouraged for treatment of CSP. Embolization therapy can reduce the risk of bleeding but is associated with potential complications and more costly, and should be performed with caution. Open or trasnvaginal surgery can be considered in difficult cases of CSP, and its combination with interventional therapy is an option to better preserving the uterus.</p>

BACKGROUND:Platelet-derived growth factor-B (PDGF-B) is an effective pro-angiogenic growth factor, and adeno-associated virus type 9 (rAAV9) has a strong cardiomyocyte targeting affinity, which is an ideal vehicle for ischemic heart disease gene therapy. OBJECTIVE:To explore the PDGF-B gene transfection of in vitro neonatal rat myocardial cells mediated by rAAV9 against ischemia and hypoxia-induced cardiomyocytes apoptosis. METHODRat neonatal myocardial cells were isolated and cultured, and then transfected by rAAV9-PDGF-B and empty virus, rAAV9 with enhance green fluorescent protein (eGFP), under multiplicity of infection (MOI) of 105, 106 and 107, respectively. We observed the expression of eGFP under fluorescence microscopy every day, and used flow cytometry to measure transfection efficiency of vector rAAV9. Western blot and immunofluorescence were used to evaluate protein expression of PDGF-B. Myocardial ischemia and hypoxia injury model was established in vitro on the 5th day of transfection of rAAV9-eGFP and rAAV9-PDGF-B with 107 MOI. The number of myocardial apoptosis was measured by TUNEL assay. Western blot was employed to detect the protein expression of Bax and Caspase-3 which were related apoptosis, and the effect and its possible mechanism of PDGF-B gene overexpression against myocardial apoptosis were explored. RESULTS AND CONCLUSION:rAAV9 vector can efficiently transfect neonatal rat myocardial cells. eGFP and PDGF-B protein expressed in myocardial cells correctly and efficiently, and the expression intensity increased gradual y with the increasing of time course and MOI. The expression became stable on the 5th day, and the transfection efficiency showed significant difference among these groups (P<0.01). Myocardial apoptosis rate was significantly reduced in the rAAV9-PDGF-B group than the rAAV9-eGFP group (P<0.05), and protein levels of Bax and Caspase-3 in the rAAV9-PDGF-B group were significantly lower than those of the rAAV9-eGFP group (P<0.05). These data indicate that overexpression of PDGF-B gene can effectively reduce ischemia and hypoxia-induced myocardial apoptosis, and the possible mechanism might be by inhibiting Bax and Caspase-3 protein expression, which can provide evidence of rAAV9-PDGF-B vector in the gene therapy of ischemic heart diseases.

<p><b>OBJECTIVE</b>The present study describes an improved in vitro culture method for obtaining high purity, vital and fully functional cardiomyocytes from neonatal rat.</p><p><b>METHODS</b>After cutting ventricular tissue with improved method, ventricular tissues were digested with low concentrations of trypsin overnight at 4 °C, and then underwent collagenase II digestion. Thereafter, cardiomyocytes were purified by combined differential adhesion and chemical inhibition methods.</p><p><b>RESULTS</b>Adherent cardiomyocytes were seen at 12 h after culture, spontaneously beating cardiomyocytes were observed at 24 h after culture, crosslinked cardiomyocytes were found at 48 h after culture, adhesion clustered cardiomyocytes were seen at 72 h after culture, dense network formed from inter-connected was evidenced together with radial arranged cell clusters and cell pseudopodia 96 h the mutual contact woven into and formed radically ordering cell clusters and island-like beating cardiomyocytes at 96 h after culture. The cell survival rate and purity were more than 98%.</p><p><b>CONCLUSION</b>Fully functional spontaneous beating cardiomyocytes can be obtained by the use of this improved primary neonatal rat cardiomyocytes culture method.</p>
